Dysfunction of Retinal Cell and Optic Nerve by Continuous Cerebroventricular Infusion of Glucosamine

  • We have investigated the effect of glucosamine on the retinal cells after continuous infusion into cerebroventricle by using osmotic minipump to avoid peripheral effect. Continuous intracerebroventricular (i.c.v) infusion of glucosamine with the rate of 0.1 ${\mu}mol$/10 ${\mu}l$/hr for 7 days resulted in morphological changes of the optic nerve in electron microscopic level as well as morphological changes of the retina in light microscopic level. Retinal sections were immunostained for the detection of morphological changes of astrocytes. GFAP immunoreactivity appeared not only in the Muller cells but also many of the radial processes of Muller cells. The optic nerve showed deformed axon and slight lamellar separation of myelin sheath after continuous infusion of glucosamine in observing with electron microscope. Interestingly, vacuoles were observed in deformed axons and retinal layers were folded and detached. These results suggested that glucosamine plays a role in induction of morphological dysfunction in retina and optic nerves.

멜라토닌이 허혈-재관류 손상에 의한 골격근의 형태학적 변화에 미치는 효과 (The Effect of Melatonin on Morphological Changes of Rat Skeletal Muscle after Ischemia-Reperfusion Injury)

  • The effect of melatonin on morphological changes after ischemia-reperfusion injury was investigated in rat skeletal muscle. Dimethyl-sulfoxide(DMSO) was also tested for comparison. Muscle injury was evaluated in 4 groups as a single laparotomy group(control), ischemia-reperfusion group, DMSO group, melatonin group. Left hind limb ischemia was induced for 4 hours by vascular clamping of the common femoral artery and followed by 24 hours of reperfusion. The midportion of gastrocnemius muscle was taken for histological evaluation. In light microscopic study, ischemia-reperfusion group showed severe neutrophil infiltration, interstitial edema, and partial loss or degeneration of muscle fibers. The muscle tissue of melatonin group showed relatively normal architecture with mild inflammatory cell infiltration. In electron microscopic study, dilated cisternae of sarcoplasmic reticulum, dilated mitochondria with electron loose matrix and dilated cristae, disordered or loss of myofilament, indistinct A-band and I-band, intracytoplasmic vacuoles, and markedly decreased glycogen granules were observed in ischemia-reperfusion group. But relatively well maintained A-band, I-band, Z-line, M-line, and mildly dilated mitochondria with well preserved cristae were observed in melatonin group. The DMSO group showed intermediately attenuated ultrastructural changes. The results show that melatonin improves morphologically ischemia-reperfusion injury more effectively than DMSO. In conclusion, melatonin seems to be a promising agent that can salvage the skeletal muscle from severe ischemia-reperfusion injury.

혈관내에 배양한 신경줄기세포의 이식이 말초신경 재생에 미치는 영향 (Effect of Transplantation of Intravascular Cultured Neural Stem Cell upon Peripheral Nerve Regeneration)

  • 흰쥐의 좌골신경을 절단한 후 혈관내에서 배양한 신경줄기세포를 이식하여 말초신경에서도 수초의 재생이 일어나는지를 형태 학적으로 규명하고 배양한 신경줄기 세포들로부터 분화한 Schwann cell들이 회복할 수 있는지를 조사하여 다음과 같은 결론을 얻었다. 이식한 20일 후 동맥내 배양한 신경줄기세포는 Schwann cell로 분화하여 신경섬유의 재생이 일어나기 시작하였다. Schwann cell은 증식 후 재수초화를 형성하기 위하여 다른 Schwann cell들로부터 여러 가지를 자극을 받고 있었으며 NGF 소견으로 볼 때 신경외막으로부터 기존의 Schwann cell로부터 신경줄기세포의 분화가 유도되었으며 PCNA 반응으로 볼 때도 기존의 신경섬유의 Schwann cell주위에서부터 증식이 일어났다. 미세구조적으로는 Schwann cell의 재수초화, 축삭내 사립체와 미세소관의 수의 증가를 관찰할 수 있었다.

