Pseudomonas aeruginosa Exotoxin A Induces Apoptosis in Chemoresistant YD-9 Human Oral Squamous Carcinoma Cell Line Via Accumulation of p53 and Activation of Caspases (항암제에 저항성을 가지는 YD-9 human oral squamous carcinoma cell line에서 Pseudomonas aeruginosa exotoxin A의 p53 단백질 누적과 caspase를 활성화 경로를 통해 유도된 세포자멸사)

  • Oral squamous carcinoma (OSC) cells present resistance to chemotherapeutic agents-mediated apoptosis in the late stages of malignancy. Advances in the understanding of bacterial toxins have produced new strategies for the treatment of cancers. It was demonstrated here that Pseudomonas aeruginosa exotoxin A (PEA) significantly decreased the viability of chemoresistant YD-9 cells in the apoptosis mechanism. Apoptotic manifestations were evident through changes in nuclear morphology and generation of DNA fragmentation. PEA treatment induced caspase-3, -6 and -9 cleavage, and activation. These events preceded proteolysis of the caspase substrates poly (ADP-ribose) polymerase (PARP), DNA fragmentation factor 45 (DFF45), and lamin A in YD-9 cells. The reduction of mitochondrial membrane potential, release of cytochrome c and SmacjDlABLO from mitochondria to cytosol, andtranslocation of AlF into nucleus were shown. While p53, p21 and $14-3-3{\gamma}$ were upregulated, cyclin Band cdc2 were downregulated by PEA treatment. Taken together, PEA induces apoptosis in chemoresistant YD-9 cells via activation of caspases, mitochondrial events and regulation of cell cycle genes.

Semi-active Control of a Seismically Excited Cable-Stared Bridge Considering Dynamic Models of MR Fluid Damper (MR 유체 댐퍼의 동적모델을 고려한 사장교의 반(半)능동제어)

  • This paper examines the ASCE first generation benchmark problem for a seismically excited cable-stayed bridge, and proposes a new semi-active control strategy focusing on inclusion of effects of control-structure interaction. This benchmark problem focuses on a cable-stayed bridge in Cope Girardeau, Missouri, USA, for which construction is expected to be completed in 2003. Seismic considerations were strongly considered in the design of this bridge due to the location of the bridge in the New Madrid seismic zone and its critical role as a principal crossing of the Mississippi River. In this paper, magnetorheological(MR) fluid dampers are proposed as the supplemental damping devices, and a clipped-optimal control algorithm is employed. Several types of dynamic models for MR fluid dampers, such as a Bingham model, a Bouc-Wen model, and a modified Bouc-Wen model, are considered, which are obtained from data based on experimental results for full-scale dampers. Because the MR fluid damper is a controllable energy-dissipation device that cannot add mechanical energy to the structural system, the proposed control strategy is fail-safe in that bounded-input, bounded-output stability of the controlled structure is guaranteed. Numerical simulation results show that the performance of the proposed semi-active control strategy using MR fluid dampers is quite effective.

Effects of Neutral Dammarane Saponin from Panax ginseng on the in vitro Function of Polymorphonuclear Leukocytes (인삼의 중성 Dammarane계 사포닌의 다형핵 백혈구 기능에 미치는 영향)

