The present study concerned the identification and characterization of a novel bacterial strain isolated from entomopathogenic nematodes collected from different regions in Korea. The bacterial isolate M1021 was Gramnegative, bioluminescent, and produced red colonies on MacConkey agar medium. A rod-shaped structure was confirmed by the electron micrograph. Fatty acid composition was analyzed by using the Sherlock MIDI system. The identification was further supported by 16S rDNA sequence analysis, which revealed 96-99% sequence homology with strains of Photorhabdus temperata. The location of the isolated strain of P. temperata in the phylogenetic tree was confirmed and it was named P. temperata M1021. P. temperata M1021 exhibited catalase, protease, and lipase activities when grown on appropriate media supplemented with respective substrates. The culture of P. temperata M1021 exhibited insecticidal activity against the larvae of Galleria mellonella and the activity was the highest after 3-4 days of cultivation with agitating at $28^{\circ}C$ under 220 rpm. Antibacterial activity was also observed against Salmonella Typhimurium KCTC 1926 and Micrococcus luteus KACC 10488.

Laboratory tests were conducted to determine the susceptibility of the alfalfa weevil, Hypera postica to Korean entomopathogenic nematodes, Steinernema carpocapsae GSN1 strain (ScG), S. glaseri Dongrae strain (SgD), Heterorhabditis bacteriophora Hamyang strain (HbH), and Heterorhabditis sp. Gyeongsan strain (HeG) at the petri dish assay. The larval mortality of H. postica was significantly different depending on nematode species and treatment concentration. SgD and HeG strain were more effective against H. postica larva than ScG and HbH strain. When SgD and HeG strain were treated with the rate of >20 infective juveniles (ijs) per alfalfa weevil larva, mortality was ca. 77.5-100% at the late instars of H. postica in 3 days. The number of established nematode was significantly different depending on nematode species, whereas number of progeny was not significantly different. The mean number of established us of SgD strain in a host was the highest at 80 ijs by 30.2. The highest progeny number of HeG strain was 2,671.5 with 80 ijs. All nematode strains were not parasitic entirely to H. postica adults. These results show that alfalfa weevil late larva is highly susceptible to Korean entomopathogenic nematodes in the laboratory.
The chestnut brown chafer, Adoretus tenuimaculatus Waterhouse, is serious insect pests in golf courses. Adults feed on the leaves of latifoliate trees but larvae feed on roots of turfgrases such as bentgrass, Agrostis spp. Damage of A. tenuimaculatus larvae was observed at the Jinju golf club which showed damage symptom on perennial ryegrass, Lolium perenne in tees and fairways in July, 2000. Damaged turf by A. tenuimaculatus larvae became yellowish and wilted. Symptom of laval damage of A. tenuimaculatus was similar to summer depression in warm season turfgrasses but not recovered by irrigation when Korean isolates of entomopathogenic nematodes were evaluated for the control of A. tenuimaculatus larvae in laboratory and field as a possible biological control agent. The nematodes used were Heterorhabditis bacteriophora Jeju strain, Hererorhabditis sp. Gyeongsan strain, Steinernema carpocapsae Pocheon strain, S.glaseri Dongrea strain, and S.longicaudum Nonsan strain. In the laboratory test H.bacreriophora Jeju strain and Heterorhabditis sp. Gyeongsan strain were highly effective for 3rd instars with 95% mortality. In the field test reduction rates of A.tenuimaculatus larvae were higher by ranging from 28 to 57% by H. bacteriophora Jeju strain, Heterorhabditis sp. Gyeongsan strain, and S.carpocapsae Pocheon strain compared to 7% by natural cause.

The 21 strains of Korean entomopathogenic nematodes, {Heterorhabditis bacteriophora Hamyang strain(HbH), Heterorhabditis sp. 202, 205, 217, Heterorhabdiris sp. KCTC 0991BP strain, Steinernema carpocapsae Pocheon(ScP), S. longicaudum Gonaju, S. longicaudum Nonsan, Steinernema sp. 7,24, 52, 55, 60, 64, 206, 207, 209, 210, 219, and 227 strain} were evaluated for the control of a turfgrass insect pest, Exomala orientalis. Heterorhabditis spp. showed higher pathogenicity than Steinernema spp. against 3rd instar larvae of E. orientalis with $55\%$ mortality by Heterorhabditis sp. 202 strain and $50\%$ by HbH and Heterorhabditis sp.205 strain at the rate of 200 infective juveniles per larva 14 days later after treatment. The number of infective juveniles of Korean entomopathogenic nematodes in 3rd instar larvae of E. orientalis was higher in Heterorhabditis spp. than in Steinernema spp.. In general, numbers of produced infective juveniles of three species were much higher, i.e., Heterorhabditis sp.202 strain produced 273,064 infective juveniles, S. carpocapsae Pocheon strain 273,043, and Heterorhabditis sp. 217 strain 248,887, respectively.
Entomopathogenic nematodes were effective in the control of the cotton caterpillar, Palpita indica Saunder. P. indica mortality was significantly different depending on nematode species, treatment concentration, and instar. S. carpocapsae Pocheon strain was more effective against P. indica than S. glaseri Dongrae strain, S. longicaudum Nonsan strain, Heterorhabditis sp. Gyeongsan isolate, and H. bacteriophora Hamyang strain. When S. carpocapsae was treated with the rate of > 20 infective juveniles (ijs)/larva, mortality was ca. 100% at the 1st-4th instars of P. indica in 72h. $LC_{50}$ of entomopathogenic nematodes were significantly different depending on nematode species. The lowest $LC_{ 50}$ value was obtained by S. carpocapsae with 4.9-8.2 ijs in the 1st-prepupa while the highest $LC_{50}$ by Heterorhabditis sp. with 5.5-21.9 ijs, the shortest LT$_{50}$ value of P. indica was induced by S. carpocapsae when 20 ijs/larva were inoculated. The $LT_{50}$ s were 3.4-9.2h against the 1st-prepupa. The mortality of P. indica in fields was significantly different depending on field condition, nematode concentration, and leaf location. Mortality of P. indica by S. carpocapsae was higher at greenhouse,$ 3$\times$10^{9}$ ijs/ha and upper leaves than at field, 1$\times$10$^{9}$ ijs/ha and lower and middle leaves, respectively.

