Polyphenol 고함유 식물의 간편 PCR 분석 (A Simple and ]Reliable Method for PCR-Based Analyses in Plant Species Containing High Amounts of Polyphenols)

  • Polyphenol 화합물이 다량 함유된 식물종의 유연관계 분석이나 형질전환 유전자 확인 등을 위해 PCR을 이용할 경우 다량의 재료로부터 신속 간편하게 분리한 DNA를 이용할 수 있는 조건을 설정 하였다. 폴리페놀 함량이 높은 포도, 사과, 복분자와 같은 과수류에서 간편법에 의해 추출된 DNA를 이용한 PCR 반응액에 2%의 BLOTTO를 첨가함으로서 DNA의 재현적 증폭이 가능하였다. 간편 추출 DNA를 이용한 PCR에서 의 BLOTTO효과는 primer, 품종, 식물종에 관계없이 일반적으로 발현되었다. 상추의 형질전환 유전자 검색을 위한 PCR에서 도 BLOTTO 효과가 확인되었다. 따라서 PCR 반응액에 2% BLOTTO를 첨가하면 간편 법 에 의해 추출된 polyphenol 화합물 고함유 식물종의 DNA를 이용하여서도 PCR에 의한 유전배경 및 특정 유전자의 대량 신속 분석이 가능할 것이다.

  • Supercritical fluid extraction (SFE) technique was applied to extract cytotoxic substances from five medicinal plants including Angelica gigas, Angelica acutiloba, Aralia cordata, Spirodela polyrhiza, Bupleurum falcatum, and Acanthopanax sessiliflorus. The cytotoxicities against P388, A549, and HL-60 cell lines were determined for the supercritical carbon dioxide extracts of five plant materials employed and were compared with those of the conventional organic solvent extracts such as n-hexane, $CHCl_{3}$, and MeOH to evaluate the SFE as an alternative method to conventional organic solvent extraction. In most cases, the SFE extracts of plant materials showed enhanced cytotoxicities when compared with those of other organic solvent extracts. In addition, the optimum temperature and pressure of SFE for extraction of the cytotoxic substances were largely affected by both the plant species and the cell lines tested. These results suggested that SFE could be an alternative to the conventional organic solvent method for the selective extraction of cytotoxic compounds from plants.

Purification and Biochemical Analysis of Rice Bran Lipase Enzyme

  • A simple procedure for the extraction of the lipolytic enzyme from rice bran has been developed. High activity of lipolytic enzyme was obtained by first defatting the rice bran to remove lipid components with various extraction conditions. Then, after rove cycles of aqueous extraction, rice bran lipolytic enzyme was purified using micro- and ultrafiltration apparatus. Lipolytic enzyme activity was estimated by its hydrolytic action of tributyrin. The result indicated that the standard activity curve of butyric acid showed that the potential rice bran enzyme is a hydrolytic lipase enzyme. In addition, it showed higher lipolytic activity and specific enzyme activity with further purification by micro- and ultrafiltration. The size of rice bran lipase enzyme was identified through 15 % SDS-PAGE. The molecular weight of the rice bran lipase enzyme was 41 kDa.

  • Natural goods, especially therapeutic plants, are abundant in the World. Because they have the ability to provide all humanity with countless advantages as a source of medicines, medicinal plants are presently receiving more attention than ever. These plants' therapeutic efficacy is based on bioactive phytochemical components that have clear physiological effects on the human body. The drying process is crucial for the preparation of plant materials prior to extraction since freshly harvested plant materials include active enzymes that create active components, intermediates, and metabolic processes. Many of the phytoconstituents may be extracted using the semi-polar solvent methanol. The goal of the current work is to use the GC-MS gas chromatography- mass spectrometry technology to identify the phytochemicals and review their biological activity. In methanol leaf extract, 5 phytocompounds were found in Ricinus communis, 5 phytocompounds in Pongamia pinnata, 12 phytocompounds in Datura metal, 7 phytocompounds in Azadirachta indica, 11 phytocompounds in Acalypha indica.

  • Plants are very crucial for life on Earth. There is a wide variety of plant species available, and the number is increasing every year. Species knowledge is a necessity of various groups of society like foresters, farmers, environmentalists, educators for different work areas. This makes species identification an interdisciplinary interest. This, however, requires expert knowledge and becomes a tedious and challenging task for the non-experts who have very little or no knowledge of the typical botanical terms. However, the advancements in the fields of machine learning and computer vision can help make this task comparatively easier. There is still not a system so developed that can identify all the plant species, but some efforts have been made. In this study, we also have made such an attempt. Plant identification usually involves four steps, i.e. image acquisition, pre-processing, feature extraction, and classification. In this study, images from Swedish leaf dataset have been used, which contains 1,125 images of 15 different species. This is followed by pre-processing using Gaussian filtering mechanism and then texture and color features have been extracted. Finally, classification has been done using Multiclass-support vector machine, which achieved accuracy of nearly 93.26%, which we aim to enhance further.

