Effect of water temperature on embryonic development and larval survival of an intertidal snail, Nassarius festivus (Powys, 1835)

  • This study was carried out to determine the optimal water temperature for the embryonic development and laboratory culture of larvae of an intertidal mud snail, Nassarius festivus. The embryos and hatched veliger larvae of N. festivus were incubated at six different temperatures (5, 10, 15, 20, 25 and $30^{\circ}C$). Developmental time for each stage decreased as water temperature increased. The elapsed time to develop to the veliger larva at 15, 20, 25 and $30^{\circ}C$ was 559, 155, 131 and 103 hrs, respectively. At 5 and $10^{\circ}C$, embryo developed to veliger larvae but failed to hatch out of the egg capsule. In contrast, all embryos successfully hatched in the temperature range from 15 to $30^{\circ}C$. The biological minimum temperature during the embryonic development of N. festivus was estimated to be $9.5{\pm}0.4^{\circ}C$. The cumulative water temperatures for blastula, gastrula and veliger stages were calculated as $111{\pm}84$, $486{\pm}185$, $1,164{\pm}72^{\circ}C$, respectively. Temperature also affected the larval survival. Five days after hatching, more than 84% of larvae survived at all experimental temperatures. However, survival began to decrease after 6 days. It was 0% at $30^{\circ}C$. Survival of larvae incubated for 8 days was higher at 15 and $20^{\circ}C$ than other experimental temperatures. We therefore suggest that the optimal range of temperature for embryonic development and larval survival of N. festivus is $15-20^{\circ}C$.

Study on Anti-estrogenic Activity of DEHP as an Endocrine Disruption Chemical (내분비 교란성 DEHP의 항-에스트로젠 활성에 관한 연구)

  • Di-2-ethylhexyl phthalate (DEHP), is a widely used plasticizer known to be a suspected endocrine disrupter, but its exact effects on aquatic organisms are not yet known. When Japanese medaka (Oryzias latipes) were exposed from the time of hatching to 3 months of age to an aqueous DEHP solution at nominal concentrations of 1, 10, and 50 $\mu\textrm{g}$/l, DEHP treated female fish showed distinct reproductive effect. And the midge (Chironomus riparius.). an aquatic invertebrate, was exposed to DEHP to evaluate the effects on reproductive processes via sediment toxicity. The test endpoints included emergence, sex ratio, fecundity, and the viability of F1 offspring egg ropes. The result implied that the normal developmental and/or reproductive processes in C. riparius had been disrupted when exposed to DEHP, the effect also being displayed in the next generation. In summary, DEHP hinders the development of reproductive organs in the female Japanese medaka and C. riparius.

Management and Development of Seed Rearing in Red Spotted Grouper Epinephelus akaara (양식 붉바리(Epinephelus akaara) 종자의 개발과 사양관리)

  • We investigated seed management and feeding regime of larval red spotted grouper Epinephelus akaara using live food organisms. Fertilized eggs were produced in March 2020 by manipulating photoperiod and water temperature. Broodstock (F0) produced approximately 0.30×106 eggs, of which 0.25×106 were fertilized; first-generation broodstock (F1) produced 2.00×106 eggs, of which 1.90×106 were fertilized. Larvae were reared at temperatures of 21.0-24.5℃. The feeding regime consisted of rotifers Artemia, and commercial pellet feed. From 5-8 days after hatching (dah), rotifers collected from high-density incubation tanks were fed to grouper larvae at a density of 10-20 individuals/mL. From 9-30 dah, rotifers collected from low-density incubation tanks were fed to larvae at a density of 10-20 individuals/mL. Newly hatched Artemia were provided from 20-24 dah and enriched Artemia from 25-50 dah. Commercial pellet feed was given starting at 13 dah. The average mouth size in hatchlings was 263.4±3.7 ㎛ at 5 dah, 406.7±21.0 ㎛ at 11 dah, and 856.9±136.6 ㎛ at 21 dah. At 1 dah, larvae had a mean total length of 1.95±0.06 mm. At 66 dah, the mean TL was 4.0 cm. The final number of surviving red spotted grouper larvae was 218,380 fishes.

  • Ecology and life History of the mudskipper, Boieophthaimus pectinirostris were investigated based on the specimens collected from the Korean roasts from 1978 to 1994. The spawning of this species takes place during the period from June to August. Prolarva hatched from egg was 3.3mm in total length, and began to bottom life in TL 16.0mm of 40 days after hatching. The stomach contents were principally diatoms. In the foraging behavior, this species were conducted at the wet soft mud on the upper tidal zones. The burrowing observed in the intertidal mud flat was YL type. B. pectinirostris is restricted to western and southwestern coast of Korea, but their habitats and individuals are being reduced by the result of reclimation to tide land.

