Numerical Analysis of Soil Vapour Extraction Remediation System using Prefabricated Vertical Drain (토목섬유 연직배수재를 활용한 토양증기추출복원시스템의 수치해석)

  • Soil vapor extraction (SVE) is an effective and cost efficient method of removing volatile organic compounds (VOCs) and petroleum hydrocarbons from unsaturated soils. However, soil vapor extraction becomes ineffective in soils with low gas permeability, for example soils with air permeabilities less than 1 Darcy. The aim of this study is to investigate numerically the performance of a prefabricated vertical drain (PVD) as a SVE well, and the pattern of the induced air flow. A validated numerical model for a single PVD extraction well is developed based on the result of a well-designed laboratory model test. The validity of the simple analytical approach to determine air permeability based on the results of model tests is also discussed.

  • The purpose of this study is to present criteria of selection of optimum grout materials through analyzing the limitation of permeability of each materials(MSG-N, OPC), in various ground conditions by comparing presentation of strength and permeability of MSG method and LW method(or SGR method). To do that, physical and chemical characteristics of grout materials were analyzed and compressive tests of homogel, mixed coagulation materials and hardening materials in certain mixing ratio, and of milk paste. In addition, permeability tests for each ground soil, each injection pressure, and each materials in combined stratum were performed with massive chamber. The results of tests showed that ultra fine grout materials like grout of MSG is necessary to construct effective grouting in sand and silty sand ground. Also, it is expected to become chemical grouting guide data to layout construction engineers because presented proper injection pressure by kind of object ground in case using ultra-fine grout material.

Characterization of valacyclovir transport mechanism across the intestinal epithelium

  • Valacyclovir is a L-valyl ester prodrug of acyclovir which is a highly effective and selective antiviral agent in the treatment of herpes virus diseases. Valacyclovir is rapidly and almost completely converted to acyclovir and increases the oral bioavailability of acyclovir three to five fold. However, the intestinal absorption mechanism of valacyclovir is not clear. If the improved absorption mechanism of valacyclovir is fully understood, it will provide a rationale of designing the amino acid ester prodrugs of polar drugs containing hydroxyl group. The main objective of our present study is to characterize the membrane transport mechanism of valacyclovir. Methods : Intestinal absorption of valacyclovir was investigated by using in-situ rat perfusion study and its wall permeability was estimated by modified boundary layer model. The membrane transport mechanism was also investigated through the uptake study in Caco-2 cells and in CHO-hPepTl cells. Results : In the rat perfusion study, the wall permeability of valacyclovir was ten times higher than acyclovir and showed concentration dependency, Valacyclovir also demonstrated a D,L stereo-selectivity with L-isomer having an approximately five-fold higher permeability than D-isomer. Mixed dipeptides and cephalexin, which are transported by dipeptide carriers, strongly competed with valacyclovir for the intestinal absorption, while L-valine did not show any competition with valacyclovir. This indicated that the intestinal absorption of valacyclovir could be dipeptide carrier-mediated. In addition, the competitive uptake study in Caco-2 cells presented that dipeptides reduced the valacyclovir uptake but valine did not. Also, in IC$\sub$50/ study, valacyclovir showed strong inhibition on the $^3$H-gly-sar uptake in CHO-hPepTl cells over-expressing a human intestinal peptide transporter. Taken together, the result from our present study indicated that valacyclovir utilized the peptide transporter for the intestinal absorption.

The Magnetic Properties of FeBSiNb Alloy Ribbons with High Glass forming Ability (고 비정질 형성능을 가진 FeBSiNb 합금 리본의 자기적 특성)

  • Amorphous FeBSiNb alloy ribbons having bulk glass forming ability and high saturation magnetic flux density were produced by single-roller melt spinning apparatus in the thickness range of 22∼102㎛. With the increase of thickness, the coercive force and squareness ratio decreased, while maximum permeability and AC permeability increased. However electrical resistivity was almost constant. Furthermore refined and complex magnetic domain structure was observed in thicker ribbons owing to the change in internal magnetic anisotropy. For the alloy with the thickness of 81㎛, small coercive force of 24 mOe and high effective permeability of 12,000 at 1㎑ were obtained, those are considered to be better comparing to the conventional soft magnetic amorphous alloys (∼20 ㎛). The good soft magnetic properties of the thick FeBSiNb amorphous alloys were attributed to the decrease in surface pinning effect during wall motion, appearance of perpendicular anisotropy and resulted domain refinement.

