Functional cosmetic properties and antimicrobial activity of extracts from Saururus chinensis by elicitor treatment (Elicitor 처리에 의한 삼백초 추출물(Saururus chinensis)의 미용식품활성 및 피부상재균 억제효과)

  • In this study, the functional cosmetic activity of water and ethanol extracts from Saururus chinensis by elicitor treatment are examined. Anti-oxidative activity 1,1-diphenyl-2-picrylhydrazyl and antioxidant protection factor of elicitor treated S. chinensis extract increased in concentration dependent manner as concentration increased from 1.5 mg elicitor/leaf 1 g (group A) to 3.0 mg elicitor/leaf 1 g (group B). Also, 70% ethanol extract showed higher anti-oxidative activity than water extract. Elicitor treated S. chinensis extract showed similar inhibitory effect on elastase activity and whitening effect indicating low effect of elicitor on elastase inhibition and whitening effect. However, it showed high anti-inflammatory and anti-microbial activity compared to control group. Both group A and B showed high anti-inflammatory and anti-microbial activity then control group indicating that elicitor treatment is suitable for biomass method used for industrializing useful compounds. Therefore, elicitor treatment during S. chinensis cultivation will increase bioactivity in the plant source which will yield high quality for industrialization.

  • This study was designed to assess the change of the antioxidative and biological enzyme activities [tyrosinase, elastase, collagenase, and hyaluronidase (HAase)] of extracts from elicitor-treated Oplismenus undulatifolius. Elicitor-treated O. undulatifolius showed higher total phenolic content than the non-treated extract. As a result of comparing the anti-oxidant activity of elicitor-treated O. undulatifolius extract and non-treated extract, the elicitor-treated group showed high activity. Elicitor-treated O. undulatifolius showed higher elastase and collagenase inhibitory activities as anti-wrinkle effect, tyrosinase inhibitory activity as whitening effect, and anti-inflammation effect to induced as HAase inhibition than the non-treated extract. Therefore, elicitor-treatment during O. undulatifolius cultivation in outdoors will elevate total phenolics content in the plant and elevate of various bioactivities, which will yield high quality for industrialization.

  • Effects of various elictitors were investigated to enhance the production of berberine in plant cell suspension cultures of Thalicrtrum rugosum. Treatments of yeast elicitor, 15 different types of abiotic elicitors, 16 kinds of fungal elicitors from three species of fungi were performed. Cell growth and berberine production were examined and compared for both normal and elicitor treated cultures. No distinguished increases in berberine yield by the addition of elicitors could be attained.

  • Elicitor treatment was performed to increase the anti-inflammatory activity of useful biological sources. The result showed that elicitor-treated Saururus chinensis leaf extracts positively affected nitric oxide (NO) production, and the expression of inducible NO synthase and cyclooxygenase-2 compared to extracts not exposed to elicitor treatment. This finding identified elicitor treatment as a suitable strategy for increasing the biological activity of S. chinensis. Therefore, elicitor-treated S. chinensis is useful both as health functional and medicinal materials.

  • Extracellular capsidiol, sesquiterpenoid phytoalexin, in the medium of pepper (Capsicum annuum L.) suspension cells was not identified from control cells, but highly accumulated in the elicitor-induced cells within 6 hours after the addition of 0.05$\mu\textrm{g}$/$m\ell$ cellulase. Capsidiol production in elicitor-induced cells was markedly suppressed by cytochrome P450 inhibitors, such as ancymidol and ketoconazole demonstrating that biosynthesis of capsidiol is catalyzed by at least on hydroxylation enzyme in the biochemical pathway. Based on protein electrophoresis, two bands, 23.0kDa and 27.5kDa, were identified as newly synthesized polypeptides in the elicitor-induced suspension cells, suggesting that pepper cells which were subjected to elicitor treatment activate specific gene(s) for capsidiol biosynthesis in cultured cells.

  • In order to improve anthocyanin production, effects of fungal elicitation in hairy root culture were investigated. fungal elicitor prepared from Fusarium moniliforme was the best in enhancement of anthocyanin production among the eight fungal elicitors tested. The optimum treat-ment time and concentration of treated elicitor for anthocyanin production were 12 hours and 3.28 mg carbohydrates per liter medium. Also, fungal elicitor was treated to hairy root culture in flat-bottomed fluidized-bed bioreactor. The anthocyanin production of elicited culture was enhanced 227% than non-treated.

