Detection of Endocrine-Disrupting Chemicals in Fish and the Use of Fish Vitellogenin as a Biomarker (어류를 이용한 내분비계 장애물질 검출 및 Biomarker로서 Vitellogenin의 이용)

  • Fish vitellogenin produces in female liver during oogensesis through estradiol cycle, and produces even in male liver by endocrine-disrupting chemicals (EDCs) such as alkylphenols. The resulting effects of EDCs lead to the low fecundity of female and the feminization (eg. shrinkage of testis) in male. Especially, the production of vitellogenin in male indicates the environmental contamination of EDCs, resulting in the modulation of gene expression profiles and the monitoring of environmental contamination at specific area. In this paper, we suggest that fish vitellogenin is useful for biomonitoring for environmental contamination and would be substantially useful as a biomarker for a detection of EDCs in aquatic environment.

  • Bisphenol A(BPA), known as a typical endocrine disruptor, has been used commercially and widely for plastics and epoxy resins. BPA-based plastic is used extensively for the production of water bottles, food containers, CDs, DVDs, and panels that can be applied in construction. Epoxy resins containing BPA are used for coatings on the insides of water pipes, food cans, and thermal papers that are used in sales receipts. As its estrogenic effects and other adverse health effects have published, BPA has been regulated in many countries, and there have been efforts made to replace BPA. Other bisphenols substitutes such as bisphenol S(BPS) and bisphenol F(BPF) have been used. Currently, BPS- and BPF-based products labeled BPA-free products have been widely consumed. Because of structural similarities with BPA, however, these alternatives also show endocrine disruption effects like BPA, and many studies on adverse health effects of these alternatives are being reported. In this review, we describe the adverse health effects of bisphenols and the current status of regulation.

Development of Quantitative Vitellogenin ELISAs for Goldfish(Carassius auratus) used in Endocrine Disruptor Screening (내분비 장애물질 측정을 위한 붕어 비탈로제닌 정량 분석 ELISA 시스템 개발)

  • 난황단백질인 비탈로제닌을 성숙한 암컷 붕어 혈청으로부터 음이온 교환 크로마토그래피를 이용하여 정제 하였다. 정제한 비탈로제닌을 BALB/c mice를 이용하여 폴리크로날 항체를 생산하였고 이를 protein A column을 사용하여 정제하였다. 또한 이렇게 정제된 폴리크로날 항체를 이용한 붕어 비탈로제닌 측정용 효소면역측정법을 개발하였으며 그 측정 범위는 2∼l,000ng/mL이고 recovery 변동 범위는 88∼112%였다. 또한 이 효소면역측정법을 평가하기 위해 성숙한 수컷 붕어를 1,000ng/L ethinylestradiol(EE$_2$) 에 4주 동안 노출시켜 유도되어지는 비텔로제닌을 측정하였다. 그 결과 성숙한 수컷 붕어의 경우 비탈로제닌이 3주 만에 암컷 붕어의 평균수치만큼 유도됨을 알 수 있었다.

  • Diesel exhaust is suspected to cause acute and chronic adverse effects on health. In recent. the effect of estrogenic endocrine disruptor in diesel particulate matter was little studied. Therefore, we examined the estrogenic activity of respirable diesel exhaust particulate matter derived from diesel engine motor. PM2.5 diesel exhaust of vehicle was collected using a high volume samples equipped with a cascade impactor. (omitted)

  • Dibutyl phthalate (DBP) is used extensively in the plastic industry and has been known as an endocrine disruptor. Present study was undertaken to examine whether DBP can induce oxidative stress in mice. In this study, oxidative stress was measured in terms of the modification of lipid peroxidation and gamma-glutamyl transferase (GGT) activity. The serum toxicity index, level of lipid peroxidation and triglyceride (TG), and activity of GGT were measured in male ICR mice after a single administration of DBP (5 g/kg, po). DBP did not alter serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine, glucose and cholesterol level. However, the treatment with DBP was found to significantly increase the level of lipid peroxidation in liver and lung. The TG content and activity of GGT in the liver of DBP-exposed animals was also increased. These results indicate that DBP can induce mild oxidative stress in mice. The GGT activity is considered to be increased as one of the adaptive defense mechanisms to oxidative stress induced by DBP.

  • Many of thyroid hormones disrupting chemicals induce effects via interaction with thyroid hormone and retinoic acid receptors and responsive elements intrinsic in target cells. We studied thyroid hormones disrupting effects of food additives and contaminants including BHA, BHT, ethoxyquin, propionic acid, sorbic acid, benzoic acid, CPM, aflatoxin B1, cadmium chloride, genistein, TCDD, PCBs and TDBE in recombinant HeLa cells containing plasmid construct for thyroxin responsive elements. The limit of response of the recombinant cells to T3 and T4 was $1\times10^{-12}\;M$. BHA. genistein, cadmium and TBDE were interacted with thyroid receptors with dose-responsive pattern. In addition, BHA, BHT, ethoxyquin, propionic acid, benzoic acid, sorbic acid, and TBDE showed synergism while cadmium chloride antagonism for T3-induced activity. This study elucidates that recombinant HeLa cell is sensitive and high-throughput system for the detection of chemicals that induce thyroid hormonal disruption via thyroid hormone receptors and responsive elements. Also this study raised suspect of BHA. BHT, ethoxyquin, propionic acid, benzoic acid, sorbic acid, TBDE, genisteine and cadmium chloride as thyroid hormonal system disruptors.

  • 최근 산업사회의 발전에 따라 수계 오염문제가 심각하게 대두되고 있다. 따라서 이들 오염물질에 대한 모니터링을 위한 방법들이 강구되고 있으며, 이 중에서 어류 간의 cytochrome P450 호소계는 지방친화성 물질의 오염상태를 평가할 수 있는 물질로 알려지고 있다(Payne et al., 1987). PCBs(Polychlorinated biphenyls)와 HCB(Hexachlorobenzene)는 유기염소계 화합물로서 환경중에 널리 분포하고 있으며, 이중 PCBs는 신경계, 면역계 및 내분비계 등 생체내에 미치는 영향이 크므로 내분비장애물질(endocrine disruptor)로 분류되고 있고, HCB는 독성이 강한 화합물로서 주로 농업에서 fungicide로서 사용되어 왔으며. 다양한 산업 연소과정의 부산물로 발생되고 있다.   • The estrogenic potency of di -2-ethylhexyl phthalate (DEHP) using reverse transcriptase-PCR respouse of liver vitellogenin mRNA in male African clawed frog (Xenopus laevis) was studied. Male frogs were injected with DEHP at dose of 300$\mu\textrm{g}$/kg and 300 mg/kg body weight through the dorsal lymph sac. After 4 days, using suitable pair of RT-PCR primers, vitellogenin mRNA induction in the liver was measured and DEHP showed vitellogenin mRNA induction in only the group treated with 300 mg/kg. Any significant histological abnormalities by the exposure of DEHP was not shown in both testis and liver.