Killing Effects of Different Physical Factors on Extracorporeal HepG2 Human Hepatoma Cells

  • Objective: To determine the killing effects on extracorporeal HepG2 cells under different temperatures, pressures of permeability and lengths of treatment time. Method: According to different temperatures, pressures of permeability and lengths of treating time, extracorporeal HepG2 cells of human hepatoma cell-line were grouped to 80 groups. Cell index (CI) as the measurement of killing effect were calculated by monotetrazolium (MTT) methods, i.e., CI =1- (the OD value in treated group - the OD value in blank control group) / (mean of untreated control group - mean of blank control group). According to the factorial design, data were fed into SPSS 10.0 and analyzed by three-way ANOVA (analysis of variance). Result: Temperature, pressure of permeability and length of treating time all had effects on the CI (cell index) level. Length of treating time was the most influential factor of the three. Additionally, any two of them all had statistically significant interactive effects on the CI level. When treated for 5-30 min, destilled water at $46^{\circ}C$ stably generated the highest CI. Conclusion: The "$46^{\circ}C$-destilled water-60 min" was considered as the optimal combination of conditions which lead to highest CI. We suggest exerting celiac lavage for 15 min with stilled water at $40^{\circ}C-43^{\circ}C$ in surgical practice as a hyperthermia treatment to achieve ideal killing effects on free cancer cells, which is feasible, practical, and clinically effective.

A Study on the Minimization of Water Damage for the Asphalt Inlay of Old Concrete Pavement (노후 콘크리트 포장 절삭 덧씌우기의 침투수에 의한 파손 최소화 방안 연구)

  • PURPOSES: The purpose of this study was to investigate the disintegration mechanism of concrete due to the infiltration of the moisture to the milling overlay pavement and to come up with a method to minimize the disintegration as well as verifying the effectiveness of the edge sealing and Fogseal method. METHODS : This study investigated the distress mechanism due to the infiltrated moisture remaining in the milling overlay pavement through chloride freezing test and verified the effectiveness of the sealing of the milling edge and fog seal methods, which have been devised to minimize the moisture infiltration, through laboratory water permeability test. Additionally, long-term pavement performance was compared for the effectiveness of the proposed method through under loading test, and field water permeability test was carried out to verify the field applicability of the proposed method. RESULTS: The result of the research confirmed that chloride deteriorates the concrete surface through disintegration and lowers its strength and that the laboratory moisture infiltration test verified the effectiveness of the milling edge sealing and fog seal methods in the deterrence of moisture infiltration to the overlay pavement with excellent long-term performance of the pavement treated with the proposed method. Although the field water permeability test revealed some deterrence of moisture infiltration of the milling edge sealing and fog seal methods to a certain extent, the difference was a little. CONCLUSIONS: The milling edge sealing and fog seal methods are limited in their effectiveness for the cases of improvident compaction management or mixture with large void, and it is believed that installation of subsurface drainage is more effective in these cases.

Development of Luteinizing Hormone Releasing Hormone (LHRH) Delivery Systems for Vaginal Mucosal Route

  • The objective of this study was to find a rational dosage form for vaginal mucosal delivery of LHRH. Vaginal absorption of LHRH was estimated by measuring its ovulation inducting effect in rat and in vitro vaginal membrane permeation study in rabbit. THe effects of different hydrogel bases, such as Polycarbophil and Pemulen compared with solutions on vaginal membrane permeation of LHRH were investigated. Sodium laurate, disodium ethylenediamine brane permeaiton of LHRH were investigated. Sodium ethylenediamine tetraacetate (EDTA) and sodium tauro-24, 25-dihydrofusidate (STDHF), which are effective peptidase inhibitors were chosen as additives to a LHRH hydrogel delivery system and LHRH solutions. A Polycarbophil compared with a solution formulation 3.4 times increase in LHRH vaginal membrane permeability compared with a solution formulation. Vaginal membrane permeability from the Polycarbophil was greater than that from Pemulen hydrogels. This may be due to the larger bioadhesive values. LHRH solution with EDTA(2%), STDHF(1%) and sodlaurate(0.5%) showed 4.1 times, 4.8 times and 6.0 times of ovulation inducing activity compared with control. These results suggest that enzyme inhibition effect of EDTA, STDHF and sod, laurate may be result in substantial enhancement of vaginal absorption. By administraiton of Polycarbophil hydrogels containing LHRH the ovulation inducing activity was 3.3 times greater than the solutions. This result indicates the bioadhesive hydrogels as well as peptidase in hibition significantly improved absorption of LHRH. By coadministration with these inhibitors the ovulation inducing activity of Polycarbophi hydrogel containing LHRH was comparable with subcutaneous administration in ovulation inducing activity.

