Deastringent Peel Extracts of Persimmon (Diospyros kaki Thunb. cv. Cheongdo-Bansi) Protect Neuronal PC-12 and SH-SY5Y Cells against Oxidative Stress

  • The peel of astringent persimmon (Diospyros kaki Thunb. cv. Cheongdo-Bansi) is a by-product of dried persimmon (gotgam). We investigated if deastringent peel extracts of persimmon cv. Cheongdo-Bansi had antioxidative and neuroprotective properties. Two different extracts were prepared: thermally and nonthermally treated persimmon peel extracts (TPE and NTPE, respectively). Both TPE and NTPE were fractionated sequentially in n-hexane, chloroform, ethyl acetate, n-butanol, and water. The TPE and NTPE ethyl acetate fractions had the highest total phenolic and flavonoid contents as well as antioxidant capacities among all the fractions. Pretreatment of neuronal PC-12 and SH-SY5Y cells with the TPE and NTPE ethyl acetate fractions increased cell viability after exposure to oxidative stress. The ethyl acetate fraction of TPE attenuated oxidative stress inside both PC-12 and SH-SY5Y cells more effectively than that of NTPE. Furthermore, the TPE and NTPE ethyl acetate fractions inhibited acetylcholinesterase and butyrylcholinesterase. Analysis of ultra-high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry results revealed gallic acid, kaempferol, kaempferol-3-O-galactoside, kaempferol-3-O-glucoside, quercetin, quercetin3-O-galactoside, quercetin-3-O-galactoside-2'-O-gallate, and quercetin-3-O-glucoside as the major phenolics of the TPE and NTPE ethyl acetate fractions. Taken together, these results suggest that the ethyl acetate fraction of deastringent persimmon peel is rich in antioxidants and has potential as a functional food to reduce oxidative stress.

Antioxidative Activity of Ulmi cortex Extract (유백피(Ulmi cortex) 추출물의 항산화 활성)

  • The Ulmi corex extract was prepared using various solvents to investigate the availability as a natural antioxidant. The extracts were added to lard emulsion and the antioxidant activities were compared. The extract that had a greater antioxidant activity was fractionized. Then the antioxidant activity and substrate specificity of the fraction were examined and optimum concentration of addition was determined. To observe the antioxidative effect of the fraction in vivo, an inhibition rate of lipid peroxidation from which might be derived was measured using a microsome in rat's liver. Among the extracts of Ulmi cortex, the extract from water had the best antioxidant activity, and the addition of 0.05% (w/w) of ethyl acetate fraction showed similar antioxidant activity to a synthetic antioxidant, butylated hydroxyanisole(BHA). Ethyl acetate fraction (0.05%, w/w) also presented the antioxidative effect in lard, soybean oil, palm oil, and com oil. The inhibition of lipid peroxidation in liver microsome showed feater in the ethyl acetate fraction than caffeic acid in both nonenzymatic peroxidation (Fe$\^$++/ascorbate system) and enzymatic peroxidation (Fe$\^$++/-ADP/NaDPH system).

Antioxidant Activity of Ethyl acetate Fraction of the Guzeunggupo-procossed Platycodon grandiflorum A. De Candolle roots in Caenorhabditis elegans (구증구포 맥문동 Ethyl acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과)

  • Using the Caenorhabditis elegans model system, the antioxidant activity of methanol extract of the guzeunggupoprocessed Liriope platyphylla F. T. Wang (Liliaceae) tuber was calculated. Between the methanol extracts of guzeunggupo-processed and non-processed L. platyphylla tuber, the processed L. platyphylla tuber showed higher DPPH radical scavenging effect than the non-processed one. The ethyl acetate soluble fraction of the methanol extract of the guzeunggupo-processed L. platyphylla tuber showed the best DPPH radical scavenging activity. The ethyl acetate fraction of the processed sample was measured for the activities of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species level. In addition, to verify the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction of the processed sample, SOD-3 expression was measured using a transgenic strain (CF1553). Consequently, the ethyl acetate fraction of the processed sample, increased SOD and catalase activities, and decreased ROS accumulation in a dose-dependent manner. Furthermore, the ethyl acetate fraction of the processed sample-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

Inhibitory Effect of Mugwort Extracts on Tyrosinase Activity (쑥 추출물의 Tyrosinase 효소활성에 미치는 억제효과)

  • To determine the inhibition of mushroom tyrosinase activity, fresh and dried mugwort, Artemisia princeps was extracted initially with water and ethyl alcohol, and subsequently with hexane, chloroform and ethyl acetate in that order. The highest yield was obtained from the ethyl acetate (15.2%) and hexane (15.5%) fraction of the ethanolic extract of fresh and dried mugwort, respectively. For all fractions tested, the inhibition of tyrosinase activity by fresh mugwort was higher than that of dried mugwort, and the inhibition ratio of tyrosinase activity was 98.9% in the chloroform fraction and 96.7% in the hexane fraction.

