Physiological Properties of Jeju Traditional Doenjang (제주 재래식된장의 생리적 특성)

  • The antioxidant activities of water extracts from wild vegetables such as Ligularia fischeri (GC), Capsicum annuum L. (GCY), Aster scaber (CNM), Petasites japonicus S. et Z. Max (MYD), Ipomoea batatas L. (Lam) (GGM) were evaluated and compared with water extracts from freeze dried block. The antioxidant properties of water extracts from wild vegetables and their freeze dried block were evaluated using different antioxidant tests; 2.2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging, hydroxyl radical scavenging and nitrite scavenging activities. The water extracts from wild vegetables were found to have a higher total phenolic content than water extracts from freeze dried block. Total phenolic contents of water extracts from GC, GCY, CNM, MYD, and GGM were $471.66{\pm}3.52\;{\mu}g/mg,\;141.33{\pm}2.51\;{\mu}g/mg,\;177.33{\pm}2.88\;{\mu}g/mg,\;238.66{\pm}9.50\;{\mu}g/mg\;and\;122.67{\pm}3.51\;{\mu}g/mg$, respectively. At the concentrations of 1000 ppm, water extracts from GC, GCY, CNM, and GGM showed higher activities than water extracts from their freeze dried block on DPPH radical scavenger activity. The activity of water extracts from CNM, GC, GCY, MYD, and GGM were 90.9%, 89.9%, 76.6%, 71.1%, and 57.4%, respectively. When 10000 ppm of GC, GCY, CNM, MYD, and GGM water extracts tested for hydroxyl radical scavenging activity, activities were increased by 38.8%, 33.4%, 35.9%, 34.3%, and 33.8%, respectively and a similar effect was found with water extracts from GCY, CNM, and GGM freeze dried block at 10000 ppm concentration. However, the water extracts from GC and MYD was slightly more effective than freeze dried block extracts. The water extracts from wild vegetables and their freeze dried block had effective DPPH radical scavenger activity and hydroxyl radical scavenging activity at all tested concentrations. Nitrite scavenging activity of GC water extract significantly increased in a dose-dependent manner and the extract had higher nitrite scavenging activity than extracts freeze dried block extracts. We found that freeze dried block maintained antioxidant activities of the wild vegetables.

Antibacterial and Antioxidative Activities of Inula britannica Flower Extract (금불초 꽃 추출물의 항균 및 항산화 활성)

  • In this study, the antibacterial activity, antioxidative effects, inhibitory effects on tyrosinase of Inula britannica flower extracts were investigated. MIC values of ethyl acetate fraction from Inula britannica flower on P. acnes 0.25 %, respectively. The results showed that the antibacterial activity of the ethyl acetate fraction was the highest in the P. acnes. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities ($FSC_{50}$) of ethyl acetate fraction of Inula britannica flower was $8.55{\mu}g$/mL. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of some fInula britannica flower extracts on ROS generated in $Fe^{3+}$- EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The order of ROS scavenging activities were ethyl acetate fraction $0.24{\mu}g$/mL. Ethyl acetate fraction showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of Inula britannica flower on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The Inula britannica flower extracts suppressed photohemolysis in a concentration dependent manner ($5{\sim}100{\mu}g$/mL), particularly deglycosylated flavonoid aglycone fraction exhibited the most prominent celluar protective effect ($\tau_{50}$, 164.15 min at $25{\mu}g$/mL). The inhibitory effect of Inula britannica flower extracts on tyrosinase was investigated to assess their whitening efficacy. Inhibitory effects ($IC_{50}$) on tyrosinase of some Inula britannica flower extracts were high. Ethyl acetate fraction has $IC_{50}$ of $87.03{\mu}g$/mL. These results indicate that extract/fractions of Inula britannica flower can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. And inhibitory activity on tyrosinase of the ethyl acetate fraction and high potential as bactericide against the skin pathogenic bacteria could be applicable to new functional cosmetics for antioxidant, antiaging, antibacterial activity.