백내장 환자의 연령에 따른 수정체낭 Type IV 아교섬유의 분포와 수정체낭 상피세포 자연사 및 전자현미경적 변화에 대한 연구 (Electron Microscopic Studies on Distribution of Collagen IV of Lens Capsule and Apoptosis of Lens Epithelium in Age-related Cataractous Human)

  • 백내장 환자의 수정체낭에서 연령차이에 따른 아교질과 type IV 아교섬유의 면역화학 반응 양상, apoptosis현상 및 수정체 상피세포의 전자현미경적인 변화를 관찰하여 다음과 같은 결과를 얻었다. Van Gieson염색소견은 수정체낭의 앞층과 앞아래층에서 짙은 염색반응이 있었으나 나이가 많을수록 반응이 감소되었다. Type IV 아교섬유 면역화학반응은 수정체낭의 앞층과 상피세포 주위에서 현저하게 일어났으며 나이가 많을수록 반응이 감소되었다. Periodic acid skiff-alcian blue반응의 변화는 수정체낭의 앞층에서 나이가 많을수록 현저하게 감소되었다. 세포자연사 현상은 수정체낭 상피세포의 핵내에서 일어났으며 나이가 많을 수록 반응이 일어나는 세포의 수가 감소되는 경향을 보였다. 수정체 상피세포의 전자현미경적인 변화는 40대까지는 외측 세포막의 folding (이하 주름)과 세포질내 공포가 증가되고 골지복합체가 발달되었으나 60대에서는 부분적으로 돌기는 내지만 골지복합체는 관찰되지 않았다. 수정체 상피세포의 바닥막부분에서는 나이가 들수록 거친 섬유들이 수정체 상피세포와 수정체낭을 격리시키고 있었으며, 수정체낭와 중간층은 40대군은 특징적으로 과립상의 구조물들이 집단적으로 축적된 부분들이 관찰되었고 60대군에서는 선상의 섬유성구조들이 나타났으며 짙은 전자밀도를 가진 과립의 수가 감소하였다.

  • The purpose of this study was to investigate the irradiation effects on the secretory granules of the rat submandibular gland acinar cell. For this study, Sprague-Dawley strain rats were irradiated to their neck region with the dose of 5Gy and l0Gy by 6MV X-radiation, and sacrificed on the experimental periods after irradiation. The authors observed the ultrastructural changes of the secretory granules of the rat submandibular gland acinar cell under a trasmission electron microscope. The results were as follows: In the transmission electron microscopic examination, secretory granules were blurred in limiting membrane on the 3hours groups after irradiation. And they showed decrease in number, irregularities in shape and distributional pattern, and inhomogeneous internal electron density on the Iday and 3days groups. After then, these changes were recovered in shape and distributional pattern on the 14days groups, and changes of internal electron density and limiting membrane were recovered on the 28days groups after irradiation. Among the intracellular organelles, rough endoplasmic reticulum was scattered, but golgi complex was not changed. And such pathologic changes were earlier and more prominent in 10Gy irradiated groups than in 5Gy irradiated groups.

방사선 조사가 백서 악하선 미세혈관계에 미치는 영향에 관한 실험적 연구 (AN EXPERIMENTAL STUDY OF THE IRRADIATION EFFECTS ON THE MICROVASCULATURE OF THE RAT SUBMANDIBULAR GLAND)

