The Inhibitory Effect of Ferulic Acid and Related Phenolic Compounds against Cancer Cell Lines (Ferulic Acid와 관련 페놀화합물의 암세포주에 대한 독성억제효과)

  • The inhibitory effect of ferulic acid and related phenolic compounds on the growth of normal cell lines and can­cer cell line was evaluated by the MTT and XTT methods. Ferulic acid decreased the cell viability of human skin melanoma cells by the MTT method and the cell adhesion activity of human oral epithelioid carcinoma cells by the XTT method. These results suggest that ferulic acid has a potential anticancer activity.

  • A 14-year-old female Gill's bottle-nosed dolphin died after having 1 month history of depression, anorexia, and respiratory insufficiency. At necropsy, numerous well-demarcated tan firm nodules in variable sizes ranging from 2 m to 3 cm in diameter were scattered throughout the lung lobes. Histologically, the pulmonary nodules formed typical granulomatous inflammation. Center of the granulomatous foci consisted of necrotic center with gram positive cocci surrounded by eosinophilic Splendore-Hoeppli material then by macrophages, epithelioid cells, multinucleated giant cells, a few lymphocytes and fibroblasts. The is the first report of pulmonary botryomycosis in dolphin in Korea.

Comparative studies of histopathologic pulmonary lesion of cattle and deer tuberculosis by Mycobacterium bovis (Mycobacterium bovis에 의한 소 및 사슴 결핵의 폐 병리조직학적 소견 비교연구)

  • Comparative studies of histopathologic lesions from 23 purified protein derivative (PPD) positive cattle, 21 slaughter cattle found with tuberculosis, and 11 tuberculosis-positive elk (Cervus elaphus) were performed. PPD positive cattle did not show specific histopathologic lesions in all 23 heads that were no visible lesion reactor. Slaughter cattle found with tuberculosis revealed microscopically classical granulomatous lesion (tubercle) with central caseous necrosis surrounded by mantle of epithelioid cells and Langhan's giant cells capsuled by connective tissue in lung. Elk was noted with some different lesion patterns with classical granulomatous lesion and suppurative abscesses that was composed of fibrin, degenerated cells without having connective tissue. In addition, many Langhan's giant cells infiltration in alveoli at peripheral lesion were observed in some cases of classical granulomatous lesion and suppurative abscesses.

  • A stable cell line, BSBS (black seabream spleen), was established from the cells in spleen of black seabream, Acanthopagrus schlegeli, and characterized. Subculture maintained more than 60 passages and mophologically, BSBS cell was epithelioid cell. The cells grew optimally at 20℃ in Leibovitz's L-15 medium supplemented with 10% fetal bovine serum with incubation temperature of 20℃. BSBS cells supported the growth of marine birnavirus (MABV Y-6), chum salmon reovirus (CSV), spring viremia of carp virus (SVCV) and hirame rhabdovirus (HIRRV). Thus, the new cell line may be useful for studying wide range of fish viruses.

Bovine tuberculosis found at slaughtered Korean indigenous cattles (도축 한우에서 발견된 결핵병)

  • There were eight Korean indigenous cattles affected with bovine tuberculosis (BTB) detected by inspectors at slaughterhouses located in Chungbuk province from May 2006 through July 2007. Postmortem finding of BTB cases was characterized by the presence of several caseous or calcified nodules encapsulated by connective tissue from the pleural/peritoneal surface, livers, lungs and regional lymph nodes On micro-scopic examinations, the characteristic lesion of BTB was the formation granulomatous nodules, which contains central calcified necrotic zone surrounded by epithelioid cells, macrophages and a few Langhans' type giant cells. In addition, mononuclear cells and fibroblasts were also infiltrated. At the periphery, encapsulation was formed that protect the neighboring healthy tissues.

  • A pulmonary epithelioid hemangioendothelioma (PEH) is a rare tumor with a vascular origin. A PEH can arise in many organ systems, such as the lung, liver, bone and soft tissues. It is a borderline malignancy but the clinical course is usually benign. In this report, we describe three cases of PEH. Case 1, a 61-year-old man, had nonspecific chest discomfort and the chest X-ray showed a solitary lung nodule. This nodule was diagnosed by an open lung biopsy. The pathologic findings including abundant necrosis, mitosis and hyperchromatic and pleomorphic nuclei, indicated a malignancy. The electron microscopic study showed Weibel-Palade bodies and the immunohistochemical stain for CD31 showed a positive reaction in the tumor cells, and linear staining along the vascular lumina. Case 2, a 34-year-old man, was admitted for an evaluation of multiple small nodules, incidentally detected a screening chest X-ray. The nodules were diagnosed by a immunohistochemical stain for the factor VIII-related antigen. Case 3, a 34-year-old woman, complained of left pleuritic chest pain. A simple chest film and the chest CT scan revealed multiple bilateral nodules and a left pleural effusion. An immunohistochemical stain for the factor VIII-related antigen was used to diagnose the nodules. Forth-one months after the diagnosis, she died of pulmonary insufficiency.

