Cytoprotective Effect of Cinnamomum japonicum Siebold Branch Extracts via Blocking Oxidative Stress in Hepatocytes (간세포에서 산화적 스트레스 억제를 통한 생달가지 추출물의 세포보호 효과)

  • Objectives : Native to East Asia, Japan, and Korea, Cinnamomum japonicum Siebold (CJ) is renowned for its aromatic leaves and bark. We previously assessed the antioxidant activity of fractionated CJ branches (CJB:70% EtOH extract), including hexane (CJB1), chloroform (CJB2), ethyl acetate (CJB3), butanol (CJB4), and water (CJB5). Our findings revealed that CJB3 exhibited the highest antioxidant activity. Here, we aimed to investigate whether CJB3 possesses cytoprotective effects and induces the activity of antioxidant enzymes in hepatocytes. Methods : As HepG2 cells were the first to exhibit the key characteristics of hepatocytes, we investigated the hepatoprotective effects of CJB3 on HepG2 cells. Results : Before conducting the cell experiment, we checked that CJB3, up to a concentration of 100 ㎍/mL, did not exhibit cytotoxicity toward HepG2 cells. ROS production increased because of t-BHP treatment decreased in a concentration-dependent manner upon CJB3 treatment. We confirmed that CJB3 inhibited t-BHP-induced cell death. CJB3 was found to reverse the expression of proteins associated with t-BHP-induced apoptosis. We also observed that CJB3 induced Nrf2 phosphorylation and the nuclear translocation of Nrf2. And, CJB3 treatment caused a time-dependent enhancement of GCL and NQO1 protein expression. We further confirmed that CJB3 increased the expression of Nrf2 target genes, and this effect was associated with the activation of JNK, p38, and AMPK. Conclusion : CJB3 prevents t-BHP-induced oxidative stress and apoptosis and enhances the expression of Nrf2 target genes via JNK, p38, and AMPK activation. These results suggest that CJB3 is a promising candidate for the treatment of liver diseases.

Effect of Astragalus membranaceus-postbiotics Polysaccharide Changed by Lactic Acid Bacteria on Macrophage (유산균에 의해 변화된 황기-포스트바이오틱스 다당류가 대식세포에 미치는 영향)

  • To increase industrial applicability of Astragalus membranaceus (AM) as immunostimulating materials, hot-water extract (AME) was prepared from AM and fermented with Kimchi-lactic acid bacteria (Lactobacillus sakei & Leuconostoc mesenteroides) to prepare fermented AM-postbiotics (FAME). Although FAME prepared from AM-postbiotics did not show a significant enhancement in macrophage stimulating activity compared to non-fermented AME, crude polysaccharide (FAME-CP) fractionated by EtOH precipitation from FAME showed significantly higher macrophage stimulating activity than AME-CP. Compared to AME-CP, FAME-CP showed dramatic changes in component sugar and molecular weight distribution. FAME-CP was a polysaccharide with a major molecular weight distribution of 113.4 kDa containing Man (44.2%), Glc (19.3%), Gal (10.2%), GalA (10.2%), and Ara (7.4%) as sugar components. FAME-CP with enhanced macrophage stimulatory activity not only increased expression levels of mRNA genes encoding macrophage-activated factors (iNOS, TNF-α, MCP-1, IL-6, and COX-2), but also led the nuclear translocation of activated p65 and c-Jun. In conclusion, crude polysaccharide from AM-postbiotics fermented with lactic acid bacteria could increase industrial applicability as a functional material with enhanced immunostimulating activity than AME-CP.

Anti-Oxidant, Anti-Melanogenic, and Anti-Inflammatory Activities of Zanthoxylum schinifolium Extract and its Solvent Fractions (산초 추출물 및 분획물의 항산화, 미백 및 항염증 활성)

