Chemical Modification of Brain Glutamate Dehydrogenase Isoproteins with Phenylglyoxal

  • Incubation of two types of glutamate dehydrogenase isoproteins from bovine brain with the arginine-specific dicarbonyl reagent phenylglyoxal resulted in a biphasic loss of enzyme activity. Reaction of the glutamate dehydrogenase isoproteins with phenylglyoxal caused a rapid loss of 53~62% of the enzyme activities and modification of two residues of arginine per enzyme subunit. Prolonged incubation of the glutamate dehydrogenase isoproteins with phenylglyoxal resulted in the modification of an additional four residues of arginine per enzyme subunit without further loss of the residual activities. Partial protection against inactivation was provided by the coenzyme NADH or substrate 2-oxoglutarate. The most marked decrease in the rate of inactivation was observed by the combined addition of NADH and 2-oxoglutarate, suggesting that the first two modified arginine residues are in the vicinity of the catalytic site. However, inactivation of the glutamate dehydrogenase isoproteins by phenylglyoxal appears to be partial with approximately 40% activity remained after an extended reaction time with excess reagent, suggesting that the modified arginine residues may not be directly involved in catalysis. The lack of complete protection by substrates also suggest the possibility that the modified arginine residues are not directly involved at the active site, and the partial loss of activity by the modification of arginine residues may be due to a conformational change. There were no significant differences between the two glutamate dehydrogenase isoproteins in sensitivities to inactivation by phenylglyoxal, indicating that the microenvironmental structures of the glutamate dehydrogenase isoproteins are very similar to each other.

  • This paper evaluates the performance of various distribution assumptions in regression settings for estimating insurance loss. The gamma distribution is commonly used to handle the asymmetry property of loss distribution. However, recent studies highlight the significance of heavy-tailedness in loss distribution. Through an analysis of real home fire insurance data, we compare the effectiveness of different distribution assumptions in regression methods. Our findings show that the choice of parametric distributional assumption is crucial in determining premiums for various insurance products, including "excess of loss insurance" and "limit insurance". Additionally, we discuss practical considerations for applying our results in home fire insurance.

  • A cylindrical constant volume combustion bomb is used to investigate the combustion characteristics and to analyze the heat quantity of homogeneous charge methane-air mixture under various initial pressures, excess air ratios and ignition times. As the overall pressure increase, the values of maximum combustion pressure, maximum heat release rate and cumulative heat release have been increased. But it is not very meaningful to compare with some values such as maximum combustion pressure, maximum heat release rate and cumulative heat release for different overall pressure due to the different heat energy of supplied fuel. So the each value is needed to be compared with normalized value, which is divided by the entered fuel energy. To analyze the heat quantity, some definitions including the CHR ratio, the UHC ratio and the HL ratio are needed and are calculated. As the overall pressure increase, the CHR ratios and the UHC ratios have been decreased, while the HL ratios have been increased. The CHR ratio of 300 ms has the higher value than that of 10000ms, and the HL ratios of 300 ms have a lower value.

  • A conventional flame type gas combustion major portion of heat is transferred to the body by convection due to small radiant ability of the gas flame. Increasing the radiation component of heat flux in the combustion zone allows to augment the efficiency of gas utilization. Such effect can be reached by using radiative gas burner applied to metal mesh combustion. Basically the gas radiant burner consists of metallic mesh of high heat resisting steels. In terms of this regards, we have made the burner consisted of metal mesh and measured the radiative flame stability of natural gas/air mixture on the metal mesh burner. The pressure loss through the metal mesh is defined by pressure-velocity slope. The more increased the pressure-velocity slope of the metal mesh is, the wider the stable zone of radiave flame on the metal mesh burner is. And the augmentation of mixture flowrate through the metal mesh make narrow the permissible range of equivalence ratio.