Developmental and Morphological Characterization of the Wild Silkmoth, Actias gnoma, in Korea

  • The wild silkmoth, Actias gnoma was firstly collected at Suwon located in the middle part of Korea. The developmental and morphological characteristics of A. gnoma reared under the laboratory conditions were analysed. The egg shape was shorter elliptic. The major and minor diameter of the eggs were 2.04 mm and 1.83 mm, respectively. White mucous material was remained inside the eggs after hatching. Also, the body color of the larvae was completely changed from dark brown to light yellowish-green at the 3rd instar, but it did not occur until pupation. In the feeding test on several plants, the oak tree, Quercus acutissima, was newly identified as a host plant. The final larval instar was mostly 6th, but in some larvae it was 7th. A few of larvae ate their own casts just after ecdysis. The whole larval duration ranged from 45 to 59 days. The single cocoon weight, cocoon shell weight and cocoon shell percentage were 1.65 g,31.3 cg and 18.9%, respectively, The cocoon shape and color were spindle and light brown, respectively. The morphology of the silk gland was greatly different from those of Bombyx mori, Antheraea yamamai and Antheraea peryi: the thickness of the middle and posterior silk glands was almost identical.

Acute and Chronic Ecotoxicity Assessment of Ambient and Effluent Water Discharged to the Lake Shihwa (시화호로 유입되는 지표수 및 방류수의 급·만성 생태독성평가)

  • The acute and chronic toxicity of ambient and effluent water discharged to Lake Shihwa were investigated by using Vibrio fischeri, Daphnia magna and Oryzias latipes. Physicochemical characteristics including biochemical oxygen demand (BOD) and nine heavy metals in a total of 15 water samples were evaluated and were satisfied with relevant Korean Water Quality Standards (KWQS) except for Hg in one sample. Acute toxicity was observed in five samples collected from three sampling locations. When impacts on reproduction and growth after chronic exposure were evaluated with D. magna, all the samples showed significant chronic effects. Reproduction appeared relatively more sensitive endpoint. In 21 days chronic tests on O. latipes, survival, mean egg number per female per day, hatching success rate and time to hatch were affected by increasing sample concentration. The organ-level changes such as gonadosomatic index (GSI), and hepatosomatic index (HSI), and molecular biomarker of vitellogenin (Vtg) induction that evaluated with O. latipes increased as exposure concentrations increased. It is noteworthy that the samples that did not exceed the KWQS resulted in acute and chronic toxicities. The results suggested that numeric criteria based on physicochemical parameters may not be protective of aquatic ecosystem. Acute and chronic toxicity tests with organisms representing different trophic groups should be supplemented in order to provide adequate level of environmental protection.

Growth and Spawning Ecology of Cobitis lutheri (Teleostei: Cobitidae) in the Mangyeong River, Korea (만경강 삼천에 서식하는 점줄종개 Cobitis lutheri의 성장과 산란상태)

  • Growth and spawning ecology of Cobitis lutheri (Teleoste: Cobitidae) were investigated in the Samcheon Stream, Mangyeong River, Jeollabuk-do, Korea from 2005 to 2006. Age group of C. lutheri estimated from the total length indicated that the 40 ~ 60 mm group is 1 year old, the 60 ~ 80 mm group is 2 years old, and the 80 ~ 110mm group is 3 ${\leq}$ years old. The sex ratio (Male/Female) was 0.78, and the female was 20 ~ 30 mm larger than the male. Lamina circularis at the base of pectoral fins in the males as a secondary sexual character was created 12 months after hatching. A change of lateral coloration of males was observed from April to September. The spawning season was early to middle July with water temperatures of $24{\sim}26^{\circ}C$. The average number of eggs was $1,127{\pm}453$ with the egg diameter of $1.09{\pm}0.02$ mm.

