Antioxidant and Anti-Proliferative Activities of Rubus Fruits in Korea (국내산 나무딸기류 과일의 항산화 및 암세포 항증식 활성)

  • This study was conducted to determine the polyphenols, flavonoids and antioxidant activity (FRAP) of the extracts (crushed by hand or a homogenizer) of Rubus fruits (blackberry, Korean raspberry, black raspberry, boysenberry and golden raspberry) produced in Korea. In addition, their nitric oxide (NO) scavenging activity in RAW 264.7 cells and anti-proliferative activity in HT-29 and KATO-3 cells were investigated. Polyphenol and flavonoid contents in the Rubus fruits ranged from 0.6 to 8.9 and from 0.1 to 7.9 mg/g fresh fruit, respectively. Black raspberry had the highest polyphenol and flavonoid contents among the Rubus fruits. The homogenized extracts of blackberry, Korean raspberry and golden raspberry fruits showed significantly higher polyphenol and FRAP values than the hand-crushed extracts. FRAP values of the Rubus fruit extracts were significantly correlated with their polyphenol (R=0.995) and flavonoid (R=0.967) contents. The Rubus fruit extracts suppressed the NO secretions in LPS-treated RAW264.7 cells. There were no significant differences between extracts obtained by crushing by hand and those obtained using a homogenizer. Proliferation rates of HT-29 and KATO-3 cancer cells treated with the Rubus fruit extracts at 0.1, 0.25 and 0.5 mg/mL were reduced by 3~32% and 0~57%, respectively. The homogenized extracts of blackberry and Korean raspberry fruits had significantly higher anti-proliferation activity against HT-29 cancer cells than the hand-crushed extracts. However, extraction method did not show any significant difference on proliferation of KATO-3 cancer cells. The NO scavenging activity of the Rubus fruit extracts were significantly correlated with the anti-proliferation activities of the HT-29 (R=0.602) and KATO-3 cells (R=0.498).

Development of Cloud and Shadow Detection Algorithm for Periodic Composite of Sentinel-2A/B Satellite Images (Sentinel-2A/B 위성영상의 주기합성을 위한 구름 및 구름 그림자 탐지 기법 개발)

  • In the utilization of optical satellite imagery, which is greatly affected by clouds, periodic composite technique is a useful method to minimize the influence of clouds. Recently, a technique for selecting the optimal pixel that is least affected by the cloud and shadow during a certain period by directly inputting cloud and cloud shadow information during period compositing has been proposed. Accurate extraction of clouds and cloud shadowsis essential in order to derive optimal composite results. Also, in the case of an surface targets where spectral information is important, such as crops, the loss of spectral information should be minimized during cloud-free compositing. In thisstudy, clouds using two spectral indicators (Haze Optimized Tranformation and MeanVis) were used to derive a detection technique with low loss ofspectral information while maintaining high detection accuracy of clouds and cloud shadowsfor cabbage fieldsin the highlands of Gangwon-do. These detection results were compared and analyzed with cloud and cloud shadow information provided by Sentinel-2A/B. As a result of analyzing data from 2019 to 2021, cloud information from Sentinel-2A/B satellites showed detection accuracy with an F1 value of 0.91, but bright artifacts were falsely detected as clouds. On the other hand, the cloud detection result obtained by applying the threshold (=0.05) to the HOT showed relatively low detection accuracy (F1=0.72), but the loss ofspectral information was minimized due to the small number of false positives. In the case of cloud shadows, only minimal shadows were detected in the Sentinel-2A/B additional layer, but when a threshold (= 0.015) was applied to MeanVis, cloud shadowsthat could be distinguished from the topographically generated shadows could be detected. By inputting spectral indicators-based cloud and shadow information,stable monthly cloud-free composited vegetation index results were obtained, and in the future, high-accuracy cloud information of Sentinel-2A/B will be input to periodic cloud-free composite for comparison.

Monitoring of a Time-series of Land Subsidence in Mexico City Using Space-based Synthetic Aperture Radar Observations (인공위성 영상레이더를 이용한 멕시코시티 시계열 지반침하 관측)

