Objectives : We compared the inhibitory effects of herb-combined remedies, which were recorded on "Yooam" prescription of Dongeuibogam, on cell migration and invasion, two critical cellular processes that are often deregulated during metastasis, in B16F10 melanoma cells. For this purpose, water extracts of Sipyukmiryukieum (SYMRKU), Danjacheongpitang (DJCPT), Cheongganhaeultang (CGHUT) and Jipaesan (JPS) were used. Methods : Cytotoxicity was assessed by an MTT assay. Wound healing and matrigel transwell assays were used to examine on B16F10 cell migration and invasion. The levels of mRNA and protein expression of matrix metalloproteinases (MMPs) and tissue inhibitor of metalloproteinases (TIMPs) were analyzed by RT-PCR and Western blotting. Results : Our data showed that DJCPT showed the strongest inhibitory effect among the four prescriptions in inhibiting cell motility of B16F10 melanoma cells within the concentration range that was not cytotoxic. The inhibitory potential of colony formation was higher in DJCPT and SYMRKU compared to the other two types of prescriptions, and the inhibitory effect of invasiveness is shown in order of DJCPT, SYMRKU, CGHUT and JPS. DJCPT, and SYMRKU strongly inhibited the activity and expression of MMP-2 and MMP-9, which are important mediators in cancer invasion, compared to CGHUT and JPS, and the increased expression of TIMP-1 and TIMP-2 was also more effective in these two prescriptions. In conclusion, DJCPT is expected to exhibit the most potent blocking effect on migration and invasion among four herb-combined remedies compared in B16F10 melanoma cells. Conclusion : Overall, the results of this study will be used as an important source to validate these prescriptions in animal models and to understand the mechanism of action of herbal remedies recorded in Dongeuibogam.
Background: Swiprosin-1 was identified in human CD8+ lymphocytes, mature B cells and non-lymphonoid tissue. We have recently reported that swiprosin-1 is expressed in mast cells and up-regulated in both in vitro and in vivo. Methods: The expression of cytokines and swiprosin-1 were determined by by real time PCR and conventional PCR. Pharmacological inhibitors were treated to investigate potential mechanism of swiprosin-1 in mast cell activation. Actin content was evaluated by confocal microscopy and flow cytometry. Results: The swiprosin-1 augmented PMA/A23187-induced expression of cytokines and release of histamine. However, knock-down of swiprosin-1 showed only a modest effect on PMA/A23187-induced cytokine expression, suggesting that swiprosin-1 has gain-of-function characteristics. Swiprosin-1 was found in microvilli-like membrane protrusions and highly co-localized with F-actin. Importantly, either disruption of actin by cytochalasin B or inhibition of PI3 kinase, an enzyme involved in actin remodeling, by wortmannin blocked cytokine expression only in swiprosin-1-overexpressing cells. Conclusion: These results suggest that swiprosin-1 modulates mast cell activation potentially through actin regulation.
This study was carried out to identify and quantify the flavonoids from 6 different plant parts of Allium victorialis var. platyphyllum (AVP), including the flower, leaf, root, stem, flower stalk, and flower seed, using liquid chromatography/ mass spectrometry. Two major flavonoids were structurally identified as quercetin (3,5,7,3'4,'-pentahydroxyflavone) and kaempferol (3,5,7,4'-tetrahydroxyflavone) at contents of 11.8-25.8 and $6.0-64.4\;{\mu}g/mL$, respectively. In particular, the flower and root plant parts contained the highest amounts of quercetin and kaempferol compared to the other parts. We also assessed the recovery effects of each plant-part extract of AVP on gap junctional intercellular communication (GJIC) in WB-F344 cells by the scrape-loading and dye transfer (SL/DT) method. According to the results, GJIC was reduced by approximately 70.2% ($62.3{\pm}12.5$ cells) compared to the control ($209{\pm}9.5$ cells, 100%) when 12-O-tetradecanoylphorbol-13-acetate (TPA) was treated alone in the WB-F344 rat liver epithelial cells. However, the stem extract (0.2 mg/mL) restored GJIC to basal levels (92%, $204{\pm}2.3$ cells, p<0.01) and the flower extract (0.2 mg/mL) stimulated GJIC to 82.5% ($172.6{\pm}8.3$ cells, p<0.05), when applied together with the TPA.
The anticarcinogenic effects of various food components on human cancer cells have received much attention in recent years. The precise effect and mechanisms of anticarcinogens in food materials on cancer cells have rarely been investigated. This study was carried out to determine the effects of Daucus carota L. leaf (DCL) extracts on cytotoxic and chemopreventive effect on human cancer cells. The experiment was conducted to determine cytotoxicity of Daucus carota L. leaf extracts on HepG2, Hela and MCF-7 cells by MTT assay. Among various partition layers of Daucus carota L. leaf, the ethylacetate partition layer (DCLMEA) at 500 $\mu\textrm{g}$/mL was shown to be most effective on MCF-7 cell lines. The four partition layers which are DCLM, DCLMH, DCLMB and DCLMH were less effecitve in inducing cytotoxicity than DCLMEA was. We also determined the induction of intracellular quinone reductase (QR) activity by adding DCL extracts on HepG2 cells. Among various partition layers of DCL extracts, DCLMH and DCLM were tested to be most effective with results such as 4.9 and 4.73 with a control value of 1.0.
