Beam-column joints are the key structural elements, which dictate the behavior of structures subjected to earthquake loading. Though large experimental work has been conducted in the past, still various issues regarding the post-yield behavior, ductility and failure modes of the joints make it a highly important research topic. This paper presents experimental results obtained for eight beam-column joints of different sizes and configuration under cyclic loads along with the analytical evaluation of their response using a simple and effective analytical procedure based on nonlinear static pushover analysis. It is shown that even the simplified analysis can predict, to a good extent, the behavior of the joints by giving the important information on both strength and ductility of the joints and can even be used for prediction of failure modes. The results for four interior and four exterior joints are presented. One confined and one unconfined joint for each configuration were tested and analyzed. The experimental and analytical results are presented in the form of load-deflection. Analytical plots are compared with envelope of experimentally obtained hysteretic loops for the joints. The behavior of various joints under cyclic loads is carefully examined and presented. It is also shown that the procedure described can be effectively utilized to analytically gather the information on behavior of joints.
High-performing architecture should be designed by taking into account the mutual dependency between the new building and the local context. The performative architecture plays an important role to avert any unforeseen failures after the building has been built; particularly ones related to the microclimate impacts that affect the human comfort. The use of the concept of solar envelopes helps designers to construct the developable mass of the building design considering the solar access and the site obstruction. However, the current analysis method using solar envelopes lack in terms of integrating the detailed information of the existing context during the simulation process. In architectural design, often the current site modelling not only absent in preserving the complex geometry but also information on the surface characteristics. Currently, the emerging applications of point clouds offer a great possibility to overcome these limitations, since they include the attribute information such as XYZ as the position information and RGB as the color information. This study particularly presents a comparative analysis between the manually built 3D models and the models generated from the point cloud data. The modelling comparisons focus on the relevant factors of solar radiation and a set of simulation to calculate the performance indicators regarding selected portions of the models. The experimental results emphasize an introduction of the design approach and the dataset visibility of the 3D existing environments. This paper ultimately aims at improving the current architectural decision of support environment means, by increasing the correspondence between the digital models for performance analysis and the real environments (context of design) during the conceptual design phase.
In earthquake engineering, dynamic analyses are usually conducted by using a nonlinear analytical model of the entire building in order to identify the performance against earthquakes. At the same time, a large number of dynamic analyses are required to consider uncertainties on analytical models and ground motions. Therefore, it is necessary for the analytical model to be adequate, that is to say, the runtime should not be too long as the entire building is modeled to be in much detail, or the nonlinear model should not yield outputs very far from the actual ones by excluding important behaviors too much. The analytical model is usually developed based on experimental results, which have been already conducted for reinforced concrete columns with relaxed details. Therefore, this study aimed at making analytical models to be able to simulate the hysteretic behavior of the columns simply and easily. The analytical model utilizes a lumped hinge model to represent nonlinear moment-rotation hysteretic behavior of RC columns, which is feasible for nonlinear dynamic analyses usually conducted in earthquake engineering and for matching the analytical model to test results. For the analytical model, elements and material models provided by OpenSees are utilized. The analytical model can define the envelope curve, pinching, and unloading stiffness deterioration, but shortcoming of this model is not to be able to consider axial force-moment interaction directly and to simulate strength deterioration after post-capping completely. However, the analytical model can still represent test results well by considering that the goal of this study is to propose a general way to represent the hysteretic behavior of RC columns with relaxed details, not to provide parameters for a refined hysteretic model that can be just applied case by case.
Vesicular stomatitis virus G glycoprotein (VSV-G) has been widely used for pseudotyping retroviral, lentiviral, and artificial viral vectors. The objective of this study was to establish a potential approach for large-scale production of VSV-G. To this end, VSV-G was cloned with an N-terminal His-tag into Pichia pastoris expression vector pPIC3.5K. Three clones ($Mut^s$) containing the VSV-G expression cassette were identified by PCR. All clones proliferated normally in expansion medium, whereas the proliferation was reduced significantly under induction conditions. VSV-G protein was detected in cell lysates by western blot analysis, and the highest expression level was observed at 96 h post induction. VSV-G could also be obtained from the condition medium of yeast protoplasts. Furthermore, VSV-G could be incorporated into Ad293 cells and was able to induce cell fusion, leading to the transfer of cytoplasmic protein. Finally, VSV-G-mediated DNA transfection was assayed by flow cytometry and luciferase measurement. Incubation of VSV-G lysate with the pGL3-control DNA complex increased the luciferase activity in Ad293 and HeLa cells by about 3-fold. Likewise, incubation of VSV-G lysate with the pCMV-DsRed DNA complex improved the transfection efficiency into Ad293 by 10% and into HeLa cells by about 1-fold. In conclusion, these results demonstrate that VSV-G could be produced from P. pastoris with biofunctionalities, demonstrating that large-scale production of the viral glycoprotein is feasible.