Extraction Characteristics of Saponin and Acidic Polysaccharide Based on the Red Ginseng Particle Size (홍삼의 입자크기에 따른 사포닌 및 산성다당체의 추출 특성)

  • Effect of pulverization on total solid, crude saponin, and acidic polysaccharide contents of dried red ginseng main root were tested. Several particle size samples, including red ginseng main root (non pulverized), $10{\sim}40$ mesh powder, $40{\sim}100$ mesh powder, and >100 mesh powder were used in the extraction. The sequential solvent extraction method (1st: 70% EtOH at $70^{\circ}C$ for 12 hr, 2nd: 70% EtOH at $70^{\circ}C$ for 12 hr, 3rd: water at $70^{\circ}C$ for 12 hr) was applied to extract the saponins and acidic polysaccharide. Extraction yield of total solid of pulverized red ginseng ($10{\sim}40$ mesh size) was increased to 20% compared with that of non-pulverized. Especially, the crude saponin content of pulverized red ginseng ($10{\sim}40$ mesh size) showed an increase of 47% over non-pulverized. No difference in the component ratio was observed by pulverization, when the individual ginsenosides were quantified by HPLC. Also, extraction yield of acidic polysaccharide of pulverized red ginseng ($10{\sim}40$ mesh size) was increased 57% compared with that of non-pulverized. The results suggested that pulverization might be useful for increasing the extraction yield of red ginseng components.

Effect of antioxidation and antibacterial activity on crude extract and Characterization of American Cockroaches (Periplaneta americana L.) in Korea (국내 서식 미국바퀴(Periplaneta americana L.)의 특성 및 추출물의 항산화·항균 효과)

  • The American cockroaches, Periplaneta americana L. was the most important worldwide pest species. It has been an public health problems. We were determinated life cycle and extraction of crude extracts by chemical reagents from cockraches (P. americana L.). The extracted crude solution has been antibacterial activity to gram negative bacteria (Pseudomonas aeruginosa, $6.44{\pm}1.03mm$), gram positive bacteria (Bacillus subtilis, $1.88{\pm}0.40mm$), and fungus (Candida albicans, $5.61{\pm}0.57mm$) using radial diffusion assay. We were analysed of up-regulation of Glutathione-S-transferases (GSTs) stimulation, indicating that antioxidantial protein from various classes are simultaneously expressed in a single insect upon infection or injury. The gene from Periplaneta americana L. were cloned, analysed sequence, and measured protein expression by Real Time PCR (Polymerase Chain Reaction).

Effect of Antioxidant, Antimutagenicity and Anticancer of Root Extract from Ixeris dentata Nakai (씀바귀 뿌리 추출물의 항산화성, 항돌연변이원성 및 항암활성 효과)

  • Ixeris dentata root were extracted with methanol and then fractionated with n-hexane, EtOAc and BuOH to get active fractions. and their antioxidant and antimicrobial activities in each fraction were determined. Ethyl acetate fraction of Ixeris dentata root showed strong antioxidant activities, but aqueous fraction did not show any activities. But in the antimicrobial test, aqueous fraction showed strong antimicrobial activities except to Escherichia coli. especially, aqueous fraction showed the strongest activities against Hypocrea nigricans. and butanol fraction showed the strongest activities against Cladosporium herbarum. This study was performed to determine the antimutagenic and cytotoxic effect of Ixeris dentata root methanol extract on Salmonella typhimurium TA98, TA100 and cancer cell lines using ames test and cytotoxicity assay, respectively. Cancer cell lines include human lung carcinoma(A549), human breast adenocarcinoma(MCF-7) and human hepatocellular carcinoma (Hep 3B). Futher fractionations with hexane, ethyl acetate, butanol and water from methanol extract of Ixeris dentata root were performed to obtain effective fraction, methanol extracts showed 79.94% inhibitory effect on the mutagenesis induced by N' -methyl- N' -nitro-N-nitrosoguanidine(MNNG) against TA100, while 89.99% inhibition was observed on the mutagenesis induced by 4-nitroquinoline-1-oxide(4NQO) against TA98. In the meanwhile, butanol fraction showed 89.92% and 71.01% inhibitory effect on the mutagenesis induced by benzo(a)pyrene(B(a)P) against TA98 and TA100, respectively. Ethyl acetate fraction showed the strongest effect against A549, MCF-7 and Hep3B at the same concentration compared to those of other fration.

