Acquired JAK-2 V617F Mutational Analysis in Pakistani Patients with Essential Thrombocythemia

  • Background: Essential thrombocythemia (ET) is a clonal hemopoietic stem cell myeloproliferative neoplasm characterized by persistent thrombocytosis along with megakaryocytic hyperplasia. In the last decade following the identification of an acquired JAK2 V617F mutation, there has been acceleration in our understanding of this disease. The rational of this study was to determine the mutational profile of JAK2 V617F in Pakistan patients with ET. Materials and Methods: In this retrospective cross sectional study, 21 patients with ET were enrolled from January 2011 to December 2014. Patients were diagnosed based on WHO criteria for essential thrombocythemia. Complete blood count was done on an automated hematology analyzer, while JAK2 V617F expression was evaluated by polymerase chain reaction. Results: The mean age was $56.7{\pm}19.0$ years (range 18-87) and the male to female ratio was 1:1.1. The frequency of JAK2 V617F positivity in our ET patients was found to be 61.9%. The mean hemoglobin was $11.7{\pm}2.4$ g/dl with a total leukocyte count of $13.3{\pm}8.1{\times}109/l$ and a platelet count of $1188{\pm}522{\times}109/l$. Positive correlations for JAK2 V617F mutation were established with high TLC count and raised LDH (P<0.05). No correlation of JAK2 V617F could be established with age and gender (P>0.05). Conclusions: JAK2 V617F mutation frequency in our ET patients was similar to those reported previously. Screening for the mutation in all suspected essential thrombocythemia cases could be beneficial in differentiating patients with reactive and clonal thrombocytosis.

A Study on Microwave Dielectric Properties of Low-Temperature Sintered (Zn0.8Mg0.2)TiO3 Ceramics (저온소결 (Zn0.8Mg0.2)TiO3 세라믹의 마이크로파 유전특성에 관한 연구)

  • The effects of sintering additives such as B $i_2$ $O_3$ and $V_2$ $O_{5}$ on the microwave dielectric and sintering properties of (Z $n_1$$_{-xM}$ $g_{x}$)Ti $O_3$ system were investigated. Highly dense samples were obtained for (Z $n_{0.8}$M $g_{0.2}$)Ti $O_3$ at the sintering temperature range of 870~90$0^{\circ}C$ with B $i_2$ $O_3$ and $V_2$ $O_{5}$ additions of 〈1 wt.%, respectively. The microwave dielectric properties of (Z $n_{0.8}$M $g_{0.2}$)Ti $O_3$ with 0.45 wt.%B $i_2$ $O_3$ and 0.55 wt.% $V_2$ $O_{5}$ sintered at 90$0^{\circ}C$ were as follows : Q$\times$ $f_{o}$ = 50,800 GHz, $\varepsilon$$_{r}$ = 22, and $\tau$$_{f}$ = -53 ppm/$^{\circ}C$. In order to improve temperature coefficient of resonant frequency, Ti $O_2$ was added to the above system. The optimum amount of Ti $O_2$ was 15 moi.% when sintered at 87$0^{\circ}C$, at which we could obtain following results: Q$\times$ $f_{o}$ = 32,800 GHz, $\varepsilon$$_{r}$ = 26, and$\tau$$_{f}$ = 0 ppm/$^{\circ}C$.EX>.EX>.EX>.EX>.EX>.EX>.EX>.

  • Microwave dielectric properties of $Ca(Li_{1/4}Nb_{3/4})O_3-CaTiO_3$ ceramic systems were investigated with calcination temperatures and amounts of $CaTiO_3$ in the range of 0.2 to 0.4mol. $Ca(Li_{1/4}Nb_{3/4})O_3$ ceramics having orthorhombic crystal structure could be synthesized at $750^{\circ}C$ and sintered well at $1250^{\circ}C$. They showed the dielectric constant of 26, quality factor($Q{\times}f_o$) of 13,000 and temperature coefficient of resonant frequency(${\tau}_f$) of $-49{\pm}2ppm/^{\circ}C$ With adding the $CaTiO_3$ amount the dielectric constant and ${\tau}_f$ increased due to the solute of $CaTiO_3$ but the quality factor decreased. The 0.7$Ca(Li_{1/4}Nb_{3/4})O_3-0.3CaTiO_3$ ceramic showed the dielectric constant of 44, quality factor($Q{\times}f_o$) of 12,000 and ${\tau}_f$ of $-9{\pm}1ppm/^{\circ}C$.

