청결 영양강화미 제조를 위한 코팅시스템 개발 (Development of a Coating System for Producing Clean Enriched Rice)

  • This study was conducted to develop a coating system for producing clean enriched rice. The nutritional components of such as brown rice, white rice, clean white rice, enriched rice of US, black rice of China, and rice with perfume of China, were analyzed and compared. Total protein lipid md minerals in brown rice were higher than those in the other rices. A nutrition premix with lysine, thiamine, niacine, calcium lactate, ferrous lactate, and rice starch was made for enriching white rice. A coating device consisted of a tumbler, two spraying nozzles, a rice mixer, etc was developed with the capacity of 1 ton/h. And far-infrared dryers of batch and continuous types were developed to dry hygienically the wet enriched rice. The percentages of lysine, thiamine, amino acid nitride, and Ca and Fe of the enriched rice samples produced in this study were, respectively, 4, 3, 3. 2, and 10 times higher than those of white rice, respectively.

Resveratrol-Enriched Rice Down-Regulates Melanin Synthesis in UVB-Induced Guinea Pigs Epidermal Skin Tissue

  • Synthetic compounds that are used in the clinic to regulate skin hyperpigmentation, such as arbutin, hydroquinone, and kojic acid, are only moderately effective. But, their use is limited by side effects. As part of an effort to overcome the limitations, we developed resveratrol-enriched rice (RR) using genetic engineering technique. Each of resveratrol and rice has been reported to produce anti-melanogenic effects. Therefore, we hypothesized that RR would show more anti-melanogenic effects than those of resveratrol or rice alone. Anti-melanogenic effect of RR was done by using melan-a mouse melanocytes. The depigmenting efficacy was then observed following topical application of the RR to UVB-stimulated hyperpigmented dorsal skin of guinea pigs. Treatment with RR extract resulted a $21.4{\pm}0.7%$ decrease in tyrosinase expression at melan-a cells. Colorimetric analysis showed a significantly lower depigmenting value by day 9 following treatment with RR in UVB-irradiated guinea pigs the dorsal skin (p<0.01), indicating that RR produced a depigmentation effect. By staining with Fontana-Masson stain, we found that the RR-treated group had more effect histopathologically in epidermal melanin production than resveratrol or rice alone-treated group. RR was associated with reduction in the levels of microphthalmia-associated transcription factor (MITF), and downregulation of tyrosinase and tyrosinase-related protein (TRP-2) expression, leading to inhibit epidermal melanin production by western blot analysis. This study suggests that the resveratrol-enriched rice may be a promising candidate in regulating skin pigmentation with UVB exposure.

논과 갯벌에서 톨루엔의 혐기성 생분해에 미치는 전자수용체의 영향 (Effect of Electron Acceptor on Anaerobic Toluene Biodegradation in Rice Field and Tidal Mud Flat)

  • In oil-contaminated environments, anaerobic biodegradation of toluene depended on the concentration and distribution of terminal electron acceptor as well as the physicochemical properties such as DO concentration, redox potential and pH. This study showed the anaerobic biodegradation of toluene in two different soils by using nitrate reduction, ferric iron reduction, sulfate reduction and methanogensis. Toluene degradation rates in the soil samples taken from rice filed and tidal mud flat by nitrate reduction were higher than those by other processes. Tho soil samples from the two fields were enriched for 130 days by providing toluene as a sole carbon source and nitrate or sulfate as a terminal electron acceptor. The toluene degradation rates in the enriched denitrifying consortia obtained from the rice field and tidal mud flat soil were 310.7 and 200.6 $\mu$mol$ L^{-1}$ / $d^{-1}$, respectively. The toluene (legradation rates in the enriched sulfate-reducing consortia from the fields ranged fi-om 149.1 to 86.1$\mu$mol $L^{-1}$ / $d^{-1}$ .

고령 소비자를 위한 식이섬유 강화식단 개발 및 관능적 이화학적 특성 분석 (Development of Dietary Fiber Enriched Menu for the Aged and Analysis of Sensory and Physicochemical Characteristics)