Paeonia lactiflora Inhibits Cell Wall Synthesis and Triggers Membrane Depolarization in Candida albicans

  • Fungal cell walls and cell membranes are the main targets of antifungals. In this study, we report on the antifungal activity of an ethanol extract from Paeonia lactiflora against Candida albicans, showing that the antifungal activity is associated with the synergistic actions of preventing cell wall synthesis, enabling membrane depolarization, and compromising permeability. First, it was shown that the ethanol extract from P. lactiflora was involved in damaging the integrity of cell walls in C. albicans. In isotonic media, cell bursts of C. albicans by the P. lactiflora ethanol extract could be restored, and the minimum inhibitory concentration (MIC) of the P. lactiflora ethanol extract against C. albicans cells increased 4-fold. In addition, synthesis of $(1,3)-{\beta}-{\small{D}}-glucan$ polymer was inhibited by 87% and 83% following treatment of C. albicans microsomes with the P. lactiflora ethanol extract at their $1{\times}MIC$ and $2{\times}MIC$, respectively. Second, the ethanol extract from P. lactiflora influenced the function of C. albicans cell membranes. C. albicans cells treated with the P. lactiflora ethanol extract formed red aggregates by staining with a membrane-impermeable dye, propidium iodide. Membrane depolarization manifested as increased fluorescence intensity by staining P. lactiflora-treated C. albicans cells with a membrane-potential marker, $DiBAC_4(3)$ ((bis-1,3-dibutylbarbituric acid) trimethine oxonol). Membrane permeability was assessed by crystal violet assay, and C. albicans cells treated with the P. lactiflora ethanol extract exhibited significant uptake of crystal violet in a concentration-dependent manner. The findings suggest that P. lactiflora ethanol extract is a viable and effective candidate for the development of new antifungal agents to treat Candida-associated diseases.

Brain Delivery of $^{99m}Tc$-Diethylene Triamine Pentaacetic Acid and Phenytoin by Transient Osmotic Opening Method in Rats (흰쥐에서 삼투개열법에 의한 $^{99m}Tc$-Diethylene Triamine Pentaacetic Acid 및 페니토인의 뇌로의 송달)

  • The blood-brain barrier (BBB) of rats was modificated opening reversibly by infusing a hyperosmotic solution of arabinose (1.6 molal) into the right external carotid artery. Pre vious studies demonstrated that permeability was increased maxmmally in the first 15 min and remained slightly elevated at 1 hr. As control reference, saline was used. In the present study, to evaluate the effects of osmotic BBB opening on the BBB trasport according to hydrophilic or hydrophobic characteristics of drugs. And the differences of the uptakes of these compounds to right (treated osmotic opening) and left (untreated) hemispheres in same rats were compared each other following injection of 8 mCi per rat of $^{99m}Tc$-ethylene triamine pentaacetic acid (DTPA) as hydrophilic drug or 5mg/kg of phenytoin as hydrophobic drug mto the right external carotid artery of rats between two groups (1.6 molal arabinose vs saline). The uptakes of $^{99m}Tc$-DTPA and phenytoin in the right cerebral hemispheres were increased to about thirty three times and twice rather than those in the left cerebral heimspheres, respectively. And PAs (permeability X capillary surface area) were also increased from a control mean of 2.11${\times}10^{-4}$ (Untreated) to 6.98${\times}10^{-3}\;sec^{-1}$ (treated osmotic opening for $^{99m}Tc$-DTPA and 0.29 to 0.17 $sec^{-1}$ for phenytoin, respectively. From the results of present study, it is noted that osmotic opening of BBB is more effective in the brain delivery of hydrophilic drugs rather than that of hydrophobic drugs.

Air-tightness Evaluation of Tube Structures for Super-speed Tube Railway Systems: I. Analytical Modeling and Material Test (초고속 튜브철도 시스템을 위한 튜브 구조물의 기밀성 평가 : I. 해석모델 수립 및 재료 기밀성)

  • This paper presents a preliminary study for air-tightness evaluation of vacuum tube structures for super-speed tube railway systems. The formula for flow rate of the air caused by the pressure difference of the inside and outside of the tube structure is derived based on Darcy's law. A test is then performed to measure the air-permeability of concrete with various compressive strengths, the result of which is used for analytical simulation of the air intrusion for a tube structure with a preliminarily defined section. It has been shown that concrete with the compressive strength of at least more than 50MPa is recommended for effective operation and maintenance of the vacuum pump systems, as the air-permeability of concrete is inversely proportional to the exponent of its compressive strength.
