Growth, Fecundity, Egg Size and Recruitment of Palaemon serrifer (Decapoda: Caridea: Palaemonidae)

  • Growth and reproduction of Palaemon serrifer were described and analyzed in a population inhabiting tide pools in warm temperate waters in Korea. The water temperature varied greatly in the tide pools, ranging from 8$^{\circ}C$ to 27.8$^{\circ}C$ Population structure and growth were investigated using size frequency distribution data collected from January to December 2003. Sex ratios fluctuated, but were almost equal during the breeding period. Growth was continuous and size increased gradually throughout the year. Adult females were larger and grew faster than males. von Bertalanffy growth parameters for a one-year sample of females and males were estimated as $L_{i\ddot{A}}$ = 11.32, K = 0.311, $t_0$ = -0.4115 and $L_{i\ddot{A}}$ = 8.36, K = 0.228, $t_0$ = -0.9693 respectively. Breeding was seasonal, starting in May, peaking in August, and finishing by the end of August. The species showed continuous production of successive broods. Laboratory observation showed that females with embryos near hatching had ovaries filled with vitellogenic oocytes ready for spawning. The reproductive output (effort) of each female (mean number of eggs: $552{\sim}1355$) was not high. The mean embryo volume, $0.078mm^3$, is relatively small, indicative of low energy allocation to each embryo. Recruitment of juveniles was closely linked to the breeding period, beginning in September.

Ferritin Light Heavy Chain 유전자가 도입된 인삼형질전환체의 단일배발생을 통한 식물체의 기내증식 (In vitro Propagation of Transgenic Ginsengs Introduced with Ferritin Light Heavy Chain Gene through Single Embryo Culture)

  • Ferritin light heavy chain (FLHC) gene는 일부 중금속과 결합, 저장 및 운반하여 무독화 시킬 수 있는 것으로 알려져 있다. Fe 관련 유전자인 FLHC유전자를 식물 발현용 promoter인 35S promoter와 Tnos를 사용하여 식물 형질전환용 vector를 재조합하였다. 식물세포형질전환용 binary vector는 상기 cassette vector가 조립이 매우 양호하며 border sequence를 가지고 있는 pRD400 binary vector를 사용하여 최종적으로 가나마이신 내성 유전자 (NPT II gene)와 tadpole ferritin heavy chain gene 및 human ferritin light chain gene를 함유하고 있는 binary vector를 재조합하였다. Binary vector의 아그로박테리움에 도입은 triparental mating 방법에 의하여 수행하여 AB배지 및 가나마이신 함유 배지에서 disarmed Ti-vector를 가지고 있는 Agrobacterium tumefaciens MP90/FLHC을 선발하였다. FLHC 유전자 도입된 식물형질전환용 binary vector를 이용하여 형질전환방법을 변형하여 많은 embryo를 유도하였으며 유도된 embryo들은 GA 10mg/L가 첨가된 배지에 지상부를 유도하였다. 형질전환체식물체의 정상적인 생장을 유도하기 위해 최적의 배양조건을 조사하였던 바, 비교적 1/3 MS배지에서 뿌리의 생장과 지상부의 생장이 균일하게 생장하는 경향을 보였으며, 뿌리와 줄기가 잘 발달된 약 7cm의 유식물체를 대량으로 증식하여, 모래와 흙이 1:1로 혼합된 토양에 옮겼다.

Effect of Heparin-binding Epidermal Growth Factor (HB-EGF) on Integrin $\alpha_{\nu}-\betaFe_3$ Expression in Preimplantation Mouse Embryos

