Determination of PCDD/Fs in Disposable Diaper for Infant (유아용 일회용 기저귀의 다이옥신 분석)

  • This study analyzed and standardized the harmful substances of organic compounds(dioxins) in disposable diapers for infant's clothing. Commercial samples obtained from Korea, Japan, America and German were determined by HRGC/HRMS. Experimental Methods were consisted of comparision of extracting solvent, analysis of PCDD/Fs, and analysis of PCDD/Fs of extraction in disposable diaper. The results were as follows. No high TEQ values in TCDD(1), PCDD(0.5) and HxCDD(0.1) were found for commercial disposable diapers from country Korea, Japan, U.S.A and German. In extraction for 6hrs experiments, HpCDD and OCDD were detected in disposable diapers of U1, U2 and G2, respectively. In extraction fur 24hrs experiments, HpCDD, OCDD and OCDF were detected in disposable diapers of J3, U1, U2 and U3. So we need to set standards of measurement for harmful materials contained in textile goods. This will make a safe and comfortable clothing environment for all users, regardless of age.

  • Jindo Hongju is a traditional liquor in Jindo island of Korea. The characteristics of Hongju are its unique flavour by fermetation and red color of gromwell(Lithospermum erythrorhizon) root. However, the evaluation of red pigment is different from one manufactures to other manufactures and from place to place, also the standard method is not established. An attempt has made to compare the quality of gromwell root from different places and to standardize the extracting method. The results obtained from this study are summerized as follow, The chemical properties and composition of gromwell root from Jindo and other areas were compared. There were no difference among the samples in moisture content, content of naphtoquinone derivatives and absoption spectra. These results indicate that the pigments from Jindo and other region products seems to be the same quality. For efficient extraction of gromwell pigment, more than 40% ethanol as solvent and at least 10 hours extraction time was required. According to the visual test for Hongju pigment, the most preferable color was that it shows absorbance of 1.0 (contents of shikonin was 3.90 mg/45% EtOH 20 ml). From this visual test it can be proposed that the may be applied absorbance at 1.0 for the quality control of pigment.

Comparing of 5-Nonylsalicylaldoxime and Salicylaldehyde Characterization Using Magnesium Salt Formylation Process

  • 5-Nonylsalicylaldoxime and salicylaldehyde are two derivatives of phenolic compounds which are very applicable materials in industries. Formerly the formylation of phenolic derivatives were carried out by Rimer-Tiemann method. In this work both of these two materials were synthesized by magnesium meditated formylation technique and their structural characterizations were compared by instrumental analysis technique. In order to achieve a selectively orthoformylated product, the hydroxyl group of nonylphenol (or phenol) was first modified by magnesium methoxide. The nonylphenol magnesium salt was then formylated by paraformaldehyde. The oximation reaction was finally applied to the prepared nonylsalicylaldehyde magnesium salt by liquid extracting via water and acid washing and other extractions. The solvent was finally removed by evaporation under reduced pressure. Some instrumental analysis such as $^1H$-NMR, GC/MS and FT-IR spectra were taken on the product in order to interpret the reaction characterization quantitatively and qualitatively. The formaldehyde and oxime functional groups of two compounds were investigated through $^1H$-NMR and FT-IR spectra and were compared. The yield of methoxilation was very good and the yields of formylation and oximation reactions were about 90%and 85% respectively. The orthoselectivity of formylation reaction were evaluated by comparing of the relevant spectra. The GC/MS spectra also confirmed the obtained results.

Studies on the Extracting Methods of Ginseng Extract and Saponins in Panax Ginseng (유출조건에 따른 인삼중의 Ginseng Extract와 Saponin 합량변화에 관한 연구)

