Study on the Erythrocyte Sedimentation Rate by Angled Tube Method during Pregnant Period in Holstein (Holstein의 임신기간별 경사시험관법에 의한 적혈구 침강율 연구)

  • The erythrocyte sedimentation rate (ESR) test could be applied well in clinic of men, horses or dogs. But the ESR test was not applied to clinic ruminants except for buffaloes since it was too low measure. To overcome this problem angled tube method was developed. This method was found to be useful for making diagnosis and prognosis. The results were summarized as follows. 1. In the early pregnancy condition, corrected ESR did not changed when compared with normal valures. 2. In the middle pregnancy condition, corrected ESR was increased when compeared with normal valures. 3. In the latter pregnancy condition, corrected ESR was increased when compeared with normal valures. 4. After parturition condition, corrected ESR did not changed when compeared with normal valures.

  • Young mae rats were fed fro 4 weeks on different experimental diets containing 8%(w/w) fat ; sardine oil, soybean oil and lard. In the sarine oil group, total cholesterol and triglyceride concentrations inplasma were significantly lower than in the soybean oil and lard group. The proportion of $\omega$-3 polyunsaturated fatty acids in erythrocyte membrane phospholipids were significantly higher in the sarine oil-fed animals. by sardine oil ingestion, lipid peroxide levels inthe plasma and liver were elevated while $\alpha$tocopherol concentrations were lowered, compared to the other groups. However, erythrocyte osmotic fragility was unaffected by the different fat diets.

Detection and Property of Leptospiral Hemolysin (렙토스피라용혈소의 검색과 성질)

  • To detect lepotospiral strains which produce hemolysin and determine the optimal condition for assaying hemolysin, we screened reference strains and observed some property of hemolysin. Hemolysin activity was assayed with cell free culture liquids and erythrocyte suspension. The production of hemolysin by local strains isolated in Korea was assayed and compared with that of reference strains. The hemolysin was produced by 18 strains among 38 reference strains and 3 local strains isolated in Korea. The production of hemolysin began with growth of Leptospira cultured in EMJH medium and reached maximum at stationary phase. The optimum temperature for hemolytic activity was $37^{\circ}C$. At lower temperature the activity of hemolysin was decreased progressively. The hemolytic activity was completely inactivated after :30 minutes' exposure at $56^{\circ}C$. Hemolysis pattern was "hot-cold type" which showed increased hemolysis after cold incubation. The hemolysin was most active on sheep erythrocyte and less active on ox, goat, human and guinea pig erythrocyte with the decreasing order.

Observations on the Cholinesterase Activity in Normal Blood of Korean Male Cattle (한국모우(韓國牡牛)의 Cholinesterase 활성도(活性度)에 대(對)하여)

  • The serum and erythrocyte cholinesterase activity and their seasonal variations were determined with 8 males of healthy Korean cattles during one year period from September, 1966 to august, 1977. The results obtained were as follows : 1. The serum cholinestrase activity showed a range of 0.64mg/hr/ml to 8.26mg/hr/ml with a mean value of $3.69{\pm}0.25mg/hr/ml$. There was highly significant differences in the seasonal variations. 2. The erythrocyte cholinesterase activity ranged from 29.78mg/hr/ml to 48.88mg/hr/ml with a mean value of $38.37{\pm}1.11mg/hr/ml$. Seasonal variations was not found to be significant. 3. The results showed that the serum cholinesterase activity of Korean cattle had great variations among individual values and significant seasonal variations. On the other hand, cholinesterase activity of erythrocyte showed a little variations among individual values and no significant seasonal variations.

  • The use of erythrocyte as drug carrier has been reviewed, Carrier erythrocytes have proven to offer many advantages for delivery of therapeutic agents, especially in the treatment of inherited enzyme deficiency and cancer. Carrier erythrocytes are biodegradable and nonimmunogenic. Encapsulated drugs may be protected from premature degradation, inactivation and excretion. Carrier erythrocytes may be used as a slow-release system. Targeting of encapsulated drugs directly to a site of action is another possibility. Methods for encapsulating drugs into erythrocytes, the fate of carrier erythrocytes in vivo, the strategies of targeting carrier erythrocytes to special organs and in vivo applications of erythrocytes have been discussed. The encapsulation of drugs in erythrocytes has shown attractive possibilites in future use.

