This work was conducted to obtain a rapid, accurate, and precise procedure for free amino acids analysis in Doenjang with HPLC-OPA (high performance liquid chromatography using-phthalaldehyde) and AAA (automatic amino acid analyzer) methods. Different sample extraction procedures among water, 0.1 M perchloric acid, and 0.1% meta-phosphoric acid were also compared. The optimal extraction solvent was 0.1% meta-phosphoric acid for both the HPLC-OPA and AAA methods. Good recoveries for glycine and methionine were observed using the 0.1% meta-phosphoric acid extraction with HPLC-OPA method. Method precisions (% relative standard deviation) for the free amino acids ranged for 1.62% to 8.27%, in which the HPLC-OPA method with water extraction showed the lowest value at 1.62%. Inhibition rates of the free amino acids in Doenjang were greatest with an addition of NaCI at a 1% concentration.
시각신경계의 특정추출 기구인 수용영역 즉, RF(Receptive Field)모델을 이용하여 한자의 획의 분리 및 추출에 관한 방법을 제안한다. 한자의 복잡한 정보에 대한 분리 추출과 데이터베이스화 등은 더욱 명백한 처리과정을 필요로하고 있다. 본 기법의 특 징은 망막과 대뇌 시각영역의 특징추출 기구인 수용영역을 모델링 하였고 신경세포 입력 방식에 따라 국소적인 처리에서 얻어진 정보를 대국적인 처리로 통합 추출하는 것으로서 그 기능성과 유효성을 확인할 수 있었다.
A simultaneous determination method of BTEX (benzene, toluene, ethylbenzene, o,m.p-xylene) and TPH (kerosene, diesel, jet fuel and bunker C) in soil with gas chromatography/flame ionization detection (GC-FID) was described. The effects of extracti on method, extraction solvent, solvent volume and extraction time on the extraction performance were studied. A sonication method was simpler and more efficient than Soxhlet or shaking methods. Sonication with 10 mL of acetone/methylene chloride (1 : 1, v/v) for 10 min was found to be optimal extraction conditions for 20 g of soil. Peak shapes and quantification of BTEX and TPH were excellent, with linear calibration curves over a wide range of 1-500 mg/L for BTEX and 10-5000 mg/L for TPH. Good reproducibilities by sonication were obtained, with the RSD values below 10%. By using about 20 g of soil, detection limits were 0.8 mg/L for BTEX and 10 mg/L for TPH. The advantages of this procedure are the use of simple and common equipment, reduced volumes of organic solvents, rapid extraction periods of less than 20 min, and simultaneous analysis of volatile and semivolatile compounds.
This study has been conducted to establish the optimal extraction process and HPLC analysis method for the determination of marker compounds as a part of the materials standardization for the development of health functional food materials from Astragali radix. Five extraction conditions including the shaking extraction at room temperature and the reflux extraction at $85^{\circ}C$ with 30%, 50% and 95% ethanol were evaluated. Reflux extraction with 50% ethanol showed the highest extraction yield as $27.27{\pm}2.27%$, while the extraction under reflux with 95% ethanol showed significantly the lowest yield of $10.55{\pm}0.24%$. The quantitative determination methods of calycosin-7-O-${\beta}$-D-glucoside and calycosin as marker compounds of Astragali radix extracts were optimized by HPLC analysis using a Thermo Hypersil column ($4.6{\times}250mm$, $5{\mu}m$) with the gradient elution of water and acetonitrile as the mobile phase at the flow rate of $0.8mLmin^{-1}$ and a detection wavelength of 230nm. The HPLC/UV method was applied successfully to the quantification of two marker compounds in Astragali radix extracts after validation of the method with the linearity, accuracy and precision. The contents of calycosin-7-O-${\beta}$-D-glucoside and calycosin in 50% ethanol extracts by reflux extraction were significantly higher as $1,700.3{\pm}30.4$ and $443.6{\pm}8.4{\mu}g-1$, respectively, comparing with those in other extracts. The results indicate that the reflux extraction with 50% ethanol at $85^{\circ}C$ is optimal for the extraction of Astragali radix, and the established HPLC method are very useful for the evaluation of marker compounds in Astragali radix extracts to develop the health functional material from Astragali radix.
기체크로마토그래피/질량분석법(GC/MS)을 이용하여 물시료 중에 미량으로 함유된 19종의 여러고리방향족탄화수소류에 대한 액체-액체 추출법, disk추출법 및 고체상 추출법을 이용한 시료 매트릭스로부터 분리, 추출하는 방법을 조사하였다. GC/MS의 오븐온도프로그램을 $80{\sim}310^{\circ}C$까지 단계적으로 변화시킨 결과 19종의 PAH류를 분리할 수 있었다. 액체-액체 추출법에 의한 물시료 중 PAH류의 회수율은 71.3~109.5% 범위로 나타났고, filtration에 의한 $C_{18}$ disk추출법의 회수율은 80.7~94.9% 범위로 높게 나타났다. 그러나 $C_{18}$ Sep-Pak을 사용한 고체상 추출법의 회수율은 51.8~77.9%범위이었다. 이상의 추출법에 대한 추출 회수율, 재현성 및 실험의 편리성 등 장단점을 비교한 결과, $C_{18}$ disk를 사용한 거르기법이 PAH류의 추출에 가장 적합한 방법임을 확인하였다. 각 추출법의 검출한계(S/N=5)는 액체-액체 추출법 및 고체상 추출법의 경우 0.25~6.25 ppb, disk추출법은 0.05~1.25 ppb 범위이었다.