  • Heparin-bindin epidermal growth factor (HB-EGF) is one of the EGF family to be expressed at the time of implantation in the mouse uterus. Although HB-EGF has been shown to stimulate the development of embryo and uterus in the mouse, its correlation between cell adhesion molecules remains undefined. Integrin $\alpha$$_{ν}$$\beta$$_3$, one of the cell adhesion molecules, is an important mediator of cell-substratum and cell-cell adhesion in implantation. In the present studies, we investigated the effects of HB-EGF on the embryonic development, initiation of implantation and expression of integrin $\alpha$$_{ν}$$\beta$$_3$ in in vitro culture, blocking of HB-EGF, RT-PCR and immunofluores cence analysis. The results showed that HB-EGF significantly improved the developmental rate of hatched embryos (24.1%, p<0.01) and outgrowth embryos (42.5%, p<0.01). On the other hand, this growth factor showed no offset before the hatching embryonic stage. Analysis of RT-PCR showed that HB-EGF upregulated the expression level of integrina $\alpha$$_{ν}$$\beta$$_3$ subunit genes on the preimplantation embryo and outgrowth of blastocyst (120hr and 144hr after hCG injection). Immunofluorescence analysis showed that the integrin $\alpha$$_{ν}$$\beta$$_3$ subunits localized at the pericellular borders and cell-cell contact areas. Increase in fluorescence intensity was observed in the HB-EGF treated embryos. Intrauterine injection of an anti-HB-EGF antiserum at day 3 significantly decreased the number of implantation sites (14.4, p<0.01) and significantly increased the number of recovered embryos(6.4, p<0.05) at day 5. From these results, it imply that HB-EGF improve the embryo development and accelerated the expression of integrin $\alpha$$_{ν}$$\beta$$_3$ in the preimplantation mouse embryos.

STUDIES ON THE TISSUE CULTURE OF PANAX GINSENG

  • Unlike the tissue culture in animals and human being, in higher plants various parts of the plant are cultured for varied purposes, and they are named variously depending on which parts are used as explants or what purposes they are cultured for. Followings are some of the names of culture used frequently: organ culture, tissue culture, callus culture, single cell culture, meristem culture, mericlone culture, ovary culture, ovule culture, embryo culture, endosperm culture, anther culture, pollen culture, protoplast culture, etc.. As the names of the culture indicate, in some kinds of culture the explants used for culture are actually not tissues, but organs, single cells, or protoplasts. It seems, however, convenient to call all of the above-mentioned cultures grossly as tissue culture. Several kinds of tissue culture were attempted using Panax ginseng as material and some of the results were summarized below. 1. Callus culture After dormancy of the sed was broken, whole embryo or parts (hypocotyl, cotyledon and epicotyl) of partly grown embryo were cultured in the media supplemented with growth regulators. Rapid swelling occurred in a few weeks, but most of the swelling was observed only in the basal part of epicotyl, changes in the other parts of embryo appearing in much later stages. The swelling or increase in size, however, was resulted not from the divisions of cells, but from the mere expansion of cell. Real calli were formed about two months after inoculation of explants. Callus tissues developed from cortex, pith, and vascular bundle in the cases of hypo- and epicotyl, from mesophyl tissue in the case of cotyledon. Shoots developed more easily from cotyledons regardless of whether they are detached from or attached to the embryo proper. 2. Culture in the Knudson C medium When cotyledons, detached from or attached to the embryo proper, were cultured in the growth regulator-free Knudson C medium comprision only several kinds of mineral compounds and sucrose, shoot primordium or callus developed profusely and finally plantlets were produced directly from shoot primordium or indirectly through callus. In this medium epidermal cells as well as mesophyl cells of the cotyledon became meristematic and divided, changing into multinucleate cells or multicellular bodies, developing eventually into either shoot primordia or calli. 3. Anther culture Anthers were cultured in the media supplemented with various growth regulators applied singly or in combinations. Callus was formed mostly in the connective tissue of anther. Cells of anther wall layers changed in appearance, but no division occurred. Microspores of all stages in development were not changed, ruling out the possibility that microspore-originated callus might be formed. 4. Isolation of protoplast Protoplasts were isolated from young root, leaf, and epicotyl, using 0.7M D-mannitols as osmoticum and using macerozyme and cellulase respectively for maceration and digestion of the cell wall. Production in large number of naked intact protoplast was rather difficult as compared with other plant species. Fusion of protoplasts occurred infrequently mainly due to the fewer number of naked protoplasts in the solution.