Zinc oxide (ZnO) has been widely studied for its practical applications such as transparent conduction electrodes for flat panel displays and solar cells. Especially, ZnO films show good chemical stability against hydrogen plasma, absence of toxicity, abundance in nature, and then suitable for photovoltaic applications. However, the fabrication process of thin film solar cells require a high substrate temperature and/or post heat treatment. Therefore, the layers have to withstand high temperatures, requiring an excellent stability without degrading their electronic and optical properties. In this paper, we investigated the stability of zinc oxide (ZnO) films doped with aluminum and hydrogen. Doped ZnO films were prepared by r.f. magnetron sputter and followed by heat treatment at different temperatures and for various times.

A synthetic naphthoquinone alkaloid. 2-amino-3-ethoxycarbonyl-l-methyl pyrolo (3,2-b) naphtho-4,9-dione (compound 1), showed a potent cytotoxicity in a panel of cancer cell lines with an IC50 ranged from 0.1 to 0.3 microgram/mL. Prompted by a potent cytotoxic activity, the mechanism action study was performed with cultured A549 of human lung cancer cells. Flow cytometric G2-M cell cycle arrest and microscopic investigation was also characterized with apoptotic morphological features. (omitted)
Binary vectors, pBI-ActR1, pBI-ActF1 and pBSH-ActR1, were constructed using pGA642, the replication origin of pTi12 and the rice actin promoter. The sizes of pBI-ActR1, pBI-ActF1 and pBSH-ActR1 were 12.9 kb, 13.2 kb and 11.95 kb, respectively. These vectors containing a rice actin promoter followed by a GUS structural gene could induce stronly the expression of GUS gene in transformed rice cells. Rice explants from 3-4 day old seedlings after germinatin were cocultured with A. tumefaceins harboring pBI-ActR1, pBI-ActF1 or pBSH-ActR1, and then GUS expression in the explants was assayed. Transformation of rice explants by these binary vectors was tissue-specific, such that the meristematic regions of shoot apex, root and hypocotyl were transformed by these binary vectors.
사람의 정상 골모세포 FOB에서 아미노산 수송계 L의 발현 및 이들 수송계 L을 통한 아미노산 수송특성을 밝히기 위하여, FOB 세포에서 RT-PCR, western blot 분석 및 아미노산 uptake 실험 등을 수행하여 다음과 같은 결과들을 얻었다. FOB 세포에서 아미노산 수송계 L의 두 아형인 LAT1, LAT2및 그들의 보조인자 4F2hc의 발현을 확인할 수 있었다. FOB 세포에서 $_{L}-leucine$의 수송은 $Na^+$-비의존적이었다. FOB 세포에서 $_{L}-leucine$의 수송은 아미노산 수송계 L의 선택적 억제제인 BCH에 의해 완전히 차단되었다. FOB 세포에서 여러 아미노산들에 의한 L-leucine수송억제 실험결과는 Xenopus oocyte에서 시행되어 보고되어진 LAT1과 LAT2 수송억제 실험 결과의 특성을 모두 포함하였다. 본 연구의 결과로 사람의 정상 골모세포주인 FOB에서 는 세포성장 및 증식을 위한 중성 아미노산의 수송에 아미노산 수송계 L의 두 아형인 LAT1과 LAT2가 중요한 역할을 하고 있음을 확인할 수 있었다.
Objective: Lactobacilli in rabbit intestine is rare and its function in rabbit gut health is not fully understood. The present study aimed to evaluate in vivo the probiotic potential of Lactobacillus casei for suckling rabbits. Methods: Two healthy 5-day-old suckling rabbits with similar weights from each of 12 New Zealand White litters were selected and disturbed to control group and treatment group. All rabbits were artificially fed. The treatment group had been supplemented with live Lactobacillus casei in the milk from the beginning of the trial to 13 days of age. At 15 days of age, healthy paired rabbits were slaughtered to collect intestinal samples. Results: i) Oral administration of Lactobacillus casei significantly increased the proportion of Lactobacilli in the total intestinal bacteria (p<0.01) and obviously reduced that of Escherichia-Shigella (p<0.01); ii) treatment increased the length of vermiform appendix (p<0.05); iii) a higher percentage of degranulated paneth cells was observed in the duodenum and jejunum when rabbits administered with Lactobacillus casei (p<0.01); and iv) the expression of toll-like receptor 9, lysozyme (LYZ), and defensin-7-like (DEFEN) in the duodenum and jejunum was stimulated by supplemented Lactobacillus casei (p<0.05). Conclusion: Orally administered Lactobacillus casei could increase the abundance of intestinal Lactobacilli, decrease the relative abundance of intestinal Escherichia-Shigella, promote the growth of appendix vermiform, stimulate the degranulation of paneth cells and induce the expression of DEFEN and LYS. The results of the present study implied that Lactobacillus casei exhibited probiotic potential for suckling rabbits.
It has been well documented that dermal irradiation by ultraviolet A (UVA) locally decreases the number of Langerhans cells and suppresses contact hypersensitivity of the skin. We found that topical irradiation of UVA to the eye systemically decreased the number of Langerhans cells (LC) in the dorsalskin and lymph nodes and elicited lymphocyte apoptosis in the latter tissues but not in the thymus. Optic nerve resection, but not ciliary ganglionectomy, eliminated the UVA-induced decrease in dermal Langerhans cells by a mechanism that was partially inhibited by hypophysectomy. The immunosuppressive effect of UVA was not observed in knockout mice lacking inducible-type of nitric oxide synthase (iNOS). These results suggested that topical irradiation of UVA to the eye induced immunosuppression via NO-dependet neuronal pathways.
