Studies on the Extraction Condition of Ssang Wha Tea (쌍화차의 추출조건에 관한 연구)

  • During the preparation of Ssang Wha tea it was observed that the highest yield was obtained when it was extracted with 50% ethanol, of which guantity was added 10 times more then that of Sample. The most effective extraction temperature and time was $90^{\circ}C$ and 8hours on the water bath, which was resulted in saving of production cost. From the HPLC Analysis, it was also observed that product extracted with 50% ethanol give higher yield than that of exfrac ted with water.

Yellow Color Extraction from Gardenia jasmonoides Ellis for Development of Natural Food Color (천연식용색소 개발을 위한 치자에서 황색소의 추출)

  • In order to make natural food color from Gardenia, we investigated optimal conditions of color extraction, and thermal stability and light stability of color extracted compared with Yellow-4. In case of ethanol extraction, optimal conditions for color extraction were substrate 10%, 4$0^{\circ}C$, pH 7.0 and 42rs, respectively. In case of water extraction, optimal conditions for color extraction were substrate 10%, 7$0^{\circ}C$, pH 7.0 and 48hrs, respectively. Extraction yield in the optimal conditions was 75% in ethanol and 63% in water. The thermal stability and light stability of Yellow-4 were both upper 98%, but those of Gardenia yellow color were 62 and 90%, respectively.

Oleoresin Content and Physiological Activities of Fresh Red pepper by microwave-Assiated Extraction (마이크로웨이브 추출공정에 의한 홍고추 올레오레진의 함량 및 기능적 특성)

  • Water and ethanol extracts of fresh red peppers were obtained by two methods, conventional extraction (CE) and microwave-assiated extraction (MAE), under different extraction conditions. Red pepper oleoresins extracted by CE and MAE were examined in oleoresin yield and physiological activities. The proper extraction time of MAE was about 5 minutes, whereas that of CE was 2 hours. therefore extraction time was decreased drastically by MAE but there was no significance in oleolesin yields. the electron donating abilities also showed negligible difference between two extracts obtained by CE and MAE, and 80% level in all extracts . the nitrite scavenging effect was reduced by increased of ph , and showed a high elimination effect over 85% at ph 1.2. All extracts had a high tyrosinase inhibitory effects of 100%. The angiotensin I-conventing enzyme effect showed higher activity with over 80% in MAE than 70% level in CE. the capsanthin was extracted with ethanol and was 11.4 and 12.9 ${\mu}$moles per 1 g of fresh red pepper by CE and MAE, respectively.

Phospholipids Isolation from Squid Viscera Residues After Supercritical Carbon Dioxide Extraction (오징어 내장의 초임계 이산화탄소 추출 잔류물로부터 인지질의 분리)

  • Phospholipids were recovered from squid viscera residues by ethanol extraction after supercritical carbon dioxide($SCO_2$) extraction and from squid viscera was not processed $SCO_2$ by various organic solvent extraction. $SCO_2$ extraction were performed at $45^{\circ}C$ and 20 MPa for removal of non polar lipid molecules from freeze dried squid viscera sample. Phospholipids were extracted from freeze dried squid viscera sample by chloroform, hexane, methanol, and ethanol and from $SCO_2$extracted squid viscera sample by ethanol. The pH was fixed at 5.7 for all phospholipids extraction conditions. Phospholipid classes were analyzed by HPLC equipped with evaporative light scattering detector (ELSD). Phosphatidyl choline(PC) extracted by ethanol from $SCO_2$ extracted residues was higher than that of extracted by ethanol from squid viscera. But phosphatidyl ethanolamine(PE) and phosphatidic acid(PA) were extracted higher percentage in raw squid viscera. The fatty acid compositions in phospholipids extract by ethanol extract from $SCO_2$ extracted residues were analyzed by gas chromatography(GC). Docosahexanoic acid(DHA) was found in highest percentage in phospholipid extract.

Optimization of Extraction of Effective Components from Vitis coignetiae, the Crimson Glory Vine (산머루 유용성분 추출공정의 최적화)

