Marked effect of Cuscuta on puerarin accumulation in cell cultures of Pueraria tuberosa grown in shake flasks and a bioreactor

  • Isoflavonoid production in cell cultures of Pueraria tuberosa as influenced by an angiospermic parasite, Cuscuta reflexa, was studied. During the time course, maximum isoflavonoid content was recorded when Cuscuta elicitor was added on day 15 of culture. Among various concentrations of elicitor tried, $1g\;l^{-1}$ of Cuscuta elicitor was found to be the most effective. The optimized elicitation conditions were used in vessels of varying capacity where maximum yield of ${\sim}91mg\;l^{-1}$ of isoflavonoid was recorded in a 2-l bioreactor which was about 19% higher than the control cultures. In this case, puerarin content increased up to $11mg\;l^{-1}$ which was 580% higher that the value recorded in the control cultures. In the bioreactor, 8 days of elicitation was optimal for the high accumulation of isoflavonoid, giving productivity of ${\sim}4mg\;l^{-1}\;day^{-1}$. The study showed persistent high isoflavonoid yield even during scale-up. Use of a preparation of Cuscuta reflexa as an elicitor is reported for the first time. The increase in isoflavonoid content was elicitor dose-dependent and can be explored to trigger high yields of isoflavonoid/secondary metabolites in production.

  • The discovery of tacit knowledge from domain experts is one of the most exciting challenges in today\`s knowledge management. The nature of decision knowledge in determining the quality a firm\`s short-term liquidity is full of abstraction, ambiguity, and incompleteness, and presents a typical tacit knowledge extraction problem. In dealing with knowledge discovery of this nature, we propose a scheme that integrates both knowledge elicitation and knowledge discovery in the knowledge engineering processes. The knowledge elicitation component applies the Verbal Protocol Analysis to establish industrial cases as the basic knowledge data set. The knowledge discovery component then applies fuzzy clustering to the data set to build a fuzzy knowledge based system, which consists of a set of fuzzy rules representing the decision knowledge, and membership functions of each decision factor for verifying linguistic expression in the rules. The experimental results confirm that the proposed scheme can effectively discover the expert\`s tacit knowledge, and works as a feedback mechanism for human experts to fine-tune the conversion processes of converting tacit knowledge into implicit knowledge.

  • SaaS is an emerging paradigm for software development and deployment, expected to able to reduce cost. SaaS is also considered as a crucial technology for implementation of cutting-edge technology, such as distributed computing, green computing, and cloud computing. SaaS is requested to be configurable software to satisfy various customers, therefore it is very important to consider every configurability requirement during requirement elicitation. Our research suggests a classification technique to secure completeness of configuration requirement.

  • Many studies have addressed 'Adaptive User Interface (AUI)', wherein the user interface changes in accordance with the situation and the environment of each user at runtime. Nevertheless, previous papers for AUI barely reflected the viewpoints from requirements engineering field, since most of them focused on proposing the architecture and design. In this study, we outline AUI with the perspective of requirements engineering and propose the requirements elicitation and specification method based on concepts which have been researched in the area of self-adaptive system. Step by step, we first redefine and reinterpret the well-known concepts of self-adaptive software, after which the AUI requirements are elicited and specified. Finally, we illustrate a case study, which demonstrates the effectiveness of our method.

The Effect of Taglisodog-eum Extract on Lipopolysaccharide-induced Otitis media

  • Objectives : It has been known that immune reaction plays an important role in the pathogenesis of otitis media (OM). We investigated the change of middle ear mucosal inflammation induced by lipopolysaccharide (LPS) following administration of Taglisodog-eum (TSE) in experimental animals. Materials and Methods : Otitis media was induced by injecting 1 mg/kg of Pseudomonas aeruginosa LPS transtympanically in 60 rats. These rats were divided into two groups, the LPS group (n=30, remained naive after OM elicitation) and the TSE group (n=30, treated with TSE after OM elicitation) and 6 additional rats were kept as a normal control group (n=6, remained naive until tissue collection). The rats were killed at the 1st, 3rd, and 7th days after challenge. The temporal bones in each group were harvested and examined histopathologically by hematoxyline-eosin stain. Middle ear mucosa were taken at the 1st, 3rd, and 7th days after challenge. The levels of spicing variants of TNF-${\alpha}$ transcription were evaluated by Realtime-PCR. Results : TSE suppressed LPS-induced TNF-${\alpha}$ mRNA expression and thickness of the submucosal layer and infiltration of inflammatory cells in rat middle ear epithelium. Conclusion : The results suggest that TSE may be effective in decreasing inflammation with particular application to mucosal metaplasia in OM.

Effect of Sihogeji-tang Extract on Indomethacin-Induced Gastric Mucosal Lesions in Rats (Indomethacin으로 유발된 백서의 위점막 손상에 대한 시호계지탕의 효과)

  • Objectives : This study was carried out to investigate the effects of sihogeji-tang on indomethacin-induced gastric mucosal lesions in rats. Methods : Experimental rats were classified into three groups. The normal group had no inflammation elicited. The control group was rats administered water after elicitation of gastro-inflammation. The sample group were administered sihogeji-tang after gastro-inflammation elicitation. Results : In the common morphology and histochemical change, the control group were observed to have various injuries by hemorrhagic erosion, while the sample group had noticeably fewer injuries than the control. In the immunohistochemical change, the distribution of HSP-70, PCNA treated with sihogeji-tang noticeably increased over the control group. The distribution of NF-kB, COX-2 treated with sihogeji-tang noticeably decreased compared to the control group. (p<0.05) Conclusions : Sihogeji-tang had significant effects on indomethacin-induced gastric mucosal lesions in rats.
