감마선을 이용한 고기능성 히알루론산/연어추출물 제형개발 (Development of High-Functional Hyaluronic Acid/Salmon Extract Formulation Using Gamma-Ray)

  • 음주 후 숙취는 사람에 따라 증상과 정도가 상이 하지만 일반적으로 갈증, 피로감, 두통, 전신권태, 위장장애, 구토, 설사, 비타민 결핍 등이 나타난다. 이러한 숙취현상은 간세포와 체내에 축적된 알코올 및 알코올에 함유되어 있는 발효 부산물인 에틸아세테이트, 아세트알데히드 등의 전구물질들의 작용에 의해 발생한다. 이에 숙취현상을 해소하기 위한 연구로 연어 추출물 중 정액 또는 정소 추출물인 핵산-당-인산으로 이루어진 고분자이며 다당체인 Polydeoxyribonucleotide(PDRN)과 동일한 Origin, 동일 구조를 가지며 PDRN에 비해 가격이 저렴한 수용성 연어 추출물 분말을 사용하였다. 그리고 D-Glucuronic acid와 N-Acetyl glucosamine으로 구성된 고분자 다당체로 뛰어난 생체 적합성, 점탄성 및 보습력을 보유하고 있어서 숙취현상 중 나타나는 생체수분 감소량 및 피부수분 함유량저하에 대한 부분에 효과가 있어 항산화 및 피부보습효과를 제공하는 히알루론산(Hyaluronic acid)을 감마선으로 조사하여 사용하였다. 연어추출물 분말과 히알루론산 등을 이용하여 조성물을 제조한 후 체내 에탄올, 아세트알데히드 감소량, 혈중 아세트알데히드 농도와 초산 농도를 측정하여 알코올 분해효과를 평가하였고, 피부 수분 증발량과 피부 수분량을 조사하여 피부개선을 평가하고 항산화 및 피부보습효과를 제공하는지 여부를 판단하였다. 이에 에탄올은 연어추출물을 첨가하였을 때가 무첨가에 비해 5배에서 7배의 감소량을 보였고 아세트알데히드 3배에서 5배 이상의 감소량을 보였다. 또한 혈중 아세트알데히드 농도와 초산 농도의 변화에서는 무첨가 대조군에 비해 급격한 저하 변화를 보여 알코올 분해효과가 있음을 확인하여 숙취해소 효과가 있음을 확인할 수 있었다. 그리고 히알루론산 원료를 첨가하여 사용하였을 때 피부 수분함유량이 높았으며, 피부 수분 증발량이 감소됨이 확인되었다. 이에 고분자 다당체인 히알루론산은 뛰어난 점탄성 및 보습력을 보유하고 있어서 숙취현상 중 나타나는 생체수분 감소량 및 피부수분 함유량저하에 효과가 있어 항산화 및 피부보습효과를 제공하는 것으로 사료되어 연어추출물 분말과 히알루론산을 주원료로 하는 조성물이 음주 후 나타나는 숙취현상에 효과가 있다고 할 수 있다.

염료감응태양전지에서 Pd 촉매층의 전해질과의 반응에 따른 특성 저하 (Degradation of the Pd catalytic layer electrolyte in dye sensitized solar cells)

  • 기존 DSSC의 상대전극을 TCO-less로 하여 도전성과 촉매기능을 동시에 가지고 있는 Pd의 안정성 확인을 위해 열증착기를 채용하여 유리기판 전면에 Pd를 90nm 두께로 증착하고 전해질과의 반응 안정성을 확인하였다. $0.45cm^2$급 면적을 가진 glass/FTO/blocking layer/$TiO_2$/dye/electrolyte(10 mM LiI + 1 mM $I_2$ + 0.1 M $LiClO_4$ in acetonitrile solution)/Pd/glass 구조의 DSSC 소자를 만들고, 시편제작 1시간, 12시간 후의 변화를 육안분석, 광학현미경과 FESEM을 이용하여 미세구조 분석을 진행하고, 전기적 분석은 각각 C-V(cyclic voltammetry measurements), I-V(current voltage) 분석을 통해 확인하였다. 미세구조 분석을 통하여 시간이 지남에 따라 확연히 Pd과 전해질이 반응하여 부식되는 것을 확인하였고, 전기적으로도 시간이 지남에 따라 촉매활동도와 효율이 감소하는 것을 확인하였다. 최종 효율은 1시간 후에는 0.34%의 광전효율을 보였으나 12시간 후에는 0.15%를 나타내어 약 44%로 감소하였다. 따라서 염료감응태양전지에 Pd촉매를 채용하기 위해 $I^-/I_3{^-}$ 전해질이 아닌 다른 전해질을 사용하거나 Pd 전극이 아닌 다른 촉매재를 사용해야 함을 확인하였다.

