In Vivo Suppression of Bisphenol A on Estradiol 2- and 4-Hydroxylase Activities in Hepatic Microsomal Fractions of Male and Female Sprague-Dawley Rats

  • This work was conducted to investigate the effect of bisphenol A (BPA) on estradiol (E2) 2-and 4-hydroxylase activities in the liver, kidney and lung tissues of male and female rats. After intraperitoneal administration of BPA to male and female rats for 4 days at 0, 10, and 50 mg/kg, the conversion of the substrate for hepatic and extra-hepatic enzyme activities was measured by GC/MSD. The result showed decreases of body and organ weights at 50 mg/kg BPA of male and female rats. Male hepatic E2 2-hydroxylase activity was inhibited by 68% at 10 mg/kg and by 82% at 50 mg/kg BPA. Female hepatic E2 2-hydroxylase activity was decreased by 46% at 10 mg/kg and by 56% at 50 mg/kg to the control. E2 4-hydroxylase was inhibited by 57 and 57% at 10 mg/kg and 54 and 78% at 50 mg/kg in liver of female and male, respectively. The urinary excretion rate of 2-hydroxyestradiol (2-OHE), androsterone and testosterone in urine of female rats with 50 mg/kg BPA were decreased significantly. The results showed that 50 mg/kg BPA was decreased E2 2-and 4-hydroxylase activities in liver, but not in other tissues. The urinary excretion rates of 2-OHE, androsterone and testosterone were also decreased. In liver, estrogenic enzyme activity were higher in male than female. These results suggest that BPA can disrupt estrogen metabolism by suppressing E2 2-and 4-hydroxylase activities in the liver of male and female rats.

Verification of Estrogenic Activity in Ethanol Extracts of Marine Organisms Using in vitro Test System. (In vitro 검출시스템을 이용한 해양생물 추출물로부터 에스트로겐 활성 검증)

  • In order to verify the occurrence of an estrogenic compound in natural products, the estrogenic activity was measured using an in vitro detection system. For this system, human breast cancer cell line MCF7 was transfected using an estrogen responsive CAT reporter plasmid. Estrogenic activities of photosynthetic algae spirulina and sea lettuce were evaluated using this system. Estrogenic activities of a $500\mug/ml\; and\; 50 \mug/ml$ ethanol extracts of spirulina were as much as that of $10^{-8}$M standard solution (17$\beta$-estradiol) and activity of $5\mug/ml$ ethanol extract of spirulina was as much as that of $10^{-10}$ M standard solution. However, no significant estrogenic activity was observed using sea lettuce extract. Estrogenic activities of marine animals, such as star fish and shrimp, were also evaluated using this system, however, no significant estrogenic activity was observed in these extracts. In this result, it is confirmed that spirulina extract possesses estrogenic compound.

Inhibitory Effect of Osteoclastogenesis and Estradiol Activity of Myelophycus simplex Extract (바위수염 추출물의 파골세포 분화 억제 및 에스트라디올 활성 평가)

  • In the present study, the estrogenic activity and anti-osteoclastogenic activity of the Myelophycus simplex extract were evaluated using T47D-Kbluc cells and bone marrow-derived macrophages (BMMs). As a result of the measurement of the estrogenic activity in the T47D-Kbluc cell line, the Myelophycus simplex extract showed increased estrogenic activity in a dose-dependent manner in association with its concentration. To confirm the regulatory effect of the Myelophycus simplex extract on the estrogen-responsive gene, the Myelophycus simplex extract showed a similar tendency to estradiol: the expression of estrogen receptor 1 (ESR1) was significantly decreased while the expression of estrogen receptor 2 (ESR2) was increased. Furthermore, the Myelophycus simplex extract exhibited an inhibitory effect on osteoclast differentiation. In conclusion, these Myelophycus simplex extracts might be regarded as candidates for further studies or the development of functional food products or medicine to prevent or avoid postmenopausal symptoms for women.

Electron Microscopic Observations of Oviductal Epithelium of the Rats Treated with Hormone (호르몬을 투여한 Rat 난관상피세포(卵管上皮細胞)의 전자현미경적 관찰)