  • Although Saponin A from Panax ginseng has previously been shown to inhibit carageenin induced edema. a paucity of information exists on the effects of components from ginseng on the cellular inflammatory response. specifically polymorphonuclear leukocyte (PMNL) function. The purpose of this study was 10 determine the effects of isolated neutral dammarane saponins from ginseng (i.e..glycosidic derivatives of 20(S)-protopanaxadiol [ginsenoside $Rb_1,\;Rb_2$ and Rc] and 20(S)-protopanaxatriol [ginsenosides Re and $Rg_1$]) on in vivo PMNL function and to compare their effects with those produced by a steroidal anti-inflammatory agent (dexamethasone) and commercially available saponin. Dexamethasone. the ginsenosides and saponin were all shown to he potent inhibitors of PMNL chemotaxis using the $^{51}Cr$ assay with $5{\times}10^{-8}M$ f-met-leu-phe [FMLP] as the chemoattractant. Inhibition or PMNL chemotaxis by dexamethasone. the ginsenosides and saponin were all shown to be both time-and dose-dependent and these agents did not affect cellular viability at the concentrations tested Saponin and the ginsenosides were more potent inhibitors of chemotaxis than was dexamethasone. while oxidant generation (as measured by the luminol-enhaneed chemil-uminescence of PMNL using FMNL $[10^{-6}]$ as the stimulus) was inhibited by dexamethasone. the ginsenosides $(Rb_1\;Rb_2\;Rc\;Re\;and\;Rg_1)$ and saponin at a concentration of 1 ${\mu}M$ had no significant effect on PMNL chemiluminescence. Thus. the neutral dammarane saponins are potentially important modulators or PMNL function and their inhibitory effects may he differentiated from those of the Steroidal anti-inflammatory agents.

A New Tongil-type Glutinous Rice Variety 'Hangangchal 1' of Multi-Diseases and Insect Resistance (중생 복합내병성 통일형 찰벼 품종 '한강찰 1호')

  • 'Hangangchal 1' is a new glutinous rice cultivar of second generation Tongil-type with a mid-maturing ecotype that developed by the rice breeding team of Yeongnam Agricultural Research Institute (YARI), RDA. in 2006. This cultivar was derived from a cross between 'Hangangchal', a Tongil-type glutinous cultivar and 'YR8208-20', a high yield potential in 1986/1987 winter season. 'Hangangchal 1' was selected by pedigree and bulk breeding methods from $F_3$ to $F_6$ populations. A promising line, YR10498-8-1-3, was selected and designated as 'Milyang 167' in 1997. The local adaptability test of 'Milyang 167' was carried out at seven locations during 3 years in 1998, 2005, and 2006. It has tolerance to lodging with good canopy architecture as 87cm of culm length. This cultivar is resistant to bacterial blight $K_1$, $K_2$, and $K_3$ race, rice stripe virus, rice dwarf virus, and leaf blast disease. The milled rice endosperm of 'Hangangchal 1' is glutinous and its whiteness was almost similar compared to 'Shinseonchalbyeo'. The yield of milled rice of 'Hangangchal 1' was average 5.97 MT/ha at ordinary cultivation of 9 kg/10a N fertilizer level in local adaptability test. This cultivar would be adaptable to the mid and southern plain of Korea.

Comparison of Characteristics of Electrodeposited Lithium Electrodes Under Various Electroplating Conditions (다양한 전착조건에서 제작된 리튬 전극의 특성 연구)

  • A lithium is the lightest metal on the earth. It has some attractive characteristics as a negative electrode material such as a low reduction potential (-3.04 V vs. SHE) and a high theoretical capacity ($3,860mAh\;g^{-1}$). Therefore, it has been studied as a next generation anode material for high energy lithium batteries. The thin lithium electrode is required to maximize the efficiency and energy density of the battery, but the physical roll-press method has a limitation in manufacturing thin lithium. In this study, thin lithium electrode was fabricated by electrodeposition under various conditions such as compositions of electrolytes and the current density. Deposited lithium showed strong relationship between process condition and its characteristics. The concentration of electrolyte affects to the shape of deposited lithium particle. As the concentration increases, the shape of particle changes from a sharp edged long one to a rounded lump. The former shape is favorable for suppressing dendrite formation and the elec-trode shows good stripping efficiency of 92.68% (3M LiFSI in DME, $0.4mA\;cm^{-2}$). The shape of deposited particle also affected by the applied current density. When the amount of current applied gets larger the shape changes to the sharp edged long one like the case of the low concentration electrolyte. The combination of salts and solvents, 1.5M LiFSI + 1.5M LiTFSI in DME : DOL [1 : 1 vol%] (Du-Co), was applied to the electrolyte for the lithium deposition. The lithium electrode obtained from this electrolyte composition shows the best stripping efficiency (97.26%) and the stable reversibility. This is presumed to be due to the stability of the surface film induced by the Li-F component and the DOL effect of providing film flexibility.

