This study was investigated discharge and water quality for rainy days at the inflow streams (Gog-Kyo, Mu-Han, Sap-Kyo and Nam-Won) into Lake Sap-Kyo from June to November of 1999. The eventual aim of this study was to improve the water quality of Lake Sap-Kyo. Therefore, in order to reduce inflow pollutants loading into Lake Sap-Kyo, we investigated characteristics of stormwater runoff and estimated pollutant loading for the rainy season. The results of that Sap-Kyo stream is $5.75\;m^3/sec$ flow and influences water quantity of Lake Sap-Kyo greatly during the dry weather. Gog-Kyo stream influence to quantity of Lake Sap-Kyo greatly for the rainy days. Gog-Kyo stream and Sap-Kyo stream are $12.13\;m^3/sec$ and $8.49\;m^3/sec$, respectively. The reason is that the watershed area of Gog-Kyo stream is larger than other streams. When we prepare to plan management to improve the water quality of Lake Sap-Kyo, we have to consider that runoff characteristics by land use. Among the annual total pollutants loading inflow Lake Sap-Kyo, the ratio of non-point pollutants loading estimated that SS, BOD, T-N and T-P are 62%, 43%, 41% and 27%, respectively. Therefore, in order to improve water quality of Lake Sap-Kyo, we have to reduce non-point pollutants loading.

Due to emissions of greenhouse gases caused by increased use of fossil fuels, the climate change has been detected and this phenomenon would affect even larger changes in temperature and precipitation of South Korea. Especially, the increase of temperature by climate change can affect the amount and pattern of snowfall. Accordingly, we tried to predict future snowfall and the snowfall pattern changes by using the downscaled GCM (general circulation model) scenarios. Causes of snow varies greatly, but the information provided by GCM are maximum / minimum temperature, rainfall, solar radiation. In this study, the possibility of snow was focused on correlation between minimum temperatures and future precipitation. First, we collected the newest fresh snow depth offered by KMA (Korea meteorological administration), then we estimate the temperature of snow falling conditions. These estimated temperature conditions were distributed spatially and regionally by IDW (Inverse Distance Weight) interpolation. Finally, the distributed temperature conditions (or boundaries) were applied to GCM, and the future snowfall was predicted. The results showed a wide range of variation for each scenario. Our models predict that snowfall will decrease in the study region. This may be caused by global warming. Temperature rise caused by global warming highlights the effectiveness of these mechanisms that concerned with the temporal and spatial changes in snow, and would affect the spring water resources.
For the detection of human immunodeficiency virus (HIV), ultra-rapid real-time PCR methods were developed. The target DNA sequences were used 495 bp HIV-1-specific env gene (gi_1184090) and 294 bp HIV-2-specific env gene (gi_1332355). Ultra-rapid real-time PCR was peformed by $Genspector^{TM}$ (Samsung, Korea) using microchip-based, $6\;{\mu}l$ of reaction volume with extremely short running time in only 2 steps (denaturation, annealing/extension) in each cycle of PCR. Total reaction for 30 cycled ultra-rapid PCR detection including melting temperature analysis was completed in 7 min and 30 sec. The HIV-1-specific 117 bp-long or HIV-2-spe-cific 119 bp-long PCR products were successfully amplified from the minimum of template, $2.3{\times}10^3$ copies of each euv gene using 30 cycled two-steps ultra-rapid PCR. This kind of ultra-rapid real-time PCR method would be useful not only for the rapid-detection of HIV, but also rapid-detection of other pathogens.

This research was performed to evaluate the feasibility of in situ soil flushing for TPH-contaminated soil remediation. It was conducted in batch test as fundamental research for in situ soil flushing. The 30% of initial TPH concentration was removed by shaking only in batch test. The removal efficiency of TPH in case of groundwater as surfactant dilution solution was approximate 2~6% lower than that of distilled water. Mixing ratio of soil to surfactant solution did not practically effect on the TPH removal efficiency. In the experiment of using single or mixed surfactant solution with 0.1~4.0 wt%, Tween-80, SWA-1503, SWA-1503+SDS showed averagely over 80%. It was determined that the optimum surfactant concentration was 0.1 wt% because there was no significant difference between concentrations of 0.1~4.0 wt%.