  • This study was designed to explore new nutraceutical and cosmetic resources possessing biological activities from the plant kingdom. To fulfill this purpose, we analyzed the anti-oxidative, anti-melanogenic, and anti-inflammatory activities of Zanthoxylum schinifolium extract (ZSE) and its solvent fractions using in vitro assays and cell culture model systems. Three kinds of ZSE treated with methanol, ethanol, and water exhibited potent anti-oxidative activities through DPPH radical scavenging capacity, and inhibited in vitro DOPA oxidation. Furthermore, Z. schinifolium methanol extract (ZSME) inhibited the ${\alpha}$-melanocyte stimulating hormone, which induces melanin contents in B16F10 cells. Its anti-melanogenic activity originates from the inhibition of tyrosinase enzyme activity and melanogenesis related protein expression. Moreover, lipopolysaccharide induced nitric oxide production in the RAW 264.7 cell line was also ameliorated by ZSME treatment in a dose dependent manner. Among the four solvent fractions of ZSME treated with dichloromethane, ethyl acetate, n-butanol, and water, three fractions, except water, showed significant anti-melanogenic and anti-inflammatory activities. Taken together, these results provide important new insights into Z. schinifolium, indicating that it possesses numerous biological activities such as anti-oxidative, anti-melanogenic, and anti-inflammatory activities. Therefore, it may well serve as a promising material in the field of nutraceuticals and cosmetics.

Analysis of Ingredients Using 5 Species of Native Plants from Mt. Jiri. for the Development of Cosmetic Raw Materials, DPPH and ABTS Activity-II (지리산 자생식물 5종의 성분 분석 및 DPPH, ABTS 활성 실험을 통한 화장품 원료개발-II)

  • Five species of plants (Pimpinella brachycarpa (Kom.) Nakai, Hylotelephium erythrostictum (Miq.) H. Ohba, Magnolia sieboldii K. Koch, Sanguisorba hakusanensis Makino, Agastache rugosa (Fisch. & Mey.) Kuntze) native to the clean area of Mt. Jiri were selected. The collection period was from May to September 2021, and the five species plants were collected in their native habitats with flowers in full bloom. The collected plants were extracted with 70% EtOH and 17 kinds of polyphenol components were analyzed. Next, flowers, leaves and roots were separated from plants, extracted with 70% EtOH for each part and experiments were conducted on DPPH, ABTS, total polyphenols, and total flavonoids. The results are as follows. 1. There were a total of 13 polyphenol components contained in the 5 species of plants native to Mt. Jiri. Among them, the total content of the most detected substance was Sanguisorba hakusanensis Makino, which was 126.2 ppm, and the main substance was ellagic acid (=ELA), which was 122.4 ppm. 2. As a result of examining the DPPH radical scavenging activity, the flowers, stems, and roots of Sanguisorba hakusanensis Makino showed the best scavenging activity, while the flower extracts of other plants showed good scavenging activity. 3. The highest ABTS radical scavenging activity was Sanguisorba hakusanensis Makino flower extract at 13.5 ㎍/㎖, followed by roots at 16.8 ㎍/㎖ and leaves at 22.6 ㎍/㎖;. Although such a large ABTS radical scavenging activity could not be confirmed in other plants, the flower extract was overall better than the leaf and root extracts of other parts. 4. The highest total polyphenol content was found in the leaves of Magnolia sieboldii K. Koch, at 161.4mg GAE/g, followed by the highest content in the roots of Magnolia sieboldii K. Koch and the roots of Agastache rugosa (Fisch. & Mey.) Kuntze, at 130.0mg GAE/g. Also, the lowest was found to be Sanguisorba hakusanensis Makino. 5. The total flavonoid content was 186.2mg CAE/g in the roots of Agastache rugosa (Fisch. & Mey.) Kuntze, 166.9mg CAE/g in the flowers, and the lowest was 116.1mg CAE/g in the leaves. As shown in the results above, Sanguisorba hakusanensis Makino has excellent antioxidant efficacy and has a high polyphenol content, so it is expected to be fully utilized in the cosmetics industry in the future.

