Antibacterial Activity against Pathogens of Acne and Tyrosinase Inhibitory Activity of Extract and Fractions from Bark of Prunus sargentii (산벚나무 수피 추출물 및 용매 분획물의 여드름 원인균에 대한 항균활성과 Tyrosinase 저해 활성)

  • In this study, we investigated on antioxidative activity, antibacterial activity against pathogens of acne and tyrosinase inhibitory activity in 75% EtOH extract and its fractions from bark of Prunus sargen tii. The total polyphenol and flavonoidcontent of the EtOAc fraction were found to be 378.0 mg/g and 67.5 mg/g as the highest content. In the measurement of DPPH radical scavenging ability, EtOAc and BuOH fraction were exhibited stronger scavenging ability than the other fractions and 75% EtOH extract. In antibacterial activity by disc diffusion assay against pathogen of acne, antibacterial activity of the EtOAc fraction and 75% EtOH extract was stronger than the other fractions. Especially, the EtOAc fraction was the highest effective fraction in the antibacterial activity. Moreover, tyrosinase inhibitory activity of EtOAc fraction, BuOH fraction and 75% EtOH extract was higher than the other fractions. In particular, tyrosinase inhibitory activity of the EtOAc fraction showed higher activity than ascorbic acid used as positive control.

Evaluation of the EtOAc Extract of Lemongrass (Cymbopogon citratus) as a Potential Skincare Cosmetic Material for Acne Vulgaris

  • This study evaluated the biological properties of lemongrass (Cymbopogon citratus) extracts. The EtOAc extract of lemongrass had DPPH, TEAC, and nitric oxide-scavenging activity assay results of 58.06, 44.14, and 41.08% at the concentration of 50, 10, and 50 ㎍/ml, respectively. The EtOAc extract had higher elastase and collagenase inhibitory activities than the 80% MeOH, n-hexane, BuOH, and water extracts and comparable whitening activity toward monophenolase or diphenolase. Also, the EtOAc fraction had higher lipase inhibitory and antimicrobial activities against Cutibacterium acnes among extracts which is known to an important contributor to the progression of inflammatory acne vulgaris, and an opportunistic pathogen present in human skin. Total phenolic and flavonoid concentrations in the EtOAc extract were 132.31 mg CAE/g extract and 104.50 mg NE/g extract, respectively. Biologically active compounds in lemongrass extracts were analyzed by LC-MS. This study confirms that lemongrass extracts have potential use as cosmetic skincare ingredients. Thus, lemongrass can be considered a promising natural source of readily available, low-cost extracts rich in antioxidant, skincare, and antimicrobial compounds that might be suitable for replacing synthetic compounds in the cosmeceutical industry.

Effects of Ethyl Acetate Extract of Poncirus trifoliata Fruit for Glucocorticoid-Induced Osteoporosis

  • Poncirus trifoliata fruit (PTF) affects the digestive and cardiovascular systems, and kidney function. The authors studied the effects of ethyl acetate (EtOAc) extract of PTF on the activities of osteoblasts and in an animal model. The main compounds of the EtOAc extract, naringin and poncirin have been confirmed by HPLC and NMR analysis. Effects of osteoblastic differentiation were measured by alkaline phosphatase (ALP) activity, osteopontin (OPN) protein expression and osteoprotegerin (OPG) mRNA expression in MC3T3-E1 cells. Also, osteoclast differentiation was measured by multinucleated cells (MNCs) formation through tartrate resistance acid phosphatase (TRAP)-positive staining. Bone mineral density (BMD) was measured before and after treatment with EtOAc extract of PTF in prednisolone-induced osteoporotic mice. Dexamethasone (DEX) decreased OPN and OPG expression level in MC3T3-E1 cells and ALP activity was decreased by DEX dose-dependently. EtOAc extract of PTF recovered the levels of ALP activity, and the expression of OPN and OPG in MC3T3-E1 cells treated with DEX. In osteoclast differentiation, multinucleated TRAP-positive cell formation was significantly suppressed by the EtOAc extract of PTF. Total body BMD was restored by EtOAc extract of PTF in prednisolone-induced osteoporotic mice. In conclusion, EtOAc extract of PTF recovered DEX-mediated deteriorations in osteoblastic and osteoclastic functions, and increased BMD in glucocorticoid-induced osteoporosis.

Studies on the Antimicrobial Activities of Prunus mume (매실의 항미생물 활성 연구)

  • This study was conducted to investigate the antimicrobial activity of Prunus mume extracts. Prunus mume extracts have growth inhibitory activity to various microorganisms. Gram positive bacteria such as Bacillus cereus, Bacillus subtilis, Micrococcus leteus, Staphylococcus epidrimidis were more easily inhibited than Gram negative bacteria tested Vibrio parahaemolyticus, Salmonella typhimurium, Escherichia coli, Proteus vulgaris and Yeasts by Prunus mume extracts. Among several fractions of methanol extract, EtOAc & BuOH fractions were showed strong antibacterial activities, but those fractions were not showed on fungi.

Effects of Ethyl Acetate Extract from Ulmus davidiana var. japonica on Melanogenesis (느릅나무의 에틸 아세테이트 추출물에 의한 Melanin생성 효과)

  • Melanogenesis is a physiological process resulted in the synthesis of melanin pigments, which has a role in protecting skin front the damaging effect of ultra-violet (UV) radiation. The main aim of the present study was to examine the effect of Ulmus davidiana var. japonica(UL) on Melanogenesis. Cells were cultured in the presence of various concentrations of Ulmus davidiana var. japonica for 48 h, and there were estimated total melanin contents as a final product and activity of tyrosinase, a key enzyme, in Melanogenesis. Among the four solvent extracts tested, EtOAc extract mostly increased tyrosinase activity, And EtOAc extract increased the melanin contents and tyrosinase activity in a dose-dependent manner. Especially It was observed that 100$\mu\textrm{g}$/ml EtOAc extract promotes melanin secretion in B16/F10 melanoma cells by 140% at 48 h treatment and activity of tyrosinase increased by 180% in the presence of same concentration. In conclusion, as for EtOAc extract treatment, there was no effect on the viability of B16/F10 cell, only to stimulate Melanogenesis.

