Production and Characterization of Monoclonal Antibodies to a Generic Hapten for-Class-Specific Determination of Organophosphorus Pesticides

  • Monoclonal antibodies have been generated against a generic hapten, ο,ο-diethyl ο-(5-carboxy-2-fluorophenyl) phosphorothioate, for the determination of organophosphorus (OP) pesticides in a class-specific manner. In an indirect competitive enzyme-linked immunosorbent assay (ELISA) format, employing a heterologous coating antigen, these monoclonal antibodies showed desirable properties for use in the class-specific determination, i.e., broad specificity and high sensitivity. The IC50 values of four commonly used ο,ο-diethyl OP pesticides were fairly uniform ranging from 0.1 to 0.3 ㎛/mL. The IC50 values of three ο,ο-dimethyl derivatives were between 0.3 and 1.4 ㎛/mL. These values, together with the limits of detection (LOD), were better, in terms of the specificity and sensitivity, compared with the values obtained previously with polyclonal antibodies.

한우송아지에서 ELISA를 이용한 소 바이러스성 설사병 바이러스 항원 검출 (Seroprevalence of Antigens to Bovine Viral Diarrhea Virus in Korean Calves of the Shown Healthy, Digestive and Respiratory Symptom)

  • The aim of this study was to investigate the prevalence of bovine viral diarrhea virus (BVDV) infection in Chonbuk province. Blood samples were taken from 92 korean calves to determined their serological status against BVDV, Capture enzyme-linked immunosorbent assay (ELISA) was used to test for antigen. The number of seropositive calves ranged from 3.3% to 12.9%. Antigens against BVDV were detected in 3.3% of healthy calves, 6.4% of digestive symptom calves, 12.9% of respiratory symptom calves, respectively. Sex and age of calves had no significant differences on the prevalence of BVDV. The results indicate that transmission of BVDV may have become exposed as a result of contact with acute infected or persistently infected cattle.

  • The aim of this study was to detect bovine viral diarrhea virus (BVDV) from calves in Chonbuk province. Blood samples were taken from ninety-two dairy calves. Capture enzyme-linked immunosorbent assay (ELISA) was used to detect BVDV. BVDV were detected in eight out of ninety-two (8.6%) dairy calves. BVDV were detected in one of twenty five of female calves and one of twenty three of male calves of 4 months old, whereas in the 5 months age group, BVDV were detected in low of twenty three of female calves and two of twenty one of male calves. There were no significant differences (p>0.05) in the detection rate of BVDV on the basis of sex. On the other hand, ages of calves had significant differences (p<0.05) on the prevalence of BVDV.

  • In order to supplement a diagnostic method for detection of infectious cattle to bovine tuberculosis, performed ELISA for detection of antibody to if bovis in serum and milk. The diagnostic efficacy of the established ELISA was compared with test of the tuberculin skin test for bovine tuberculosis. The positive corresponding rate of serum ELISA and tuberculin skin test showed 84.3%, milk ELISA and tuberculin skin test showed 75.0%, milk ELISA and serum ELISA showed 75.0% respectively. Comparison of the serum and milk to tuberculin antibody concentration in tuberculin positive cattle, the milk contained 1/100-1/150 concentration compared serum tuberculin concentration. The established ELISA was considered efficient for detection of antibodies to M bovis in serum and milk.

Enzyme-linked immunosorbent assay for detection of Trichinella spiralis antibodies and the surveillance of selected pig breeding farms in the Republic of Korea

  • Trichinellosis is a parasitic zoonosis of public health importance. It is caused by Trichinella spiralis which has a wide host range including humans. In the present communication, the ELISA technique was employed on a total of 803 blood samples from 7 selected pig breeding farms in 1996 for diagnosis and surveillance of trichinellosis. Out of the entire 803 samples, nine were found to be suspected while one was positive by ELISA. But western blot analyses employed for further confirmation have shown that all of 10 samples did not react to larval excretory-secretory product antigens. These results indicate that pig breeding farms included in the present study are free from trichinellosis . However, it does not mean Korea is free from trichinellosis since human trichinellosis has recently been reported. The necessity of continued surveillance for trichinellosis in both pigs and wild animals was discussed.