  • Four dietary fiber enriched menus for the aged were developed and physicochemical were analyzed. Major foods enriching the dietary fiber were as following; kimchi & bean sprouts soup and sweet potato for menu 1, chamcheunamul and grape for menu 2, mushroom and kimchi for menu 3, dried radish leaves and green pepper for menu 4. One portion of calorie, carbohydrate, protein and fats for the aged were calculated as 567 kcal, 92.8 g, 21.3 g and 12.6 g, respectively based on recommended daily allowances for adults. Dietary fiber content increased in the menus 1, 2, 3 and 4 were 17.27%, 20.84%, 16.91%, and 23.64%, respectively. The menu 1 had significantly the highest acceptance scores among the rice, soups, side dishes and overall desirability with the acceptance sores of 4.77, 4.58, 4.62 and 4.69, respectively(p<0.05). Female showed significantly the higher acceptance scores of all of the enriched fiber menu than those of male. Moisture contents of the boiled rice with barely in menu 3 had significantly the highest value of all(p<0.05) and the side dishes of all menus did not show any significant differences at p<0.05. The pH value of boiled rice with barely in menu 3 was significantly the highest and menu 1 of kimchi & bean sprouts soup had significantly the lowest pH values(p<0.05). Hunter L value was significantly the highest in boiled rice with barley. The dietary fiber enriched menus were successfully developed for application and standardization in the meal service planning for the aged as one of trials of basic data collection.

고령소비자를 위한 단백질 강화 식단 개발 및 기호도 조사 (Development and Acceptance Test of Protein Enriched Menu for the Aged)

  • The purpose of this study was to develop protein enriched menu for the aged and running an acceptance test on the protein enriched menu. Four kinds of the menu were developed with emphasis on protein enriched food materials of chicken and beans for menu1, chunggukjang for menu2, pork for menu3, beef and mackerel for menu4. one portion of calorie, polysaccharide, protein and fats for the aged were calculated at 567kcal, 92.8g, 21.3g and 12.6g respectively based on recommended daily allowances for adults. Protein content increased in the menus 1, 2, 3 and 4 were 13.2%, 17.5%, 12.4%, and 13.3%, respectively. In the result of the acceptance test, the cooked rice with black beans and chicken soup of menu1 had significantly the highest acceptance score among the rice and soup groups, respectively (p<0.05). In the side dish, bean curd, cucumber and onion salad in menu1 had relatively higher acceptance scores than the others with significant difference at p<0.05. Overall acceptance of menu 1 had significantly the highest acceptance scores of all. Female showed significantly the higher scores of overall acceptance on menu 2 and 4 than those of male from the t test results of acceptance test.

Production of $\beta$-Carotene-Enriched Rice Bran Using Solid-State Fermentation of Rhodotorula glutinis

  • This work was aimed at utilizing rice bran as a substrate for $\beta$-carotene production by Rhodotorula glutinis DM 28 under optimized conditions of solid-state fermentation. The biomass and $\beta$-carotene content of Rhodotorula glutinis DM 28 grown on rice bran as a sole substrate under solid-state fermentation were 54 g/kg rice bran and 1.65 mg/kg rice bran, respectively. Its biomass and $\beta$-carotene content, however, could be improved by 60% and 30%, respectively, using the Central Composite Design for the optimization of its cultivation conditions. The optimized conditions obtained were a pH of 5, a moisture content of 70% (w/w), and a carbon-to-nitrogen ratio of 4. Under these conditions, rice bran containing R. glutinis DM 28 had nutritional values of $\beta$-carotene, protein, and fat higher than those of rice bran alone. Yeast-grown rice bran could be suitable, therefore, to use as a $\beta$-carotene-enriched supplement in animal feeds.

전통증편의 단백질보강에 관한 연구 (Processing Conditions for Protein Enriched Jeung-Pyun (Korean Fermented Rice Cake))

  • 쌀을 주원료로 한 재래식 증편에 단백질을 보강하기 위하여 콩단백질(볶은 콩가루, 콩단백추출물)을 첨가한 증편의 최적 가공조건을 구명하고 제품의 전분 및 단백질 품질을 평가 하였다. 쌀가루에 대하여 콩가루 25%(w/w), 설탕 20%(w/w), 막걸리 50%(v/w)로 섞어 $35^{\circ}C$에서 3시간 발효 시켰을 때 부피팽창이 가장 컸으며 설탕을 첨가하지 않으면 거의 발효는 진행 되지 않았고 설탕 첨가량이 많아질수록 반죽이 질어져 성형이 곤란 하였다. 또한 콩가루 대신 soy protein isolate(SPI)를 첨가 하였을 때는 쌀가루에 대하여 SPI 15%, 설탕 20%, 막걸리 60%, 발효시간 2시간, 발효온도 $30^{\circ}C$에서 가장 크게 부풀었다. 발효된 반죽을 수증기로 찌면 발효 부피의 50%정도 부피가 줄어들며 발효시간이 긴것일수록 부피가 크게 줄어 들었다. 부피가 가장 크게 팽창된 조건에서 만든 발효반죽을 15분간 쪄서 제조한 증편의 전분품질(소화율, 환원력) 및 단백질 품질 (소화율, 단백효율비)은 가장 우수 하였으나, 관능검사 및 textrometer에 의한 질감검사의 결과는 콩가루나 SPI첨가 비율이 높을수록 불리하였다. 그러므로 최적 부피 팽창 발효조건과는 달리 관능 및 질감이 좋은 증편은 쌀가루에 대하여 콩가루 20%, SPI 5% 정도의 수준으로 첨가하여 제조 가능할 것으로 생각되었다.