Abstract White spot disease (WSD), resulting in more than 90% mortality of aquacultured penaeid shrimp, has been reported off the southern and western coasts of Korea since 1993. The pafuogen of WSD has been identified as being a virion wifu an envelope around a central nucleocapsid, and with an average size of 167 nm in diameter and 375 nm in length. In the present study, an in situ hybridization technique was developed as a rapid. sensitive, and specific diagnostic assay for the WSD viros infection in shrimp. Furthermore. the pathological changes ofWSD, in shrimp experimentally infected with WSD viroses. were investigated. Using a biotinylated 643 bp probe obtained from a peR using primers specific to the rod-shaped virus of Penaeus japonicus (RV-PJ), positive signals were detected in both naturally and experimentally infected shrimps. The in situ hybridization revealed positive reactions in the nuclei of the stromal matrix cells in the lymphoid organ, epithelia of the gills, foregut. epidermis, and hematopoietic cells of the interstitial tissues, suggesting the presence of WSD virus. Tills result indicates that the in situ hybridization method can be useful for a rapid and sensitive detection of WSD viruses in shrimp.shrimp.

An acid phosphatase (HppA) activated by $NH_4Cl$ was purified 192- and 34-fold from the periplasmic and membrane fractions of Helicobacter pylori, respectively. SDS-polyacrylamide gel electrophoresis revealed that HppA from the latter appears to be several kilodaltons larger in molecular mass than from the former by about 24 kDa. Under acidic conditions (pH${\leq}$4.5), the enzyme activity was entirely dependent on the presence of certain mono- and/or divalent metal cations (e.g., $K^+$,$ NH_4{^+}$, and/or $Ni^{2+}$). In particular, $Ni^{2+}$ appeared to lower the enzyme's $K_m$ for the substrates, without changing $V_{max}$. The purified enzyme showed differential specificity against nucleotide substrates with pH; for example, the enzyme hydrolyzed adenosine nucleotides more rapidly at pH 5.5 than at pH 6.0, and vice versa for CTP or TTP. Analyses of the enzyme's N-terminal sequence and of an $HppA^-$ H. pylori mutant revealed that the purified enzyme is identical to rHppA, a cloned H. pylori class C acid phosphatase, and shown to be the sole bacterial 5'-nucleotidase uniquely activated by $NH_4Cl$. In contrast to wild type, $HppA^-$ H. pylori cells grew more slowly. Strikingly, they imported $Mg^{2+}$ at a markedly lowered rate, but assimilated urea rapidly, with a subsequent increase in extracellular pH. Moreover, mutant cells were much more sensitive to extracellular potassium ions, as well as to metronidazole, omeprazole, or thiophenol, with considerably lowered MIC values, than wild-type cells. From these data, we suggest that the role of the acid phosphatase HppA in H. pylori may extend beyond 5'-nucleotidase function to include cation-flux as well as pH regulation on the cell envelope.
In the past, there was a theory that influenza wasn't transmitted directly from birds but was infected to humans via swains. Recently, molecular level research has progressed, and it was confirmed that the avian influenza virus can directly infected to human lung and intestinal epithelial cells. Three pandemicsin the past 100 years were also infected to humans directly from birds. In view of such scientific background, we are developing a method for screening sick birds by monitoring the physiological characteristics of birds in a contactless manner with sensors. Here, the movement of respiratory muscles and abdominal muscles under autonomic innervation was monitored using a magnet and Hall sensor sewn on the thoracic wall, and other multimedia devices. This paper presents and discusses the results of experiments involving continuous periodic noise discovered during flight experiments with a data logger mounted on a Japanese pheasant from 2012 to 2015. A brief summary is given as the below: 1. Magnet and Hall sensor sewn to the left and right chest walls, bipolar electrocardiograms between the thoracic walls, posterior thoracic air sac pressure, angular velocity sensors sewn on the back and hips, and optical reflection of LEDs (blue and green) from the skin of the hips allow observation of periodic vibrations(fasciculations) in the waves. No such analysis has been reported before. 2. These fasciculations are presumed to be derived from muscle to maintain and control air sac pressure. 3. Since each muscle fiber is spatially Gaussian distributed from the sympathetic nerve, the envelope is assumed to plot a Gaussian curve. 4. Since avian trunk muscles contract periodically at all time, we assume that the sympathetic nerve dominates in their control. 5. The technique of sewing a magnet to the thoracic wall and measuring the strength of the magnetic field with a Hall sensor can be applied to screen for early stage of avian influenza, with a sensor attached to the chicken enclosure.