  • In order to investigate the optimal conditions which affects to extraction of ginseng extract and saponin in ginseng extract, experiment was carried out varing with ethanol percentage, extraction time, temperature, sol$.$vent and Plant Parts. The results art as follows: 1. The amounts of ginseng saponin was increased according to increanation of ethanol Percentage while the amounts of ginseng extract was decreased. 2. The amounts of ginseng extract was increased as the prolongation of extraction time, on the ether hand, ginseng saponin contents increased lentil 40hr. and decreased after that. 3. By the raise of extract temperature, both of the amounts of ginseng saponin and ginseng extract was increased two times and four times. respectively. 4. The total amounts ginseng extract was obtained 22.86u when the water used as the extraction solvent, 11.28% on ethanol and 11.04U on methanol, in the order. and the saponin contents gained when the extraction solvents of water, methanol and ethanol 7.47%, 12.36% and 12.77%, respectively. 5. It showed 9.23% of ginseng extract in epidermis and 8.4% of ginseng saponin in tail Part of raw ginseng and in the case of dried ginseng, ginseng extract and saponin showed the most amounts in epidermis of 18.28% and 19.35%, respectively. 6. The ratio of panaxadiol and panaxatriol contents of ginseng saponin was almost same when it was extracted varing with ethanol percentage and extraction time (duration), and the more alcohol percentage and the longer extraction time increased, the more fractional content of ginseng saponin was extracted.

A Study on the Production of Ginseng Extracts 1. Production of extracts from fresh ginseng (인삼엑기스 제조에 관한 연구 제1보, 수삼 엑기스 제조)

  • In order to establish effective extracting method of ginseng extracts from fresh ginseng, the yield, chemical composition, physical properties and organoleptic quality of the extracts, which are extracted with various concentrations of ethanol, were investigated. The results are as follows : 1. The yield of the extracts was increased with decreasing the concentration of ethanol as solvent. As in case of water as a solvents, the highest yield was achieved when 23.64% of water was used. The yield were 12.3% and 9.05%, when 70% and 90% of ethanol were used, respectively lively. 2. Crude protein content is the highest level and nitrogen·free extracts content is the lowest at the concentration of 50% ethanol. Lipid was increased linerly while ash was decreased as increment of ethanol concentration. 3. Viscosity and residue of the extracts also decreased in accordance with the increament of ethanol concentration and the transmittance value and pH of extract solutions were almost similar except transmittance value of the water extracts. 4. The extracts extracted with 70% ethanol gave the best result of sensory test. The total sensory test score of each extracts (70%, 90%, 50%, 0% and 30%) were 70, 65, 50, 46 and 41, respectively.

  • Investigations were carried out on a $\beta$-glucosidase produced by Aspergillus niger under solid-state fermentation conditions as a model of enzyme recovery from fermented wheat bran. The leaching efficiency of distilled water to recover the enzyme from the fermented bran was higher than acetate buffer, citrate buffer, citrate-phosphate buffer and 5% methanol; thus, the conditions were further optimized with distilled water as the extracting agent. After fermented bran was washed three times with distilled water for 1.5 hr each under shaking conditions at 1:5 solid to solvent ratio, a maximum recovery of 0.025 U/g of wheat bran was obtained.

  • This study investigated an efficient method for the extraction of astaxanthin from the red yeast Xanthophyllomyces dendrorhous. The extraction process comprised three steps: 1) cultivating the yeast; 2) treating the yeast culture suspension with microwaves to destroy the cell walls and microbodies; and 3) drying the yeast and extracting the astaxanthin pigment using ethanol, methanol, acetone, or a mixture of the three as the extraction solvent. Ultimately, various treatment tests were performed to determine the conditions for optimal pigment extraction, and the total carotenoid and astaxanthin contents were quantified. A frequency of 2,450 MHz, an output of 500 watts, and irradiation time of 60 s were the most optimum conditions for yeast cell wall destruction. Furthermore, optimal pigment extraction occurred when using a cell density of 10g/l at $30^{\circ}C$ over 24 h, with a 10% volume of ethanol.

