Effects of fermented ginseng on memory impairment and β-amyloid reduction in Alzheimer's disease experimental models

  • This study examined the effect of fermented ginseng (FG) on memory impairment and ${\beta}$-amyloid ($A{\beta}$) reduction in models of Alzheimer's disease (AD) in vitro and in vivo. FG extract was prepared by steaming and fermenting ginseng. In vitro assessment measured soluble $A{\beta}42$ levels in HeLa cells, which stably express the Swedish mutant form of amyloid precursor protein. After 8 h incubation with the FG extract, the level of soluble $A{\beta}42$ was reduced. For behavioral assessments, the passive avoidance test was used for the scopolamine-injected ICR mouse model, and the Morris water maze was used for a transgenic (TG) mouse model, which exhibits impaired memory function and increased $A{\beta}42$ level in the brain. FG extract was treated for 2 wk or 4 mo on ICR and TG mice, respectively. FG extract treatment resulted in a significant recovery of memory function in both animal models. Brain soluble $A{\beta}42$ levels measured from the cerebral cortex of TG mice were significantly reduced by the FG extract treatment. These findings suggest that FG extract can protect the brain from increased levels of $A{\beta}42$ protein, which results in enhanced behavioral memory function, thus, suggesting that FG extract may be an effective preventive or treatment for AD.

Analgesic and antipyretic actions of Muntingia calabura leaves chloroform extract in animal models

  • The present study was carried out to elucidate the potential of Muntingia (M.) calabura leaves chloroform extract (MCCE) as antinociceptive, anti-inflammatory and antipyretic agents using various animal models. The dried powdered leaves of M. calabura (20 g) were soaked in chloroform for 72 h and the supernatant obtained was then evaporated to dryness. The crude dried extract (0.912 g), dissolved in dimethyl sulfoxide (1:20; w/v) and considered as a stock solution (100% concentration/strength), was then diluted to the concentrations of 10 and 50% and used together in all experimental models. The MCCE was found to show significant (P < 0.05) antinociceptive and antipyretic activities, but less remarkable anti-inflammatory activity. Only the antinociceptive activity of MCCE measured using the abdominal constriction test and in the first phase of the formalin test occurred in a concentration-dependent manner. The anti-inflammatory activity of 50 and 100% concentrations MCCE was observed only at the range of time interval of 60 - 120 and 60 min, respectively. Based on the results, we conclude that the M. calabura leaves chloroform extract possessed remarkable antinociceptive and antipyretic, but less effective anti-inflammatory, activities and thus justifies the Peruvian folklore claims of its medicinal values.

헛개나무(Hovenia dulcis var. koreana) 과병 열수추출분획물의 간보호 및 혈중 알콜 저하 작용 (Hepatoprotective and Blood Alcohol Lowering Effects of Fruit Peduncle Extract of Hovenia dulcis var. Koreana in the In Vitro and in Vivo Animal Models)

  • Hovenia dulcis which is distributed in Korea, China and Japan is known to show hepatoprotctive effect and reduce the acute alcohol toxicity. In this study, the hepatoprotective effect against the chemically induced experimental liver injury models and lowering effect of blood alcohol level in animal models acutely administered alcohol by the peduncle extracts of Hovenia dulcis var. koreana were investigated. HdfHW-1, and HdfM-1 which are the extracts of fruit peduncles and young branches with hot water or 70% methanol and followed with 100% methanol, were significantly reduced the $CCl_4$ or D-galactosamine/LPS induced damage in sliced liver. The hot water or methanol extracts of fruit peduncle protected dose-dependently against $CCl_4$ induced toxicity in primary hepatocyte culture and particularly, the amount of LDH release was reduced to the control level at 500 $\mu\textrm{g}$/ml of hot water extracts. HdfHW-1 also decreased the hepatotoxicity induced by $CCl_4$ in rats. The active components of HdfHW-1 seemed to be high molecular weights because 0.2 M NaCl HdfHW-1 fraction was the most effective among NaCl fractions of HdfHW-1 eluted with various concentrations of NaCl on DEAE 650C column chromatography. HdfM and HdfHW were significantly reduced the levels of blood alcohol in rats and mice administered 40% alcohol. These results indicated that the hot water or methanol extracts of fruit peduncle of Hovenia dulcis var. koreana have hepatoprotective effect and may be reduce alcohol toxicity.