  • Recently, nonthermal bioplasma has been attracted by researchers due to their potentials to modulate cellular functions resulting in changes of biomolecular electron band structures as well as cell morphologies. We have investigated the secondary electron emission characteristics from the surface of the erythrocyte, i.e., red blood cell (RBC) with and without the nonthermal bioplasma treatment in morphological and biomolecular aspects. The morphologies have been controlled by osmotic pressure and biomolecular structures were changed by well known reactive oxygen species. Ion-induced secondary electron emission coefficient have been measured by using gamma-focused ion beam (${\gamma}$-FIB) system, based on the quantum mechanical Auger neutralization theory. Our result suggests that the nonthermal bioplasma treatment on biological cells could result in change of the secondary electron emission coefficient characterizing the biomolecular valence band electron energy structures caused by the cell morphologies as well as its surface charge distributions.

Effects of Calcium Channel Blockers on Human Erythrocyte Ghost Membranes

  • The effects of calcium channel blockers (CAB's) verapamil, diltiazem and nicardipine, on erythrocyte ghost membranes have been studied. Using the fluorospectroscopic method, it was observed that the fluidity of the inner layer of ghost membranes was increased with an increase of drug concentrations but did not any changes in the fluidity of the outer layer. These drugs showed protectuve effect against hypotonic hemolysis of erythrocytes. Thus, the expansion of surface area in response to corpuscular volume of erythrocytes in the presence of CAB's is seemed to play an important role in protecting hypotonic hemolysis of erythrocytes.

  • In order to evaluate the effectiveness of KOK(kyungokko) experimentally used for longevity hematology, erythrocyte hemolysis and TBA(thiobarbituric acid) were measured, after LKOK(low concentration KOK) and HKOK(high concentration KOK) were administrated to mice pretreated with AAPH (2,2'-Azobis(2-amidinopropane), hydrocholoride, free radical inducer. In hematology WBC and hemoglobin were reduced significantly, while the number of platelet was significantly incresed as compared with control. Erythrocyte hemolysis by damage of free radical on cell membrane of RBC and TBA(Thiobarbituric acid) were significantly inhibited in both sample groups as compared with control. These results suggest KOK had antioxidant effect.

A Study on the Absorption Mechanism of Drugs through Biomembranes

  • The effect of lipophilicity on the mechanisms of drug absorption through biomembranes was investigated empolying HPLC system and the fluorescence technique. Human erythrocyte ghost membranes were used as a model biomembrane. A series of four parabens (methyl, ethyl, rpopyl, and butyl) and p-hydroxybenzoic acid were used as the model drugs for lipophilicities and their partition coefficients were measured in Sorensen's phosphate buffer solution (pH 5)/octanol system. Absorption amount of parabens through erythrocyte ghost membranes increased with an increase of lipophilicity resulted from the addition of methylene group to the n-alkyl chain of parabens. And the effect of parabens on the fluidity of ghost membrane also increased with an increase of their lipophilicities.

  • To assess the clastogenic effects of the diglyceride preparation containing conjugated linoleic acid (DG+CLA) in vivo micronucleus test was performed using ICR mice. Each of the groups consisted of three doses of DG+CLA (500, 1,000 and 2,000 mg/kg, p.o.), Mitomycin C (positive control, 2 mg/kg, i.p.) and negative control (olive oil, 10 mL/kg, p.o.). A slide preparation was made at 24 hours after 1st treatment with DG+CLA. As a result of counting the icronucleated polychromatic erythrocyte (MNPCE) of 2,000 polychromatic erythrocyte (PCE), the number of aberrant cells was not increased in any of the three doses of DG+CLA orally administered. There was no clinical sign connected with administration of DG+CLA. These results indicate that DG+CLA is not capable of inducing micronuclei in vivo mice cells and thus has no genotoxicity in micronucleus.
