Comparison of Photostimulated Luminescence, Thermoluminescence, and Electron Spin Resonance Spectroscopic Analyses on Dried-spices Irradiated by Gamma Ray and Electron Beam (감마선 및 전자선 조사 처리 건조향신료에 대한 광자극발광, 열발광 및 전자스핀공명의 분광학적 분석 비교)

  • This study was conducted to determine the effect of gamma-ray and electron-beam irradiation on dried spices (black pepper, red pepper, parsley, and basil) using the photostimulated luminescence (PSL), thermoluminescence (TL) and electron spin resonance (ESR) methods. The spices were irradiated at 0, 1, 5, and 10 kGy. All non-irradiated spices had photon counts (PCs) less than 700 PCs. The PCs of three irradiated spices (red pepper, parsley, and basil) were clearly distinguishable from those of non-irradiated ones, exhibiting PSL signals higher than 5000 PCs. However, negative PSL counts (<700 PCs) were obtained for most irradiated black pepper, except those irradiated with 5 kGy gamma rays and 10 kGy electron-beams. TL glow curves of the irradiated spices showed a higher peak at $150-250^{\circ}C$. TL ratios were found to be less than 0.1 for non-irradiated spices and higher than 0.1 for irradiated ones. No ESR signal was observed for any irradiated spice except red pepper, which displayed cellulose-based ESR spectra. Therefore, the results suggest that the PSL, TL, and ESR methods are effective detection techniques for dried spices irradiated with electron beams as well as gamma rays.

  • Electron spin resonance(ESR) spectroscopy was used to investigate the effect of irradiation dose on the ESR signal intensity of irradiated spices and to identify the stability of radicals after storage. Red, white and black peppers, and garlic powders were irradiated with doses of 0, 1, 5, 10, 20 and 30 kGy at room temperature using a Co-60 irradiator. Triplet ESR signals were observed in irradiated pepper powders, while singlet ESR signals were observed in irradiated garlic powders. Those characteristic signals were not detected in non-irradiated samples. The strength of ESR signals linearly increased with the applied doses(1~3 kGy). Highly positive correlation coefficients ($R^2$=0.9757~0.9933) were obtained between the irradiation doses and the corresponding ESR signal intensities. The signal intensities of irradiated samples were stable even after 97 days of storage at room temperature.

  • For the research of the natural marine antioxidant, an antioxidant-producing marine actinomycetes was isolated from sea water in Jeju coastal area. The strain was identified based on 16S rDNA sequencing, the morphology by a method of scanning electron microscopy, physiological and biochemical characteristics and cellular fatty acid analysis. The isolated strain ACT-18 was gram positive, aerobic, non-motile spores. Substrate mycelia are dark green and yellow gray aerial mycelia. The cell size of the strain was $0.5{\sim}1.0\;{\mu}m$. 16S rDNA sequence analysis showed that were Gram-positive bacteria grouped on Streptomyces sp. Results of cellular fatty acid analysis showed that major cellular fatty acids were $C_{15:0}$ anteiso (39.33%), $C_{16:1}$ cis 9 (11.96%), $C_{16:0}$ (13.08%) and $C_{17:0}$ anteiso (10.99%). The antioxidant activity of methanol extract from Streptomyce sp. ACT-18 was evaluated by measuring 1,1-diphenyl- 2-picrylhydrazyl (DPPH), hydroxyl, and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. DPPH radical scavenging activity of SBME (Streptomyces Broth Methanol Extract) A-18 was 46% at 0.1 mg/mL. Hydroxyl radical scavenging activity of SBME A-18 was 63% at 0.1 mg/mL. Alkyl radical scavenging activity of SBME A-18 was 39% at 0.1 mg/mL.

Identification Characteristics of Irradiated Dried Red Pepper during Storage by the Analysis of Electron Spin Resonance and Hydrocarbons (전자스핀공명 및 Hydrocarbon 분석에 의한 방사선처리 건고추의 저장 중 검지특성)

  • Electron spin resonance (ESR) and hydrocarbon characteristics were analyzed to establish identification conditions for irradiated dried red pepper. The ESR spectroscopy for 4 different parts (powder, pericarp, seed, stem) of the samples showed that irradiated samples signaled (g=2.024, 2.006, 1.987) a pair of peaks from a cellulose radical at intervals of 6 mT, which were not found on the non-irradiated samples. The ESR signals increased in directly proportion to the irradiation doses, which were still detectable after 12 weeks of storage at room temperature. The GC-MS analysis of hydrocarbons after fat extraction and separation by florisil column chromatography revealed that hydrocarbons, such as 1-tetradecene (14:1), 1,7,10-hexadecatriene (16:3), 1,7-hexadecadiene (16:2), 1-hexadecene (16:1), 6,9-heptadecadiene (17:2), and 8-heptadecene (17:1), were detected only from the irradiated samples immediately after irradiation and 8 months of storage. They linearly increased with the dose of irradiation, suggesting them as radiation-induced markers for irradiated dried red pepper.