  • Morphological changes of the oviductal epithelium of the rat treated with hormones ($17{\beta}$-estradiol ${\mu}g$/day and progesterone 2.5mg/day) for ten days were observed transmission and scanning electron microscopically. The results obtained were as follows: 1. The cilia formation of ciliary cell(CC) was more accelerated by the treatment of estradiol than progesterone, but the balance of estrogen and progesterone was required for the maintenance of CC. The effect of hormone was different between the segments for the maintenance of CC. 2. The short secretory cell(SSC) was severely inhibited in the formation of secretory granules with single hormonal treatment but the activity of secretion was more inhibited by progesterone than by estradiol. 3. The long secretory cell(LSC) had not a great difference between estradiol and progesterone treatments as compared with the normal sexual cycle, but the formation of secretory granules was somewhat accelerated by progesterone treatment. 4. The formation of secretory granules of junctional cell (JC) was severely accelerated by estradiol treatment as compared with the normal sexual cycle. The formation of secretory granules during progesterone treatment, on the other hand, was inhibited completely, but the numbers of pinocytotic vesicles appeared at the cytoplasmic apical portion. 5. Three types of secretory cells, SSC, LSC and JC, on the rat oviductal epithelium could be suggested to have different cell tapes respectively from the morphological changes by hormone treatment.

Studies on Immunity and Sex Hormone Activity of Bis(tributyltin) oxide (Bis(tributyltin) oxide가 랫드의 면역 및 성호르몬 활성에 미치는 영향)

  • The purpose of this study finds out the effect of red ginseng extract(1.0 g/kg) on bis(tributyltin)oxide(TBTO)(10, 20 and 40 mg/kg) which poisons against some organs like thyroid gland, liver, kidney, testis, ovary. Serum immunity and sex hormone activity of rats are examined by gastric tubing for 3 weeks. The weights of each in treated group were increased, especially liver in females and those of testis in males were signignificantly increased at 10, 20 and 40 mg/kg (P<0.05, P<0.01). In group treated with TBTO(10, 20 and 40 mg/kg) for females, the serum IgG level was significantly reduced in comparison with control group(P<0.05). The group of females which were poisonned by TBTO and rGe was significantly recovered in TBTO 10 mg/kg + rGe 1.0 g/kg and TBTO 20 mg/kg + rGe 1.0 g/kg. In case of sex hormone activity of each sex, the estradiol activity of females and testosterone activity of males were signignificantly decreased rather than the control group. And control group by bis(tributyltin) oxide and red ginseng extract is just increased estiradiol activity.

Influences of Hydrocortisone, DHEA, Estradiol and Testosterone on the Hepatic and Intestinal Polyamine Metabolism of Castrated Mice (Hydrocortisone, DHEA, Estradiol 및 Testosterone에 의하여 나타나는 마우스-간 및 소장 Polyamine 대사의 변동에 관한 연구)

  • Hydrocortisone 50 mg/kg (HC), dehydroepiandrosterone 250 mg/kg (DHEA), ${\beta}-estradiol$ 5 mg/kg (E2), and testosterone 20 mg/kg (TS) were subcutaneously injected into the castrated ICR mice at noon for four days, and the animals were sacrificed at 10-12 A.M. of the fifth day. The intestinal DAO activity was significantly decreased by HC, but it was rather increased by E2 and TS, respectively. And DHEA did not change the DAO activity. But the hepatic MAO activity was not affected by anyone of HC, DHEA, E2, and TS. Aminoguanidine 25 mg/kg produced the marked decrease of the intestinal DAO activity and the significant increases of the intestinal PT and SD contents, but it did not change the hepatic polyamine contents. HC and DHEA induced the significant increase of the intestinal PT content. E2 induced the marked increase of the hepatic PT content and the moderate increase of the intestinal PT content. TS little affected the polyamine contents of the liver and intestine. These results suggest that the E2-induced increase of the hepatic PT content is rather ascribed to the greater enhancement of PT synthesis than the inhibition of polyamine catabolism, and that the HC-induced increase of the intestinal PT content is due partly to the inhibition of polyamine catabolism via DAO.

Effects of Progesterone and 17β-Estradiol under Presence or Absence of FBS on Plasminogen Activators Activity in Porcine Uterine Epithelial Cells

  • The present study was conducted to investigate the regulatory mechanism of plasminogen activators (PAs) activation by $17{\beta}$-estradiol ($E_2$) and progesterone ($P_4$) in porcine uterine epithelial cells (pUECs). pUECs were collected from porcine uterine horn and cultured at 80% confluence. Then, 0.1% (v/v) DMSO, 20 ng/mL $E_2$, and $P_4$ with or without fetal bovine serum (FBS) treated to cultured cells for 24 hours. The supernatants were used for measurement of PAs activity and expression of urokinase-type PA (uPA), tissue-type PA (tPA), uPA specific receptor (uPAR), and type-1 PA inhibitor (PAI-1) mRNA were analyzed by real-time PCR. The expression of PAs-related genes was not affect by steroid hormones in both of serum treatment groups. However, PAs activity was increased by treatment of $E_2$ compared to 0.1% DMSO treatment in serum-free group (p<0.05). Then, $E_2$ and $P_4$ were diluted with 0.002% (v/v) DMSO for reduction of its effect and treated to cultured cells without FBS. Only tPA mRNA was significantly increased by $E_2$ treatment (p<0.05). PAs activity was enhanced in $E_2$ treated group compared to control groups (p<0.05). These results indicate that serum-free condition is more proper to evaluate effect of steroid hormones and activation of PAs in pUECs was mainly regulated by estrogen. These regulation of PAs activation may be associated with uterine remodeling during pre-ovulatory phase in pigs, however, further studies are needed to investigate precise regulatory mechanism.