  • The purpose of this study was to investigate the effects of irradiation on the microvascular structure of the submandibular gland in rats. For this study, 110 male rats were singly irradiated with the dose of 10Gy or 20Gy to their neck region by 6MV X-irradiation and sacrificed on the 1st, 3rd, 7th, 14th and 28th day after irradiation. The author observed distribution and structural changes of the microvasculature in rat submandibular glands using a scanning electron microscope by forming vascular resin casting. The author observed ultrastructural changes of the endothelial cells using a transmission electron microscope, and also histologic changes using a light microscope at Hematoxylin and Eosin staining and PAS staining process. The results of the irradiation effects on the microvasculature in rat submandibular gland were as follows: By light microscopic examination, the dilation of small vessels were observed until the 7th day after irradiation. After then, the vascular constriction and decrease in number of small vessels were noticed. Changes were greater on 20Gy irradiated group than on lOGy irradiated group. The reaction to PAS staining at acinar cells was decreased just after irradiation, but gradually recovered with days. There was no specific difference between two irradiated groups. By scanning electron microscopic examination, general findings on the two irradiated groups were similar. The dilation of conduits and meandering were observed on the 3rd day after irradiation. Decrease of capillary density and blunt ended small vessels were appeared on the 7th day after irradiation. After that, findings of the tortuous and twisted vascular running and coarseness of capillary lumen were increased. Changes were greater on 20Gy irradiated group than on l0Gy irradiated group. By transmission electron microscopic examination, increase of the formation of cytoplasmic process was observed on the 3rd day after irradiation. After that, swelling of endothelial cell and bridge formation of cytoplasmic processes were also observed, but destruction of endothelial cell and loss of basement membrane were observed only on 20Gy irradiated group on the 28th day after irradiation.

  • The present study is of ultra-fine structure of Cryptococcus neoformans by means of electron microscopy and reveals the following : 1) In constrast to the bacteria, the normal Cryptococcus neofrmans contains nuclear enveloped with nuclear menbrane, mitochondria, endoplasmic reticulum, distinct cell wall and cell membrane, vacuoles and storage granules as observed in the eucaryotic cells. 2) In apparent cell walls and cell membrane with the appearance of electron transparent area (ETA) and changes of cell morphology were observed in the ultra-violet ray irradiated cell. 2) In apparent cell walls and cell membrance with the appreance of electron transparent area (ETA) and changes of cell morphology were observed in the ultra-violet ray irradiated cell. 3) Morphology changes and cytoplasmic element abnormality was increased with irradiated time. 4) Increase of electron transparent area was thought to be associated with degradation of cell.

토끼의 바이러스성(性) 출혈증(出血症) 감염시(感染時) 신장(腎臟)의 광학(光學) 및 전자현미경적(電子顯微鏡的) 관찰(觀察) (Light and electron microscopy of the kidney tissues on viral haemorrhagic disease in rabbits)

  • In order to observe the pathological changes of kidney in rabbits infected with the viral haemorrhagic disease, the kidney tissues from the 91 rabbits infected with the viral haemorrhagic disease were examined by light and electron microscopy. The results observed were as follows: 1. On light microscopic observation, the kidney lesions were identified as haemorrhagic glomerular necrosis(33.0%), membranous glomerulonephritis(20.9%), thrombotic glomerulopathy(19.8%), membranoproliferative glomerulonephritis(8.8%), mesangial proliferative glomerulonephritis(8.8%) ischemic acute tubular necrosis(7.7%), and acute serous glomerulitis(6.6%). 2. On electron microscopic observation, cytoplasmic degeneration of mesangial cells, and irregular thickening of basement membranes with electron dense granular materials were observed. In podocytes swelling of mitochondria, dilatation of endoplasmic reticulum and extensive fusion of foot processes were also observed. Nonenveloped round icosahedral picornaviral particles with a diameter of 28~33nm were detected in the cytoplasm of degenerative endothelial cells, polymorphonuclear leucoytes, and monocytes.

  • The light and electron microscopic observations were carried out in order to know the morphological changes of the fat-storing cells (FSC) in the liver of rabbits given Ginseng for 30 days. The results obtained were as follows: The FSC of experimental group was shown the appearance of a few large lipid droplets, enlarged rough surfaced endoplasmic reticulum, some residual body in the enlarged rER and the nuclear body in some cases. As these findings, it could be suggested that the Ginseng affect the synthesis and accumulation of lipid on the FSC.