  • The identification characteristics of inadiated dried onion and garlic at 1, 4 and 7 kGy were investigated by analyzing their photostimulated luminescence (PSL), thermoluminescence (TL) and electron spin resonance (ESR). The PSL results showed that the photon counts of non-irradiated dried onion were less than 700 (negative) and those of irradiated samples at 1kGy or more were over 5000 (positive), making it possible to discriminate non-irradiated onions from irradiated ones. However, the intermediate counts (700-5000) were observed in irradiated garlic at 1 and 4 kGy, even though positive counts were measured in 7 kGy group. Radiation-induced TL glow curves $(TL_1)$ were measured at $150^{\circ}C$ range in both irradiated onion and garlic samples at 1 kGy or more, which could identify irradiated groups. Furthermore, determination of TL ratios $(TL_1/TL_2)$ for both samples could verify the above results. It was found, however, that ESR spectroscopy was not suitable for the detection of irradiated dried onion and garlic.

ESR Spectroscopy for Detecting Gamma-Irradiated Shellfishes (방사선 조사된 조개류의 확인을 위한 ESR Spectroscopy의 이용)

  • Electron spin resonance (ESR) spectroscopy was used to investigate the effect of irradiation dose on the ESR signal intensity and to identify the stability of radicals after 9 weeks of storage in order to detect irradiated shellfishes. The irradiated shellfishes (short-necked clam, purplish washington clam, freshwater clam, jackknifed clam, scallop and hard-shell mussel) presented an asymmetric absorption in shape at $g_{1}$=2.002~2.003 and $g_{2}$=1.998. The strength of the ESR signal increased linearly with the applied doses (1~7 kGy). A highly positive correlation coefficients ($R^{2}$=0.9136~0.9896) were obtained between the irradiation dose and corresponding ESR signal intensity. The intensity of the signals after irradiation was stable even after 9 weeks of storage at 5$\pm$1$^{\circ}C$.

Antioxidative Activity of Rice Grain using FI-CL and ESR Methods

  • Fifty-four Korean native and 28 foreign rice varieties harvested in 1998 and 1999 were examined for antioxidative activity that is measured to a chemiluminescence and superoxide radical intensity, by the flow injection chemiluminescence (FI-CL) system and an electron spin resonance (ESR) spectrophotometer, respectively. In the chemiluminescence measurement by FI-CL, radical scavenger activity did not differ significantly among rice varieties between origin types of rice varieties, and between storage periods. GinSun and Hongchoengdo, colored rice exhibited high electron scavenging effect by ESR. Therefore, these results indicate that the pigments of rice varieties may play important antioxidative roles and that it may be possible to breed rice varieties with higher antioxidative potentials.

Electron Spin Resonance Study of Manganese Ion Species Incorporated into Novel Aluminosilicate Nanospheres with Solid Core/Mesoporous Shell Structure

  • An ion-exchanged reaction of $MnCl_2$ with Al-incorporated solid core/mesoporous shell silica (AlSCMS) followed by calcinations generated manganese species, where average oxidation state of manganese ion is 3+, in the mesoporous materials. Dehydration results in the formation of $Mn^{2+}$ ion species, which can be characterized by electron spin resonance (ESR). The chemical environments of the manganese centers in Mn-AlSCMS were investigated by diffuse reflectance, UV-VIS and ESR spectroscopic methods. Upon drying at 323 K, part of manganese is oxidized to higher oxidation state ($Mn^{3+}$ and $Mn^{4+}$) and further increase in (average) oxidation state takes place upon calcinations at 823 K. It was found that the manganese species on the wall of the Mn-AlSCMS were transformed to tetrahedral $Mn^{3+}$ or $Mn^{4+}$ and further changed to square pyramid by additional coordination to water molecules upon hydration. The oxidized $Mn^{3+}$ or $Mn^{4+}$ species on the surfaces were reversibly reduced to $Mn^{2+}$ or $Mn^{3+}$ species or lower valances by thermal process. Mn(II) species I with a well resolved sextet was observed in calcined, hydrated Mn-AlSCMS, while Mn (II) species II with g = 5.1 and 3.2 observed in dehydrated Mn-AlSCMS. Both species I and II are considered to be non-framework Mn(II).

  • The chalcogenide glass (ALS, $Al_2S_3-La_2S_3$) was prepared by melting a stoichiometric mixture of aluminum powder and $La_2S_3$ under $H_2S$ atmosphere at $1200{\circ}C$. Glasses containing 0.1-0.1% of $Mn^{2+},\;Gd^{3+}$ and $Cu^{2+}$ were also prepared. The characteristic features of the ESR spectra for the transition metal containing ALS glasses are interpreted.