  • Anthropogenic activities and natural processes have been causes of land subsidence which is sudden sinking or gradual settlement of the earth's solid surface. Mexico City, the capital of Mexico, is one of the most severe land subsidence areas which are resulted from excessive groundwater extraction. Because groundwater is the primary water resource occupies almost 70% of total water usage in the city. Traditional terrestrial observations like the Global Navigation Satellite System (GNSS) or leveling survey have been preferred to measure land subsidence accurately. Although the GNSS observations have highly accurate information of the surfaces' displacement with a very high temporal resolution, it has often been limited due to its sparse spatial resolution and highly time-consuming and high cost. However, space-based synthetic aperture radar (SAR) interferometry has been widely used as a powerful tool to monitor surfaces' displacement with high spatial resolution and high accuracy from mm to cm-scale, regardless of day-or-night and weather conditions. In this paper, advanced interferometric approaches have been applied to get a time-series of land subsidence of Mexico City using four-year-long twenty ALOS PALSAR L-band observations acquired from Feb-11, 2007 to Feb-22, 2011. We utilized persistent scatterer interferometry (PSI) and small baseline subset (SBAS) techniques to suppress atmospheric artifacts and topography errors. The results show that the maximum subsidence rates of the PSI and SBAS method were -29.5 cm/year and -27.0 cm/year, respectively. In addition, we discuss the different subsidence rates where the study area is discriminated into three districts according to distinctive geotechnical characteristics. The significant subsidence rate occurred in the lacustrine sediments with higher compressibility than harder bedrock.

An Analysis of a 100-Years-Old Map of the Heritage Trees in Jeju Island (제주도 노거수 자연유산의 100년 전과 현재 분석)

  • The purpose of this study is to verify and reconstruct the record information for big old trees of Jeju on the basis of the precise map of Jeju island in 1918 which was produced 100 years ago. For the analysis of high altitude, coordinate system and georeferencing were performed by selecting representative points using ArcGIS. We extracted digitized information by using point extraction method and extracted attribute information based on legend type and relative size in map. Based on the map of the past 100 years ago, the present situation of the big old tree in Jeju was analyzed and their characteristics were analyzed. In addition, based on the information of the protected big old trees in present, we discussed the characteristics of past tree (1918), present tree (2019), and contribution of big old tree in Jeju landscape and vegetation. As a result, 1,013 individuals were distributed in Jeju Island 100 years ago. Even when it was intensive in the use of timber, the big old trees were protected, and contributed as a representative component of Jeju's unique landscape. The remaining distribution of Jeju's big old tree is 159 trees. As in the past, distribution has been confirmed around the lowlands, but declines in numbers are found throughout the island. The major factors for the decline of individuals are large-scale development projects such as reaching the limit of life, natural disturbance (typhoon, disease, pest, drought, etc.). However, it is presumed that a large number of individuals have played a leading role in shaping the current forests as contributing to important species sources in the restoration process of Jeju vegetation. However, it is presumed that a large number of individuals (405) have played a leading role in forming the present forest by contributing to the species pool in the restoration process of Jeju vegetation.

Cytotoxicity of Particulate Matter in Various Human Cells Lines (미세먼지가 다양한 사람 세포주에 미치는 세포 독성)

  • The present study investigated the cytotoxicity of particulate matter (PM) derived from car air filter (outdoor PM) and home cleaner filter (indoor PM) in the various human cell lines. Each outdoor and indoor PM were harvested by ethanol extraction method, subsequently sieved with 10 um filter paper, sterilized with autoclave and added to culture media. The half maximal inhibitory concentration ($IC_{50}$) values was significantly (p<0.05) lower in the outdoor PM, compared with indoor PM, and the significantly (p<0.05) higher $IC_{50}$ values were observed in the cancer cell lines (A-549 lung adenocarcinoma and AGS stomach adenocarcinoma), than those of normal MRC-5 fibroblasts and dental papilla tissue derived-mesenchymal stem cells (DSC). After being exposed to $100{\mu}g/ml$ outdoor PM for 7 days, the population doubling time (PDT) was significantly (p<0.05) increased in especially MRC-5 and DSC cell lines, compared with untreated cell lines. Further, the expression of senescence-associated ${\beta}$-galactosidase activity was up-regulated in all the cells exposed to outdoor PM than those of untreated control. Besides, the expression level of inflammation-associated genes, such as cyclooxygenase-2 (COX-2) and interleukin-6 (IL-6) was found to be significantly (p<0.05) increased in the outdoor PM-treated cell lines than those of untreated cell lines. Our results showed that PM induces the cytotoxicity via arrest of cell growth, cell damage and inflammation response.