Mixture Density Measurement of Biodegradable Poly(lactide-co-glycolide) Copolymer in Supercritical Solvents (초임계 용매내에서 생분해성 Poly(lactide-co-glycolide) 공중합체의 혼합물 밀도 측정)

  • The mixture density data for poly(lactide-co-glycolide) [PLGA] with supercritical $CO_2$, CHF$_3$ and CHClF$_2$ were obtained in the temperature range of 27 to 10$0^{\circ}C$ and at pressures as high as 3000 bar (PLGA$_{x}$, Where the molar concentration of glycolide in the backbone, x, range from 0 to 50 mol%). The PLA-$CO_2$, PLA-CHF$_3$, and PLA-CHClF$_2$ systems dissolve in the pressure less than 1430 below 700, and below 100 bar, respectively. The mixture density shows from 1.084 to 1.334 g/cm$^3$ at temperatures from 27 to 93$^{\circ}C$. The PLGA$_{15}$ -$CO_2$ mixture dissolves at pressures of below 1900 bar and the mixture density is in the range of 1.158 to 1.247 g/cm$^3$ at temperatures between 37 and 92$^{\circ}C$. The solubilities of the PLGA$_{25}$ for $CO_2$, CHF$_3$, and CHClF$_2$ are shown to pressure as high as 2390, 1470, and 118 bar, respectively, and the mixture density exhibits iron 1.154 to 1.535 g/cm$^3$ at temperatures from 29 to 81$^{\circ}C$. The PLGA$_{50}$-$CO_2$ system does not dissolve at 24$0^{\circ}C$ and 3000 bar while the PLGA$_{50}$-CHCIF$_2$ does easily at 5$0^{\circ}C$ and 100 bar. The mixture density for the PLGA-CHClF$_2$ system increases even at low pressures as the glycolide molar concentration increases.es.es.

  • Digestibility trial was performed with carps weighing 200 g/fish. Fecal collection was made over 7 days using a settling column. The water temperature was in the range of 27 to $30{^{\circ}C}$ during whole experimental period. Fishes were fed 5 diets (Control, $F_{24}S_{13}$, $F_{16}S_{27}$, $F_8S_{40}$ and $F_0S_{56}$) containing both 32%, 24%, 16%, 8% and 0% of fish meal (F) and 0%, 13%, 27%, 40% and 56% of roasted full-fat soybean (S), respectively. Water volume in each recirculated rearing tank was maintained at 130 l with flow rate of 10-12 l/min. Apparent digestibility coefficients (ADCs) of dry matter, protein, lipid, total carbohydrates and energy in diets showed a reduction (p < 0.05) with the increase of dietary full-fat soybean level, although there was no difference in the ADCs between Control and $F_{24}S_{13}$ (p > 0.05). The ADCs of ash, Ca and P in diets significantly increased with the increase of dietary full-fat soybean level, suggesting the high availability of phosphorus in monocalcium phosphate.

  • Equivalent conductances of ammonium salts such as $NH_4Cl,\;CH_3NH_3Cl,\;(CH_3)_2NH_2Cl,\;(CH_3)_3NHCl\;and\;(CH_3)_4NCl$ have been measured in methanol-water mixtures of various composition (0.0${\sim}$0.6-mole frcation: m. f.) in the concentration range from 0.01 to 0.04 M at $25^{\circ}C$, and limiting equivalent conductances (${\Lambda}_0$) are determined by the analysis of Fuoss Onsager's semiempirical equation.The experimental results are discussed in terms of varing the size of ${\Lambda}_0$ due to the salts species and solvent composition. It has been observed in quite common over all salts that ${\Lambda}_0$ decreases with the increase of m. f. methanol, and the maximum decrement in ${\Lambda}_0$ shows between water and 0.1 m. f., while the plot of ${\Lambda}_0$ vs. m. f. goes through a minimum at 0.4 m. f. and then turns slightly upward. On the other hand, the ${\Lambda}_0$ decrease with the increase of the size of ammonium cation and the larger values ${\Lambda}_0$ are showed in comparison with those of values observed for the corresponding $n-RNH_3Cl$ under the same experimental procedure. All these behaviors are considered to be closely correlated with the structures of solvents and solutes and the significant similarities are compared with the behaviors of partial molar volumes for amine salts in same solvent system.