Anti-pruritic Effect of Ethanol Extracts from Platycodon grandiflorum and its fermented production in Scratching Behavior Mouse Models (길경(桔梗)발효 추출물의 알레르기성 소양행동 억제효과연구)

  • Objectives : Platycodon Root is frequently used in traditional medicine to treat inflammatory diseases of the throat. The purpose of this study was to characterize the effect of the EtOH extract of fermented Platycodon grandiflorum on the ameliorative effects on the Antipruritic Effect of atopic dermatitis mouse model induced by compound 48/80 and ovalbumin (OVA)-induced allergic responses in mice. Methods : In the present study, we examined the anti allergic effect of Platycodon grandiflorum (PR) and its fermented production (FPR) in several mouse model. We measured acute ear edema in a mouse model caused by TPA and consecutively histological change of Ear tissue was observed by hematoxylin and eosin (H&E) staining. and also Scratching behaviors by compound 48/80 was investigated. The levels of allergic mediators such as immunoglobulin (Ig) E, and anti-oxidant markers such as SOD and MDA in the sera of OVA induced allergic mice were measured by enzyme-linked immunosorbent assay. Results : FPR inhibited compoud 48/80-induced scratching behavior in mice, as well as acetic acid-induced writhing in mice. The anti-scratching behavioral effect of FPR was more potent than PR. FPR extract significantly decreased the serum levels of IgE and MDA compared with those of OVA control group. Conclusions : These results indicate that Anti allergic effect of Platycodon grandiflorum is enhanced by fermentation with Saccharomyces cerevisae and FPR may be useful for protection from the itching reactions, which are IgE-mediated representative skin allergic diseases.

Inhibitory Effects and Molecular Mechanism of Adipocyte Differentiation by Rosae laevigata Fructus Ethanol Extracs (금앵자 에탄올 추출물에 의한 3T3-L1 지방세포의 분화억제 효과와 그 메커니즘 규명)

  • Obesity is caused by excess accumulation of body fat and contributes to various pathological disorders such as diabetes, hypertension, cardiovascular disease, and cancer. In this study, we investigated the effect of a 30% ethanol extract of Fructus Rosae laevigata (RLE) on adipogenesis in 3T3-L1 adipocytes, measured by triglyceride accumulation and expression of adipogenesis-related transcription factors during differentiation of pre-adipocytes into adipocytes. RLE decreased the intracellular triglyceride contents (assessed by Oil Red-O staining) in a dose-dependent manner. It also downregulated the expression of adipogenic transcription factors and inhibited cell proliferation during the mitotic clonal expansion phase of adipocyte differentiation by inducing G1 phase arrest. We investigated the alterations in the levels of G1 phase arrest-related proteins. The expression of p21 protein significantly increased, while the levels of Cyclin E, Cdk2, and phospho-Rb decreased in a dose-dependent manner in 3T3-L1 cells treated with RLE. These results suggest that RLE inhibits the differentiation of 3T3-L1 adipocytes by suppressing the expression of adipogenic transcription factors and inducing G1 phase arrest in the early stages of adipocyte differentiation.

Physiological Properties of Oak Mistletoe (Loranthus yadoriki) Extracts by Microwave Extraction Condition (마이크로웨이브 추출조건에 따른 참나무 겨우살이 추출물의 생리활성)

  • The physiological properties of oak mistletoe slice extracts were investigated. The total polyphenol content of mistletoe extracts was 50.28-109-69 mg%, depending on extraction conditions. The polyphenol content increased as extract concentration rose (25 mL/g>50 mL/g>100 mL/g). The electron-donating ability (EDA) of the 60% (v/v) ethanolic extract was 68.88% at 25 mL/g; this was the highest value of all extracts tested (p<0.05). The SOD-like activities of water, 30%, 60%, and 90% (all v/v) ethanolic extracts were 15.24-30.84% lower than those of 1.0% and 0.1% (both w/v) L-ascorbate solutions. Tyrosinase inhibitory activities of all samples were 3.40-30.92% lower than those of 1.0% and 0.1% (both w/v) L-ascorbate solutions. Nitrite-scavenging abilities measured at pH 1.2 and pH 3.0 were highest when the water, and the 30%, 60% and 90% (all v/v) ethanolic extracts, were tested. The results expand our understanding of the physiological properties of oak mistletoe slice extracts.