Characterization of Crosslinks of Maleic Anhydride-Grafted EPDM/Zinc Oxide Composite Using Dichloroacetic Acid/Toluene Cosolvent and Extraction Temperature (디클로로아세트산/톨루엔 공용매와 추출 온도를 이용한 무수말레산-그래프트 EPDM/산화 아연 복합체의 가교 특성 분석)

  • Crosslink characteristics of maleic anhydride-grafted EPDM (MAH-g-EPDM)/zinc oxide composite were investigated by weight losses after dichloroacetic acid (DCA)/toluene cosolvent extraction at different temperatures and by measurement of crosslink densities. The chemical changes were analyzed using attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR). The weight losses by extraction at high temperature ($90^{\circ}C$) were remarkably greater than those at room temperature and those by DCA/toluene cosolvent extraction were greater than those by toluene one by more than 5 times. The crosslink densities were measured after the solvent extraction, and the second crosslink densities were higher than the first ones. The first crosslink density was lower when the extraction temperature was high, and it was much lower for the toluene extraction than for the DCA/toluene cosolvent extraction. The second crosslink density of the sample extracted with DCA/toluene cosolvent was greater than that extracted with toluene. The extracted components were depending on the extraction solvents and temperatures, for example; only strong crosslinked networks were remained when extracting with DCA/toluene cosolvent at high temperature, while only uncrosslinked polymer chains were extracted when extracting with toluene at room temperature. Therefore, crosslink characteristics of the MAH-g-EPDM/zinc oxide composite can be analyzed by comparison of the extracted components according to the extraction solvents and temperatures and by measurement of successive crosslink densities.

Studies on Efficient Extraction of Limonene from Citron and Immune-modulation Activity for Development of Environmentally Friendly Material (친환경 소재 개발을 위한 유자에서의 효율적 Limonene 추출 및 면역기능 조절활성에 관한 연구)

  • The objectives of this study were to find out the best condition of extracting methods of limonene from citron and to determine effects of limonene on immune modulation activity by measuring cytokine secretion using RAW 264.7 mouse macrophage cells. When distilled water was used as a solvent instead of organic solvents to extract limonene from citron, addition of refluxing process to simultaneous steam distillation extraction method was found to be much effective in extracting limonene. However, it required longer extraction time than using other organic solvents. Limonene extracts showed increased IL-β and IL-6 but decreased the TNF-α gene expression in limonene concentration dependant manner. However oral administration of limonene extracts to mice did not influence significantly compared to control in in vivo experiment. It might be due to that the mice were kept in well controlled and complete environment. Limonene, a natural material from citron has been approved to have a immune-modulation activity in the present study and have a potential as a feed additive that is environmentally friendly and no harmful. Further study with protected limonene, for example, for the protection of limonene from oxidation or bypass the ruminal degradation in order consequently to increase immune-modulation activity might be useful as a further research.

Quality Properties of Conger Eel (Conger myriaster) Oils Extracted by Supercritical Carbon Dioxide and Conventional Methods (초임계 이산화탄소 및 유기용매를 이용하여 추출된 붕장어(Conger myriaster) 오일의 품질특성)

  • In this study, the extraction of Conger myriaster oil by using supercritical carbon dioxide (SC-CO2) and organic solvent was investigated. The extraction conditions conducted for SC-CO2 varied for pressure (25, 30 MPa) and temperature (45, 55 ℃), while the SC-CO2 flow rate was kept constant during the experiment (27 g min-1) and hexane was used as a conventional organic solvent. The extraction yield indicated that the best extraction condition would be SC-CO2 at 55 ℃ and 30 MPa, resulting in the highest yield of 37.73 ± 0.14%. The oils were characterized for their fatty acid (FAs) composition using gas chromatography, while it was revealed that the major FAs were mystric acid, palmitoleic acid, oleic acid, electroosapentaenoic acid (EPA), and docosahexaenoic acid (DHA). The oxidation stability of the extracted C. myriaster oil was evaluated by measuring the acid value, peroxide value, and free fatty acid. The best oxidative stability was obtained from SC-CO2 extracted oil at 30 MPa and 55 ℃. There was a significant difference in the color properties of the SC-CO2 and hexane extracted oils, with the SC-CO2 extracted oil showing better chromaticity than the oil extracted using hexane. Extracting oils from C. myriaster with SC-CO2 could bring better economic benefits than using organic solvents. When supercritical carbon dioxide was used, there was no post-treatment process; thus, it was confirmed that this is a more environmentally friendly oil extraction method.