Manufacturing Fermented Rapeseed Meal Compost using Two Microbial Agents and the Effect of Their Application (유용 미생물 제제 이용 발효 유채박 비료 제조 및 시용 효과)

  • Rapeseed meal, which is a byproduct of rapeseed oil extraction, improves crop productivity by supplying nutrients to the soil. The present study aimed to manufacture fermented rapeseed meal compost using two effective microbial agents and evaluate their efficiency as fertilizer. To types of fermented rapeseed meal, manufactured using either a bio-carrier or microbial agent, showed no differences in pH, electrical conductivity (EC), and total nitrogen content. However, the contents of $NH_4-N$ and $NO_3-N$ as inorganic nitrogen were increased by 5.6 times and 1.5 times, respectively, after 5 d of fermentation. Rapeseed meal fermented for 5 d was applied to tomato a basal fertilizer and after eight weeks, the plant height increased in all fermented rapeseed treatments compared to that in the chemical fertilizer treatment, and also the quantum yield of photosystem II (PS II) showed the same trend. The total nitrogen content of tomato leaves treated with a microbial fermented rapeseed meal was twice as high as that of that treated with a chemical fertilizer. It was confirmed that the increase in the tomato height was an effect of the rapeseed meal containing inorganic nitrogen, which can easily be absorbed by plants. From these results, it is considered that fermented rapeseed meal manufactured with an effective microbial agent for 5 d showed the highest inorganic nutrient content and greatest growth enhancement in tomato.

Development of Samgyetang Broth from Air-dried and Oven-roasted Chicken Feet (열풍건조 및 오븐구이 닭발로부터 추출한 삼계탕 육수 제조)

  • This study was conducted to develop and compare Samgyetang broth from extract of pre-treated chicken feet. Chicken feet were subjected to non-heating (control), heating at $70^{\circ}C$ for 12 h in a hot air dryer, and heating at $180^{\circ}C$ for 1 h in an oven. The heat-treated chicken feet were extracted at $121^{\circ}C$ for 1 h and 2 h, respectively. The extract was placed in a pouch with whole chicken carcass ($470{\pm}10g$). The sealed Samgyetang retort was made according to the industrial method. The pH of the extract from preheated chicken feet was lower than that extracted from fresh chicken feet. The Thiobarbituric Acid Reactive Substances (TBARS) value of the preheated chicken feet extract was significantly lower (P<0.05) than that of fresh chicken feet extract, but there were no significant differences among the broths. As the extraction time increased, the pH and TBARS value decreased in the extract (P<0.05) but increased in the broth (P<0.05). According to the sensory evaluation test, the extract from 1 h hot air heating and drying was significantly better in appearance, aroma, and overall preference than the other treatments (P<0.05). The GC-MS results showed that benzaldehyde and benzothiazole, which are widely known to give meat and nuts flavor, were detected in those treatments (P<0.05). The Samgyetang broths prepared from 1 h hot air heating and drying extract were significantly higher in the overall acceptability according to the sensory test (P<0.05). In summary, the quality of retort Samgyetang broth can be improved by adding chicken feet extract which is subjected to heating and drying for 1 h.

Analysis of Research Trends of 'Word of Mouth (WoM)' through Main Path and Word Co-occurrence Network (주경로 분석과 연관어 네트워크 분석을 통한 '구전(WoM)' 관련 연구동향 분석)

  • Word-of-mouth (WoM) is defined by consumer activities that share information concerning consumption. WoM activities have long been recognized as important in corporate marketing processes and have received much attention, especially in the marketing field. Recently, according to the development of the Internet, the way in which people exchange information in online news and online communities has been expanded, and WoM is diversified in terms of word of mouth, score, rating, and liking. Social media makes online users easy access to information and online WoM is considered a key source of information. Although various studies on WoM have been preceded by this phenomenon, there is no meta-analysis study that comprehensively analyzes them. This study proposed a method to extract major researches by applying text mining techniques and to grasp the main issues of researches in order to find the trend of WoM research using scholarly big data. To this end, a total of 4389 documents were collected by the keyword 'Word-of-mouth' from 1941 to 2018 in Scopus (www.scopus.com), a citation database, and the data were refined through preprocessing such as English morphological analysis, stopwords removal, and noun extraction. To carry out this study, we adopted main path analysis (MPA) and word co-occurrence network analysis. MPA detects key researches and is used to track the development trajectory of academic field, and presents the research trend from a macro perspective. For this, we constructed a citation network based on the collected data. The node means a document and the link means a citation relation in citation network. We then detected the key-route main path by applying SPC (Search Path Count) weights. As a result, the main path composed of 30 documents extracted from a citation network. The main path was able to confirm the change of the academic area which was developing along with the change of the times reflecting the industrial change such as various industrial groups. The results of MPA revealed that WoM research was distinguished by five periods: (1) establishment of aspects and critical elements of WoM, (2) relationship analysis between WoM variables, (3) beginning of researches of online WoM, (4) relationship analysis between WoM and purchase, and (5) broadening of topics. It was found that changes within the industry was reflected in the results such as online development and social media. Very recent studies showed that the topics and approaches related WoM were being diversified to circumstantial changes. However, the results showed that even though WoM was used in diverse fields, the main stream of the researches of WoM from the start to the end, was related to marketing and figuring out the influential factors that proliferate WoM. By applying word co-occurrence network analysis, the research trend is presented from a microscopic point of view. Word co-occurrence network was constructed to analyze the relationship between keywords and social network analysis (SNA) was utilized. We divided the data into three periods to investigate the periodic changes and trends in discussion of WoM. SNA showed that Period 1 (1941~2008) consisted of clusters regarding relationship, source, and consumers. Period 2 (2009~2013) contained clusters of satisfaction, community, social networks, review, and internet. Clusters of period 3 (2014~2018) involved satisfaction, medium, review, and interview. The periodic changes of clusters showed transition from offline to online WoM. Media of WoM have become an important factor in spreading the words. This study conducted a quantitative meta-analysis based on scholarly big data regarding WoM. The main contribution of this study is that it provides a micro perspective on the research trend of WoM as well as the macro perspective. The limitation of this study is that the citation network constructed in this study is a network based on the direct citation relation of the collected documents for MPA.

