한국 미기록 귀화식물: 아메리카대극과 털땅빈대 (Two Newly Naturalized Plants in Korea: Euphorbia heterophylla L. and E. hirta L.)

  • 부산광역시와 제주도에서 대극과의 미기록 귀화식물 2종을 발견하였다. Euphorbia heterophylla L. (아메리카대극)은 부산광역시 동래구 안락동 수영강변 공원에서 확인되었으며, E. dentata Michx. (톱니대극)과 비교하여 원줄기 잎은 호생하며, 소총포의 선체 입구는 원형인 특징으로 구별된다. 또한 Euphorbia hirta L. (털땅빈대)은 제주특별자치도 서귀포시 예래동 예래마을에서 확인되었으며, E. hypericifolia L. (큰땅빈대)와 비교하여 삭과에 털이 있는 특징으로 구별된다.

비단풀 추출물의 식중독세균에 대한 항균활성 (Antibacterial Activity of Euphorbia humifusa Extracts on Food-Borne Pathogenic Bacteria)

  • This study was performed to investigate the antimicrobial effects of Euphorbia humifusa ethanol-extract against food-borne pathogens. The growth inhibitory effects of the extract at a concentration of 250, 500, 1,000 or 2,000 mg/L on food poisoning microorganism were determined against Salmonella typhimurium, Listeria monocytogenes, Yersinia enterocolitica, Escherichia coli O157:H7 and Staphylococcus aureus. The microorganisms growth was not affected by the extract at the concentration up to 250 mg/L, but was significantly (p<0.05) inhibited by the extract at a concentration higher than 1,000 mg/L. The extract of Euphorbia humifusa had strong antimicrobial activity against all test strains at a concentration of 2,000 mg/L. The results in the present study demonstrate antimicrobial effects of Euphorbia humifusa ethanol-extract against food-borne pathogens, suggesting that Euphorbia humifusa could be an effective natural antibacterial agent in food.

한방약침소재로써 속수자 초임계추출물의 항산화 및 항노화에 대한 연구 (Anti-Oxidant and Anti-Aging Effect of Supercritical Fluid Extraction of Seed of Euphorbia lathyris L. as a Pharmacopuncture Material)

  • Objectives : The purpose of this study was to investigate the anti-oxidant and anti-aging effect of the seed of Euphorbia lathyris L. extracted by supercritical CO2. Methods : Human dermal fibroblast cells dosed with the extract from Euphorbia lathyris L. were harvested and the intracellular proteome was analyzed to examine the expression of proteins related collagen synthesis pathway, metalloproteinases (MMPs), extracellular matrix (ECM)-cell interaction, cytokines, and antioxidant enzymes by 2-dimensional gel electrophoresis. Results : Fatty acid analysis of the extract from Euphorbia lathyris L. showed oleic acid was 84% and linoleic acid was 4.1%. Antioxidative effect was about 53% by beta carotene bleaching assay. In 2-dimensional polyacrylamide gel electrophoresis (2-D PAGE) analysis, fifteen protein changes in five mechanisms which were collagen synthesis pathway, MMPs, ECM-cell interaction, cytokines, and antioxidant enzymes were analyzed. Conclusions : This study suggests the supercritical extraction from the seed of Euphorbia lathyris L. could be used as anti-oxidant substances for pharmacopuncture.

땅빈대 추출물의 세포 보호 효과 및 성분 분석에 관한 연구 (Cellular Protective Effect and Component Analysis of Euphorbia humifusa Extracts)

  • In this study, the cellular protective effect, antioxidative property and component analysis of Euphorbia humifusa extracts were investigated. The ethyl acetate fraction ($3.68\;{\mu}g/mL$) and aglycone fraction ($3.15\;{\mu}g/mL$) of Euphorbia humifusa extract showed prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of Euphorbia humifusa extract on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction ($0.43\;{\mu}g/mL$) and aglycone fraction ($0.35\;{\mu}g/mL$) of extract showed higher ROS scavenging activity than L-ascorbic acid ($1.50\;{\mu}g/mL$). The cellular protective effects of fractions of Euphorbia humifusa extract on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction and aglycone fraction of extract protected cellular membranes against ROS in a concentration dependent manner ($5{\sim}25\;{\mu}g/mL$), and was more effective than (+)-${\alpha}$-tocopherol, lipid peroxidation chain blocker. Aglycone fraction from Euphorbia humifusa extract showed 2 bands in TLC and 2 peaks in HPLC. In HPLC chromatogram of aglycone fraction, peak 1 and peak 2 were identified as quercetin and kaempferol, respectively. And these components are very effective as antioxidant. Thus, these results indicate that fractions of Euphorbia humifusa extracts can function as antioxidant in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Fractions of Euphorbia humifusa extracts can be applicable to new functional cosmetics for antioxidant.