  • Purpose: An animal periodontitis model is essential for research on the pathogenesis and treatment of periodontal disease. In this study, we have introduced a lipopolysaccharide (LPS) of a periodontal pathogen to the alveolar bone defect of experimental animals and investigated its suitability as a periodontitis model. Methods: Alveolar bone defects were made in both sides of the mandibular third premolar region of nine beagle dogs. Then, the animals were divided into the following groups: silk ligature tied on the cervical region of tooth group, Porphyromonas gingivalis LPS (P.g. LPS)-saturated collagen with silk ligature group, and no ligature or P.g. LPS application group as the control. The plaque index and gingival index were measured at 0 and 4 weeks postoperatively. The animals were then euthanized and prepared for histologic evaluation. Results: The silk ligature group and P.g. LPS with silk ligature group showed a significantly higher plaque index at 4 weeks compared to the control (P<0.05). No significant difference was found in the plaque index between the silk ligature group and P.g. LPS with silk ligature group. The P.g. LPS with silk ligature group showed a significantly higher gingival index compared to the silk ligature group or the control at 4 weeks (P<0.05). Histologic examination presented increased inflammatory cell infiltration in the gingival tissue and alveolar bone of the P.g. LPS with silk ligature group. Conclusions: An additional P.g. LPS-saturated collagen with silk ligature ensured periodontal inflammation at 4 weeks. Therefore, P.g. LPS with silk ligature application to surgically created alveolar bone defects may be a candidate model for experimental periodontitis.

Effects of probiotics on the prevention of atopic dermatitis

  • Atopic dermatitis (AD) is an immune disorder that is becoming increasingly prevalent throughout the world. The exact etiology of AD remains unknown, and a cure for AD is not currently available. The hypothesis that appropriate early microbial stimulation contributes to the establishment of a balanced immune system in terms of T helper type Th1, Th2, and regulatory T cell (Treg) responses has led to the use of probiotics for the prevention and treatment of AD in light of various human clinical studies and animal experiments. Meta-analysis data suggests that probiotics can alleviate the symptoms of AD in infants. The effects of balancing Th1/Th2 immunity and enhancing Treg activity via the interaction of probiotics with dendritic cells have been described in vitro and in animal models, although such an effect has not been demonstrated in human studies. In this review, we present some highlights of the immunomodulatory effects of probiotics in humans and animal studies with regard to their effects on the prevention of AD.

MILK YIELD AND ITS REPEATABILITY IN JAPANESE BLACK COWS

  • Daily milk yield estimates were obtained on 74 lactations of 35 Japanese Black cows on weeks 1 to 9, 11, 13, 17, 21 and 26 postpartum using the weigh-suckle-weigh method. The data obtained were analyzed by least-squares procedures according to the models including the following effects; dam, lactation number, season of calving, week of experiment and partial regressions on the lactation number. The overall mean and standard error for daily milk yield was $4.55{\pm}0.04\;kg$. The milk yield declined essentially linearly throughout the experimental period. The main effects of lactation number, season of calving and week of experiment were highly significant. Differences in daily milk yield among cows were significant, and the repeatability was estimated as $0.60{\pm}0.06$. The overall means(kg) and repeat-abilities of cumulative milk yield for 1, 4, 9, 13, 17, 21 and 26 weeks postpartum were 41, 0.08; 166, 0.40; 361, 0.61; 503, 0.66; 632, 0.67; 749, 0.65 and 884, 0.58; respectively. The means of calf weight at birth and 26 weeks weight for different lactations ranged from 25.5 to 33.6 kg and 145.6 to 185.4 kg, respectively. Calf daily gains between experiment weeks were low in earlier stages of lactation.