Assessment of Practical Use of Recycling Oil from the Pyrolysis of Mixed Waste Plastics (혼합폐플라스틱의 열분해를 통한 회수오일의 이용가능성 평가)

  • In Korea, although the generation of waste plastic has been increasing, the rate of recycling is considerably low and moreover, there is no suitable method for the treatment of waste plastics. However, pyrolysis, which is appropriate for the treatment of highly polymerized compounds, such as plastics, has recently gained much interest. In this study, a property of the products from the pyrolysis of mixed waste plastics, with a possible practical use for the recycling oil produced, were assessed. First of all, in order to investigate the pyrolysis characteristic of waste plastics, TGA (Thermogravimetric analysis) and DCS (Differential Scanning Calorimetry) were performed on a number of different plastics, including PP, LDPE, HDPE, PET and PS, as well as others. According to the result, it appeared that PP was the most efficiently pyrolyzed by changing the temperature, followed by LDPE, HDPE, PET, PS and the other plastics, in that order. From the results, the optimum conditions f3r pyrolysis were set up, and the different waste plastics pyrolyzed. The recycling oil produced from the flammable gases generated during the pyrolysis was com-pared with fuel oil by an analysis using the petroleum quality inspection method on KS(Korea industrial Standard). The results of the analysis showed the recycling oil was of a similar standard to fuel oil, with the exception of the ignition point, with a quality somewhere between that of paraffin oil and diesel fuel. With respect to these results, the quality of the recycling oil produced by the pyrolysis of waste plastics was suf-ficient for use as fuel oil.

Production of the melittin antimicrobial peptide in transgenic silkworm (멜리틴 항균펩타이드를 생산하는 형질전환누에)

  • Melittin is the main component of Bee Venom and has antibacterial activity against several bacteria. To produce the melittin antimicrobial peptide, we constructed transgenic silkworm that expressed melittin gene under the control BmA3 promoter using piggyBac vector. The use of the 3xP3-driven EGFP cDNA as a marker allowed us to rapidly distinguish transgenic silkworm. Mixtures of the donor vector and helper vector were micro-injected into 300 eggs of bivoltin silkworms, Baegokjam. In total, 131 larvae (G0) were hatched and allowed to develop into moths. The resulting G1 generation consisted of 36 broods, and we selected 4 broods containing at least 1 EGFP-positive embryo. The rate of successful transgenesis for the G1 broods was 11%. We identified 12 EGFP-positive G1 moths and these were backcrossed with wild-type moths. With the aim of identifying a melittin as antimicrobial peptide, we investigated the Radical diffusion Assay (RDA) and then demonstrated that melittin possesses high antibacterial activities against gramnegative bacteria.

Achievements in the Creativity Education through Freshmen Engineering Design (대학 신입생 공학설계과목을 통한 창의성 교육의 성과)

  • This study is a part of the research on measuring and developing the creativity of college students, especially of engineering college students. This study was done in the class of imaginative design engineering which was a creativity training program integrated with engineering majors, including idea generation skills, creative problem solving, patent applications, design, manufacturing and marketing. Participants in this program were 75 freshman strudetns in engineering college. The achievements in this program were measured by the figural form of TTCT(Torrance Tests of Creative Thinking), as well as MBTI(Myers-Briggs Type Indicator). The results are as follows. First, TTCT scores of the participants in imaginetive design engineering increased significantly. Second, type indicator scores of MBTI varied significantly, to the directions of extrovert, feeling, and perception. Therefore, according to the results of this study, we can conclude the imaginative design engineering class had significant positive effects on the creativity of the engineering students.

