Quality Characteristics of Tofu Added with Loquat (Eriobotrya japonica Lindl.) Leaf Powder (비파잎 분말을 첨가한 두부의 품질특성)

  • This study was conducted to examine the quality characteristics of tofu prepared with Loquat (Eriobotrya japonica Lindl.) leaf powder (LLP). Moisture, crude ash, carbohydrate, crude protein, and crude lipid contents of Loquat leaf powder were 10.25, 6.72, 69.54, 8.23, and 5.26%, respectively. The yield rates, turbidity, and total acidity of tofu increased according to the quantity of added LLP, whereas pH level decreased. The L- and a-values of samples decreased as the amount of LLP increased, whereas b-value increased. With regard to textural characteristics, LLP addition increased hardness, chewiness, and brittleness, as well as reduced springiness and cohesiveness. In terms of overall acceptability, the most preferred tofu samples were the control and 0.3% LLP addition groups.

  • This study compares the major nutritional components of the leaf, flesh, and seed of dry loquat (Eriobotrya japonicaLindl.). Among proximate compositions, the crude fat, crude ash, and dietary fiber of the leaf exceeded those of the seed and flesh, whereas the carbohydrate content of the leaf was lower than that of the seed and flesh. The main component of free sugars in the leaf, flesh, and seed was fructose. Total amino acids of the leaf, flesh, and seed were 552.43, 63.00, and 260.29 mg%, respectively. Although the amino acid composition of the leaf, flesh, and seed varied, glutamic acid and ${\gamma}$-aminobutyric acid were the major amino acids in the leaf, flesh, and seed. Major fatty acids of total lipid were oleic acid and stearic acid in the leaf and seed, and the major acid was linoleic acid in the flesh. Major organic acids were oxalic acid in the leaf, maleic acid in the flesh, and citric acid in the seed. Vitamin C content was higher in the seed than in the leaf and flesh.

Effects of Loquat(Eriobotrya japonica Lindl.) Extracts in Different Aerial Components on Differentiation of 3T3-L1 Cells and Pig Preadipocytes (비파 부위별 추출물이 3T3-L1 세포와 돼지 지방전구세포의 분화에 미치는 효과)

  • The current study was undertaken to determine the effects of the ethanol extracts of loquat (Eriobotrya japonica Lindl.) seeds, flesh or leaves on the differentiation of 3T3-L1 cells and male pig preadipocytes. The cell number was measured with the MTT assay after trypsin digestion. The cell differentiation was determined by measuring the glycerol-3-phosphate dehydrogenase (GPDH) activity and triglyceride(TG) content. No cytotoxicity was observed from the loquat flesh and leaf ethanol extracts at concentrations of 5, 10, 25, 50, 100 or $200{\mu}g/mL$ in 3T3-L1 cells and pig preadipocytes. However, the cell viability of neither cell line were affected by up $50{\mu}g/mL$ of loquat seed ethanol extract. Treatment with the loquat seed and leaf ethanol extracts significantly suppressed the terminal differentiation of both cell lines in a dose-dependent manner, as confirmed by the decrease in the glycerol-3-phosphate dehydrogenase(GPDH) activity and TG content. Treatment with the loquat seed and leaf ethanol extracts inhibited the GPDH activity and reduced the TG content of both cell types more effectively than that with the loquat flesh ethanol extract. The most potent anti-adipogenic effect was obtained in the case of the ethanol extract of loquat seeds.

  • This study was conducted to investigate the effects of loquat (Eriobotrya japonica Lindl.) leaf powder (LLP) on quality characteristics of dumpling shells. Dumpling shell samples were prepared with wheat flour containing different amounts of LLP, and their characteristics were then investigated. Amylography revealed that the gelatinization temperature of the LLP-wheat flours composite increased, while the initial viscosity at $95^{\circ}C$, viscosity at $95^{\circ}C$ after 15 minutes, and maximum viscosity, all decreased with increasing LLP content. As increasing amounts of LLP were added, the L and a values were reduced, whereas the b value was increased. Moreover, the addition of LLP increased hardness, cohesiveness, and adhesiveness, while it reduced springiness. Overall, sensory evaluation revealed that dumpling shells with 5% LLP were preferred more than other samples.

  • The purpose of this study is to investigate the quality characteristics of cookies prepared with Loquat (Eriobotrya japonica Lindl.) leaf powder (0, 1, 2, 3, and 4%) for flour. The pH of cookie dough decreased significantly in response to the addition of all levels of Loquat leaf powder (LLP). The spread factor of the cookies also increased significantly with more LLP content added to the cookies recipe. In addition, Hunter's color L and a values significantly decreased with increasing LLP content, whereas the b value was increased. According to hardness measurements, the substitution of 1~4% for LLP resulted in increased hardness when samples were compared to the control. Moreover, the addition of LLP resulted in increased DPPH free radical scavenging activity when compared to the control. The results of sensory evaluation showed that the cookies containing 2% had the highest scores.

