  • The seasonal variation of two flavonol glycosides, icariin and epimedoside A, in the aerial parts and underground parts of Epimedium koreanum from June through September was investigated. The icariin concentration was decreased with time in both parts. Epimedoside A concentration was fluctuated, being highest in June and lowest in July in the underground parts. In the aerial parts, however, it was almost same in concentration. Determinations were made of the occurrence of two new flavonol glycosides, 2‘-O-rbamnosyl ikarisoside A and 2’-O-rhamnosyl icarisid II in the aerial parts and their changes in concentration were similar to those in the underground parts, being highest in July in both parts.

  • Effects of Lycium chinensis, Epimedium koreanum and tuguaconitine which is isolated from Aconitum sibiricum on primary culture chicken embryonic brain cells were studied by microscopic observation and determined of the activity of pyruvate dehydrogenase complex(PDHC). Brain cells were prepared from the brain of 10-day-old chicken embryo and cultured with a medicine consisted of 90% Dulbecco's Modified Eagle Medium(DMEM) and 10% horse serum. It was observed that all substances studied seemed to show the tendency to stimulate the neurite outgrowth of brain cells which were cultured with a deficient medium under microscopic observation. The activity of PDHC in brain cells cultured with a deficient medium was increased by Lysium chinensis and Epimedium koreanum. However, tuguaconitine had not influence on the activity of PDHC.

  • Icariside II, a flavonol glycoside, was isolated from the aerial part of Epimedium Koreanum Nakai by the anti-hepatotoxic acitivity guided fractionation technique employing $CCl_4-in-toxicated$ primary cultured rat hepatocytes as an assay system. Its anti-hepatotoxic activity was evaluated by measuring activity of glutamic pyruvic transaminase released from the $CCl_4-in-toxicated$ primary cultured rat hapatocytes. Icariside II significantly reduced the activity of glutamic pyruvic transaminase released from the $CCl_4-in-toxicated$ primary cultured rat hepatocytes and resulted in 78% recovery of the toxicity at the concentration of $200{\;}\mu\textrm{m}$. The anti-hepatotoxic activity of icariside II on the $CCl_4-in-toxicated$ primary cultured rat hepatocytes was as potent as that of silybin.

  • To investigate how the aqueous extracts of Epimedium koreanum Nakai (EK) affects pigmentation of skin, the aqueous extract of EK at various concentrations were incubated with 1 $\times$ 10$^{5}$ melanoma cells per well for 72 h. The morphology and number of cell line were not changed, but the aqueous extract of EK increased the tyrosinase activity and the content of melanin polymer in the cell line. These results indicate that the aqueous extract of EK promotes melanogenesis of Bl6 mouse melanoma cell line.

  • This study was designed to evaluate the antioxidant activities and protective effects on PC12 cells of the extract of Epimedium koreanum and its main constituents icariin and icariside I. After screening the seven identified flavonoid glycosides from E. koreanum through DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) assay, E. koreanum, Icariin and Icariside I exhibited significant effect on radical scavenging activity. E. koreanum, icariin and icariside I were examined using DPPH, ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) and FRAP (ferric reducing ability power) assay. In all antioxidant assays, E. koreanum, icariin and icariside I showed high radical scavenging activities in a dose-dependent manner. Protective effects against $H_2O_2-induced$ PC12 cells were assessed with MTT assay. The results indicated that cell viability and protection on PC12 cells of icariside I and icariin increased dose dependently. These study results suggest that E. koreanum, icariin and icariside showed high antioxidant capacities and cell protective effects. Icariside I, one of the metabolites of icariin, may be a new and effective flavonoid compound as a functional component.

  • Near infrared reflectance spectroscopy (NIRS) has become widely accepted for rapid quantitative analysis of components in some crops. Our object was to determine icariin contents in whole plant of Epimedium koreanum by using an NIRS system. Total 150 plant samples previously analyzed by HPLC were scanned by NIRS and 68 samples were selected for calibration and validation equation. A calibration equation calculated by MPLS(modified partial least squares) regression technique was developed and a coefficient of determination in calibration and validation sets were 0.95 and 0.82, respectively. A comparison between NIRS estimation and HPLC value was performed with the remaining samples not included in the calibration and validation sets. Most of samples also showed a positive correlation like a validation set. Our results demonstrate that this developed NIRS equation can be practically used as a mass screening method for rapid quantification of icarin contents in Epimedium koreanum N.

  • The efficiency of regeneration of callus and explants from leaf and stem disks of Epimedium koreanum was examined on the MS media containing 2,4-D, NAA, Kinetin, BA and TDZ. Calli were formed on the 2mg/l 2,4-D media at the rate of 32% from leaf discs and 52% from stems. No callus was produced on the media which are containing BA or TDZ alone. The combination of 2,4-D and BA showed the effect on the formation of callus. The combination of 2mg/l 2,4-D and 0.lmg/l BA in the MS media had produced the highest percentage of callus formation, 50% from leaf discs and 40% from stems, respectively. The combination of 2mg/l 2,4-D and 1mg/1 BA in the MS media had affected the formation of callus in the rate of 40% from leaf discs and 25% from stems. The combined plant growth regulators of 2,4-D and BA increased the formation of calli from leaf discs, but single treatment of 2,4-D showed the highest callus formation from stems. Multiple shoots from leaf discs were formed on the media containing NAA, BA, kinetin, and TDZ. The highest number of multiple shoots were obtained 0.1mg/l NAA combined with 1mg/l kinetin. As a result, leaf discs or stems can be used for the mass propagation of Epimedium koreanum, but stem elongation of shoots from calli was not easy.

  • The air dried of Epimedium koreanum Nakai was extracted with MeOH and its extractives were concentrated with a vacuum evaporator. The extractives were fractionated with a series of n-hexane, chloroform (${CHCl}_3$), butanol (BuOH), ethyl acetate (EtOAc) and water on a separately funnel. Each fraction was freeze dried to give some dark brown powder. The EtOAc and BuOH soluble fractions were chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-n-hexane mixture as eluents. The isolated compounds were tested with a cellulose TLC developed with TBA and 6% acetic acid and then visualized on UV lamp or sprayed with vanillin-HCl-EtOH. The purified compounds were flavonoids and their glycosides, and organic acid as follows : (+)-catechin, icariin, hyperoside, Ikarisoside A and caffeic acid. The structures of each compounds were confirmed by $^1H-NMR,\;^{13}C-NMR$ and Mass spectra. Also, executed qualitative analysis as use GC/MS(Libraries search) about ${CHCl}_3$ soluble compounds of each part.