Effects of Ixeris dentata extract on the production of pro-inflammatory cytokines in the LPS stimulated rat and Raw 264.7 cells (씀바귀 추출물이 LPS투여 흰쥐 및 Raw 264.7세포에서 전염증성 cytokines 생성에 미치는 영향)

  • The purpose of this study was to investigate anti-inflammatory effect of Ixeris dentata ethanol extract in lipopolysaccharide-exposed rats and Raw 264.7 cells. Plasma concentrations of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ were lower in Ixeris dentata-treated groups than in control group. Concentration of plasma IL-10 was higher in Ixeris dentata-treated groups than in control group. Concentrations of liver IL-$1{\beta}$ and IL-6 were lower in the Ixeris dentata-treated groups than in control group. However, concentrations of liver TNF-${\alpha}$ and IL-10 were not significantly different among all treatment groups. In the study using lipopolysaccharide-exposed Raw 264.7 cells, concentrations of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ tended to be decreased, but concentration of IL-10 tended to be increased in Ixeris dentata-treated groups. Plasm concentrations of total protein and albumin appeared to be increased in Ixeris dentata-treated groups.

Leonuri Fructus Ameliorates Acute Inflammation via the Inhibition of NF-${\kappa}B$-mediated Nitric Oxide and Pro-inflammatory Cytokine Production (NF-${\kappa}B$ 조절(調節)을 통한 충울자(茺蔚子)의 염증억제효과)

  • Leonuri Fructus, a semen of Leonuri Herba, has been used for the treatment of menstrual disorders such as amenorrhea, dysmenorrhea and leukorrhea and for the remedy of hyperemia. The present study was conducted to evaluate the anti-inflammatory effects of the Leonuri Fructus extract (Leonurus japonicus Houtt. EtOH extract; LJE) in vivo and in vitro. In vitro study, the MTT assay for cell viability was conducted to determine the non-cytotoxic concentration of LJE treatment in media. The levels of NO were measured with Griess reagent. Pro-inflammatory cytokines were detected by ELISA method. The inflammation-related proteins of this study were detected by immunoblot anlaysis. The increases of NO production and iNOS expression were detected in LPS-treated cells compared with control, but LJE attenuated the increases of NO and iNOS by LPS. LJE reduced the production of TNF-${\alpha}$ and IL-$1{\beta}$ induced by LPS stimulation. LJE suppresses the signaling pathways of NF-${\kappa}B$ and MAPKs in LPS-induced macrophage cells. In vivo study, carrageenan-induced hind paw acute edematous inflammation rat model was used for evaluation of anti-inflammatory activity of LJE. LJE significantly inhibited the increases of hind paw swelling, skin thicknesses and inflammatory cell infiltrations, and decreased the numbers of mast cell induced by carrageenan injection. These results suggest that LJE has an anti-inflammatory therapeutic potential, which is mediated through modulating NF-${\kappa}B$ activation and MAPK phosphorylation. Inhibition of the rat paw edema induced by carrageenan is considered as direct evidence that LJE may be a useful source to treat inflammation.

Tyrosinase Inhibitory Activities of Meso-dihydroguaiaretic Acid from Machilus thunbergii (후박나무에서 분리한 Meso-dihydroguaiaretic acid의 tyrosinase 저해활성)

  • Machilus thunbergii (Lauraceae) is an evergreen tree cultivated in Korea and Japan. M. thunbergii has long been used as a traditional medicine in Korea, China, and Japan to treat various diseases, including edema, abdominal pain, and abdominal distension. In this study, dried stem bark of M. thunbergii extracted in methanol and extract was partitioned into n-hexane, CHCl3, and BuOH. The CHCl3-soluble extracts chromatographed on silica gel column using a CHCl3/acetone and n-hexane/EtOAc mixture to afford Compound 1 and 2. Two dibenzylbutane lignans, macelignan (1) and meso-dihydroguaiaretic acid (2), were isolated from the CHCl3-soluble extract of M. thunbergii stem bark. The structures of 1 and 2 were determined by 1H- and 13C-NMR spectroscopic data analyses and a comparison with literature data. The tyrosinase inhibitory activity of the isolated compounds was evaluated. Among these compounds, Compound 2 strongly inhibited the monophenolase (IC50=10.2 μM) activity of tyrosinase. A kinetic analysis showed that Compound 2 was a competitive inhibitor. The apparent inhibition constant (Ki) for Compound 2 binding to free enzyme was 4.8 μM. Based on these results, it can be concluded that meso-dihydroguaiaretic acid (2) is a potential candidate for the treatment of melanin biosynthesis-related skin diseases.