수질오염의 효율적 관리를 위한 오염총량관리제(TMDL)의 적용에는 하천 수질모델링이 우선 되어야 하며, 이러한 모델링은 하천중심선의 추출이 필수이다. 반면, 현재는 수질모델링을 수행하는 기관들이 제각기 다른 기준으로 하천중심선을 제작하여 정확도가 높지 않으며, 이로 인하여 수질모델링의 수행기관에 따라 통일 유역임에도 수질모델링 결과가 달라지는 문제점이 발생하고 있다. 따라서 본 연구에서는 정확한 하천중심선 추출방법의 개발을 주요 목표로 하였다. 이를 위하여 최대내접원을 활용한 중심선 추출방법을 GIS 기술과 결합하여 자동화 모듈을 개발하였다. 연구 결과 기존 방법의 하천중심선 추출보다 개발된 모듈을 이용한 하천중심선의 추출이 하천형상의 변화를 보다 세부적으로 표출하는 것이 가능하였으며, 정확도 역시 향상되었다. 또한, 기존 방법의 한계점이었던 섬을 포함한 하천의 중심선 추출도 가능하여 보다 정확한 수질모델링의 지원이 가능하였다.
영국의 고원지대 (Moorland)의 이탄토양 (blanket bog peat soil에 서식하는 애지렁이 정량조사를 위한 수분갈때기추출법의 효율에 미치는 수온, 물의 량, 추출시간의 복합적인 효과를 조사하였다. 이탄 토양에서 애지렁이 추출을 위한 최적조건은 물의 양과 초기 수온에 관계없이 표면수의 최종 온도는 35$^{\circ}C$이고, 추출시간은 6시간이었다. O'Connor의 방법 (최종 온도 40-45$^{\circ}C$,열의 점진적인 증가, 3시간 추출)은 비교적 늪은 온도와 짧은 추출시간으로 인해, 추출효율에 변이가 심하였다. 열을 가하지 않고 장시간 추출하는 방식은 열을 가하는 방식에 비해 추출효율이 현저히 낮았다.
An ionic liquid-based ultrasonic-assisted extraction method has been successfully applied to the effective extraction of phenolic compounds from Laminaria japonica Aresch. Three kinds of 1-alkyl-3-methyl-imidazolium with different cations and anions were evaluated for extraction efficiency. The results showed that both the characteristics of anions and cations have remarkable effects on the extraction efficiency. In addition, the ionic liquid-based ultrasonic-assisted extraction procedure was also optimized on some extraction parameters, such as ultrasonic power, extraction time and solid-liquid ratio. Compared with the conventional solvent, the optimum approach gained the highest extraction efficiency within the shortest extraction time. Average recoveries of phenolic compounds were from 75.5% to 88.3% at three concentration levels.
This study systematically investigated a method for extraction of safflower (Carthamus tinctorius Linnaeus) colorants by ultrasonic treatment. Compared to pigments productivity and cell wall structures of safflower after general and ultrasonic method, the ultrasonic method showed high extraction efficiency of safflower pigments due to destruction of safflower cell wall caused by high vibration energies. Microscopic analysis confirmed the hypothesis that the ultrasonic treatment of safflower caused its cell wall structure loosened and made efficient extraction of safflower pigments. And also, LC-MS/MS analysis revealed that productivities of the yellow and red safflower pigments by ultrasonic method were 21.9% and 14.6% higher, respectively, than those of pigments extracted by general method. The ultrasonic extracted yellow and red colorants could be used to dye not only natural fibers like cotton, silk and wool, but also synthetic fiber like nylon, and generally gave a better color tone than the general extracted colorants from safflower due to the affinities of red and yellow colorant on different fibers. As the yellow and red colorant were extracted by ultrasonic treatment in water, the K/S value on of 550/440nm of cotton and rayon was increased but in the case of silk and wool the change of this value was almost not detected. Finally, this technique might provide a solution to establish reproducibility and standardization for the extraction and dyeing methods on fabrics.
본 논문은 지능형 로봇 시스템을 위한 인간 몸의 하이브리드 실루엣 추출 기법을 제안한다. 지능형 로봇은 내부적인 진동과 낮은 해상도로 인해 강인한 실루엣 추출을 필요로 한다. 이를 극복하기 위해서 본 논문에서는 하이브리드 실루엣 추출 기법을 제안하였다. 하이브리드 실루엣은 영상간의 공간차 및 시간차 정보를 고려 생성되며 움직임 영역 모델을 통해 두 정보간의 중요성에 가중치를 준다. 최종적으로 실험결과를 통해 제안된 기법의 우수성을 확인하였다.