Five strains of Korean entomopathogenic nematodes(EPN), steinernematids and heterorhabditids(Steinernama carpocapsae GSN1, Steinernema sp. GSNUS-10, Steinernama sp. GSNUS-14, Heterorhabditis bacteriophora Hamyang, Heterorhabditis sp. GSNUH-1) were evaluated and tried in petri dish, pot, and vegetable greenhouses for environmentally friendly control of beet armyworm, Spodoptera exigua. $LC_{50}$ values of five EPN strains against beet armyworm was different depending on nematode strain and beet armyworm instar. $LC_{50}$ value of Steinernema carpocapse GSN1(GSN1) was 3.8-5.1 infective juveniles(Ijs) in 2nd to 4th instars of beet armyworm. Pathogenicity of five EPN strains against beet armyworm different in nematode strain, concentration, application times, and vegetable species in pot and greenhouse. Steinernema spp. was more effective than Heterorhabditis spp. against beet armyworm. Two or three times of applications of EPN were found to be effective regardless of nematode strain and concentration in pot and greenhouse. ENP showed different reactions on vegetable species. Efficacy of EPN was higher on Chinese cabbage than that on cabbage and kale. GSN1 was one of the most effective nematodes and 100,000 infective juveniles per $m^2$(720,000 Ijs/$7.2m^2=1{\times}10^9$Ijs/ha) resulted in higher mortality in greenhouse.
- Turfgrass insect pests and natura.l enemies for biological control were investigated to develop pest management effectively in golf courses at several golf clubs. Twenty eight insect pest species of 10 families in 6 orders were collected from golf courses. The zoysiagrass mite, Eriophyes zoysiae and root-knot nematode, Meloidogyne incognita were also collected from zoysiagrass. White grubs of several scarab beetles and cutworms (Agrotis spp.) damaged seriously at most surveyed golf clubs. In addition, bluegrass webworm (Crambus sp.), Japanese lawngrass cutworm (Spodoptera depravata), scale insects, Tipula sp., and ants (Camponitus japonicus, Formica japonica, and Lasins japonicus) damaged turfgrasses directly or indirectly in golf courses. The entomopathogenic nematodes, Heterorhabditis spp., Steinernema glaseri, and S. longicaudum, entomopathogenic fungi, Beauveria bassiana and Metarhizium anisopliae, and milky disease, Paenibacil/us popil/iae were isolated from white grubs or turfgrass soil as microbial control agents. Besides, dipteran predators, Cophinopoda chinensis, Philonicus albiceps, and Promachus yesonicus and hymenopteran parasitoid, Tiphia sp. were also collected. The P. yesonicus was the most active in golf courses. The root-knot nematode, M. incognita was found from Zoysia japonica, Z. matrella. and Cynodon dactylon.

Toxicological studies of two potential biological control agents, the entomopathogenic nematode (Steinernema carpocapsae) and the symbiotic bacteria (Xenorhabdus nematophilus) were conducted against two beneficial insects and one mammal species. Two microbial agents varied in their toxicities between two insect species: an ant, Pristomyrmex pungens, and silkworm, Bombyx mori. In oral toxicity test, the symbiotic bacteria resulted in significant lethal [half lethal concentration of $1.4$\times$10^3$colony-forming units (cfu)/ml] on the ants, while they gave little lethal effect (half lethal concentration of more than $10^{8}$ cfu/ml) on the silkworms. The nematodes, however, gave significant lethal effect [half lethal concentration of 4 infected juveniles (IJs)/ml] on the silkworms, while they did little lethal effect (half lethal concentration of 150,000 IJs/ml) on the ants in topical assays. Both the nematodes and the bacteria did not give lethal effect to the albino rats, Rattus norvegicus, when they were fed orally into the rats. Also, any of these microbial agents were not detected in the internal organs of the treated rats.

Three Korean isolates of entomopathogenic nematodes, Steinernema carpocapsae KCTC 0981BP (ScK), S. glaseri Dongrae (SgD), and Heterorhabditis sp. KCTC 0991BP (HsK), were evaluated for the control of a turfgrass pest, Popillia quadriguttata. Three days after treatment, all nematodes showed high pathogenicity to adult P. quadriguttata with $97.5\%$ mortality by ScK, $90.8\%$ by HsK, and $80\%$ by SgD at the concentration of 900 infective juveniles per adult. Nematode attachment and infection rate to adult P. quadriguttata were various depending on nematode species and inoculation density. The rate of nematode attachment was $90.8\%$ in HsK, $90.6\%$ in SgD, and $35\%$ in ScK, resfectively at the concentration of 900 infective juveniles per adult. The infection rate that represents the rate of detected nematode from inside insect of body was $97.5\%$ in ScK, $ 80\%$ in both HsK and SgD at the 900 concentration of infected juveniles. The infection rate, however, was decreased to $27.5\%$ in ScK, $72.5\%$ in SgD, but no nematodes was detected in HsK at the concentration of 90 infective juveniles.