한국 미기록 귀화식물: 톱니대극(Euphorbia dentata Michx.)과 왕관 갈퀴나물(Securigera varia (L.) Lassen) (Two new naturalized species from Korea, Euphorbia dentata Michx. and Securigera varia (L.) Lassen)

  • 경북 영천시와 서울 여의도에서 미기록 귀화식물 2종을 발견하였다. 톱니대극으로 이름 붙인 Euphorbia dentata Michx.(대극과)는 경북 영천시 대창면 대창리 중부고속도로에서 확인되었으며, 동속식물인 포인세티아 (E. pulcherrima)와 비교하여 초본이면서 정상엽들이 한결같이 녹색인 특징으로 구별된다. 또한 왕관갈퀴나물로 이름 붙인 Securigera varia (L.) Lassen(콩과)는 서울 여의도 한강둔치에서 확인되었으며, Securigera속은 국내 미기록속이다. 형태적 유사종으로 묏황기속(Hedysarum L.) 식물에 비해 꽃이 담홍색이고 길이 5-10 cm의 꽃자루 끝에 20개 내외의 꽃이 산형으로 붙는 특징으로 구별된다.

  • Eight compounds were isolated from the whole plants of Euphorbia supina (Euphorbiaceae) through repeated silica gel, YMC gel and Sephadex LH-20 column chromatography. Their chemical structures were elucidated as 7-hydroxy-6-methoxycoumarin (scopoletin) (1), p-hydroxybenzaldehyde (2), methyl gallate (3), gallic acid (4), quercetin (5), quercetin $3-O-{\alpha}-L-arabinofuranoside$ (avicularin) (6), kaempferol $3-O-{\alpha}-L-arabinofuranoside$ (juglanin) (7) and kaempferol $3-O-{\beta}-D-glucopyranoside$ (astragaline) (8) by spectroscopic (NMR and MS) analysis.

포인세티아 줄기조직배양에 의한 재분화체 제조. (Somatic Embryogenesis and Plant Regeneration from Poinsettia (Euphorbia pulcherrima L.) Stem Explants)

  • 포인세티아식물체의 줄기조직을 이용한 재분화조건을 확립하였다. MS배지에 종류 및 농도별 식물성장 조절제를 첨가하여 포인세티아의 잎, 줄기, 엽병조직으로부터의 배구조의 발생을 조사하였다. 잎과 엽병조직에서는 callus의 형성은 실험조건에서 매우 활발하였으나 배구조로의 발달은 전혀 이루어지지 않았다. 줄기조직의 경우에서는 1.5 mg/L의 BA가 첨가되는 경우 6-8주 정도의 경과 후 엽초의 발생이 관찰되었다. 이들을 식물생장조절제를 무첨가한 MS고체배지로 이동시 뿌리의 발달이 관찰되었다.

땅빈대 추출물의 항산화 활성 및 세포독성 효과 (Antioxidant Activity and Cytotoxic Effect of Extracts from Euphorbia humifusa)

  • The antioxidant activities, anti-inflammatory activity and cytotoxic effects of methanol extract from Euphorbia humifusa were evaluated in this study. Total phenolic compound contents were $68.35{\pm}0.16$ mg/g and total flavonoid compound contents were estimated as $38.74{\pm}1.26$ mg/g. EC50 values for DPPH radical scavenging activity of methanol extract was $56.26{\pm}0.66{\mu}g/mL$ and those of positive controls as ascorbic acid, ${\alpha}$-tocopherol and BHA were $8.38{\pm}0.14{\mu}g/mL$, $16.45{\pm}0.89{\mu}g/mL$ and $21.18{\pm}1.01{\mu}g/mL$ respectively. NO scavenging activity increased in depending on concentration of extract. Treatment of RAW 264.7 cells with extract caused inhibition of LPS-induced nitric oxide production. The cell viability showed that the methanol extract had cytotoxicity in the growth of breast cancer cell ($66.54{\pm}1.91%$ at $400{\mu}g/mL$ conc., $43.98{\pm}3.35%$ at $800{\mu}g/mL$ conc.). Based on the results, It was suggested that the methanol extract of Euphorbia humifusa has a potential candidate for functional cosmetic and medicine.