Physiological Activity of Supercritical Poria cocos back Extract and Its Skin Delivery Application using Epidermal Penetrating Peptide (초임계 복령피 추출물의 생리활성 및 경피투과 펩티드를 이용한 경피 약물전달의 응용)

  • In this study, Poria cocos bark were extracted by supercritical process, and anti-inflammatory, whitening, and antioxidant effects were measured in comparison with ethanol extract. Also, An effective percutaneous permeation method using a selected formulation of the extract and a drug delivery peptide was proposed. Pachymic acid, known as the anti-cancer and anti-inflammatory compound of the ventricle, is an indicator component and the HPLC analysis shows that the supercritical extract of the pericardium is more than twice that of the Poria cocos bark extract. In order to confirm antioxidative effect of Bombyx mori, DPPH scavenging ability and ABTS scavenging ability test showed that the ethanol extract of Poria cocos Back had lower concentration than the supercritical extract of Poria cocos back. However, RAW 264.7 Measurements of Nitric oxide (NO) production in cells showed lower NO production at the same concentration than the Poria cocos back ethanol extract. In addition, after 72 hours of processing of $20{\mu}g/mL$ of the Poria cocos back extract in B16 melanoma cells, both the intracellular and extracellular melanin extract were effective and the supercritical extract was lower melanin content. No toxicity was observed at the concentration of $800{\mu}g/mL$ in RAW 264.7 cells used in NO production experiments. However, in B16 melanoma cells, even at $50{\mu}g/mL$, both Poria cocos back ethanol extract and supercritical extract showed a survival rate of less than 60%. The liposome formulation and drug delivery peptides were shown to be useful for percutaneous permeation of Supercritical Extract of Poria cocos back using a liposome formulation and a drug delivery peptide. it is expected that there will be great potential for development as a variety of cosmetic materials for Poria cocos back.

Enhancement of Anti-inflammatory Activity of Lactobacillus plantarum Fermented by Achyranthes japonica on Extraction Solvents (추출 용매에 따른 Lactobacillus plantarum 발효 우슬의 항염증 효과 증진)

  • In this study, we used extracts obtained from five different solvents (water, ethanol, hexane, ethyl acetate, butanol) of Achyranthes japonica (AJ) and also AJ fermented with Lactobacillus plantarum (LP) to confirm effects on the anti-inflammatory activity in RAW264.7 cells. Experiments of measuring nitric oxide (NO) and cytokine production were performed in lipopolysaccharide (LPS)-induced RAW264.7 cells, and the expression of both cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) was observed by a western blot method. The cytotoxicity of RAW264.7 was confirmed by the cell counting kit (CCK) assay at a concentration of $100{\mu}g/mL$, which has no toxicity. As a result of the inhibition of NO production, the inhibition rate of AJ-LP extracted with ethanol samples was about 74% higher than that of using the control group. Interleukin-6 (IL-6), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and Interleukin-$1{\beta}$ (IL-$1{\beta}$), which are inflammatory cytokines, also showed an excellent efficacy with inhibition rates of about 57, 70, and 74%, respectively. Comparing to the results of COX-2 and iNOS expression in the AJ group, the inhibition rate of 20-hydroxyecdysone was the highest than others. On the other hand, the COX-2 expression level of AJ-LP group decreased about 16% compared to that of the control group, and the iNOS expression level was also decreased about 7%. These results suggest that the extract of AJ fermented from L. plantarum can be used as an anti-inflammatory natural material.