  • A central composite design was used to investigate the effects of the three independent variables of extraction temperature ($X_1$), ethanol concentration ($X_2$), and extraction time ($X_3$), on dependent variables including yield ($Y_1$), total phenol levels ($Y_2$), electron-donating ability ($Y_3$), brownness ($Y_4$), and reducing sugar content ($Y_5$) of Vitis Coignetiae. Yield was affected by extraction temperature and time. The maximum yield was obtained at $91.62^{\circ}C(X_1)$, and, 25.37% (w/v) ethanol ($X_2$), after 317.70 min of extraction ($X_3$), evident as a saddle when displayed graphically. Total phenol levels were essentially unaffected by extraction temperature or ethanol concentration, but were highly influenced by extraction time. The maximum total phenol levels was 4,763.46 GAE mg/100 g obtained at $88.20^{\circ}C(X_1)$, and 47.79% (w/v) ethanol ($X_2$), after 349.32 min ($X_3$) of extraction. Electron-donating ability (EDA) was affected by extraction temperature and time. Maximum EDA was 55.90% at $86.72^{\circ}C(X_1)$, 50.61% (w/v) ethanol ($X_2$), and 265.96 min ($X_3$) of extration time, again shown by a graphical saddle. Brownness was affected by extraction time. The maximum extent of brown coloration was obtained at $82.66^{\circ}C(X_1)$, 99.27% (w/v) ethanol ($X_2$), and 252.63 min of extraction time ($X_3$), once again shown by graphical saddle. The maximum reducing sugar content was obtained at $96.24^{\circ}C(X_1)$, 22.59% (w/v) ethanol ($X_2$), and 216.06 min extraction time($X_3$).

Anti-inflammatory Effects, Analgesic Effects, Antipyretic Effects, Anti-oxidative Effects, and Histamine Permeation Inhibition Effects of Modified Gamgil-tang per Extraction Method (추출 방법에 따른 감길탕가미방(甘桔湯加味方)의 항염증(抗炎症), 진통(鎭痛), 해열(解熱), 항산화(抗酸化) 및 Histamine 투과 억제효과)

  • Objectives : Modified Gamgil-tang is a prescription commonly used for respiratory diseases. This thesis was carried out to check the treatment effects and diversity of drug formulation by comparing extraction method of ethanol and water of modified Gamgil-tang. Methods : All experiments were carried out with water and 50% ethanol extraction for comparison. In vivo experiment, hyaluronidase inhibitory effects and trypsin inhibitory effects were tested to measure the anti-inflammatory effects activity. Scavenging effects of DPPH free radical, xanthine oxidase inhibitory effects and inhibition on TBA-RS formation were experimented to measure anti-oxidative effects. With the in vivo experiment, ICR group mice and SD group rats were used as experimental animals. An anti-inflammatory effects experiment were carried out to measure the action on carrageenin-induced hind paw edema: analgesic effects were measured using writhing syndrome induced by 0.7% acetic acid in mice: antipyretic effect was measured using endotoxin, and inhibitory effects of increase vascular permeability induced by 0.5% histamine were measured. Results : For extraction of glycyrrhizin contents, ethanol extract was extracted 2 times of that of water extract. Anti-inflammatory effects showed high in ethanol extract. Anti-oxidative effects measured high in ethanol extract. No significant result was found in inhibition on TBA-RS formation. Analgesic effects were found to be similar in water and ethanol extract. Antipyretic effects were found to be stronger in water extract. Inhibitory effects of increase vascular permeability induced by 0.5% histamine showed stronger in ethanol extract. Conclusion : By measuring anti-inflammatory effects, analgesic effects, antipyretic effects, anti-oxidative effects, and histamine permeation inhibition effects both in water extract and ethanol extract after adding agents such as Mentha Herba, Gardenias Fructus, and propolis to existing Gamgil-Tang, ethanol extract was found to be more effective in anti-inflammatory effects, analgesic effects, anti-oxidative effects, and histamine permeation inhibition effects. The converse was found for antipyretic effect.

Optimization of Extraction Conditions for Dried Jujube by Response Surface Methodology (반응표면분석에 의한 건대추의 추출조건 최적화)

  • Extraction characteristics of dried jujube and functional properties of corresponding extract were monitored by response surface methodology. Maximum extraction yield of 53.69% was obtained at extraction temperature of $50.35^{\circ}C$, extraction time of 16.69 hr, and ethanol concentration of 72.88%. At extraction temperature, extraction time, and ethanol concentration of $45.80^{\circ}C$, 15.47 hr, and 73.12%, respectively, maximum cyclic adenosine monophosphate content was 8.20 mg/100 g. Maximum total polyphenol content was 18.85 mg/g at extraction temperature, extraction time, and ethanol concentration of $64.91^{\circ}C$, 20.84 hr, and 66.91%, respectively. Maximum total flavonoid content was 0.48 mg/g at extraction temperature, extraction time, and ethanol concentration of $57.36^{\circ}C$, 15.14 hr, and 71.08%, respectively. $IC_{50}$ value of electron donating ability showed maximum level of 32.34 mg/mL at extraction temperature of $48.46^{\circ}C$, extraction time of 19.25 hr, and ethanol concentration of 65.36%. Maximum ascorbic acid equivalent antioxidant capacity was 3.58 mg ascorbic acid equivalent per gram sample at extraction temperature, extraction time, and ethanol concentration of $56.09^{\circ}C$, 21.86 hr, and 65.36%, respectively.