Physicochemical Properties and Physiological Activities of Rhus verniciflua Stem Bark Cultured with Fomitella fraxinea (장수버섯 균사체가 배양된 옻피의 이화학적 특성 및 생리활성)

  • The contents of proximate composition, free amino acids and phenolic acids in the Fomitella fraxinea cultivated-Rhus verniciflua stem bark(FRVSB), and its adipogenesis effect were investigated. The proximate composition(%) of FRVSB was as follows: moisture(7.64), ash(6.30), crude fat(3.86), crude protein(3.59) and sugar(not detected); while Rhus verniciflua stem bark(RVSB) contained 1.64, 8.09, 7.28, 6.48 and 5.39, respectively. The total free amino acids concentration was 97.41 mg% in FRVSB and 71.91 mg% in RVSB. Phosphoserine(55.06 mg%), ammonia(17.84mg%) and aspartic acid(6.05mg%) were predominant amino acids. The content of total phenolic acids was 422.89 ppm in ethanol extract and 283.86 ppm in water extract, with syringic and gallic acid as the main component. The FRVSB extracts showed a potent free radical scavenging activity for DPPH(2,2-diphenyl-1-picrylhydrazyl hydrate) with $IC_{50}$ of $28.54\;{\mu}g$(EtOH) and $54.70\;{\mu}g$(water), respectively, whereas $IC_{50}$ value of gallic acid was $1.84\;{\mu}g$. The protective effect of both ethanol and water extract the extracts against UV-induced oxidative stress in NIH3T3 was observed. The water extracts of FRVSB may promote adipogenesis in 3T3-L1 cells.

Protective effect of ethyl acetate extract of Ishige okamurae against carbon tetrachloride-induced acute liver injury in rats

  • Several compounds and extracts isolated from a brown alga, Ishige (I.) okamurae, exhibit anti-oxidant and anti-inflammatory effects. The present study investigated whether the ethyl acetate (EtOAc) fraction of I. okamurae (EFIO) could ameliorate carbon tetrachloride ($CCl_{4}$)-induced hepatotoxicity in rats. Sprague-Dawley rats were intraperitoneally (i.p.) administered with EFIO at 10 or 50 mg/kg per day for 2 consecutive days before $CCl_{4}$ injection (3.3 mL/kg, i.p.). Twenty four hours later, the rats were anesthesized with diethyl ether and dissected. Pretreatment with EFIO significantly reduced the increased serum levels of alanine aminotransferase and aspartate aminotransferase in $CCl_{4}$-treated rats. Pretreatment with EFIO also significantly inhibited the reduced activities of superoxide dismutase and catalase in the $CCl_{4}$-injured liver. Histopathological evaluations showed that hemorrhage, hepatocyte necrosis, inflammatory cell infiltration, and fatty degeneration induced by $CCl_{4}$ treatment were ameliorated by the administration of EFIO. Additionally, liver immunohistochemical analyses revealed the marked reduction in ED1-positive monocyte-like macrophages in EFIO-pretreated rats given $CCl_{4}$. These results suggest that EFIO ameliorates $CCl_{4}$-induced liver injury, possibly through the inhibition of oxidative stress.

