Purification and Characterization of Six Fibrinolytic Serine-Proteases from Earthworm Lumbricus rubellus

  • The six lumbrokinase fractions (F1 to F6) with fibrinolytic activities were purified from earthworm Lumbricus rubellus lysates using the procedures of autolysis, ammonium sulfate fractionation, and column chromatography. The proteolytic activities on the casein substrate of the six iso-enzymes ranged from 11.3 to 167.5 unit/mg with the rank activity orders of F2 > F1 > F5 > F6 > F3 > F4. The fibrinolytic activities of the six fractions on the fibrin plates ranged from 20.8 to 207.2 unit/mg with rank orders of F6 > F2 > F5 > F3 > F1 > F4. The molecular weights of each iso-enzyme, as estimated by SDS-PAGE, were 24.6 (F1), 26.8 (F2), 28.2 (F3), 25.4 (F4), 33.1 (F5), and 33.0 kDa (F6), respectively. The plasminogen was activated into plasmin by the enzymes. The optimal temperature of the six iso-enzymes was $50^{\circ}C$, and the optimal pH ranged from pH 4-12. The four iso-enzymes (F1-F4) were completely inhibited by PMSF. The two enzymes (F5 and F6) were completely inhibited by aprotinin, TLCK, TPCK, SBTI, LBTI, and leupeptin. The N-terminal amino acid (aa) sequences of the first 20 to 22 residues of each fraction had high homology. All six isoenzymes had identical aa residues 2-3 and 13-15. The N-terminal 21-22 aa sequences of the F2, F3, and F4 isoenzymes were almost the same. The N-terminal aa sequences of F5 and F6 were identical.

Isolation of the Anti-tumor Promoters from Citrus Peels (감귤과피로부터 발암 promotion 억제활성성분의 분리)

  • This study was carried out to isolate the possible anti-tumor promoters from the citrus peel (Citrus natsudaidai Hayata). We fractionated the cold-pressed oil of citrus peel by column chromatography, HPLC and TLC. The analysis on column chromate-graphy yielded seven peaks $(F-I{\sim}F-VII)$, all of which showed single spot on TLC analysis ($R_f$ for $F-I{\sim}VIII$; 0.31, 0.13, 0.13, 0.78, 0.79, 0.69 and 0.84). Among the seven fractions, three fractions (F-I, -II and F-IV) were re-analyzed on HPLC, also showing single peak except for one fraction (F-IV) which was divided two peaks. The retention times $(R_f)$ of F-I and F-II was 3 min. and 2.5 min., respectively, but these of two peaks from F-IV were 2 min. and 4.5 min., respectively. Since the area of the latter peak (4.5 min.) was very smaller than that of the former one (2 min.), it is considered that the latter one did not appear on TLC analysis. The inhibitory effect on tumor promoter 12-O-tetradecanoylphorbol-13-acetate(TPA)-induced Epstein-Barr virus activation in Raji cells was tested for the seven fraction obtained. It decreased in order of F-VI (82.3+1.3%) > F-I (80.4+1.6%) > F-II (77.2+0.9%) > F-III (75.0+1.2%) > F-IV (74.1=1.0%) > F-V (71.0+1.1%) > F-VII (70.2+1.2%). These results imply that some constiuents contained in citrus peels have the inhibitory activity of TPA-induced tumor promotion.

QUADRATIC (ρ1, ρ2)-FUNCTIONAL EQUATION IN FUZZY BANACH SPACES

  • In this paper, we consider the following quadratic (ρ1, ρ2)-functional equation (0, 1) $$N(2f({\frac{x+y}{2}})+2f({\frac{x-y}{2}})-f(x)-f(y)-{\rho}_1(f(x+y)+f(x-y)-2f(x)-2f(y))-{\rho}_2(4f({\frac{x+y}{2}})+f(x-y)-f(x)-f(y)),t){\geq}{\frac{t}{t+{\varphi}(x,y)}}$$, where ρ2 are fixed nonzero real numbers with ρ2 ≠ 1 and 2ρ1 + 2ρ2≠ 1, in fuzzy normed spaces. Using the fixed point method, we prove the Hyers-Ulam stability of the quadratic (ρ1, ρ2)-functional equation (0.1) in fuzzy Banach spaces.

Crossability and Chromosome Variation in the Early Generation of the Crosses between the Hexaploid Triticale and Diploid Rye (6배체 트리티케일 2배체 호밀과의 잡종 초기세대에서 교잡 친화성 및 염색체 변이)

  • This experiment was carried out to obtain the information on the crossability, variation of chromosome number in pollen mother cell (PMC) and somatic cell of the progeny from the cross between hexaploid triticale cv. Sinkihomil and two diploid rye varieties. Seed set was 39.3 to 41.6% (averaged 40.5%) in the cross between triticale (P$_1$) and rye(P$_2$), which resulted in 0.33% in F$_2$(selfed F$_1$), 2.69% in F$_1$/P$_1$ 5.47% in F$_1$/P$_2$ respectively. However, seed set was extremely low in both reciprocal crosses when triticale was used as male. Germination rate of the crossed seed was 94.0% in F$_1$ 40.8% in F$_2$(selfed F$_1$), 59.5% in F$_1$/P$_1$ and 65.9% in F$_1$/P$_2$ from the cross between triticale and rye, respectively. Pollen fertility of F$_1$ plant was averaged 18.7% in the cross between triticale and rye. Number of Uni-, Bi-, and Trivalent in PMC was 12. 6, 6.94, and 0.53, respectively, in the F$_1$ between the triticale and rye. There were 28 chromosomes in F$_1$, 21 to 34 in F$_2$, 34 to 38 in F$_1$/P$_1$ and 19 to 23 in F$_1$/P$_2$ from the cross between the triticale and rye, respectively.

