Inhibitory Effects of the Extracts from Eleutherococcus senticosus Maxim. on Histamine-release from Rat's Mast Cell

  • Eleutherococcus senticosus Maxim. has been successfully used as an oriental medicine for various diseases including allergic disorders. Histamine is a major factor on various allergic responses and it is reported that histamine was released from mast cells by sensitization of allergens. In this study, ethanol extracts from E. senticosus Maxim. were prepared and the composition was analyzed by high performance liquid chromatography. The eleutheroside B as a primary effective component of E. senticosus was contained approximately 225 mg/kg in root bark extracts. The extracts were found to significantly inhibit compound 48/80-induced histamine release form mast cells in dose dependent manner. However the extracts had low cytotoxicity on the mast cells with MTT assay. These results showed that E. senticosus Maxim. extracts may be the effective materials on inflammatory disorders.

Antimutagenic and Cytotoxicity Effects of Extracts of Eleutherococcus senticosus Maxim fruits (가시오갈피 열매 추출물의 항돌연변이원성 및 세포독성 효과)

  • This study was earned out to investigate the antimutagenic and cytotoxic effects of Eleutherococcus senticosus Maxim fruits ethanol extract on Salmonella typhimurium TA98, TA100 and cancer cell lines using Ames test and SRB assay, respectively. They were extracted with ethanol and then fractionated with hexane, chloroform, ethyl acetate, butanol and water to get active fractions. In the Ames test, most of the extracts had strong antimutagenic effects against the mutagenesis induced by MNNG, 4NQO, B(${\alpha}$)P and Trp-P-l. The ethanol extract (200 $\mu\textrm{g}$/plate) of Eleutherococcus senticosus fruits showed 87.2% inhibitory effect on the mutagenesis induced by MNNG against TA100. And also, The suppression ratio against B(${\alpha}$)P and Trp-P-l in the TA100 showed 96.1% and 95.5%, respectively. In the cytotoxic effects against human cancer cell lines (A549, AGS, MCF-7, Hep3B), the value of inhibition were mostly above 60% for each fraction (1 mg/mL). Hexane fraction (1 mg/mL) against showed the strongest cytotoxic effects of 92.7% compared to those of other fraction and butano fraetion against Hep3B was relatively high growth inhibitory effect of 82%.

Effects of LED on Growth, Morphogenesis and Eleutheroside Contents of in vitro Cultured Plantlets of Eleutherococcus senticosus Maxim (가시오갈피 기내 식물체의 생장, 형태형성 및 eleutheroside 함량에 미치는 발광다이오드의 효과)

  • The effects of red, blue, and far-red light by illumination of light emitting diodes (LEDs) on growth, morphogenesis and eleutheroside contents of in vitro plantlets of Eleutherococcus senticosus were examined. As a control, plantlets were grown under a broad spectrum white fluorescent lamp (16/8 h illumination). The length of plantlets grown under the red/blue LEDs was taller than those under fluorescent lamps. Leaf area, root length and fresh weight of plantlets were highest under blue light compared to other kinds of light sources. Chlorophyll contents in plantlets grown under fluorescent lamps were higher than those in plantlets grown under LED illumination. Production of eleuthroside B and E in plantlets was highest under blue LED. However, production of eleuthroside E1 was highest under fluorescent lamps. These results suggest that plant growth and eleuthroside accumulation can be controlled by wave length of light under LED illumination system.

Cutting Propagation of Eleutherococcus senticosus MAXIM (가시오가피의 거목번식방법(揷木繁殖方法))

  • Eleutherococcus senticocus MAXIM containing compounds for a cordial and lumbago is a native plant in this country. However it is generally difficult to propagate them by seed. This study was carried out to improve propagation efficacy by cutting method using Eleutherococcus senticosus MAXIM collected Mt. Odae for three years$('90{\sim}'92)$. Rooting was delayed by 3 to 12 days but callus formation and rooting percentage was high by 75%, 30% respectively in the treatment of Rooton-F powder, a chemical for rooting promotion, compared with non-treatment. Rooting percentage and rooting characters were not differenced by treatment for remove rooting inhibition compounds and soil-media mixured by 1:1 of vermiculite and perlite was suitable for elevating rooting ratio. A. chiisanensis and A. sieboldianum were rooted well in any time to be cut, while rooting activity of E. senticosus was different by time to be cut. Rooting ratio of E. senticosus was 26%, 36% and 60% when cutten March 20, July 20 and september 20, respectively.

  • Eleuthero(Eleutherococcus senticosus Maxim.) cortex is well known as a herb medicine for tonic. This study was performed to improve the quality of dried E. senticosus cortex. Investigation of quality factor and contents of efficient compounds under different steaming times and drying methods were performed to determine the proper processing and drying conditions of Eleuthero cortex harvested on March in annual stems. The proper steaming time for peeling bark to make high quality Eleuthero cortex took less than 20 mins. Eleutheroside B and E contents among drying methods were significantly different at 5% level DMRT. The $50^{\circ}C$ heat drying was the most advisable condition for drying, when drying and keeping contents of effective compounds.