  • Polymerase chain reaction (PCR) is a powerful technique in molecular biology and is widely used in various fields. By amplifying DNA fragments, PCR has facilitated gene cloning procedures, as well as molecular genotyping. However, the extraction of DNA from samples often acts as a limiting step of these reactions. In particular, the extraction of PCR-compatible genomic DNA from higher plants requires complicated processes and tedious work because plant cells have rigid cell walls and contain various endogenous PCR inhibitors, including polyphenolic compounds. We recently developed a novel solution, referred to as AnyDirect, which can amplify target DNA fragments directly from whole blood without the need for DNA extraction. Here, we developed a simple lysis system that could produce an appropriate template for direct PCR with AnyDirect PCR buffer, making possible the direct amplification of DNA fragments from plant leaves. Thus, our experimental procedure provides a simple, convenient, non-hazardous, inexpensive, and rapid process for the amplification of DNA from plant tissue.

  • There are roughly a hundred types of cables in power plants. The distribution of circuits in a nuclear plant is comprised of 20% instrument cables, 61% control cables, 13% AC power cables, 1% DC power cables, and 5% communication lines. In the nuclear power plant, medium voltage cables are generally included in the scope of systems reviewed for safety and are included in a plant's maintenance program. Medium voltage cables provide power to many critical components in plants, including feed water pumps, circulating water pumps, and condensate pumps. Among these cables, high temperature sections of cables feeding electrical power to the circulating water pump and the condenser extraction pump were found. The evaluation for these cables is performed to find the maximum allowable current and temperature. The result shows that the load current flowed about 85% of the allowable current ampacity, and the temperature of conductor at full load current did not exceed the limited temperature. Therefore, existing cables for circulating water pump and condenser extraction pump system are going to be used during design life.

다섯 가지 DNA 추출방법에 의한 옥수수 원료 및 가공시료의 DNA 추출 효율의 비교 (Comparison of the Efficiency from Raw and Processed Corns by Five Different DNA Extraction Methods)

  • 본 연구는 일반적으로 사용되는 DNA 추출방법들에 대한 효율을 비교하기 위해 수행하였다. 원료 옥수수 및 이를 가공한 시료들로부터 DNA추출을 하였으며, 추출된 DNA의 형상, 농도 및 순도 측정 그리고 PCR 분석 결과를 비교하였다. 5가지 방법으로 옥수수의 DNA를 추출한 결과, 추출방법에 따른 DNA 형상의 차이가 거의 없는 것을 확인하였으나, 각각의 시료들로부터 추출된 DNA의 양은 시료 g당 $0.25{\mu}g$부터 $234.0{\mu}g$까지 매우 다양하게 나타났다. 5가지 방법으로 옥수수 시료들의 DNA를 추출한 결과, CTAB법과 DNeasy plant Maxi 키트를 이용한 DNA 추출방법이 높은 수율을 보였다.

추출법에 따른 참당귀의 부위별 정유성분 비교 (Comparison of Volatile Compounds in Plant Parts of Angelica gigas Nakai by Extracting Methods)

  • Volatile flavor compounds from the shoot and root of Angelica gigas Nakai were extracted by HE (Hydrodistillation extraction), SDE (Simultaneous steam distillation & extraction), and SFE (Supercritical fluid extraction system), and analyzed by GC-MS. The amount and the number of chemical components in essential oils from shoot and root by SFE was the higher than those by other extraction methods. Respectively, thirty one constituents were identified from the essential oil of the shoot and root by HE, twenty seven and twenty three constituents were identified from the essential oil of shoot and root by SDE, thirty one and forty five constituents were identified from the essential oil of shoot and root by SFE. The result showed large differences in extraction methods and in plant parts of Angelica gigas Nakai. Also, the bioactive compounds in root part was identified as nodakenin and decursinol (11.95% and 8.42%, respectively) by SFE. These results suggested that SFE was the best extraction method for the increasing of extraction yield, the determination of volatile components and the increasing of bioactive compounds in the shoot and root of Angelica gigas Nakai.