Measurement of Apron Shielding Rate for X-ray and Gamma-ray (X선 및 감마선에 대한 apron의 차폐율 측정)

  • This research measured the shielding rates of apron 0.25 and 0.5 mmPb for X-ray energy in diagnosis radiation system and gamma-ray energy of $^{99m}Tc$-MDP and $^{18}F$-FDG. X-ray energies were measured on effective energy of $26.2{\sim}45.6\;keV$ when additional filtering plate of 0, 2 mmAl is used within the range of tube voltage $40{\sim}120\;kVp$, and at this time, apron 0.5 mmPb has shown about 5.5% of increase in its shielding rate over 0.25 mmPb at the highest quality. Besides, the aprons of the two types have shown high shielding rate of over 90% for direct X-ray and spatial dose rate. And, in case 0.25 and 0.5 mmPb aprons were used at 140keV of $^{99m}Tc$-MDP, the shielding effects were between 30 and 53%, and at high energy of 511 keV, $^{18}F$-FDG, the shielding effects of apron, $1.3{\sim}3.6%$, were very small.

  • Changes of nucleic acid related substances and their enzymes during rice makkulli koji making were observed and enzymological properties of crude enzymes were examined. The results obtained were as follows : (1) The amounst of acid soluble phosphorus were increased, while no remarkable changes were observed in the component of total phosphorus during koji making. (2) AMP and IMP were increased, while ADP and ATP were decreased gradually in the course of process. (3) Activities of nucleic acid degrading enzymes were increased with the lapse of time. (4) In the crude enzyme solution extracted from rice makkulli koji, the optimal pH of RNase was 4.0~5.0 and those of PDase PNase were 5.0. (5) RNase and PMase were stable at the range of pH 4.0~5.0 and PDase was stable at the pH 4.0. (6) The optimal temperature of RNase was 55$^{\circ}C$, and that of PDase was at the range of 50~55$^{\circ}C$, and 5$0^{\circ}C$ for PMase. (7) Among the three enzymes, the heat stability was in order RNase, PDase and PMase, and especially PMase was so heat labile that it was almost inactivated at 7$0^{\circ}C$ for 10 min. (8) Inhibition by metal ions and other inhibitors was disclosed : C $u^{++}$ and Z $n^{++}$ inhibited the activity of RNase, and C $u^{++}$, NaF and N $a_2$HP $O_4$ inhibited that of PDase, while C $u^{++}$ and NaF inhibited the PMase activity.ctivity.

  • A Contact-type Linear Encoder-like Capacitive Displacement Sensor (CLECDiS) has been developed to measure displacements at high accuracy within a long measurement range. In this paper, we have worked on improving the performance and reliability of the sensor. The performance increase can be done by introducing the smaller electrode patterns of $4{\mu}m$ width. In order to improve the reliability of the sensor we have changed the electrode layers from chrome-gold to chrome-gold-chrome and re-design its supporting structure. The newly-designed sensor is fabricated and tested to show that its sensitivity is $35pF/{\mu}m$, which implies that its resolution may be 0.36nm if SNR (Signal-to-Noise-Ratio) is 80.1dB. It is about ten times of that $(3.14pF/{\mu}m)$ of its previous version with 10${\mu}m$ electrodes. The total measurement range remains the same as the previous one; 15mm. The calibration experiments show its improved performance and reliability.

  • This study was to examine the change of vowel acoustic characteristics of the temporomandibular joint disorder patients by maintaining normal vocalization pattern of the temporomandibular joint through increasing the range of motion, that was, the oral cavity sonorant cavity of the temporomandibular joint, related to vowel articulation through temporomandibular training using the physical therapy. The subjects of this study were 3 male adults in 20-30s that were diagnosed with temporomandibular joint disorder. As a result of conducting temporomandibular training program using the physical therapy, the $1^{st}$ Formant Frequency(F1), $2^{nd}$ Formant Frequency(F2), and Fundamental Frequency(F0) of the temporomandibular joint disorder patients were increased compared to before and this showed the change of the $1^{st}$ Formant Frequency(F1) related to the open mouth grade of a vowel, as well as the $2^{nd}$ Formant Frequency(F2), and Fundamental Frequency(F0) related to the front-back of a vowel which shows the relationship between the temporomandibular joint, vowels and voice calculation.