Analysis of Life Cycle on Spodoptera exigua by Various Temperatures and Larval Susceptibility against Two Pesticides in Quarantine Pest Research Facility (검역해충실험동에서 파밤나방의 온도별 생활사 및 2종 살충제에 대한 유충 감수성 조사)

  • A Quarantine Pest Research Facility (QPRF) had been build in Animal and Plant Quarantine Agency and designed to ensure alien pests are safety contained inside the laboratory. Functions of QPRF including insect rearing, research, and physical containment facility were verified to check suitability as a research laboratory using the beet armyworm, Spodoptera exigua. As insect rearing and laboratory facility, S. exigua completed its development for 32.2 days at 25℃. Egg hatching, pupation, and adult eclosion showed 80, 86.6, and 90%. Two pesticides, fluxametamide and hydramethylnon showed high susceptibility against S. exigua larvae by spraying and dipping methods. As physical containment facility, male adults, which were artificially released from laboratory, did not capture in delta trap installed sex pheromone lure during winter season. Based on these results, QPRF will be used for study on alien insect species.

Survey the Occurrences and Establishment of Environment-friendly Control System of Ricania shanthungensis in Jeonnam Province (전남지역 갈색날개매미충 발생현황과 친환경 방제)

  • This study was carried out to establish of environment-friendly control system and to survey the occurrence status of Ricania shanthungensis in Jeonnam province from 2016 to 2017. R. shanthungensis occurred at 1,344 ha in 61 towns of 15 cities in Gwangju and Jeonnam province. At four sites in the stationary area, the densities of eggmass and pupae were lower in 2017 than in 2016, the hatching rate decreased, and the first hatching time, emergence of adults, and the start of spawning tended to be slower. Telenomus sp., an egg parasitoid of R. shanthungensis, was found in most areas. Sunchon had the highest rate of 46.1%, followed by Muan 40.6, Goksung 29.2, Gurye 25.8, and Gwangju 17, respectively. The optimum spraying time for the control of R. shanthungensis was early March. The spraying material and the dilution multiples are 10 times of the machine oil at environmental friendly cultivation and 500 times of the chlorpriphosphate wp. at normal cultivation. When sprayed these materials, 95% of egg prohibited to hatch. There was more than 80% of the insecticidal effects on the organic materials of the machine oil emulsion, the Sophora root extrect + microbial extract agent, and the castor emulsion. For adults, Sophora root extrect + microbial extract, neem extract, and machine oil were better. Four kinds of chemical pesticides such as dinotefuran wp were effective for nymph and adult control. We have developed an adult catching device using the most preferred daylight color and behavioral habits of R. shanthungensis. The capture device consisted of two daylight compact lamps (30W and 20W), a yellow plate, and a catcher using water, and caught about 700 individuals a day. Based on the above results, we have established a system for controlling and life cycle of R. shanthungensis in Jeonnam province.

Toxicity of Persistent Organic Pollutants, PAHs and TBT, in Zooplankton and Influence on Their Viability (동물플랑크톤에 대한 지속성 유기오염물질 PAHs와 TBT의 독성 및 생존능력에 미치는 영향)

  • We conducted three experiments to estimate the toxicity of POPs (persistent organic pollutants) on two copepod species (Acartia erythraea and A. omorii) and Artemia sp.; (1) 48 h-LC$_{50}$ of A. omorii with the five PAHs [polycyelic aromatic hydrocarbons anthracene, benzo〔a〕pyrene, fluoranthene, phenanthrene, pyrene〕 which were often detected in the Gwangyang Bay, (2) toxicity of benzo〔a〕pyrene and TBT on Artemia in different temperatures (1$0^{\circ}C$, 15$^{\circ}C$, 2$0^{\circ}C$), (3) effects of benzo〔a〕pyrene and TBT on egg Production rate, hatching rate and fecal Pellet Production of two copepod species (A. erythraea and A. omorii) fed on Heterocapsa triquetra (dinoflagellate) exposed in benzo〔a〕pyrene. Toxic chemicals which were most effective to A. omorii were flueranthene (48 h-LC$_{50}$ 19.20 $\mu\textrm{g}$ L$^{-1}$ ) and benzo〔a〕pyrene (48 h-LC$_{50}$ 29.89 $\mu\textrm{g}$ L$^{-1}$ ). The toxi- city of chemicals to Artemia increased when temperature increased. The toxicity of TBT was about 100 times higher than that of benzo〔a〕pyrene at 15$^{\circ}C$. Food materials (Heterocapsa triquetra) exposed in benzo〔a〕pyrene, affected negatively the rate of egg production, hatching rate and the fecal pellet production of the copepods at the high concentration. It is suggested that an increase in the concentration of benzo〔a〕pyrene might offset the production of copepods in marine ecosystems. This study suggests that copepods may be used as n indicator for early warning of the risk of POPs in marine ecosystems.