냉습적(冷濕積)에 따른 은행나무종자내(種子內) 생장조정물질(生長調整物質)의 변화(變化) (Alteration of Endogenous Growth Substances in Cold-moist Stratified Seeds of Ginkgo biloba L.)

  • 휴면기간(休眠期間)이 긴 은행나무 종자(種子)의 냉습적처리(冷濕積處理)에 따른 생장촉진물질(生長促進物質)과 생장억제물질(生長抑制物質)의 증감(增減)을 구명(究明)하기 위하여 본(本) 연구(硏究)를 수행(遂行)하고 얻어진 결과(結果)를 요약(要約)하면 다음과 같다. 1. 무처리종자(無處理種子)는 배(胚), 배유(胚乳), 종피(種皮)의 각추출물(各抽出物)에서 생장억제물질(生長抑制物質)을 볼 수 있었으며, 특(特)히 종피(種皮)에서 가장 현저(顯著)하게 나타났다. 2. 냉습적처리종자(冷濕積處理種子)의 배(胚)와 종피(種皮)에서 생장억제물질(生長抑制物質)의 현저(顯著)한 감소(減少)가 있었으나, 생장촉진물질(生長促進物質)은 배부분(胚部分)에서 증가(增加)가 약간 나타났다. 3. 이상(以上)의 결과(結果)에서 은행나무 종자(種子)의 휴면(休眠)에는 종피(種皮), 배(胚), 배유(胚乳)의 각부분(各部分)이 생장억제물질(生長抑制物質)이 존재(存在)하며 발아(發芽)에 관계하는 것으로 추측(推測)된다. 4. 은행나무 종자(種子)와 발아촉진(發芽促進)과의 관계는 생장촉진물질(生長促進物質)의 증가(增加)보다는, 배(胚)와 종피(種皮)의 생장억제물질(生長抑制物質)의 감소(減少)에 영향된다고 추측(推測)된다. 5. 생장촉진물질(生長促進物質)인 Gibberellin은 자외선형광검사등검정(紫外線螢光檢査燈檢定)에서 Rf 0.26, 에서 나타났고, 생장억제물질(生長抑制物質)인 Abscisic acid는 Rf 0.62, Rf 0.70, Rf 0.78에서 각각(各各) purple, gray, blue의 형광색(螢光色)으로 나타났다.

혈관현성 및 혈관내피세포의 성장에 미치는 모유의 영향 (Effect of human milk on the angiogenesis and endothelial cell growth)

  • Human milk was examined for antiangiogenic activities by using the chick embryo chorioallantoic membrane (CAM) assay and endothelial cell growth. The low molecular weight (20 KD>$\sim$ ) fraction of human milk stimulated the angiogenesis and increased the endothelial cell growth. These results suggest that the increase of angiogenesis and endothelial cell growth might be attributed to several growth factors and/or angiogenic factors in low molecular fraction (20 KD>$\sim$) of human milk.

가토의 수정란이식에 관한 연구 I. PMSG와 HCG투여에 따른 난소반응 (Studies on Embryo Transfer in Rabbit I. Ovarian response to PMSG and hCG administration)

  • As a preliminary experiment to establish the process of embryo transfer in rabbit, present sutdies were carried out with 75 mature Japanese of ovary to pregnant mare's serum gonadotropin(PMSG) and human chorionic gonadotropin(hCG) and collection rate of embryos at various times after hCG injection. Female rabbits were superovulated using 50∼100IU hCG or 75∼100IU PMSG and 50∼751IU hCG injected 83hrs apart. The results obtained were as follows: 1. The average number of growth follicles obtained from all of rabbits treated with hCG or PMSG-hCG was 28.1. PMSG-hCG treatment group (30.9) was clearly increased more than hCG treatment group (16.7). 2. In ovulation score, PMSG-hCG treatment group (21.0) was increased more than hCG treatment group (7.9), showing the same trends in the growth of follicles. 3. The ovulation rate per follicles developed was higher in the rabbits treated with 100 IU PMSG and 75 IU hCG (18.9%) than that from the other groups. 4. The oviduct score (72.9%) was inclined to higher than that from uteri (57.1%) in score of embryo collection.