ddY 마우스를 이용한 골다공증 모델에 대한 비교연구: 난소적출 및 신경절단 모델 (A comparative study on the osteoporotic animal models in ddY mouse: ovariectomized and neurectomized models)

  • In order to compare the induced time of osteoporosis between ovariectomized and neurectomized models in ddY mice. Experimental groups were divided into Sham, ovariectomized (OVX group) and neurectomized (NX group) group. The changes of body weight, tibia weight and histomorphometry of epiphyseal regions of tibia that were generally used as criteria index in osteoporosis, were evaluated at 2 and 4 weeks after operations with other generally used index-changes of serum osteocalcin. Also, calcium and phosphorus levels in the ash tibia were demonstrated with their ratio (Ca/P ratio). From the result of this study, evidences which reflect osteoporotic states of animals such as decrease of absolute and relative tibia weight, histomorphometrical index of epiphyseal region of tibia including trabecular bone volume %, and calcium and phosphorous contents in tibia, were generally detected from 4 weeks after ovariectomy and 2 weeks after neurectomy with increase of serum osteocalcin levels. In conclusion, it is considered that more rapid and favorable osteoporosis was induced in neurectomized model compared to that of ovariectomized model.

Modeling the Fate of Priority Pharmaceuticals in Korea in a Conventional Sewage Treatment Plant

  • Understanding the environmental fate of human and animal pharmaceuticals and their risk assessment are of great importance due to their growing environmental concerns. Although there are many potential pathways for them to reach the environment, effluents from sewage treatment plants (STPs) are recognized as major point sources. In this study, the removal efficiencies of the 43 selected priority pharmaceuticals in a conventional STP were evaluated using two simple models: an equilibrium partitioning model (EPM) and STPWIN$^{TM}$ program developed by US EPA. It was expected that many pharmaceuticals are not likely to be removed by conventional activated sludge processes because of their relatively low sorption potential to suspended sludge and low biodegradability. Only a few pharmaceuticals were predicted to be easily removed by sorption or biodegradation, and hence a conventional STP may not protect the environment from the release of unwanted pharmaceuticals. However, the prediction made in this study strongly relies on sorption coefficient to suspended sludge and biodegradation half-lives, which may vary significantly depending on models. Removal efficiencies predicted using the EPM were typically higher than those predicted by STPWIN for many hydrophilic pharmaceuticals due to the difference in prediction method for sorption coefficients. Comparison with experimental organic carbon-water partition coefficients ($K_{ocs}) revealed that log KOW-based estimation used in STPWIN is likely to underestimate sorption coefficients, thus resulting low removal efficiency by sorption. Predicted values by the EPM were consistent with limited experimental data although this model does not include biodegradation processes, implying that this simple model can be very useful with reliable Koc values. Because there are not many experimental data available for priority pharmaceuticals to evaluate the model performance, it should be important to obtain reliable experimental data including sorption coefficients and biodegradation rate constants for the prediction of the fate of the selected pharmaceuticals.

정신분열병의 실험적 모델 (Experimental Models of Schizophrenia)