Ultra-rapid real-time PCR was performed by $Genspector^{TM}$ using microchip-based, $1\;{\mu}l$ of reaction volume with extremely short time in each 3 step in PCR. The detection including DNA-amplification and melting temperature analysis was completed inner 15 minutes. The HIV-1 specific 117 bp-long and HIV-2 specific 119 bp-long PCR products were successfully amplified from minimum of template,2.3 molecules of each env gene. This kind of real-time PCR was designated as ultra-rapid real-time PCR in this study and it could be applied not only an alternative detection method against HIV, but also other pathogens using PCR-based detection.
Phylogenetic analysis was conducted to monitor transmission of HIV and to investigate the genetic structure of primary isolates from 12 HIV-1 subtype A infected Koreans. The individuals infected with subtype A viruses had been diagnosed as HIV-1 seropositives during the period 1987 to 1995 and blood samples have been collected from 1991 to 1997. DNA of each individual was isolated from uncultured or cultured peripheral blood mononuclear cells. V3-V5 (0.7 kb) fragment of HIV-1 env gene was amplified by nested polymerase chain reaction and the PCR products were sequenced. The mean value of the divergence of nucleotide of HIV-1 env V3-V5 fragment was $17.0{\pm}4.06%$ ($8.6{\sim}25.8%$) within HIV-1 subtype A isolates from Koreans. This diversity was higher than those of African isolates ($13.7{\pm}2.66%$). In the phylogenetic tree, Korean subtype A isolates were not grouped together, but intermingled into African isolates. The results of this study suggested that HIV-1 subtype A variants be introduced from multiple sites of Africa into Korea and the big genetic diversity of Korea HIV-1 subtype A isolates may be further influenced by the range of geographic locations in which the infection occurred rather than the elapsed time between infection and collection of samples and the disease progression.
To develop the polymerase chain reaction (PCR) for the detection of type D simian retrovirus (SRV) infection, an oligonucleotide primer pair was designed to hybridize to the sequences within env gene of SRV subtype 1 (SRV-1). The 3' proximal env sequences annealing to the primers had been rather conserved among three different subtypes of SRV, SRV-1, SRV-2, and SRV-3 (Mason-Pfizer Monkey Virus: MPMV). The PCR using the primer pair targeting an env region successfully detected and amplified all three subtypes of SRV with excellent specificity after single round of reaction. The tests with peripheral blood mononuclear cells infected either with simian immunodeficiency virus or simian T-Iymphotropic virus type 1, major immunosuppressive viral agents together with SRV in simian, verified the specificity of the PCR by excluding any cross reactivity. Semiquantitative titration PCR, amplifying serially diluted plasmid DNA of each subtype, was performed to evaluate sensitivity limits of the reaction. Based on molecular weight of each cloned SRV genome, the PCR should be able to detect one SRV-infected cell per more than $5-7{\times}10^4$ uninfected cells after simple ethidium bromide staining of resulting products. The PCR must be very efficient screening system with its quickness, certainty, and sensitivity for SRV-infected animals used in human AIDS research model. Second round amplification of the reaction products from the first PCR, or Southern hybridization by radiolabeled probes shall render to compete its efficacy to ELISA which has been the most sensitive technique to screen SRV infection but with frequent ambiguity problem.
Many reservoirs in Korea and their downstream environments are under increased pressure for water utilization and ecosystem management from longer discharge of turbid flood runoff compared to a natural river system. Turbidity($C_T$) is an indirect measurement of water 'cloudiness' and has been widely used as an important indicator of water quality and environmental "health". However, $C_T$ modeling studies have been rare due to lack of experimental data that are necessary for model validation. The objective of this study is to validate a coupled three-dimensional(3D) hydrodynamic and particle dynamics model (ELCOM-CAEDYM) for the simulation of turbid density flows in stratified Daecheong Reservoir using extensive field data. Three different groups of suspended solids (SS) classified by the particle size were used as model state variables, and their site-specific SS-$C_T$ relationships were used for the conversion between field measurements ($C_T$) and state variables (SS). The simulation results were validated by comparing vertical profiles of temperature and turbidity measured at monitoring stations of Haenam(R3) and Dam(R4) in 2004. The model showed good performance in reproducing the reservoir thermal structure and propagation of stream density flow, and the magnitude and distribution of turbidity in the reservoir were consistent with the field data. The 3D model and turbidity modeling framework suggested in this study can be used as a supportive tool for the best management of turbidity flow in other reservoirs that have similar turbidity problems.

In this study, the importance of implied strength correction for shallow depths at a region of moderate to low seismicity with primary focus on its effect upon site natural period and mean period of the ground motion is investigated. In addition to the most commonly used Modified Kondner-Zelasko (MKZ) model, this paper uses a quadratic/hyperbolic (GQ/H) model that can capture the stress - strain response at large strains as well as small strain stiffness dependence. A total of six site profiles by downhole tests are used and 1D site response analyses are performed using three input motions with contrasting mean periods. The difference between non-corrected and corrected analyses is conditional on the site period as well as mean ground motion period. The effect of periods is analyzed by correlating them with the effective peak ground acceleration, maximum shear strains and amplification factors. The comparative study reveals that the difference is more prominent in soft sites with long site periods. Insignificant differences are observed when soil profiles are subjected to ground motion with very short mean period.

In this paper, a small-scale model testing system was developed using a series of small-scale model tests to analyze the mechanism of compaction pile formation and evaluate the quality of controlled grading aggregates proposed as an alternative material to the sand compaction pile (SCP) method and granular compaction pile (GCP). These are the most typical ground improvement methods in field practice, particularly for soft grounds. However, the SCP has faced difficulties due to the supply shortage of natural sand and the corresponding price surge of sand. The GCP is limited in marine soft grounds because of the failure occurring at the pile tip caused by excessive expansion of the deeper bulbs, leading to uneven bulb formation. The uniformity of compacted pile bulbs is critical to ensuring the bearing capacity and quality of the compaction pile. This study aims to evaluate the performance of the new material and controlled grading aggregates using small-scale model tests simulating field compaction process to investigate its potential application in comparison with SCP. The compaction piles are examined in four cases according to different materials used for compaction pile and clay strength. The compaction pile materials, which are made of sand and controlled grading aggregates, used in this study were compared to reveal the mechanism of the bulb creation. The experimental data confirm that the bulb formation quality of the traditional sand and the new material, controlled grading aggregates are comparable. The compaction pile made of controlled grading aggregates presents higher bearing capacity than that of marine sand.
