Methanol induction strategy using the two-loop control-based DO-stat and its application to repeated induction in methylotrophic yeast Pichia pastoris

  • A simple control strategy of DO-stat was introduced to the recombinant rGuamerin production process in Pichia pastoris. This induction strategy consisted of two interrelated control loops ‘by which oxygen ratio of inlet gas and methanol feeding rate was controlled. Using this control strategy, over-feeding or under-feeding of methanol could be avoided in concomitance with the efficient control of dissolved oxygen level. As a result, the cell concentration reached 130 g/L and rGuamerin expression level was 450 iu/L, which was more than 40% increased result comparing with the fed-batch process using manual control of methanol feeding rate.

  • A major effect of electronic commerce on the Internet is that it reduces transaction costs and creates an efficient market. Thus, an information strategy for developing of regional economy must be changed toward 21st century of electronic commerce era. In this paper, we present an information strategy for activating the electronic commerce of regional small and medium-sized business organizations. The paper stresses creation of a network organization comprising local government, regional business organizations, universities, research institutes, and citizens' coalitions as groups of organizational actors. Necessary conditions of a successful network organization in electronic commerce are to build the public key infrastructure for developing the trust of transactions among partners, and to create an information center to manage a web of partners. In the network organization, the information center must play critical roles as a creator of value and the certification authority in order to issue and manage participants' electronic certificates.

  • This paper is aimed to propose an testing and evaluation strategy and plan in Military Information Systems, especially focusing on development an“New-generation Wargame Model”. Through this research, we concluded that the effective and efficient testing/evaluation strategy/plan can aid the productivity, maintainability, availability, etc., of the Militray Information Systems. Highlights of the proposed testing and evaluation strategy and plan for Military Information Systems are as follows. First, in the unit and module integration testing phase, hybrid of black-box and white-box testing techniques are available for Military Information Systems and progressive approach for module integration phase should be considered because of the complexity. Second, in the system testing phase, integrated module should be tested with respect to the function and performance that should be satisfied with the user requirements, specifications, risk analysis, etc., Third, in the acceptance testing phase, reliability, interoperability, maintainability, availability, integrity, etc., must be considered in the actual or mini-operational environments for testing efficiently and effectively.

  • The demand for steam generators is increasing in industrial systems in which the design strategy should be implemented for safe and efficient operation of steam generators. It is, however, difficult to design a controller by the conventional method because of the nonlinear dynamics of the steam generator and influences by the set value of disturbance. This paper presents an automatic parameter optimization technique for a multivariable fuzzy controller using evolutionary strategy, At first, we use the steady state information such as a steady state gain matrix(SSGM) and a relative gain matrix(RGM). We can obtain much information on the control inputs and the outputs of the boiler-turbine system from the matrices. In order to determine the structure of the controller by using RGM and SSGM, the fuzzy rules are trained by evolutionary strategy. The good performance of the proposed multivariable fuzzy controller is verified through simulations.

  • The optimization of fed-batch yeast fermentation process has been performed using genetic algorithm(GA). Three strategies were designed and applied to obtain the optimal feed rate profiles. Genes in the chromosome (input variables for optimization) included feed rates on fixed time intervals (strategy I), or swiching times $t_s1\;and\;t_s2$, and feed rates on singular arc (strategy II), or feed rates and the length of time interval (strategy III). Strategy III showed the best results for all initial conditions due to efficient utilization of genetic information. Simulation results using GA showed similar or better performance compared with previous results by variational caculus and singular control approach.

  • This paper proposes the strategy of ladder trades in a symbol pool and simulates a performance of it on the Multicharts' system trading tool. The ladder trading strategy is based on a multi-entry strategy which is efficient for several symbols. A symbol pool is composed of verified stocks comprising KOSPI200 and 17 symbols are selected for an examination which equity is over 10 billion. From July to December in 2009, optimum parameters are searched and the results are delta is 4% and ladder is 5. When those parameters are adopted, the profit is 1,000,000 won as a single trade and the number of trade is 188. The next study is to modify a ladder shape for increasing probability and to find the difference of optimum parameters according to trading months.

  • In this paper, we propose an efficient dynamic workload balancing strategy which improves the performance of high-performance computing system. The key idea of this dynamic workload balancing strategy is to minimize execution time of each job and to maximize the system throughput by effectively using system resource such as CPU, memory. Also, this strategy dynamically allocates job by considering demanded memory size of executing job and workload status of each node. If an overload node occurs due to allocated job, the proposed scheme migrates job, executing in overload nodes, to another free nodes and reduces the waiting time and execution time of job by balancing workload of each node. Through simulation, we show that the proposed dynamic workload balancing strategy based on CPU, memory improves the performance of high-performance computing system compared to previous strategies.

A Refolding Strategy for Recombinant Metalloprotease

  • The partial cDNA of the MT-c clone encoding snake venom metalloprotease was subcloned and expressed in E. coli. The expressed metalloprotease was purified by affinity chromatography in the presence of urea, and then successfully refolded into its functional form, retaining metalloprotease activity that hydrolyzes fibrinogen. The simple and convenient refolding strategy established in this work was highly efficient in recovering the recombinant enzyme activity. Experimental evidence suggests that the C-terminal amino acid stretch of 16 residues is a critical sequence for proper folding of the metalloprotease domain.

  • This paper develops a method of modifying the usual maximum likelihood estimators of the parameters of a two state Markov process when the trajectory of the process can only he observed at regular epochs. The method utilizes the limiting behaviors of the process and the properties of state transition counts. An efficient adaptive strategy to be used together with the modified estimator is also proposed. The properties of the new estimators and the adaptive strategy are investigated using Monte Carlo simulation.