Currently, in the most mail automatic processing centers, after facing and canceling, envelope mail is passed through an Optical Character Recognition/Barcode Sorter(OCR/BS) to read the address and 3 of 5 fluorescent(luminescent) barcode is applied. Normally, 30%∼35% of this mail is rejected. The usual reasons for read failure are poor printing quality of address and barcode, script printing and failure to locate the address. This paper describes a verification system of the postal 3 of 5 customer barcode for solving this problem. The certification system of the 3 of 5 customer barcode consists of barcode verification system and postal address database. The purpose of certification system of the customer barcode verifies the postal 3 of 5 customer barcode and tests matching of mail piece postal address, and retrieves postal code.

The aim of this study was to analyze the thermal performance through Test-Cell of TI-wall in domestic climate. This study was carried out as follows: 1) The TI-wall was studied for ability to reduce heat loss through the building envelope and analyzed to TIM properties. 2) Test models of TI-wall were designed through the investigation of previous paper and work, measured for winter and spring, and the thermal effects were analyzed. The type of the TIM used in test model is small-celled(diameter 4mm and thickness 50mm) capillary and cement brick(density $1500kg/m^3$) was used by thermal mass. 3) Test-cell of TI-wall was calibrated from measured data and the dynamic simulation program ESP-r 9.0. In these simulations, the measured climate conditions of TaeJon were used as outdoor conditions, and the simulation model of Test-cell was developed. 4) The sensitivity analysis is executed in various aspects with standard weather files and ESP-r 9.0, and then most suitable system of TI-wall are predicted. Finally, The suitable system of TI-wall was analysed according to sizes of air gap, kinds, thickness, and the surface absorption of therm wall. The result is following. In TI-wall, Concrete is better than cement brick, at that time the surface absorption is 95%, and the most efficient thickness is 250mm. As smaller of a air gap, as reducer of convection heat loss, it is efficient for heating energy. However, ensuring of a air gap at least more than 50mm is desirable for natural ventilation in Summer.

The performance and design of climate responsive buildings such as AKU requires a detailed study of attributes of climate both at micro as well as macro level. The therapeutic value of contact with nature through window view, greenery and landscape is calculated there. Method: A two prong strategy is been devised for this article, at micro level three typical morphologies are analysed by creating same environment of neighboring building on sun shading chart, radiation and temperature range. Since the analysis of local climate helps to determine the design strategies for hospital Healing Environment which is suitable for Karachi climate; in order to track the macro climatic behaviour, a considerable analysis of psychometrics chart for AKU Karachi are designed on Climate Consultant (CC) and analysed by Machine Learning. Climate Consultant proposes different design strategies suitable for Karachi. And on the other hand time wise illumination sources for clinical area which are then measured on psychrometric chart- according to singular space: multi patient admission, secondly: acute ambulatory ward, and tertiary: multi windowed space according to the mushrabiyah and sky light pattern. Result: Our findings support the hypothesis that windowed wall is 75-80% more healing wall; an accelerated evidence was found for healing at macro level if the form of the hospital is designed according to the climatologically preferences, whereas at micro level: the light resource becomes the staff attentiveness determinant. In Conclusion evidence was provided that the actual form of luminosity results consequently in satisfaction while light entering from several set of windows and other sources might be valued if design according to the healing environment. The data added on the sun shading chart to calculate rays entraining into space in patient room equal to 124416.21 Watts/ meter $m^2$ is calculated as precise healing rate-and is confirmed by questionnaire from patients belonging from each clinical stage having different illnesses.