A Case of Pulmonary Epithelioid Hemangioendothelioma (폐의 유상피성 혈관내피종 1예)

  • Epithelioid hemangioendothelioma(EH) is a rare pulmonary vascular malignancy. Clinically, EH has been considered as an indolent, generally non-aggressive tumor. We report a case of EH which was confirmed by open lung biopsy. A 34-year-old woman was admitted for further evaluation of multiple small(less than 2cm in size) nodules, incidentally detected on screening chest radiograph. The chest CT showed multiple, relatively well-marginated, variable sized nodules at both whole lung. Transbronchial lung biopsy and transthoracic needle aspiration were nondiagnostic and open lung biopsy was performed from right middle lobe of lung. On light-microscopic examination, the nodules were composed of a poorly cellular hyaline core and a more cellular peripheral zone which extended into air space in a micropolypoid fashion and obliterated blood vessels. The tumor cells at the peripheral zone had intracytoplasmic vacuoles which suggested primitive, vascular differentiation. Immuno-histochemical study revealed the cellular area which gave positive reaction to factor VIII-related antigen. She received no specific therapy after open lung biopsy and chest X-ray films had showed no change for about two years.

  • Renal angiomyolipomas (AMLs) are typically solid tumors, but there have been few reports of a rare cystic variant of AML. AML with epithelial cysts, where the epithelial cyst has a cuboidal epithelial lining, account for the majority of them. Next, epithelioid AML (EAML) with cystic changes due to hemorrhage and necrosis, which is composed of epithelioid cells with abundant eosinophilic cytoplasm, have also been reported. These rare cystic types of AML can be mistaken for other cystic tumors, such as cystic renal cell carcinoma, in preoperative imaging. We report the imaging findings of a rare case of EAML with epithelial cysts.

Hydrogen Peroxide Promotes Epithelial to Mesenchymal Transition and Stemness in Human Malignant Mesothelioma Cells

  • Reactive oxygen species (ROS) are known to promote mesothelial carcinogenesis that is closely associated with asbestos fibers and inflammation. Epithelial to mesenchymal cell transition (EMT) is an important process involved in the progression of tumors, providing cancer cells with aggressiveness. The present study was performed to determine if EMT is induced by $H_2O_2$ in human malignant mesothelioma (HMM) cells. Cultured HMM cells were treated with $H_2O_2$, followed by measuring expression levels of EMT-related genes and proteins. Immunohistochemically, TWIST1 expression was confined to sarcomatous cells in HMM tissues, but not in epithelioid cells. Treatment of HMM cells with $H_2O_2$ promoted EMT, as indicated by increased expression levels of vimentin, SLUG and TWIST1, and decreased E-cadherin expression. Expression of stemness genes such as OCT4, SOX2 and NANOG was also significantly increased by treatment of HMM cells with $H_2O_2$. Alteration of these genes was mediated via activation of hypoxia inducible factor 1 alpha (HIF-$1{\alpha}$) and transforming growth factor beta 1 (TGF-${\beta}1$). Considering that treatment with $H_2O_2$ results in excess ROS, the present study suggests that oxidative stress may play a critical role in HMM carcinogenesis by promoting EMT processes and enhancing the expression of stemness genes.

Angiolymphoid Hyperplasia with Eosinophilia (ALHE) of the Oral Cavity: a Case Report (구강내에 발생한 혈관림프증식증: 증례보고)

  • Angiolymphoid hyperplasia with eosinophilia (ALHE) is a rare benign vascular lesion that is characterized by proliferation of small to medium-sized vascular structures lined by histiocytoid or epithelioid endothelial cells and often accompanied by an inflammatory infiltrate comprising lymphocytes, plasma cells and eosinophils. A 34-year-old man without any generalized systemic conditions presented with a slowly enlarging painless swelling in the buccal area. An excisional biopsy was conducted and the diagnosis of Angiolymphoid hyperplasia with eosinophilia was confirmed. We report a case of angiolymphoid hyperplasia with eosinophilia of the buccal area in oral cavity and review the previously reported cases and literatures of angiolymphoid hyperplasia with eosinophilia.