Gene Expression Profiles of Dibutyl Phthalate and 17$\beta$-Estradiol using cDNA microarray in MCF 7 Human Breast Cancer Cell Line

  • Phthalates, suspected endocrine disruptor, are plasticizer and solvent used in industry, and some phthalates are known as potential carcinogen. Most common human exposure to this compounds may occur with contaminated food. It may migrate into food from plastic wrap or may enter food from general environmental contamination, and it has become widespread environmental pollutants, thus leading to a variety of phthalates that possibly threaten the public health. Dibutyl phthalate (DBP) may playa part of cell proliferator, which mediates changes in gene expression and the metabolism of xenobiotics. An understanding of the role of DBP in modulating gene regulation should provide insight regarding mechanisms of DBP induced xenoestrogenic impact. To elucidate the type of genes that are associated with estrogenic activity induced by DBP at the dose (10$^{-8}$ M) appeared proliferating effects, the pattern of gene expression in MCF7 cells was compared between 17$\beta$-estradiol and DBP exposure in the cDNA microarray. From the results, it showed some differences of gene expression patterns between MCF7 cells treated with 17$\beta$-estradiol and DBP, and also DBP shows estrogenic potential with changes in estrogen-related gene expression levels.

The Estrogenic Effects of Phthalates(DEHP, DBP) in Yeast Recombinant Assay (효모재조합 검색시험법을 이용한 DEHP, DBP의 에스트로젠 효과)

  • Di(2-ethylhexyl) phthalate (DEHP) and di-n-butyl phthalate (DBP) were screened for estrogenic activity using a recombinant yeast screening system consisted with estrogen receptors and ${\beta}-galactosidase$ as reporter gene. The chemicals showed estrogenic activity in ranges of $1{\times}10^{-10}\;to\;1{\times}10^{-7}M(DEHP)\;and\;of\;1{\times}10^{-9}M\;to\;1{\times}10^{-6}M(DBP)$ respectively. $17{\beta}-estradiol$, as a positive control of, showed maximal activity at $1{\times}10^{-9}M$. The concentration of half-maximal estrogenic activity was $1{\times}10^{-9}M$ for both chemicals. However, the concentration of maximal estrogenic activity was $1{\times}10^{-7}M$ for DEHP and $1{\times}10^{-8}M$ M for DBP. These results suggested that DBP was higher in relative potencies and more sensitive than DEHP. In conclusion, DEHP and DBP were both estrogenic, even though DBP was more reactive to estrogen receptor.

Estrogen Modulation of Human Breast Cancer Cell Growth

  • To gain further insight into how estrogens modulate cell function, the effects of estrogen on cell proliferation were studied inhuman breast cancer cells. We examined the effects of estrogen on the proliferation of three human breast cancer cell lines that differed in their estrogen receptor contents. Ten nM estradiol markedly stimulated the proliferation of MCF-7 human breast cancer cells that contained high levels of estrogen receptor $1.15{\pm}0.03 pmole/mg protein)$(over that of control. In T47D cells that contained low levels of estrogen receptor $0.23{\pm}0.05 pmole/mg protein)$, Ten nM estrogen slightly stimulated the proliferation over that of control. MDA-MB-231 cells, that contained no detectable levels of estrogen receptors, had their growth unaffected by estrogen. These results showed their sensitivity to growth stimulation by estrogen correlated well with their estrogen receptor content. Also we examined the effect of estrogen on cellular progesterone receptor level as well as plasminogen activator activity in MCF-7 cells. Ten nM estradiol showed maximal stimulation of progesterone receptor level as well as plasminogen activator activity in MCF-7 cells. It is not clear whether these stimulations of progesterone receptor and plasminogen activator activity by estrogen are related to the estrogen stimulation of cell proliferation of MCF-7 cells. Studies with estrogen in human breast cancer cells in culture indicate that sensitivity to growth stimulation by estrogen correlates well with estrogen receptor contents.
