Distributive Characterization of Estrogenic Activity in Sediments from Gwangyang Bay, Korea (광양만 퇴적물에서의 에스트로겐 활성분포 특성)

  • In this study, we tried to quantitatively study the distribution of estrogenic activity in sediment from Gwangyang Bay by E-screen assay. Besides, we compared the estrogenic activity and the concentration of chemical pollutants. The highest estrogenic activity was recorded at the stations(GY6 and GY8) close to industrial complex and the river mouth of Seomjin. These results obtained from the E-screen assay similar to those of simultaneous analytical detection of 310 chemicals. In particular, GY6 and GY8 sites are confirmed as the full agonist sites because of their RPE values were over 90% having strong estrogenic effect. Also, their EEQ(Estradiol Equivalency Quantity) values are 35.6 ng/g and 14.6 ng/g, low than that of other sites, and these results suggests that have relatively high estrogenic efficiency in Gwangyang Bay. From these results, we can estimate that the stations close to industrial complex and the river mouth of Seomjin are major sources of endocrine disrupter in Gwangyang Bay. On the other hand, when we tried to compare the endocrine disrupter activity and $COD_{Mn}$ value, that is not correlated.

An immunohistochemical study of the endocrine cells on the gastro-entero-pancreatic system of the African clawed toad, Xenopus laevis (아프리카발톱두꺼비(Xenopus laevis)의 위장췌내분비세포의 면역조직화학적 연구)

  • The GEP endocrine cells of the African clawed toad, Xenopus laevis, were studied immunohistochemically. Five kinds of the endocrine cells were identified in this study A moderated number of 5-HT-immunoreactive cells were detected throughout the gastro intestinal tract, being almost uniform frequency. Gas/CCK -immunoreactive cells were restricted to the basal portion of the pyloric gland and among the duodenal mucosa. A rare glucagon-immunoreactive cells were weakly reacted in the fundic region of the stomach and observed in the exocrine portions of the pancreas. Somatostatine-immunoreactive cells were distributed throughout the gastrointestinal tract with except for the rectum, and not only the periphery of the islets but also the exocrine portions in the pancreas. No CGs- and insulin-immunoreactive cells were found in the gastrointestinal tract, whereas in the pancreas, the later was seen in the central region of the islets and the exocrine portions.

An Analysis of Requisite Knowledge Body of Physiology for Nursing Education (간호학 관점에서 본 생리학 지식체 내용 분석)

  • The purpose of this study is to define requisite content of physiology for nursing education. This study classifies the subjects of physiology into 15 areas. The areas are cell and cell membrane, body fluid, nervous system, special sense, muscular system, blood, cardiovascular system, respiratory system, urinary system, digestive system, energy metabolism, body temperature, immune system, endocrine system, and reproductive system. Each subject area is further classified into subarea, resulting in a total of 194 subarea. The importance of each subarea is measured with a 3-point scale using a questionnaire. The subjects of this study were full-time professors teaching nursing in Korean universities. The analysis of the data collected from 68 respondents is as follows. 1. The areas of physiology necessary for nursing education in the order of importance are : body fluid, blood, endocrine, immune system, body temperature, urinary system, respiratory system, digestive system, reproductive system, energy metabolism, nervous system, cardiovascular system, cell and cell membrane, muscular system, and special sense. 2. Depending on the specific areas of nursing(such as pediatric nursing, maternity nursing), the importance of each physiology area may differ. For instance, the most important area for maternity nursing is reproductive physiology, whereas one for the psychiatric nursing is neuro-physiology. 3. The importance of each physiology area does not determine the importance of its subarea. For example, while the importance of cell and cell membrane was relatively very low across the respondents from different areas of nursing science, the importance of such subarea as osmosis, diffusion, and filteration reported by most respondents were high.