  • Bone is a dynamic tissue that is constantly being remodelled. Resolution of bone and formation of new bone are closely linked, so that bone mass remains constant. With age, this process becomes unlinked with an imbalance in bore resorption and formation that results in a net loss of bone. Especially, osteoporosis is a disease characterized by low bone mass with age. One form of aging-related primary osteoporosis is postulated with the reduction of circulating estrogen, rapid bone loss occurs as a result of enhanced bore remodelling with an excess of resorption over bore formation. The oxidative stress is also involved in the pathogenesis of osteoporosis. Oxidative stress by cytokines, such as IL-a and TNF-${\alpha}$, inhibits osteoblast function in vitro and stimulates osteoblast apoptosis resulting in an imbalance in bore remodelling. The present article reviews the current perspectives on the interaction between bone remodelling and factors such as estrogen and oxidative stress, providing an interpretation of bone diseases in a view of molecular mechanisms.

  • Waterjet propulsion is widely used to thrust high speed marine vessels in excess of 30-35 knots by virtue of the high propulsive efficiency, good maneuverability, and less cavitation. From the aspect of power loss, approximately $7-9\%$ of the total power is lost in intake duct due to the flow separation, nonuniformity, etc. Thus, detail understanding of flow phenomena occurring within intake duct is essential to reduce the power loss, as well as noise and vibration. The present work solved 30 incompressible RANS equations to provide complicated viscous flow features of intake duct. The numerical results were compared with experiments and good agreements were obtained for three jet velocity ratios.

Effects of CuO Addition on the Dielectric Properties of $Pb(Mg_{1/3}Nb_{2/3})O_3-PbTiO_3$ Ceramics (CuO의 첨가가 PMN-PT 세라믹스의 유전특성에 미치는 영향)

  • 95Pb(Mg1/3Nb2/3)O3-5PbTiO3 (hereinafter designated as 95PMN-5PT) system was prepared by the columbite-precursor method with 2 mol% excess PbO to compensate the PbO loss during thermal process. The amount of CuO was 1~10 mol%, and the effects of CuO addition on the dielectric properties of this system have been investigated. From the microstructures, XRD analysis and dielectric measurements, the solubility limit of CuO in 95PMN-5PT was found to be around 3 mol%. Lattice parameter and Curie temperature were found to be decreased as the amount of CuO increased up to the solubility limit. This result confirmed that the Cu2+-ions substituted the Pb2+-ions. It was revealed that the addition of CuO on 95PMN-5PT promoted the sinterability and properties. The room temperature dielectric constant, the loss factor and the specific resistivity of the specimens processed with optimum conditions were 23000, 1%, and 8$\times$1011Ω.cm, respectively.

  • In this paper, $0.10Pb(Sb_{1/2}Sn_{1/2})O_3-0.25PbTiO_3-0.65PbZrO_3$ ceramics were fabricated by the mixed-oxide method. The sintering temperature and time were $1230^{\circ}C$ and 2[hr], respectively. The structural, dielectric and piezoelectric properties with addition of NiO were studied. The crystal structure of a specimen was rhombohedral. As a result of SEM, the average grain size were decreased with increasing the contents of NiO. But the grains of the specimens doped with 0.4wt% NiO were increased, due to deposits of excess NiO at grain boundaries in the liquid phase. Relative dielectric constant and dielectric loss of the specimen doped with 0.1wt% NiO were 701 and 0.026, respectively.

  • This study revised a model for hydrologically analyzing rainwater harvesting facilities considering their rainfall-runoff properties and the data available. This model has only a few parameters, which can be estimated with rather poor measurements available. The model has a non-linear module for rainfall loss, and the remaining rainfall excess (effective rainfall) is assumed to be inflow to the storage tank. This model has been applied for the rainwater harvesting facilities in Seoul National University, Korea Institute of Construction Technology, and the Daejon World Cup Stadium. As a result, the runoff coefficients estimated were about 0.9 for the building roof as a rainwater collecting surface and about 0.18 for the playground. This result is coincident with that for designing the rainwater harvesting facilities to show the accuracy of model and the simulation results.

  • Products are assumed to be spoiled and not recoverable when the raw material below the prescribed weight level is charged in the mold. The raw material is autoscaled and their weight is normally distributed with a given variance. This paper describes an appropriate stochastic model of determining the optimum weight allowance and the associated defective rate for auto scale system. The optimum weight allowance and the defective rate which minimizes the total loss of defective and the excess weight allowance is obtained. A numerical example for the existing rubber company is also presented and evaluated.