Single Nucleotide Polymorphism (SNP) Discovery and Kompetitive Allele-Specific PCR (KASP) Marker Development with Korean Japonica Rice Varieties

  • Genome resequencing by next-generation sequencing technology can reveal numerous single nucleotide polymorphisms (SNPs) within a closely-related cultivar group, which would enable the development of sufficient SNP markers for mapping and the identification of useful genes present in the cultivar group. We analyzed genome sequence data from 13 Korean japonica rice varieties and discovered 740,566 SNPs. The SNPs were distributed at 100-kbp intervals throughout the rice genome, although the SNP density was uneven among the chromosomes. Of the 740,566 SNPs, 1,014 SNP sites were selected on the basis of polymorphism information content (PIC) value higher than 0.4 per 200-kbp interval, and 506 of these SNPs were converted to Kompetitive Allele-Specific PCR (KASP) markers. The 506 KASP markers were tested for genotyping with the 13 sequenced Korean japonica rice varieties, and polymorphisms were detected in 400 KASP markers (79.1%) which would be suitable for genetic analysis and molecular breeding. Additionally, a genetic map comprising 205 KASP markers was successfully constructed with 188 $F_2$ progenies derived from a cross between the varieties, Junam and Nampyeong. In a phylogenetic analysis with 81 KASP markers, 13 Korean japonica varieties showed close genetic relationships and were divided into three groups. More KASP markers are being developed and these markers will be utilized in gene mapping, quantitative trait locus (QTL) analysis, marker-assisted selection and other strategies relevant to crop improvement.

Risk Factors for Atrophic Renal Scar (위축성 신반흔의 위험인자)

  • Purpose : An atrophic renal scar(RS) is one of the underlying causes for childhood hyper tension and chronic renal failure. The risk factors for atrophic renal scar were evaluated. Methods : 41 children, who presented with first febrile urinary tract Infection at the Ewha Womans University Hospital between 1995 and 2003 and had generalized atrophic RS on $^{99m}Tc-DMSA$ renal scan, were retrospectively studied. Atrophic RS was divided into severe atrophic RS(n=14) if relative uptake on renal scan was below 10$\%$, or mild atrophic RS(n=27) if relative uptake on renal scan was between 10-35$\%$. RS was defined as congenital if the scar was detected on the first renal scan, and as acquired if the scar developed on the follow-up renal scan from acute pyelonephritis of the first renal scan. The control group was consisted of randomly selected 41 children with segmental RS. The risk factors for atrophic RS such as the generation time, VUR, gender and ACE gene polymorphism were evaluated. Results : The age distribution of atrophic RS and segmental RS did not differ significantly (P>0.05). The rate of congenital RS in atrophic RS was 61.0$\%$(25/41), which was significantly higher than 9.8$\%$(4/41) of segmental RS(P<0.01). Atrophic RS developed mote frequently in male children(M:F 68.3$\%$ 31.7$\%$) than segmental RS(M:F 41.4$\%$ .58.5$\%$)(P<0.05). Vesicoureteral reflux(VUR) was found in 92.7$\%$(38/41) of 4he atrophic RS, which was significantly higher than 53.7$\%$(22/41) of segmental RS(P<0.05). In children without VUR, the male to female ratio did not differ between atrophic RS and segmental RS(P>0.05) But in children with VUR, there was a higher proportion of males with severe atrophic RS than segmental RS($85.7\%:45.5\%$) ACE gene polymorphism did not differ between the atrophic and segmental RS groups, irrespective of the presence of VUR(P>0.05). Conclusion : Most atrophic RSs were congenital which could not be preventable postnatally and the major risk factors were VUR and the male gender. ACE gene polymorphism was not the significant risk factor for an atrophic RS. (J Korean Soc Pedialr Nephrol 2005;9:193-200)