A Study on the Solvent Fraction for Extraction of Bioactive Substances in Sea Cucumber (해삼의 생리활성물질 추출 용매분획에 따른 연구)

  • In this study, a study was attempted to confirm the yield of the solvent fraction for extracting physiologically active substances of polyphenols and flavonoids in sea cucumber. In the previously reported cases, the 50% ethanol extraction solvent fraction was confirmed as a result of high yield of sea cucumber antioxidants. The determination of the amount of antioxidants extracted from sea cucumber was confirmed as a result of the extraction solvent fraction applied to the extraction. In addition, the necessity of extraction of the solvent fraction was obtained through a previous study that 'there is a significant difference in the content of sea cucumber physiologically active antioxidants extracted depending on the extraction solvent.' The results of extracting high antioxidants from the phenolic substances contained in the sea cucumber extract of the 50% ethanol extraction solvent fraction were demonstrated. In the study case of extraction solvent fraction, the ethyl acetate solvent fraction, which yielded the lowest, yielded a higher phenol content than other solvent fractions, and the antioxidant effect was confirmed. Accordingly, it was confirmed that the effect on the high yield of the antioxidant extract through the yield change according to the application of the extraction solvent fraction was performed. Therefore, it was verified as an optimized sea cucumber physiologically active substance extraction solvent fraction of 50% ethanol solvent.

The effect of Plantaginis asiaticae Folium on lipid accumulation in 3T3-L1 adipocytes by extraction conditions (추출조건에 따른 차전초 추출물이 3T3-L1 세포의 지방 축적에 미치는 영향)

  • Objectives : Plantaginis asiaticae Folium (PA) has been widely used in Korean medicine for treatment of liver disease, stomach troubles and inflammation. We investigated the effect of PA on lipid accumulation in 3T3-L1 according to extraction conditions. Methods : The MTT assay was employed to evaluate the cytotoxicity of PA extracted by different solvents (water, 50% ethanol, and 95% ethanol) on 3T3-L1 preadipocytes. Oil red O staining was used to identify intracellular lipid accumulation in 3T3-L1. 3T3-L1 adipocytes were treated with PA at concentration ranging in 0.1, 0.2 and $0.4mg/m{\ell}$. PA was extracted by different extraction conditions such as extraction solvents, extraction time, and extraction temperature. In addition, UPLC analysis was used for determination of candidates of active ingredients in PA. Results : 3T3-L1 preadipocytes were treated with PA extracted by different solvents (water, 50% ethanol, and 95% ethanol) and there was no cytotoxicity. Oil red O staining was employed to identify the effect of PA on lipid accumulation in 3T3-L1. In the present study, PA water extraction at $70^{\circ}C$ for 6 hours decreased greatly in lipid accumulation. The range of concentrations was 0.1, 0.2 and $0.4mg/m{\ell}$. Concentration at $0.2mg/m{\ell}$ was the most effective one among them. Candidates of active ingredients in PA were shown plantamajoside and acteoside through UPLC. Conclusions : These results suggest that the effect of PA water extraction at $70^{\circ}C$ on lipid accumulation in 3T3-L1 is superior to other extraction conditions. We suppose that plantamajoside and acteoside may be candidates of active ingredients in PA.

Effect of Extraction Solvent on the Antioxidant Activity of Lentinula edodes GNA01 Extract (추출용매에 따른 이슬송이버섯(Lentinula edodes GNA01) 추출물의 항산화 활성)

  • Antioxidant activities of 80% methanol, water, and 70% ethanol extracts of Lentinula edodes GNA01 were compared and estimated. The yield of Lentinula edodes GNA01 was identified to be in the following order: water>70% ethanol>80% methanol, but there was no significant difference between 80% methanol and 70% ethanol extracts. The highest total phenolic content (TPC) and total flavonoid content (TFC) were found in water extract, and TPC of 80% methanol extract was higher than that of 70% ethanol extract and TFC of 70% ethanol extract was higher than that of 80% methanol extract. Water extract exhibited the strongest DPPH, ABTS radicals, and nitrite scavenging activities, $Fe^{2+}$ chelating ability, and FRAP among the three extracts. In addition, antioxidant activity of 80% methanol extract was higher than that of 70% ethanol extract in most of the experiments. As a result, antioxidant activity of Lentinula edodes GNA01 showed a difference according to extraction solvent and concentration; nevertheless, water extract exhibiting high polarity had the strongest antioxidant effect. Consequently, water extract from Lentinula edodes GNA01 is anticipated to be useful for the development of a high value-added functional product.