  • 동물모형은 정신장애의 연구와 치료에 유용한 도구를 제공할 수 있다. 정신장애의 동물모형으로서의 적합성에 대한 평가 기준은 첫째, 유도된 상태의 유사성, 둘째, 행동상의 유사성, 셋째, 공통된 근저의 신경생물학적 기전, 넷째, 임상적으로 효과적인 치료 기술에 의한 역전 등 4가지이다. 저자는 정신분열병의 연구에 유용한 도구로 사용할 수 있는 실험적 모델을 간략히 소개하였다. 정신분열병의 실험적 모델로서는 L-dopa 모델, phenylethylamine 모델, hallucinogen 모델, amphetamine 모델, phencyclidine(이하 PCP) 모델, NE 보상계병소 모델, 망상계자극 모델, 사회격리 모델, 조건화된 회피반응, catalepsy 시험, paw 시험, 자기자극 paradigm, latent inhibition paradigm, blocking paradigm, 경악반사의 prepulse inhibition, 설치류 상호작용, 원숭이의 사회적 행위, 해마손상, 선택적 breeding을 사용한 모델, 고기압 모델, 각성 모델, 감각운동 gating 모델 등이 제시되고 있다. 저자는 이중에서 특히 정신분열병의 실험적 모델로서 face validity, predictive validity, construct validity가 높은 몇 가지 방법에 대해서 중점적으로 설명하였다. 임상경험 및 동물실험에서 PCP 및 ketamine 등 PCP 유사물질은 양성증상 뿐만아니라 음성증상, 파과형, 인지장애군에 좀 더 가까운 비망상형의 정신분열병에도 적용시키기가 좋은 것으로 알려졌다. 임신이나 출생 시 합병증에 의한 해마병소화, 내재적 신경독성에 의한 NE 보상계 병소, 스테로이드 증가에 의한 해마변화 등에 의해서 정신분열병이 유도될 수 있음은 해마병소화 모델이 정신분열병의 실험적 모델로서의 첫번째 기준을 충족시킴을 입증한다. 해마 병소화 후에 정신분열병 환자에서 흔히 볼 수 있는 주의집중력 장애, 공간 및 문맥 정보의 이해결핍, 기억구성, 인식기억, 고전적 조건화, 일반화, 복합적 학습, 상동적 행동, 미신적 행동, 과잉각성, 소멸과 습관형성, 피부전도 실험 등의 이상이 관찰됨은 정신분열병의 실험적 모델로서 두 번째의 기준에 합당한 것이다. 사후 부검한 뇌조직 및 여러 신경영상화 연구에서 해마의 구조적 이상 및 신경전달물질의 수용체 변화가 보고됨은 세 번째 조건을 만족시키는 것이며, 정신분열병의 여러 치료약물을 투여함으로써 상기 변화가 반전됨은 해마병소화 모델이 정신분열병의 실험적 모델로서 타당도가 높음을 시사한다. 정신분열병의 양성증상, 음성증상, 인지장애를 모두 포괄적으로 반영할 수 있는 실험적 모델로서 ketamine 모델과 해마병소화 모델 등이 보다 유용한 도구로 제시될 수 있다. 그러나 현재까지 어떠한 정신분열병의 모형도 정신분열병 전체를 대표할 수 있는 것은 없다. 따라서 동물모형을 사용한 연구의 결과는 매우 조심스럽게 해석되어야 한다. 또한 동물모형을 발전시키고 타당화 시키려는 노력은 인간에서의 현상을 신빙성있게 측정하는 방법을 규명하는 과정과 공동으로 이루어져야 한다.

엉겅퀴 뿌리 물 추출물의 류마티스 관절염 동물 모델에 대한 개선 효과 (Improvement Effect of the Water Extract from the Root of Cirsium japonicum var. ussuriense on Type II Collagen-induced Rheumatoid Arthritis Animal Models)

  • Background: The roots of Cirsium japonicum var. ussuriense (RCJ) have been used as traditional medicine in Korea for hematuria and hematemesis. These extracts exert anti-oxidative and anti-inflammatory effects by scavenging for free radical and regulating the inflammatory response. However, the effect of RCJ on rheumatoid arthritis (RA) has not been elucidated. Thus, we evaluated the water extract of RCJ (WRCJ) using type II collagen-induced RA models. Methods and Results: RA was induced by immunization with type II collagen. All experimental materials were orally administered daily for three weeks. The positive control group was administered with 0.2 mg/kg methotrexate (n = 7), while the experimental group was administered with WRCJ (100 or 500 mg/kg, n = 7). Serum levels of TNF-alpha, Interleukin 6 (IL-6), and type II collagen IgG (CII) were measured using ELISA. Administration of 500 mg/kg WRCJ decreased the levels of TNF-alpha, IL-6, and CII. Moreover, WRCJ treatment diminished swelling of hind legs and infiltration of inflammatory cells in RA models' synovial membrane. Conclusions: These results indicate that WRCJ could improve RA, reduce inflammatory indicators and synovial inflammation. However, further experiments are required to determine how WRCJ can influence the signal transduction pathway in RA.