Validation of analytical method and antioxidant properties of Eriobotrya japonica Lindl. Leaf extract according to extraction solvent (추출용매 조건에 따른 비파 잎 추출물의 항산화 활성 및 유효성분의 분석법 밸리데이션)

  • The antioxidant properties of Eriobotrya japonica leaf extract were investigated using DPPH and ABTS radical scavenging assay. The 80% ethanol extract of leaves ($IC_{50}$ values for DPPH and ABTS were 13.9 and $10.9{\mu}g/mL$, respectively) and young leaves ($IC_{50}$ values for DPPH and ABTS were 20.7 and $17.3{\mu}g/mL$, respectively) showed high radical scavenging activity. Additionally, the quantitative method for estimation of ellagic acid and chlorogenic acid from E. japonica leaves was optimized by HPLC/DAD. This method showed high linearity of the calibration curve with a coefficient of correlation ($R^2$) equal to 0.999. The LOD values for ellagic acid and chlorogenic acid were 2.35 and $0.73{\mu}g/mL$, respectively, whereas LOQ values were 7.13 and $2.22{\mu}g/mL$, respectively. Recovery of the two compounds was 99.7-108.0% with RSD values less than 5.31%. These results suggest that 80% ethanol extract of E. japonica leaves could serve as a potential source of natural antioxidant for us in various industrial applications.

  • This study was carried out to investigate the physicochemical compositions and antioxidative effects of Eriobotrya japonoca Lindl (Loquat). The proximate compositions of the loquat leaf on a dry matter basis were 8.78% moisture content, 6.74% crude protein, 7.87% crude fat, 6.99% crude ash, 43.61% dietary fiber and 26.01% carbohydrate. In analysis of free amino acids, 16 kinds total amino acid components, 17 kinds of components were isolated from loquat. The essential amino acids contained in loquat leaf accounted for 50.15% of total amino acids, while the non-essential amino acids accounted for 49.85%. In analysis of total fatty acids, only 5 kinds of acid were detected: lauric acid, myristic acid, pentadecanoic acid, stearic acid and oleic acid. Antioxidative and Antimicrobial Activities of Eriobotrya japonica Lindl. Leaf Extracts (비파 잎 추출물의 항산화 및 항균활성)

  • Antioxidative, antimicrobial activities and Raw 264.7 cell viability as cytotoxicity of various solvent extracts from leaf of Eriobotrya japonica Lindl. dried by different methods were investigated for processing as functional ingredient. In DPPH radical scavenging activity, RLE (80% EtOH extract of raw leaf) and FLE (80% EtOH extract of freeze-dried leaf) exhibited strong scavenging effect on $300{\mu}M$ DPPH radical solution (1.71 mg/mL and 2.11 mg/mL for RLE $SC_{50}$ and FLE $SC_{50}$). Also in nitric oxide scavenging activity, RLE and FLE showed strong activities (83.9% and 82.2% in 5 mg/mL sample concentration). Total phenolic compound contents of each extracts were found to be $73.7{\sim}215.4$ mg/g and RLE was showed the highest phenolic compound content. Also, total flavonoid contents were found to be $24.85{\sim}110.3$ mg/g and RLE was showed the highest flavonoid content. In antimicrobial activity, RLE was showed higher growth inhibition effect against all microbial strains. RLE, RLW (hot water extract of raw leaf), and FLW (hot water extract of freeze-dried leaf) exhibited strong antimicrobial activities against MRSA and S. aureus. In measurement of cytotoxicity by MTT assay, Raw 264.7 cell viabilities of 80% EtOH extracts showed better effect than water extracts. Especially viability of RLE was found be over 100% in every tested sample concentration.

Effects of Loquat (Eriobotrya japonica Lindl.) Ethanol Extracts of Different Aerial Parts on Antioxidant Activity and Antiproliferation of Human Cancer Cells (비파 부위별 에탄올 추출물의 항산화 활성 및 암세포 증식 억제효과)

  • The current study was carried out to determine the effects of the seed, flesh (seedless fruit), and leaf of loquat (Eriobotrya japonica Lindle.) on antioxidative activity and anti-proliferation in human cancer cells. Total polyphenol contents of loquat seed, flesh, and leaf ethanol extracts were found to be 17.77, 32.32, and 28.08 mg/g, respectively. Also, total flavonoid contents of loquat seed, flesh, and leaf ethanol extracts were found to be 18.77, 28.73, and 21.35 mg/g, respectively. The $IC_{50}$ values of DPPH hydroxyl scavenging of loquat seed, flesh, and leaf ethanol extracts were 0.049, 0.063, and 0.042 mg/mL, respectively. Antioxidative indexes of loquat leaf and seed ethanol extracts was highly and it was similar to the BHA and BHT. The antioxidative activities in loquat seed and leaf ethanol extracts were higher in loquat flesh. The antiproliferation effect of loquat seed and leaf ethanol extracts on liver cancer cell line (H460), stomach cancer cell line (AGS) and lung cancer cell line (A549) showed higher values compared with the flesh ethanol extracts. These results indicate that loquat ethanol extracts may play a positive role in antioxidative properties and cancer prevention.

Antimicrobial and Antioxidant Activities of Various Solvent Extract from Different Parts of Loquat (Eriobotrya japonica, Lindl.) (비파 부위별 용매추출물의 항균 및 항산화 활성)

  • Antimicrobial and antioxidative activities of various solvent extracts from different parts of the loquat were investigated to process it as the functional food. The extraction yield showed higher in butanol fraction of 4.95% in peel and 4.42% in seed than others, but water fraction showed high extraction yield of 3.89% in leafs 23.6% in the seedless fruit and 21.1% in fresh, In the antimicrobial activity test, ethyl acetate fraction that leaf and fruit excluded seed inhibited higher 19, 15 mm and 16, 15 m in clear zone far Staphylococcus aureus and Escherichia coli in each. Ethyl acetate fraction that extracted from a series of polar and nonpolar solvent fractionation of methanol extracts showed better results in the hydrogen donating activity of 82% in leaf, 74% in seedless fruit, 68% in seed, 52% in peel and 30% in fresh.