식품 중 다환방향족탄화수소 분석법 연구 (A study on analytical methods for polycyclic aromatic hydrocarbons in foods)

  • This study was proceeded the analytical methods using various analytical instruments for polycyclic aromatic hydrocarbons (PAHs) in food products. Various analytical methods were developed to determine levels of PAHs including benzo[a]pyrene, benzo[a]anthracene, benzo[b]fluoranthene, and chrysene formed in various food products using gas chromatography-mass spectrometry (GC-MS), enzyme-linked immunosorbent assay (ELISA) and raman spectroscopy. Recently, the rapid on-site response for the detection of hazardous substances in food aims to develop an onsite rapid detection of a simplified technical analysis method to reduce the time and cost required for analysis of PAHs. Current PAHs detection methods have been reviewed along with new raman spectroscopy analytical method.

  • Trichoderma viride가 분비하는 cellobiohydrolase를 항원으로 사용하여 이 섬유소 분해효소에만 특이적인 항체를 얻을 수 있었다. 이 항체를 이용하여 column single immunodiffusion 방법에 의해서 cellobiohydrolase의 농도 2-10 $\mu\textrm{g}$ 범위 내에서는 직선적으로 정량 할 수 있었고, ELISA 방법을 사용할 때는 $10^6$의 항체 희석시는 40-110ng, $10^5$ 항체 희석시는 100-1,200pg 농도범위의 cellobiohydrolase 를 정량 할 수 있었다.

  • 장티푸스가 의심되는 51명의 환자 혈청내에서 Salmonella typhi의 균체항원에 대한 IgG, IgM과 IgA 항체률 효소면역측정법으로 측정하였다. 균체항원에 대한 IgG와 IgA 항체가는 세균이 분리되어 장티푸스로 확진된 환자에서 건강대조군보다 높았다. 반면 세균을 분리하지는 못하였지만 임상증상 등으로 장티프스를 의심하는 환자에서는 IgM항체가는 건강대조군보다 높았다. 균체항원에 대한 항체를 효소면역 측정법으로 측정하는 것이 Widal 검사보다 민감도가 높았다. 장티푸스를 진단하는데 균체항원을 부착시켜 효소면역측정법을 사용하는 것이 유용할 것이다.

뇌낭미충증의 혈청학적 진단에 있어서 간접 형광항체 반응 및 효소연결성 면역흡착 검사의 비교 평가 (Comparative evaluation of indirect immunofluorescent antibody test with enzyme-linked immunosorbent assay in serodiagnosis of human neurocysticercosls)

  • 뇌낭미충증의 면역혈청학적 진단에 있어서 간접 형광항체 법의 유용성을 평가하기 위하여 효소연결성 면역홉착 검사와 비교 검토하였다. 검사대상자는 확진된 뇌낭미충증 환자의 혈청 163예, 다른 뇌신경 증상 환자, 조층 및 흡충류 감염자 101예 및 건강인 대조군 100예로서 모두 364예이었다. 간점 형광항체 반응에는 인체 유구낭미충의 낭벽 항원을, 효소연결성 면역흡착 검사에는 낭액 항원을 사용하여 혈청내 특이 IgG 항체를 조사한 결과 두 방법간의 민감도 및 특이도에 큰 차이가 없었으며, 양성 및 음성의 동일한 힐청을 검사하였을 때 낭미충증 혈청의 90.8%가 서로 합치되어 밀접한 연관성을 나타내었다. 또한 장내 조충 감염증의 경우 두 방법 모두에서 높은 교차반응을 나타내었으나 간접 형광항체반응의 특이성이 더 좋았으며 특히 간접 형광항체 반응은 흡충류 감염자 혈청에서 교차반응을 나타내지 않았다. 이와 같은 결과는 혈청만을 사용하였을 경우 간접 형광항체반응의 민감도나 특이도가 효소연결성 면역홉착 검사와 차이가 없으며 뇌낭미충증의 진단에 매우 유용함을 나타내고 있었다.