  • The rates of mass transfer with the alkaline hydrolysis of ethyl acetate and n-butyl acetate were measured by using a modified Lewis cell. The rates of mass transfer with chemical reaction were independent of the speed of agitation, and the reaction enhancement factors were independent of the ionic strength. The second order reaction rate constants of ethyl acetate and n-butyl acetate could be obtained from an approximate solution of a diffusion equation by film theory, and their values were $0.041m^3/kg\;mol{\cdot}s$ and $0.338m^3/kgmol{\cdot}s$, respectively.

Antioxidant Activity of Ethanol Extract from Rhus javanica $Linn\'{e}$ on Edible Oil (식용유지에 대한 붉나무 추출물의 항산화 효과)

  • For natural antioxidant test, the Rhus javanica $Linn\'{e}$ was extracted by 75% ethyl alcohol and fractionated it by chloroform and ethyl acetate using separatory funnel and column chromatography. In the Rhus javanica L. extract, the effective material was disolved easily in ethyl acetate and chloroform. When 600 ppm of ethyl acetate fraction of the extract was added to palm oil and lard, AI(antioxidant index) of each oil was 1.60 and 3.90 respectively. The $CHCl_3$ and EtOAc fractions were more effective than whole extract of Rhus javanica L. The palm oil, lard and soybean oil containing different levels of the Rhus javanica L. extract were stored at $60^{\circ}C$ to compare their antioxidative activity. Peroxide value, acid value and TBA value of each oil were monitored. The Rhus javanica L. extract was very effective to retard oxidation of palm oil and lard. This result coincided with Ranimat test.

Convergence study on the antioxidant effect of crude extracts of Nelumbo nucifera Gaertner (연밥 조추출물의 항산화 효능에 관한 융합 연구)

  • The present study was performed to evaluate the antioxidant effect of crude extracts of Nelumbo nucifera Gaertner. Antioxidant effect was analysed by DPPH-radical scavenging activity, lipid peroxidation and superoxide dismutase (SOD)-like activity. DPPH-radical scavenging activity and SOD-like activity of linoleic acid, glutamic acid, ethyl acetate crude extract and ethyl alcohol crude extract of Nelumbo nucifera Gaertner were dose-dependently increased. However, lipid peroxidation of glutamic acid, linoleic acid, ethyl acetate crude extract and ethyl alcohol crude extract of Nelumbo nucifera Gaertner were time-dependently decreased. The data suggests that crude extracts of Nelumbo nucifera Gaertner may be a putative antioxidant substance and apply the development of medicine through convergence study.

Treatment of gas from the vent of a fine chemical plant using a pilot-scale biofilter (파일럿 규모 바이오필터를 이용한 정밀화학공장 배출가스의 처리)

  • A pilot-plant biofilter (1750 mm W ${\times}$ 2750 mm L ${\times}$ 2000 mm H) packed with polyurethane foam (20 mm W ${\times}$ 20 mm L ${\times}$ 20 mm H) was installed in an fine chemical plant emitting gas streams containing ethyl alcohol, ethyl acetate, and dichloromethane. The biofilter was successfully operated for 30 days under highly fluctuating incoming concentrations (maximum 3500 ppm) at a residence time of 36 and 60 sec. Both ethyl alcohol and ethyl acetate were removed more than 95%, but dichloromethane removal was less than 50%. Malodor was also removed more than 90% from 17 days after start up.

Antioxidant Activities of Extract with Water and Ethanol of Perilla frutescens var. acuta kudo Leaf (차조기(Perilla frutescens var. acuta kudo) 잎의 물과 에탄올 추출물의 항산화 활성)

  • This study was conducted to examine antioxidant activities of Perilla frutescens var. acuta leaf. For the this purpose, DPPH radical scavenging activity, lipid oxidation inhibition, SOD-like activity, and xanthine oxidase inhibitor activity of water extract, ethanol extracts (30, 50, 70, and 95%) and the fractions obtained from these extracts were determined. The electron donating abilities of the chloroform fraction obtained from the 70% and 95% ethanol extracts were 50%, and that of the ethyl acetate fraction for all of the extracts was above 75%. In particular, the electron donating ability of the ethyl acetate fraction of the 70% ethanol extract showed the greatest activity with 200.5 ppm of $RC_{50}$ value. The 70% ethanol extract was most effective to inhibit the automatic oxidation of linoleic acid at $40^{\circ}C$ storage. The highest inhibition effects appeared in the chloroform and ethyl acetate fractions of the water extract, and the 30, 50, and 70% ethanol extracts, and the highest lipid oxidation inhibiting effect of the 95% ethanol extract occurred in the hexane and acetate fractions. The SOD-like activity of the water extract was 30.3%, and the activities of the various concentration of ethanol extracts were 28-32% and the activity of the 70% ethanol extract was the highest. The SOD-like activity of the ethyl acetate fraction of the 70% ethanol extract was highest with 1,549.0 ppm of $RC_{50}$ value. Xanthine oxidase inhibition activity was greatest in the water extract and the activities of the ethanol extracts were 36-41.2%. The xanthine oxidase inhibition activity of the ethyl acetate fraction of the water extract was highest. In summary, we found that electron donating ability, lipid oxidation inhibition, and SOD-like activity of Perilla frutescens var. acuta leaf were greatest in the ethyl acetate fraction of the 70% ethanol extract, and xanthine oxidase inhibition activity was highest in the ethyl acetate fraction of the water extract.