Signaling Molecules at the Conceptus-Uterine Interface during Early Pregnancy in Pigs

  • The process of embryo implantation requires physical contact and physiological communication between the conceptus trophectoderm and the maternal uterine endometrium. During the peri-implantation period in pigs, the conceptus undergoes significant morphological changes and secretes estrogens, the signal for maternal recognition of pregnancy. Estrogens secreted from the conceptus act on uterine epithelia to redirect $PGF_2{\alpha}$, luteolysin, secretion from the uterine vasculature to the uterine lumen to prevent luteolysis as well as to induce expression of endometrial genes that support implantation and conceptus development. In addition, conceptuses secrete cytokines, interferons, growth factors, and proteases, and in response to these signals, the uterine endometrium produces hormones, protease inhibitors, growth factors, transport proteins, adhesion molecules, lipid molecules, and calcium regulatory molecules. Coordinated interactions of these factors derived from the conceptus and the uterus play important roles in the process of implantation in pigs. To better understand mechanism of implantation process in pigs, this review provides information on signaling molecules at the conceptus-uterine interface during early pregnancy, including recently reported data reported.

병아리 태자에 대한 엔로플록사신의 최기형 독성 연구 (Teratological Effects of Enrofloxacin on the Embryos and Fetus During the Hatching of Chicken Eggs)

  • The purpose of this study was to investigate the teratological effects of enrofloxacin, a veterinary antibiotic, on the embryos and fetus of hatching chicken eggs. A control group and four experimental fertilized egg groups were set up. The four experimental groups were injected with 0.05 ml, 0.1 ml, 0.2 ml, and 0.3 ml enrofloxacin ($50{\mu}g/ml$) respectively. During the hatching period, the weights of total eggs, inside material(yolk and white) and embryos or fetuses, embryo growth, and teratological effects were investigated. In the experimental groups (fertilized eggs injected with 0.05 ml, 0.1 ml, 0.2 ml and 0.3 ml enrofloxacin), the weights of total eggs were decreased, but the yolk and white weights were increased by the higher amount of antibiotics. The weights of embryos (fetuses) in experimental groups were 86.1%, 78.6%, 65.6% and 61.4% of the control, respectively. Retarded growth, deformity and embryo loss were observed in experimental groups. Teratological effects such as undeveloped eyes, wings and legs, and deformed head and bill were also detected. In conclusion, we found that veterinary antibiotics, enrofloxacin have made teratological effects on the embryos and fetus of hatching eggs.

랫드 전배아배양법을 이용한 2-Bromopropane의 최기형성 평가 (Teratogenicity Evaluation of 2-Bromopropane Using Rat Whole Embryo Culture)

  • Recently, we have reported that the environmental pollutant 2-bromopropane (2-BP) induces a significant embryo-fetal developmental toxicity in rats. However, the cause of developmental toxicity and the relationship between maternal and developmental toxicities could not be elucidated because the developmental toxicity of 2-BP was observed only in the presence of maternal toxicity The in vitro teratogenicity study using whole embryo culture was carried out to understand the teratogenic properties and the possible mechanism of teratogenicity induced by 2-BP in rats. Rat embryos aged 9.5 days were cultured in vitro for 48 hrs at medium concentrations of 0, 1, 3, or 10 mg/ml of 2-BP. Embryos were evaluated for growth, differentiation, and morphological alterations at the end of the culture period. At 10 mg/ml, 2-BP caused a delay in the growth and differentiation of embryos and an increase in the incidence of morphological alterations, including altered yolk sac circulation, abnormal axial rotation, craniofacial hypoplasia, open neuropore, absent optic vesicle and kinked somites. At 3 mg/ml, only a delay in the growth and differentiation of embryos was observed. There were no adverse effects on embryonic growth and development at the concentration of 1 mg/ml. The results showed that the exposure of 2-BP to rat embryos results in a developmental delay and morphological alterations at dose levels of 3 mg/ml culture media or higher and that 2-BP can induce a direct developmental toxicity in rat embryos.