  • There is a concern that chemicals in our environment are affecting human health by disrupting a normal endocrine function. Much of the concern has focused on chemicals that can interact directly with steroid hormone receptors. The ability of certain man-made chemicals to mimic the effects of natural steroid hormones and their potential to disrupt the delicate balance of the endocrine system in animals are of increasing concern.   • Endocrine disruption in crucian carp (Carassius auratus) living in the branch of Han River were examined. Vitellogenin level in plasma was measured using ELISA system and aromatase mRNA level in brain was observed using RT-PCR technique. In all female fish, vitellogenin levels were in the range of 20∼40 $\mu\textrm{g}$/ml and aromatase mRNA expression could be detected on the agarose gel after RT-PCR. However, in case of males, vitellogenin level was elevated in only one fish, while vitellogenin was hardly detected in others. Aromatase was expressed in all males although the levels were relatively lower than the level in female fish. Testis-ova and any other histological changes of reproductive organ were not shown in both sexes.

  • The safety of human exposure to an ever-increasing number and diversity of electromagnetic field (EMF) sources both at work and at home has become a public health issue. To date, many in vivo and in vitro studies have revealed that EMF exposure can alter cellular homeostasis, endocrine function, reproductive function, and fetal development in animal systems. Reproductive parameters reported to be altered by EMF exposure include male germ cell death, the estrous cycle, reproductive endocrine hormones, reproductive organ weights, sperm motility, early embryonic development, and pregnancy success. At the cellular level, an increase in free radicals and $[Ca^{2+}]i$ may mediate the effect of EMFs and lead to cell growth inhibition, protein misfolding, and DNA breaks. The effect of EMF exposure on reproductive function differs according to frequency and wave, strength (energy), and duration of exposure. In the present review, the effects of EMFs on reproductive function are summarized according to the types of EMF, wave type, strength, and duration of exposure at cellular and organism levels.

  • A large number of chemical pollutants including phthalates, alkylphenolic compounds, organochlorine pesticides and bisphenol A have the ability to disrupt endocrine function in animals. and alter cognitive function. Because hormone mediated events playa important role in central nervous system development and functions.   • The somatotropic axis plays a key role in proliferation and differentiation of avian organs during both pre- and posthatching periods. This review discusses the complexity of regulation of the endocrine system for chicken development and growth by growth hormone (GH), insulin-like growth factor (IGF), and IGF binding protein (IGFBP). In addition, the thyrotropic axis, including thyrotropin-releasing hormone (TRH) and thyroid hormones ($T_4$ and $T_3$), is also involved in the GH-secreting pattern. In mammals, IGFI and -II are always sequestered in a 150 kDa non-covalent ternary complex. This complex consists of one molecule each of IGF-I or IGF-II, IGFBP-3 or IGFBP-5 and an acid labile subunit (ALS). Chick ALS is identified in different strains for the first time, and further investigation of the expression of ALS on developmental stage and ALS effect on IGF bioavailability may be addressed in the future.

  • 인류는 1,500 만 종류에 달하는 화학물질을 합성 혹은 분리ㆍ동정 해왔다. 그리고 현재 약 10 만 종류 정도의 화학물질이 상업적으로 이용되고 있다. 이들 화학물질 중에 유기염소화합물, 공업용화합물, 농약류, 유기취소(bromine)화합물, 중금속 및 유기금속, 식물 및 합성에스트로겐 등을 포함한 수십 종의 화합물이 내분비계장애물질 (Endocrine Disruptors, EDs)로서 생각, 혹은 의심되고 있다. 그러나, 대부분의 다른 화합물에 대해서는 그것들이 내분비계장애 작용을 가지고 있는지에 대한 충분한 지견이 얻어지지 않고 있어 이들 화학물질의 내분비계장애 작용을 평가하기 위해서는 신속 또는 간편한 검출계의 확립은 매우 중요하다.

Effect of Endocrine Disrupting Chemicals on Bombina orientalis Aromatase Activity Expressed in Cultured Mammalian Cells

  • In this study we present a mammalian cell culture model that allows to study the effect of endocrine disruptors (EDCs) on aromatase activity of aquatic amphibian, Bombina orientalis. Bombina orientalis aromatase gene was subcloned into a mammalian expression vector and subsequently transfected to mammalian cells. Although the protein expression level of Bombina orientalis aromatase was low, it had a significant aromatase activity. When EDCs were added to aromatase transfected cells, aromatase activity was significantly decreased. We report here that this system may be used to monitor the effect of EDCs on aromatase activity of aquatic organisms.
