  • It is well-known that bisphenol A(BPA), an industrial raw material for polycarbonate and epoxy resins, shows estrogenic activity. Recent research from our laboratory has shown that SPA disrupts interaction between thyroid hormone and its receptor in a non-competitive manner, and alters the thyroid-hormone dependent expression of growth hormone(GH) and prolactin(PRL).   • Effect of 4-nonylphenol (4-NP), endocrine disrupting compounds (EDCs), on glutamate oxaloacetate transaminase (GOT) and glutamate pyruvate transaminase (GPT) were investigated in the plasma of juvenile rockfish, Sebastes schlegeli. Fish were injected with 4­NP (10, 50, 100 and 200 mg/kg body weight) in $70\%$ ethanol twice at 3-day intervals and plasma sampling were extracted at 7 days after the last injection. Controls received solvent only. 4-NP significant increased GOT in a dose-dependent manner. GPT was markedly elevated to $61\%$ (P<0.05) and $82\%$ (P<0.01) than that of the control at the 4-NP doses of 100 and 200mg, respectively. These results suggest that the estrogenic activity of 4-NP increase plasma GOT and GPT by toxic effect on hepatocyte.

Effects of Acute Oral Administration of Mancozeb on the Immune Function in Mice (Mancozeb의 급성노출이 마우스의 면역기능에 미치는 영향)

  • Mancozeb, a polymeric complex of zinc and manganese salts of ethylene bisthiocarbamate (EBDC), is used widely in agriculture as fungicides, insecticides, and herbicides. Mancozeb can be occupationally and environmentally exposed to human and has been reported to induce estrogenic activity, therein it is considered as an endocrine disrupter, We investigated the effects of acute exposure of Mancozeb on the immune function in mice. After single oral administration of Mancozeb to female ICR mice, the immunopathological parameters (body- and organ-weight, splenic cellularity hematological parameters), mitogen (Con A, PHA+IL-2, LPS)-induced splenocyte proliferation (SP) and splenic IgM plaque forming cell (PFC). WBC and splenic cellularity were decreased, but liver-, kidney-, and spleen-weight were increased when compared with control group. Splenic IgM PFC against SRBC was slightly lowered. Mitogen-induced proliferation of spleen cells from Mancozeb-treated mice was not signifcantly changed ex vivo, however, SP in vitro were significantly lowered in concentration dependent manner. These results indicate that Mancozeb could affect the immune function in mice.

  • Bisphenol A shares similarities in structure, metabolism and action with DES, a known human teratogen and carcinogen. Bisphenol A, a monomer of polycarbonate and epoxy resins, has been detected in canned food and human saliva. The purpose of the this study was to evaluate the cytotoxicity, cell proliferation of bisphenol A In the presence of a rat liver S9 mix, contaning cytochrome P450 enzymes, and Cu (II). In the present study, Bisphenol A in combination with Cu (II) exhibited a enhancement in cytotoxicity which were inhibited by free radical scavengers. The content of malondialdehyde, an end product of lipid peroxidation, was also found to increase with concentration of bisphenol A. Also, we examined the change of CuZn-SOD, Mn-SOD, catalase and GPx activities in the MCF-7 cells exposed to bisphenol A. The activities of CuZn-SOD, CPx, catalase were found to decrease with bisphenol A concentration. Meanwhile, the activity of Mn-SOD was unchanged. This indicated that elevated oxidative stress caused by imbalance between the production and removal of free radicals occurred in cells.

  • Isoflavones have received much attention because of their health-related and clinical benefits such as estrogenic and anti-oxidative activities as well as triggering of natural killer cell activity. However, there are few publications reporting the pharmacokinetic profiles together with physicochemical properties of main isoflavones. Therefore, the pharmacokinetic parameters of main aglycones, daidzein, glycitein and genistein after oral administration of soybean extracts were investigated and the physicochemical properties of soybean extracts were characterized. It was observed that angle of repose was $46^{\circ}$ and tap density, bulk density and porosity were 10.12, 4.3 and $0.86\;g/cm^3$ and the mean $AUC_{last}$ of daidzein, glycitein and genistein was $11.376\;{\mu}g{\cdot}h/mL$, $3.045\;{\mu}g{\cdot}h/mL$ and $0.825\;{\mu}g{\cdot}h/mL$, respectively. Cell viability was 60% at a concentration of 10 mg/mL. Taken together, it was suggested that isoflavones were contained in the soybean products and had an antioxidant activity and this study would be the basis to control the quality of soybean products and study of the bioequivalence between soybean products in future.

  • The use of underarm and body care cosmetics with oestrogenic chemical excipients (particularly the parabens) and the hypothesized association with breast cancer incidence, particularly in women. It is noted that the type of cosmetic product is irrelevant (e.g. antiperspirant/deodorant versus body lotion, moisturizers or sprays versus creams) and attention must focus on issues of actual exposure to chemicals through continued dermal application of body care products and the endocrine/hormonal activity and toxicity of the chemicals in the formulations. To evaluate the estrogenic activities of parabens such as ethylparaben, butylparaben, propylparaben, isobutylparaben and isopropylparaben, we used recombinant yeasts containing the human estrogen receptor [Saccharomyces cerevisiae ER+LYS 8127], human breast cancer MCF-7 cell lines and human estrogen receptor ${\alpha}\;and\;{\beta}$. In E-screen assays, isopropylparaben is the most estrogenic paraben, and in ER competition assay, isobutylparaben is the most estrogenic paraben. We evaluated isopropylparaben was most active in the recombinant yeast assay, followed by propylparaben, ethylparaben, isobutylparaben and butylparaben. Results from this study demonstrate that parabens are observed in human endocrine system. Therefore, we have shown that the parabens is induced the estrogenic activities similar to $17{\beta}$-estradiol and Bisphenol-A.