In vitro Adipocyte Differentiation Inhibition and in vivo Effects on Lipid Metabolism in High-Fat Diet-Induced Obesity of Euphorbia humifusa

  • Euphorbia humifusa Willd (Euphorbiaceae) is a functional raw material with various pharmacological activities. This study aimed to validate the inhibitory effect of Euphorbia humifusa extract (EHE) on adipocyte differentiation in vitro and in a high-fat-diet (HFD)-induced mouse model to evaluate the E.a humifusa as a novel anti-obesity and lipid metabolism enhancer agent. EHE effects on obesity and lipid metabolism were assessed in HFD-induced obese mice after 4-week treatments. Results were compared among four treatment groups (n = 7/group): low fat diet (LFD), high fat diet (HFD), and HFD-induced obese mice treated with either 100 or 200 mg/kg/day EHE (EHE100 and EHE200, respectively). EHE (50 to 200 ㎍/ml) and quercetin (50 ㎍/ml) significantly reduced 3T3-L1 preadipocyte differentiation (p < 0.001), in a concentration-dependent manner. EHE affected lipid metabolism, as evidenced by changes in serum lipid components. The HFD-EHE100 and HFD-EHE200 groups exhibited significantly (p < 0.05) reduced triglycerides (TG, 97.50 ± 6.56 and 82.50 ± 13.20 mg/dL, respectively) and low-density lipoprotein-cholesterol (LDL-c: 40.25 ± 4.99 and 41.25 ± 6.36 mg/dL, respectively) compared to the HFD group (TG: 129.25 ± 19.81 mg/dL; LDL-c: 51.75 ± 11.59 mg/dL). Haematoxylin and Eosin (H&E) and Oil red O staining showed that EHE markedly reduced lipid accumulation and inhibited lipogenesis in the liver. Interestingly, EHE significantly (p < 0.01) reduced the expression of adipogenic transcription factors in liver tissue. Our results indicated that EHE has the potential to be a therapeutic agent for addressing obesity and lipid metabolism.

Antimicrobial and Anti-Inflammatory Potential of Euphorbia paralias (L.): a bioprospecting study with phytoconstituents analysis

  • Objectives: The phytochemicals in the aerial parts of Euphorbia paralias (also known as Sea Spurge) and their anti-inflammatory and antimicrobial activities were investigated. Methods: The methanolic extract was characterized using GC-MS and HPLC techniques. The anti-inflammatory feature was estimated through a Human Red Blood Cell (HRBC) membrane stabilization technique, while the antimicrobial feature was evaluated by the disc diffusion agar technique, minimum bactericidal concentration, and minimum inhibitory concentration (MIC) via micro-broth dilution method. Results: The GC/MS results demonstrated the existence of various phytochemicals, such as n-hexadecenoic acid, cis-11-eicosenoic acid, and methyl stearate, recognized for their anti-inflammatory and antibacterial features. The similarity of the phytochemical composition with other Euphorbia species emphasizes the genus-wide similarity. The anti-inflammatory activity exhibited a noteworthy inhibitory effect comparable to the reference drug indomethacin. The extract's antimicrobial potential was tested against a range of microorganisms, demonstrating significant action against Gram-positive bacteria and Candida albicans. The quantification of total phenolics and flavonoids further supported the therapeutic potential of the extract. Conclusion: The methanolic extract from E. paralias emerges as a successful natural source of important active constituents with potential applications as anti-inflammatory and antimicrobial agents. This research provides a first step to valorize Euphorbia paralias insights as a source of worthwhile phytochemicals that have potential applications in the pharmaceutical industry.