Combining-Ability and Heterosis for Mutant Character of Quantitative Characters in Flue-Cured Tobacco Varieties(Nicotiana tabacum L) (황색종 연초에 있어서 변이체의 조합능력 및 Heterosis)

  • This experiment were conducted to investigate heterosis and combining ability for several mutant characters by analyzing dialled crosses of flue-cured tobacco. In a dialled cross of 3 flue-cured varieties and the mutant line 83H -5, the heterosis was somewhat higher in Fl than in F2. For growth character, the heterosis was 0.28-6.03% in plant height, leaf number, leaf shape index and yield, and was 43.2% for bacterial wilt disease index. The mutant line 83H-5 showed significantly negative GCA effect for plant height, leaf width and bacterial wilt disease index in Fl and F2, leaf length in F2, and positive GCA effect for total alkaloids, total nitrogen in Fl and days to flower in F2, respectively. Specific combining ability(SCA) in 83H-5 x Hicks was significant in negative effect for leaf length(F2), number of leaves(F2), leaf shape(F1, F2), bacterial wilt(F2) and alkaloids(F1), and in 83H-5 x NC 2326 in positive effect for leaf length(F1, F2) and leaf width(F2), and for 83H-5 x NC 82 in positive effect for plant height(F1, F2) and leaf width(F2), and for 83H-5 x NC 82 in Positive effect for Plant height(F1, F2), leaf length(F2) and yield(F1, F2).

  • If a multiplicative function f is commutable with a quadratic form x2 + xy + y2, i.e., f(x2 + xy + y2) = f(x)2 + f(x) f(y) + f(y)2, then f is the identity function. In other hand, if f is commutable with a quadratic form x2 - xy + y2, then f is one of three kinds of functions: the identity function, the constant function, and an indicator function for ℕ \ pℕ with a prime p.

Removal effect of Cadmium by Polyphenol Compound Extracted from Persimmon Leaves(Diospyros kaki folium) (감잎(Diospyros kaki folium)으로부터 분리한 Polyphenol 화합물의 카드뮴 제거효과)

  • In order to investigate the removal effect of cadmium by polyphenol compound extracted from persimmon leaves(Diospyros kaki folium), animal test was done. Two fractions such as F-1 and F-2 from persimmon leaves were compared with their safety and function. The food intake of group F-1 and F-2 considerably decreased within 1% level. Cadmium addition influenced to rat growth(a tested animal), but food efficiency ratio(FER) wasn't shown any considerable difference in F-1 and F-2, as compared to the control. Cadmium content of liver, kidney and femur considerably decreased within 1% level in F-1 and F-2, compared to the control, cadmium content of liver decreased 25% in F-1, 28% in F-2 3150 decreased 22% md 25% in kidney. In femur, also decreased 53% in F-1 and 59% in F-2 respectively. The test of cadmium content in feces indicate that the content considerably increased within 1% level in both group F-1 and F-2, as compared to the control(42% in F-1, 54% in F-2). As shown the above results, have seen the removal effect of cadmium by polyphenol compound.

Effects of F1/F2 Manipulation on the Perception of Korean Vowels /o/ and /u/ (F1/F2의 변화가 한국어 /오/, /우/ 모음의 지각판별에 미치는 영향)

  • This study examined the perception of two Korean vowels using F1/F2 manipulated synthetic vowels. Previous studies indicated that there is an overlap between the acoustic spaces of Korean /o/ and /u/ in terms of the first two formants. A continuum of eleven synthetic vowels were used as stimuli. The experiment consisted of three tasks: an /o/ identification task (Yes-no), an /u/ identification task (Yes-no), and a forced choice identification task (/o/-/u/). ROC(Receiver Operating Characteristic) analysis and logistic regression were performed to calculate the boundary criterion of the two vowels along the stimulus continuum, and to predict the perceptual judgment on F1 and F2. The result indicated that the location between stimulus no.5 (F1 = 342Hz, F2 = 691Hz) and no.6 (F1 = 336Hz, F2 = 700Hz) was estimated as a perceptual boundary region between /o/ and /u/, while stimulus no.0 (F1=405Hz, F2=666Hz) and no.10 (F1=321Hz, F2=743Hz) were at opposite ends of the continuum. The influence of F2 was predominant over F1 on the perception of the vowel categories.