Pine Needle Extracts Inhibit Contractile Responses of the Isolated Rat Aortic Strips

  • The aim of the present study was to investigate whether self-fermented pine extract for 2 years (SFPE2) and ethyl acetate (EtOAc) fraction from self-fermented pine needle extract may affect the contractility of the isolated aortic strips and blood pressure of normotensive rats. SFPE2 ($360-1440\;{\mu}g/mL$) significantly depressed both phenylephrine ($10\;{\mu}M$)- and high potassium (56 mM)-induced contractile responses of the isolated rat aortic strips in dose-dependent fashion. The EtOAc-fraction ($400\;{\mu}g/mL$) also inhibited both phenylephrine ($10\;{\mu}M$)- and high potassium (56 mM)-induced contractile responses. Also, in anesthetized normotensive rats, intravenous injection of the EtOAc fraction (1.0~10.0 mg/kg) dose-dependently elicited hypotensive responses. The EtOAc fractions (1.0 and 3.0 mg/kg/30 min) inhibited norepinehrine-induced pressor responses. Intravenous infusion of SFPE2 fraction (3.0 and 10.0 mg/kg/30 min) also inhibited norepinehrine-induced pressor responses in both anesthetized spontaneously hypertensive rats (SHRs) and normotensive rats. In conclusion, these results suggest that both SFPE2 and the EtOAc fraction cause vascular relaxation in the aortic strips isolated from normotensive rats and SHRs as well as vasodepressor responses. Based on these experimental data, it seems that SFPE2 or the EtOAc fraction possesses active antihypertensive components, which are available to prevent or treat hypertension in future.

Effects of Anthriscus sylvestris Hoffmann Extract on the Biosynthesis of Collagen in Human Dermal Fibroblasts (사람 섬유아세포에서 전호(Anthriscus sylvestris Hoffmann)추출물이 콜라겐 생합성에 미치는 영향)

  • In this study, we investigated the boosting effects on collagen biosynthesis of $Anthriscus$ $sylvestris$ ethanol extract (ASE) in human dermal fibroblasts. To obtain more effective fraction and subfraction for collagen biosynthesis, standard solvent partition and open column chromatography were performed. The EtOH extract, solvent fractions, and 8 EtOAc subfractions were tested for their collagen synthesis capacity by [$^3H$]Proline-incorporation and ELISA assay. ASE increased 25% of total collagen synthesis and 27% of procollagen biosynthesis. The total collagen biosynthesis was increased by EtOAc fraction and E6 subfraction to 28% and 50% respectively. Type I procollagens were also upregulated by EtOAc fraction and E6 subfraction to 30% and 47%, each. Taken together, our data suggest that potential anti-aging effect of ASE on skin is via increasing collagen biosynthesis and effective subtraction is E6 subfraction of EtOAc fraction.

Antibacterial Activities of Caesalpinia sappan L. Extract and Structural Analysis of Its Related Brazilin (소목 추출물의 항균활성과 Brazilin의 구조분석)

  • Caesalpinia sappan L. has long been commonly used in oriental folk medicines to treat diseases. To investigate the antibacterial effects from C. sappan L. heart wood, the MeOH soluble extract was successively fractionated by using hexane, $CHC1_3$, EtOAc, BuOH, MeOH, and $H_2O$. Among of these extracts, the EtOAc fraction which partitioned to 3.94% of the highest yields was to be the most active against all human pathogenic bacteria in this experiment. In addition, the antibacterial activities of the EtOAc fraction were more effective against Gram (+) bacteria compared to those against Gram (-) bacteria, which showed difference of the antibacterial activities against Gram (-) bacteria. To confirm the identity of the active substances, the EtOAc fraction was further separated by silica gel adsorption column, high performance liquid chromatography, and 98.48% purity of brazilin (1.67 mg)/EtOAc (10 mg) fraction was obtained from 300 g of C. sappan L. heart wood. The isolated active substance was a single compound of yellow crystalline, and was identified as brazilin ($C_{16}H_{14}O_5$) by MS, and $^lH$-NMR and $^{13}C$-NMR. These results suggest that the brazilin in the EtOAc fraction from MeOH extract of C. sappan L. has a potential as a natural therapeutic agent against human pathogenic Gram (+) bacteria such as Staphylococcus aureus.

Antioxidative Activity of Some Solvent Extract from Caesalpinia sappan L. (소목 추출물의 항산화 효과)

  • Antioxidative activity of the extract from Caesalpinia sappan L. by various solvent was compared with several commercial antioxidants, using the Rancimat method. AI (antioxidative index; induction period of oil containing extract/induction period of control oil) of all extracts were higher than commercial antioxidants, such as BHA, ${\delta}-tocopherol$ and ascorbic acid. The ethanol extract was fractionated by liquid liquid extraction. Ethyl acetate fraction showed higher AI than the whole crude extract. When comparing POV and TBA value of palm oil and lard containing different level of each fraction, the oxidative stability of ethyl acetate fraction at 200 ppm level on palm oil and lard were similar to that of BHT at 200 ppm level, and better than BHA, ${\delta}-tocopherol$ and control.