Breeding of New Cultivar 'Cheonsu' and 'Misu' for Seed Harvesting of Eleutherococcus senticosus (Rupr.&Maxim.) Maxim. (가시오갈피 채종용 신품종 '천수'와 '미수' 육성)

  • This study was executed to breed new cultivar of E. senticosus suitable for seed harvesting by selection method from 896 native plants collected from 35 regions. Basic selection points were C.V. values of characters such as filament length, blooming period, shoot length, no. of shoot and hundred fruits weight. By four steps of selection, two lines having high biomass yielding, diseases tolerant and fruiting capacities were finally selected and registered as new cultivar of Eleutherococcus senticosus at the Korea seed & variety service by regulation for seed production and marketing of plant variety.

Eleutherosides Extraction from Acanthopanax sessiliflorus Seeman and Eleutherococcus senticosus Maxim Using an Enzymatic Process (효소적 생물전환을 이용한 토종오가피와 가시오가피의 Eleutheroside B, E의 추출 함량 최적화)

  • In this study, we optimized conditions for extraction of eleutherosides B and E from stem powder of Acanthopanax. To enhance eleutheroside B and E yields, Acanthopanax sessiliflorus Seeman and Eleutherococcus senticosus Maxim were pre-incubated with the following enzymes: Celluclast, Viscozyme L, Lactozyme, Lecitase, and Novozyme 33095. Treatment with Novozyme 33095, a commercial pectinase, for 3 h resulted in the highest yields of eleutherosides B and E from A. sessiliflorus and E. senticosus. Compared with extraction at $121^{\circ}C$ for 120 min, at $121^{\circ}C$ for 15 min after Novozyme 33095 pre-incubation increased eleutheroside B and E yields from A. sessiliflorus by 7% and 17%, respectively. In the case of E. senticosus, eleutheroside B and E yields increased by 25% and 29%, respectively.

Effects of Shading treatments and Harvesting methods on the Growth of Eleutherococcus senticosus Maxim (차광처리(遮光處理) 및 수확방법(收穫方法)이 가시오갈피 생육(生育)에 미치는 영향(影響))

  • This experiment was carried out to investigate the effects of shading treatments and harvesting methods on the growth of Eleutherococcus senticosus, which was known as the medicine of anticancer, anti-stress, hepatic detoxification activity, immunoactivity, and tonic. Light transmission ratio and air temperature were decreased as $40{\sim}64%$,$1.9{\sim}2^{\circ}C$, respectively, in 30% to 70% shading net treatment compared to non-shading. Fifty percentage shading net treatment was effective for the growth and yield of Eleutherococcus senticosus. During summer Eleutherococcus senticosus was growing under shading net treatment compared to non-shading. Tunnel type was more effective for growth than vertical type in shading method. 30cm length included 2 to 3 nodes cutting from soil surface was the most effective for branching stem length, plant height and yield as harvesting methods.

Inhibitory Effect of Methanol Extract of Eleutherococcus senticosus Maxim. on the Direct Mutagen Mutagenicity (직접 돌연변이원에 대한 가시오갈피 추출물의 항돌연변이 효과)

  • Antimutagenic effects og Eleutherococcus senticosus Maxim. on the mutagenicty induced by mutagens, 4-NQO, MNNG, $\textrm{NaN}_{3}$, 2-NF, and 1-NP was studied by the Ames test with Salmonella typhimurium TA98 and TA100. The methanol extract ($500\mu\textrm{g}$/plate) of E. senticosus Maxim. showed inhibitory effect on the mutagenicty induced by 1-NP only among the tested mutagens. In S. typhimurium TA98, the methanol extracts of the root, stem and leaf showed inhibitory effects of 54.9, 29.5, and 32.9% inhibition on 1-NP mutagenicity, respectively. In S typhimurium TA100, the methanol extracts of the root, stem, and leaf showed inhibitory effects of 593, 30.2, and 43.6%, respectively. The methanol extract were further fractionated by a subsequent liquid-liquid partition technique with chloroform, butanol, and water. The chloroform ($300\mu\textrm{g}$/plate) fraction of the root, stem and leaf showed the strong antimutagenic effects on the mutagenicity induced by 1-NP in S typhimurium TA98 and TA100. But none or weak antimutagenicities were observed in the butanol and aqueous fraction. The chloroform fractions of root, stem and leaf showed the antimutagenic effects of 61.6~88.6% in a dose-dependent manner. In the antimutagenic mode test, the inhibition effect of root was mainly bio-antimutagenic, whereas stem and leaf were desmutagenic.