혐기성 소화-고온 호기법에 의한 유기성폐기물의 처리와 생성열의 재활용 검토 (Treatment of Organic Wastes and Reuse of Bio-energy from the Anaerobic Digestion - Thermophilic Oxic Precess)

  • 본 연구에서는 유기성 폐기물의 분해시 발생되는 열을 재활용하고, 혐기성 소화액의 퇴비화를 위한 혐기성 소화 - 고온 호기법(Anaerobic Digestion-Thermophilic oxic process, ADTOP)을 고안하고, 유기물 분해와 수분의 증발 그리고 생성열의 적용성을 검토하였다. 유기성 폐기물인 중화요리 잔반은 TOP에 의해 완전처리가 가능하며, 최대 용적부하는 $55kg/m^3{\cdot}d$, 투입된 수분은 거의 완전히 증발되었으며 탄소수지에 의한 탄소성 유기물의 이산화탄소 전환율은 90.5%이었다. 고온 혐기성 소화를 위한 적정온도(약$55^{\circ}C$)를 유지하기 위한 최소용적부하는 $45.0kg/m^3{\cdot}d$이었다. 혐기성 소화조의 온도는 수리학적 체류시간이 짧아짐에 따라 지수적 온도강하를 나타내었으며 고온 혐기성소화를 위한 최소 HRT는 약 10일 정도로 판단된다. 따라서 고온 호기법을 이용한 유기성폐기물의 처리시 발생되는 열에너지는 혐기성 소화와 같은 체류시간이 비교적 긴 공정에서 효과적일 것으로 판단된다. 혐기조의 유기물 부하 $1.1kg-COD/m^3{\cdot}d$, 고온 호기조 유기물의 투입량 $50kg/m^3$, 공기 유입량 $250{\ell}/m^3{\cdot}min$의 조건에서 혐기성 소화효율은 90% 이상으로 나타났다.

  • PLGA미립구는 주사제형과 같은 형태로 조직공학적 응용에 이용할 수 있다. 본 연구에서는 미립구 제조시 에밀젼 형성조건에 대한 영향과 미립구 표면에 세포를 부착시키는 방법에 대하여 연구하였다. BSA를 함유하는 PLGA미립구는 수중유형(O/W)과 수중유중수형(W/O/W) 용매증발법을 이용하여 제조하였다. 미립구의 초기방출효과제어와 PLGA분해의 과정에서 발생되는 지연시간을 개선시키기 위하며 알긴산나트륨을 수상에 용해시켜 사용하였다. 미립구에 부착된 세포의 형태를 전자주사현미경(SEM)을 이용하여 분석하였고 PLGA미립구에 배양된 인간디스크세포의 증식은 MTT분석을 이용하였으며 이를 통하여 PLGA미립구 표면에 세포가 부착되었음을 확인하였다. 본 연구는 BSA가 함유된 알긴산/PLGA미립구를 이용하여 조직공학적 응용이 가능한 주사제형으로서의 가능성을 제안하였다.

표고 재배 중 톱밥 배지의 물리적 성질과 목재 화학성분 변화 (Changes in Physical Properties and Wood Chemical Components of sawdust medium during Oak Mushroom (Lentinula edodes) Cultivation)

  • 참나무 톱밥을 이용한 표고버섯 톱밥봉지 재배는 원목재배에 비하여 많은 이점을 가지고 있어 국내 톱밥배지 재배 농가는 매년 증가하고 있는 추세이다. 그동안 표고 톱밥재배에 대한 많은 연구가 수행되어 보고된 바 있으나, 주요 연구 내용은 톱밥배지 제조 시 혼합하는 농산부산물 영양원 첨가제의 종류와 첨가효과, 톱밥 배지 재배에 적합한 신품종 개발, 참나무 발효 톱밥과 생 톱밥 간의 재배 특성 비교 등이 있으며, 배지 구성 물질 중에서 표고의 주 영양원인 참나무 톱밥의 재배 기간 중 목재 화학적 성분 변화에 대한 보고는 이루어지지 않았다. 이에 본 연구에서는 균 배양 및 버섯 수확 기간 중 배지의 물리 및 톱밥의 목재 화학 성분 변화 등에 대한 조사를 수행하였다. 그 결과, 종균 접종 후 배양 기간 중에 배지의 중량 감소가 발생하며, 이것은 배지 함유 수분의 증발이나 톱밥 목질부의 분해에 의한 현상이 아니고, 배지 중에 첨가한 저분자 수용성 물질인 쌀겨가 먼저 균사에 의해 분해, 이용되기 때문이라는 결론에 이르렀다. 또한 배양 중에 균사 생장 배지의 함수율이 꾸준히 증가하는 것을 확인하였으며, 균사 생장과 함께 유기물 분해 과정인 이화작용이 진행되어 분해 산물인 물이 생성됨으로써 배지의 함수율이 증가하는 것으로 판단하였다. 표고버섯 수확이 반복하여 진행됨에 따라 톱밥의 목재 세포벽 주성분인 홀로셀룰로오스와 리그닌 등 유기물 비율이 꾸준하게 감소하였으며, 특히 리그닌 감소 비율이 크게 나타나 백색부후균인 표고 균의 목재 세포벽 성분 분해 특징이 확인되었다. 이에 반하여 무기물 성분으로 이루어진 회분은 수확 횟수 증가와 함께 증가하는 경향을 보였는데, 이것은 홀로셀룰로오스와 리그닌의 유기물 비율이 감소함으로써 상대적으로 증가하는 결과로 나타난 것이다.