Antioxidative and Hepatoprotective Effect of Compounds Isolated from Galla Rhois(Rhus javanica Linne) (오배자 성분의 항산화 및 간보호 효과)

  • The major components were isolated from the n-hexane, EtOAc and BuOH extract of Galla Rhois(Rhus Javanica Linne). Their structures were characterized as syringic acid, gallic acid methylester, protocatechuic acid, gallic acid and 1, 2, 3, 4, $6-penta-O-galloyl-{\beta}-D-glucose$. This study was carried out to investigate the biological activities of isolated compounds. Five compounds were tested for hepatoprotective effects on CCl4-induced cytotoxicity in primary cultured rat hepatocytes and antioxidative effect on Ferric-Thiocyanate method and TBA method. As a result, isolated five compounds showed stronger antioxidative activity than tocopherol, and the antioxidative activity of gallic acid methylester, protocatechuic acid and syringic acid were similar to that of BHA on Ferric-Thiocyanate method. Specially 1, 2, 3, 4, $6-penta-O-galloyl-{\beta}-D-glucose$ showed stronger effect of lipid-peroxidation inhibition than BHA. Gallic acid appeared stronger inhibitory effect of malondialdehyde on TBA method. Hepatoprotective effect of 1, 2, 3, 4, $6-penta-O-galloyl -{\beta}-D-glucose$ was similar or even higher than that of glycyrrhizin on primary cultured rat hepatocyte cytotoxicity.

Biological activity of supercritical extraction residue 60% ethanolic extracts from Ulmus davidiana (느릅나무 초임계 추출박 60% 주정추출물의 생리활성)

  • Ulmus davidiana supercritical fluid residue EtOH extracts(USCFR) and ethyl acetate solvent fraction (USCFREA) of supercritical extraction foil were investigated in order to examine the recycling of supercritical extraction foil in the process of studying Ulmus davidiana branch supercritical extract. Experiments were performed for the determination of total phenol content. The $IC_{50}$ value(ppm) of DPPH radical scavenging activity and ABTS radical scavenging activity was $7.42{\pm}0.09$, $7.50{\pm}0.05$, $22.94{\pm}0.09$, $6.43{\pm}0.10$, and USCFREA, respectively, as compared with the positive control (vitamin C) with values $17.80{\pm}0.14$ and $5.34{\pm}0.06$, respectively. The antioxidative activities of USCFR and USCFREA were confirmed to be superior to the positive control group. In anti-allergic activity studies, both USCFR and USCFREA showed concentration-dependentanti-allergic activity, and USCFREA showed strong anti-allergic activity even at very low concentrations. Thetotal phenolic contents (ugEG, ugGA; ppm) of USCFR were $134.17{\pm}0.13$, $132.02{\pm}0.24$ and USCFREA were $154.77{\pm}1.05$ and $153.18{\pm}1.10$, respectively. Based on the above results and strong antioxidant activity, USCFR and USCFREA hold the potential to be considered as basic research materials for the development of therapeutic supplements based medicines or functional cosmetics related to chronic inflammatory skin immunity diseases.

Antioxidant and Tyrosinase Inhibition Activity Promoting Effects of Perilla by the Light Emitting Plasma (발광플라즈마 처리에 의한 들깨 부위별 항산화 및 Tyrosinase 저해 활성 효과)

  • Background: The light emitting plasma (LEP) has recently attracted attention as a novel artificial light source for plant growth and functional component enhancement. We investigated the effects of LEP on whitening and antioxidant activities of the plant parts of perilla. Methods and Results: Previously germianted seeds of perilla were cultivated under different light conditions (fluoresce lamp, LED red, blue, white, green, and LEP) in a culture room for 2 months. Parts of perilla were harvested and extracted in 70% EtOH. The extracts were used to detect total phenolic contents, total flavonoid contents, 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS), reducing power and tyrosinase inhibition activity as indicators of biological activity. Biological activity was highest in seedlings grown under LEP. The total phenolic content was highest in the stems and the total flavonoid content was highest in the roots of perilla exposed to LEP. The DPPH and ABTS radical activity in all the parts of perilla exposed to LEP were higher by approximately three-fold compared to that in the control (fluoresce lamp). The reducing power values of perilla significantly increased after treatment with LEP. In addition, all the extract of perilla plants exposed to LEP promoted the tyrosinase inhibitory activity. These results suggest that LEP can be an important artificial light source for enhancement of biological activity. Conclusions: LEP could promote whitening and antioxidant activity of perilla.
