High-Performance Liquid Chromatographic Determination of Tricyclazole Residues in Rice Grain, Rice Straw, and Soil

  • An analytical method was developed to determine tricyclazole residues in rice grain, straw, and soil using high-performance liquid chromatography (HPLC) with ultraviolet absorption detection. Tricyclazole was extracted with methanol from moist rice grain, straw, and soil samples. n-Hexane washing was employed to remove nonpolar co-extractives during liquid-liquid partition. Tricyclazole was then extracted with dichloromethane from alkaline aqueous phase, while acidic interferences remained in the phase. Dichloromethane extract was further purified by silica gel column chromatography prior to HPLC determination. Reverse-phase HPLC using an octadecylsilyl column was successfully applied to separate and quantitate the tricyclazole residue in sample extracts monitored at ${\lambda}_{max}$ 225nm. Recoveries from fortified samples averaged $95.5{\pm}3.0%\;(n=6),\;87.5{\pm}20.%\;(n=6),\;and\;84.3{\pm}2.8%$ (n=12) for rice grain, straw, and soil, respectively. Detection limit of the method was 0.02 mg/kg for rice grain and soil samples while 0.05 mg/kg for rice straw samples. The proposed method was reproducible and sensitive enough to evaluate the safety of tricyclazole residues in rice grain, straw, and soil.

  • This study was carried out to investigate the making method and properties of Hanji(Korean traditional paper) for wallpaper for the purpose of mass production and mass consumption of Hanji. Paper mulberry fiber was longer than wood fiber in length and it had also much extractives and ash in content. Bast fiber was a little damaged in the fibrilation and beating process because it was weak in flexibility and compressive properties. Proper conditions for making two-ply and three-ply Hanji were examined and six kinds of Hanji were made on the basis of the three-ply paper making method. The sample OH(Original Hanji) 2-2 and OH 3-2 among them obtained good results in quality test by appearance from wallpaper makers.

Changes in Extractives of Blue Stained Woods and Its Pulping (청변목재의 추출물함량 변화와 펄프화특성)

  • 변색목재의 화학적 성질을 조사한 결과, 처리전.후의 유기용매추출물 함량에 있어서 소나무재 및 리기다소나무재 공히 노화 및 변색균처리포 그 함량이 낮아지는 결과를 나타냈는바, fatty acids와 sterols 은 3주간 노화처리로 10.6-11.2%, 변색균처리로 36-41.1%의 높은 감소율을 나타냈다. triglycerides도 3주간 노화처리로 5.68-9.4%, 변색균처리로 38.5-40.0% 높은 감소율을 나타냈다. resin acids의 경우 3주간 노화처리로 6.1-11.3%, 변색균처리로 36-41.5%의 감소율을 나타냈다. 변색목재의 펄프화와 관련하여 소나무재 및 리기다소나무재의 크라프트펄프화결과, 알칼리펄프화에 비하여 펄프수율이 2-3% 높아졌다. 최적펄프화조건으로 활성알칼리농도 20%에서 펄프수율이 46.9-47.8%, reject율이 3.21-3.52%로 낮았다. 변색균처리한 목재도 활성알칼리 20%에서 펄프수율이 비교적 높은 43.5-45%를, reject율이 1.3-1.45%로서 변색균처리 모두에서 현저히 낮아짐을 알 수 있었다.

Characteristics of pre-extracted hemicelluloses from Korean mixed wood by hot water and alkali solution and its effect on handsheet properties (열수 및 알칼리 용액을 이용하여 국산 목재 칩으로부터 선추출한 헤미셀룰로오스의 특성과 이에 따른 수초지 물성 변화)

  • Hemicelluloses pre-extracted from Korean mixed wood chip were investigated as a wet-end additive. Hemicelluloses dissolved in hot water and alkali solution were isolated by ethyl alcohol precipitation from pre-extractives. They showed molecular weight of 9,000 ~ 27,000 g/mol as revealed by size exclusion chromatography. The reduction of molecular weight through hot water extraction was caused by autohydrolysis. Chemical composition of the hemicelluloses were analyzed with high-performance liquid chromatography and UV-Vis spectroscopy. As the surface charge of isolated hemicelluloses were negative, the adsorption of hemicelluloses onto softwood unbleached kraft pulp fiber was promoted by poly-DADMAC. The physical properties of handsheets increased as the molecular weight of hemicellulose increased. On the other hands, the optical property decreased with hemicellulose adsorption.

  • Hydrophobic property of Kapok fiber was investigated by sequential removal of its components by different extraction methods. Acetone extraction for the removal of the hydrophobic extractives, holo-cel-lulose preparation after the removal of lignin and xylan extraction by potassium hydroxide was applied. The degree of hydrophobicity of each samples were measured by the water sorption ability. The water sorption ability of Kapok fiber was increased by the sequential treatment of acetone extraction, holocellulose preparation and xylan removal. Based on holocellulose compositional analysis by $^1H$-NMR spectroscopic method, the unusual high amount of the acetyl groups in the holocellulose of Kapok partially contributed to the hydrophobicity of Kapok holocellulose fiber.