Coordination of Basic and Development Researches on Vitamins

  • Reviewed biosynthetic pathways in green leaves of thiamine, riboflavin and folate as disclosed by the authors, and the regulatory systems operating on their biosynthesis and biodegradation as revealed since a potent inhibitory activity of ATP was found for the phosphatase reactions in 1965. Physiological roles of ascorbic acid in plant were evidenced by its higher content in green and flower leaves; the content in persimmon leaves for example was found ten or more times of that in citrus fruits. A close relationship existing between basic and development researches was illustrated by examples chosen from the author's researches. Examples cited were as follows; basic researches on vitamins were extended to the creation of thiamine enriched rice, to the growth promotion of rice plant by foliar application of thiamine, and to the use of ascorbic acid to prevent the appearance of beer cloudness; histological and biochemical studies on protein bodies in rice endosperm turned out amino acid enriched rice; techniques acquired in the studies on catalase were successfully employed for the utilization of single cell protein (MIPRON) for human food; conception of hybernation was led up to under-water or under-ground storage of cereal grains.

Translocation of Seed Storage Proteins into Microsomes Isoalted from Rice Endosperm Cells

  • Developing rice endosperm cells display two morphologically distinct rough endoplasmic reticulum (ER) membranes, the cisternae ER (C-ER) and theprotein body ER (PB-ER), the latter delimiting the prolamine protein bodies. We (Li et al., 1993) have recently shown that the storage protein mRNAs are not randomly distributed on these ER types; the C-ER is enriched for glutelin mRNAs, whereas the PB-ER harbors predominantly prolamine transcripts. To address whether these ER types have differnet capacities to translate these mRNAs and translocate their proteins into the lumen, a microsomal fraction enriched in C-ER vesicles was prepared from devleoping rice seeds. When present in an in vitro translatin system, the microsomes were able to proteolytically remove the signal peptide and internalize both preproglutelin and preprolamine within the microsomal vesicles. Of the two species, preprolamine was more effectively translocated and processed. These results suggest that the C-ER has the capacity to recognize and bind both storage protein mRNAs during protein synthesis. Moreover, efficient translocation and processing of glutelin requires additional factors that are deficient or absent in the in vitro system.

Anti-Melanogenic Potentials of Nanoparticles from Calli of Resveratrol-Enriched Rice against UVB-Induced Hyperpigmentation in Guinea Pig Skin

  • We already reported that genetically engineered resveratrol-enriched rice (RR) showed to down-regulate skin melanogenesis. To be developed to increase the bioactivity of RR using calli from plants, RR was adopted for mass production using plant tissue culture technologies. In addition, high-pressure homogenization (HPH) was used to increase the biocompatibility and penetration of the calli from RR into the skin. We aimed to develop anti-melanogenic agents incorporating calli of RR (cRR) and nanoparticles by high-pressure homogenization, examining the synergistic effects on the inhibition of UVB-induced hyperpigmentation. Depigmentation was observed following topical application of micro-cRR, nano-calli of normal rice (cNR), and nano-cRR to ultraviolet B (UVB)-stimulated hyperpigmented guinea pig dorsal skin. Colorimetric analysis, tyrosinase immunostaining, and Fontana-Masson staining for UVB-promoted melanin were performed. Nano-cRR inhibited changes in the melanin color index caused by UVB-promoted hyperpigmentation, and demonstrated stronger anti-melanogenic potential than micro-cRR. In epidermal skin, nano-cRR repressed UVB-promoted melanin granules, thereby suppressing hyperpigmentation. The UVB-enhanced, highly expressed tyrosinase in the basal layer of the epidermis was inhibited by nano-cRR more prominently than by micro-cRR and nano-cNR. The anti-melanogenic potency of nano-cRR also depended on pH and particle size. Nano-cRR shows promising potential to regulate skin pigmentation following UVB exposure.