Cu 기판위에 성장한 MgO, $MgAl_2O_4$$MgAl_2O_4/MgO$ 박막의 집속이온빔을 이용한 스퍼터링수율 측정과 이차전자방출계수 측정 (Sputtering Yield and Secondary Electron Emission Coefficient(${\gamma}$) of the MgO, $MgAl_2O_4$ and $MgAl_2O_4/MgO$ Thin Film Grown on the Cu Substrate by Using the Focused Ion Beam)

  • [ $MgAl_2O_4$ ] 막은 MgO 보호막 보다 단단하며 수분 흡착 오염 문제에 상당히 강한 특성을 가진다. 본 연구에서 AC-PDP 의 유전체보호막으로 사용되는 MgO 보호막의 특성을 개선하기 위해 $MgAl_2O_4/MgO$ 이중층 보호막을 제작하여 특성을 조사하였다. 전자빔 증착기를 사용하여 Cu 기판에 MgO 와 $MgAl_2O_4$을 각각 $1000\AA$ 두께로 증착, $MgAl_2O_4/MgO$$200/800\AA$ 두께로 적층 증착 후, 이온빔에 의한 충전현상을 제거하기 위해 Al 을 $1000\AA$ 두께로 증착하였다. 집속 이온빔 (focused ion beam: FIB) 장치를 이용하여 10 kV 에서 14 kV 까지 이온빔 에너지에 따라 MgO는 $0.364{\sim}0.449$ 값의 스퍼터링 수율에서 $MgAl_2O_4/MgO$ 을 적층함으로 $24{\sim}30 %$ 낮아진 $0.244{\sim}0.357$ 값의 스퍼터링 수율이 측정되었으며, $MgAl_2O_4$는 가장 낮은 $0.088{\sim}0.109$ 값의 스퍼터링 수율이 측정되었다. g-집속이온빔 (g-FIB) 장치를 이용하여 $Ne^+$ 이온 에너지를 50 V 에서 200 V 까지 변화시켜 $MgAl_2O_4/MgO$ 와 MgO 는 $0.09{\sim}0.12$의 비슷한 이차 전자방출 계수를 측정하였다. AC- PDP 셀의 72 시간 열화실험 후 SEM 및 AFM으로 열화된 보호막의 표면을 관찰하여 기존의 단일 MgO 보호막과 $MgAl_2O_4/MgO$의 적층보호막의 열화특성을 살펴보았다.

환경조건이 표고톱밥배지의 갈변에 미치는 영향 (The effect of environmental condition to the mycelial browning of Lentinula edodes (Berkeley) Sing. during sawdust bag cultivation)

  • 표고 톱밥배지의 갈변을 촉진시키기 위하여 광을 조사한 결과 100Lux 이상의 광처리에서 원하는 수량을 얻을 수 있었으며 암상태에서의 자실체의 발생은 기형버섯율이 높았으며 수량도 저조하였다. 그러나 갈변은 200 Lux이상의 광에서 정상적으로 이루어졌으며 가장 빨리 갈변이 되기 시작하였다. 온도별 처리에서는 $25^{\circ}C$의 배양온도를 유지하여 배양한 것이 갈변시작일도 가장 빨랐고 갈변도 가장 많이 진행되었으며 수량과 개체중량도 가장 높았으며 정상적인 수량을 나타낸 처리 중 기형버섯의 발생량도 가장 적었다. 배지내에 통기성이 미치는 영향을 조사하기 위하여 솜마개의 크기를 달리하여 처리한 결과 마개의 크기가 클수록 균사의 생장량도 빨랐으며 갈변이 이루어지는 시기도 빨랐다. 그러나 자실체의 수량은 솜마개의 직경이 16mm일 때 가장 높았으며 마개가 클수록 배지내의 $CO_2$함량은 낮았으며 배양기간 중 갈변이 진행되는 8주에서 14주 사이에서 $CO_2$함량이 가장 높았다. 배양기간 중 $C_2H_4$함량은 8mm의 솜마개에서 가장 많이 발생하였으며 12, 16, 20, 0, 4mm의 솜마개으로 발생하였다. $C_2H_4$함량도 $CO_2$함량과 같이 8주에서 14주사이의 배양기간에서 가장 높았다. 침수시간에 따른 자실체의 발생효과를 구명하기 위하여 침수시간별로 자실체의 수량과 효소들의 활성을 조사하였다. 침수를 하지 않은 배지는 수량이 침수를 한 배지에 비하여 약 40%가 감소하였다. 침수시간은 4시간과 15시간 침수한 것이 각각 165g/1000ml, 175g/1000ml 이었다. 침수시 cellulose분해효소는 침수에 따른 변화가 없으나 lignin 분해효소인 laccase는 침수시간에 따라 약 4 배정도까지 효소의 활력이 증가되었다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction. Electrical stimulation for each muscle was designed to generate bipedal locomotion, so that individual muscles alternate contraction and relaxation to simulate stance and swing phases.