Characterization of Carboxylated Cellulose Nanocrystals from Recycled Fiberboard Fibers Using Ammonium Persulfate Oxidation

  • As a way of finding value-added materials from waste medium density fiberboard (MDF), this study characterized cellulose nanocrystals (CNCs) isolated by ammonium persulfate (APS) oxidation using recycled MDF fibers. Chemical composition of the recycled MDF fibers was done to quantify α-cellulose, hemicellulose, lignin, nitrogen, ash and extractives. The APS oxidation was performed at 60 ℃ for 16 h, followed by ultrasonication, which resulted in a CNC yield of 11%. Transmission electron microscope images showed that rod-like CNCs had an average length and diameter of 167±47 nm and 8.24±2.28 nm, respectively, which gave an aspect ratio of about 20. The conductometric titration of aqueous CNCs suspension resulted in a carboxyl content of 0.24 mmol/g and the degree of oxidation was 0.04. Attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopy clearly showed the presence of carboxyl group on the CNCs prepared by the APS oxidation. The change of pH of the aqueous CNC suspension from 4 to 7 converted the carboxyl group to sodium carboxylate group. These results showed that the APS oxidation was facile and CNCs had a one-step preparation method, and thus suggested an optimization of the oxidation condition in future.

  • Pulping properties of the imported mixed eucalyptus chips and the domestic yellow poplar chips were investigated for comparing two chips as the raw materials for the hardwood chemical pulp. Soda-AQ (anthraquinone) pulping was applied for this study. The pulp from yellow poplar showed higher pulp yield than pulp from mixed eucalyptus, which comes from the lower hot-water soluble extractives and the higher polysaccharides in yellow poplar chips than those of the mixed eucalyptus chips. The yellow poplar pulp had higher average fiber length and fiber width than those of the mixed eucalyptus pulp, which led to the better respond to beating and the higher tear strength than those of the pulp from the mixed eucalyptus chips.

  • The structures of six flavonoids isolated from heartwood of Prunus sargentii(Rosaceae) were analyzed by Mass and NMR spectrometry. These flavonoids were grouped into dihydroflavonol, flavanone, and flavanone glycoside, and identified as follows : 3,3',4',5,7-pentahydroxyflavanone(taxifolin) as a dihydroflavonol, 5-hydroxy-7-methoxyflavanone(pinostrobin), 4',5,7-trihydroxyflavanone(naringenin), 3',4',5,7-tetrahydroxyflavanone(eriodictyol), 5,7-dihydroxyflavanone(pinoccmbrin) as a flavanone and 7-hydroxyflavanone 5-O-${\beta}$-D-glucopyranoside(verecundin) as a flavanone glycoside.

Determination of Buprofezin Residues in Rice and Fruits Using HPLC with LC/MS Confirmation

  • A high-performance liquid chromatographic (HPLC) method was developed to determine buprofezin residues in hulled rice and fruits. The buprofezin residue was extracted with acetone and the extract was stepwise purified by liquid-liquid partition and Florisil column chromatography. For rice samples, acetonitrile/n-hexane partition was additionally employed to remove nonpolar lipids. Reversed phase HPLC using an octadecylsilyl column was successfully applied to separate buprofezin from sample co-extractives, as detected by ultraviolet absorption at 250 nm. Recovery experiment at the limit of quantitation validated that the proposed method could evidently determine the buprofezin residue at the level of 0.02 mg/kg. Mean recoveries from hulled rice, apple, pear, and persimmon samples fortified at three tenfold levels were in the range of 80.8~85.2%, 89.1~98.4%, 88.8~95.7% and 90.8~96.2%, respectively. Relative standard deviations of the analytical method were all less than 5%, irrespective of sample types. A selected-ion monitoring LC/mass spectrometry with positive electrospray ionization was also provided to sensitively confirm the suspected residue.

  • Six compounds were isolated from the EtOAc and $Et_2O$ fractions of the bark of Rhus chinensis by repeated column chromatography with $SiO_2$ and Sephadex LH-20. The structures were determined by instrumental analysis using MS and NMR spectrophotometer as: gallic acid (1), methyl gallate (2), 6, 7-dimethoxycoumarin (3), orcinol-${\beta}$-D-glucoside (4), scopoletin (5), semialactone (6). Among these compounds, 6,7-dimethoxycoumarin (3) was isolated from this plant for the first time. To measure the antioxidant activity, the DPPH radical scavenging activity test was performed. Gallic acid (1) showed the strongest activity, while orcinol-${\beta}$-D-glucoside (4), semialactone (5) and scopoletin (6) had the low activities.