Convenient Assay of O2- Generated on Potato Tuber Tissue Slices Treated with Fungal Elicitor by Electron Spin Resonance - No Secondary Oxidative Burst Induction by H2O2 Treatment

  • Since the discovery of generation of $O_2^-$ in plant, many evidence for the oxidative burst (OXB) has been accumulated in various combinations of plant and pathogen or elicitor systems. $O_2^-$ generating system responsible for the OXB was coupled with oxidation of reduced nicotinamide adenine dinucleotide phosphate (NADPH) in microsomal fraction isolated from sliced aged potato tuber slices which were treated by hyphal wall components elicitor from Phytophthora infestans (HWC). We developed new assay method for quantitative measurement of oxygen radical $O_2^-$ by using electron spin resonance (ESR) analysis during elicitor­induced OXB on the surface of plant tissues. The ESR analysis using an $O_2^-$ trapper, Tiron (1,2-dihydroxy-3,5­benzenedisulfonic acid), provided a convenient assay for detecting only $O_2^-$ during elicitor-induced OXB producing various active oxygen species (AOS) on plant tissue surface. Tiron was oxidized to Tiron semiquinon radical by $O_2^-$. Quantity of the radical signal was measured by specific spectra on ESR spectroscopy. The level of $O_2^-$ was high in from surface of potato tuber tissue treated with hyphal cell wall elicitor (HWC) from Phytophthora infestans. There was no secondary OXB induction by $H_2O_2$ treatment in plant.

  • It was investigated that systemic acquired resistance (SAR) was induced in plant treated with a elicitor, which was derived from a non-virulent fungus. The elicitor, a hyphal cell wall component derived from fungus, induced a production of phytoalexin and a generation of reactive oxygen species (ROS) in potato treated with its low level concentrations. The effect of the fungus-derived elicitor was better than that of virulent pathogen-derived elicitor, which was well known in potato. These results, therefore, suggested potentcial use of fungus-derived elicitor as a new plant protector for commercial development.

  • Effects of ultraviolet stress and elicitors, cellulase and jasmonic acid (JA), for the production of capsidiol, sesquiterpenoid phytoalexin, in seedlings and suspension cultures of pepper (Capsicum annum L. cv, Soobicho) were examined. Extracellular capsidiol in the medium of suspension cultures was absent from control cells, but accumulated in the elicitor treated cells with 0.05 $\mu\textrm{g}$/mL of cellulase or 0.1 $\mu\textrm{g}$/mL JA. Elicited cells gradually decreased their viability and eventually died within 48 hours of elicitor treatment by the toxicity of capsidiol accumulated in the culture medium. Capsidiol production in the leaves of pepper seedlings was markedly increased by the treatment of ultraviolet stress and reached maximum level at 48 hours of irradiation. Infiltration of elicitors, 0.05 $\mu\textrm{g}$/mL cellulase or 1.0 $\mu\textrm{g}$/mL JA, to the surface of leaf or fruit, stimulated the elicitation of the cells which resulted in the production of capsidiol and expansion of pathogene-like lesion around the elicitor treated region.

Production of Salidroside in Rhodiloa sachalinensis A. Bor Callus by the Elicitation and Precursor (참돌꽃 (Rhodiola sachalinensis A. Bor) 캘러스에서 elicitor와 전구체에 의한 Salidroside 생산성의 변화)

  • The effect of elicitor and precursor on salidroside production from Rhodiola sachalinensis A.Bor callus cultures was investigated. Callus cultures were treated with yeast extract, soft-ferrite ceramics powder, methyl jasmonate, ascorbic acid, jasmonic acid and $CuCl_2$/$CdCl_2$ as an elicitor. When callus cultures were treated with $0.2g/\ell$ of yeast extract, salidroside production from callus treated with yeast extract is 3.45 times higher than that of the controlled group. Among of them, callus cultures treated with yeast extract produced the highest salidroside. Callus cultures were treated with L-phenylalanine and L-tyrosine as a precursor for 4 days. The result of salidroside content analysis showed that all feeding of precursors not affected salidroside production from callus cultures. In case of L-tyrosine fed into callus cultures, both callus growth and salidroside production decreased at all concentrations.
