조선 탑재일정의 부하 평준화를 위한 유전 알고리듬 (A Genetic Algorithm Application for the Load Balancing of Ship Erection Process)

  • In this paper, we develope a genetic algorithm for the erection scheduling in shipbuilding. Erection, the final manufacturing stage of shipbuilding, involves the landing and joining of blocks at drydock. Since several ships are built simultaneously at the same drydock and they compete with the common constrained production resource such as labor, space, and crane, we should consider both ship-specifics and common resource constraints for the desirable erection scheduling. Ship erection should also satisfy the predetermined dock cycle given from higher level production planning. Thus, erection schedule of a ship can be represented as a PERT/CPM project network with its own deadline. Since production resources are shared, the erection scheduling become the so-called multi-project scheduling problem with limited resources, which can not be solved easily due to the large size of project network. We propose a function as a minimization of load index which represented the load deviation over time horizon considering the yard production strategy. For the optical parameter setting, we tried various experiments. We verified that the proposed approach was effective to deal with the erection scheduling problem in shipbuilding.

우모분해세균 Bacillus megaterium F7-1에 의한 Keratinolytic Protease의 생산 (Production of a Keratinolytic Protease by a Feather-Degrading Bacterium, Bacillus megaterium F7-1)

  • 본 연구에서는 우모의 생물학적 처리를 위하여 keratinolytic pretense를 생성함으로써 우모를 분해할 수 있는 Bacillus megaterium을 붕괴된 우모로부터 분리하였다. 본 균주에 의한 keratinolytic pretense생산 최적배지 조성 및 배양조건은 0.2% glucose, 0.8% skim milk, 0.05% NaCl, 0,01% $(K_2HPO_4$, 0.02%, $(KH_2PO_4$, 0.01% $MgCl_2$, 초기 pH 6.5 및 $25^{\circ}C$이었다. 특히, skim milk의 첨가는 효소 생산에 가장 효과적이었다. 최적조건에서 배양 5일만에 269 U/ml의 효소가 생산되었으며, 배양 6일경 98%의 우모가 분해되었다.

Characterization of Bacteriocin Production by Lactococcus lactis LAB3113 Isolated from Kimchi

  • A lactic acid bacterium LAB3113, isolated from traditionally fermented Kimchi was found to produce bacteriocin whose activity was very specific toward lactobacilli and not effective against any other bacteria. Lactobacilli affected by the inhibitory substance included Lactobacillus delbrueckii-lactis, L. johnsonii, L. gsseri, and L. curvatus. Based upon biochemical and physiological characteristics, LAB3113 was classified as Lactococcus lactis, and its bacteriocin was named as lactococcin K3113. Lactococcus lactis. LAB3113 produced bacteriocin at th early stage of growth and the concentration of the bacteriocin did not decrease even after alt stationalry phase. Optimal temperature of bacteriocin production was $25^{\circ}C$ at the initial pH 7.0. Partially purified lactococcin K3113 was completely inactivated by protease, but not affected by lipase, lysozyme and RNase. The bacteriocin was very heat-stable even after autoclaving for 20 min. It was also stable in pH changes, an was not affected by th presence of solvents. lacotococcin K3113 appeared to act in bactericidal mode against L. delbrueckii-lactis ATCC4797. Molecular weight of lactococcin K3113 was calibrated as 10,500 dal by SDS-PAGE an activity staining. Lactococcus lactis LAB3113 had four residential plasmids of 3.7kb, 11.2kb, 15.5kb, and 48kh in molecular sizes. Plasmid profile analysis of mutant strain revealed that 15.5 kb plasmid was re-sponsible for the production of lactococcin K3113 and its immunity to the bacteriocin.

  • Digital Manufacturing-based production could be very effective in shipbuilding in order to save costs and time, to increase safety for workers, and to prevent bottleneck processes in advance. Digital shipbuilding system, a simulation-based production tool, is being developed to achieve such aspects in Korea. To simulate material flow in a subassembly line at a shipyard, the product, process and resources was modeled for the subassembly process which consisted of several sub-processes such as tack welding, piece alignment, tack welding, and robot welding processes. The analysis and modeling were carried out by using the UML(Unified Modeling Language), an object-oriented modeling method as well as IDEF(Integration DEFinition), a functional modeling tool. Initially, the characteristics of the shop resources were analyzed using the shipyard data, and the layout of the subassembly line was designed with the resources. The production process modeling of the subassembly lines was performed using the discrete event simulation method. Using the constructed resource and process model, the productivity and efficiency of the line were investigated. The number of workers and the variations In the resource performance such as that of a new welding robot were examined to simulate the changes in productivity. The bottleneck process floated according to the performance of the new resources. The proposed model was viewed three-dimensionally in a digital environment so that interferences among objects and space allocations for the resources could be easily investigated

Influence of Culture Conditions on Production of NGPs by Aspergillus tubingensis

  • The filamentous fungus Aspergillus tubingensis that belongs to the black Aspergillus section has the capacity to produce high-value metabolites, for instance, naphtho-gamma-pyrones (NGPs). For these fungal secondary metabolites, numerous biological properties of industrial interest have been demonstrated, such as antimicrobial, antioxidant and anti-cancer capacities. It has been observed that production of these secondary metabolites is linked with fungal sporulation. The aim of this research was to apply osmotic and oxidative environmental stresses to trigger the production of NGPs in liquid cultures with CYB (Czapek Dox Broth). In addition, numerous parameters were tested during the experiments, such as pH value, incubation time, container geometry, and static and agitation conditions. Results demonstrate that the produced amount of NGPs can be enhanced by decreasing the water activity ($a_w$) or by adding an oxidative stress factor. In conclusion, this study can contribute to our knowledge regarding A. tubingensis to present an effective method to increase NGP production, which may support the development of current industrial processes.

Design optimization for analysis of surface integrity and chip morphology in hard turning

  • The present work addresses the surface integrity and chip morphology in finish hard turning of AISI D3 steel under nanofluid assisted minimum quantity lubrication (NFMQL) condition. The surface integrity aspects include microhardness, residual stress, white layer formation, machined surface morphology, and surface roughness. This experimental investigation aims to explore the feasibility of low-cost multilayer (TiCN/Al2O3/TiN) coated carbide tool in hard machining applications and to assess the propitious role of minimum quantity lubrication using graphene nanoparticles enriched eco-friendly radiator coolant based nano-cutting fluid for machinability improvement of hardened steel. Combined approach of central composite design (CCD) - analysis of variance (ANOVA), desirability function analysis, and response surface methodology (RSM) have been subsequently employed for experimental investigation, predictive modelling and optimization of surface roughness. With a motivational philosophy of "Go Green-Think Green-Act Green", the work also deals with economic analysis, and sustainability assessment under environmental-friendly NFMQL condition. Results showed that machining with nanofluid-MQL provided an effective cooling-lubrication strategy, safer and cleaner production, environmental friendliness and assisted to improve sustainability.

Overexpression of Mutant Galactose Permease (ScGal2_N376F) Effective for Utilization of Glucose/Xylose or Glucose/Galactose Mixture by Engineered Kluyveromyces marxianus

  • Mutant sugar transporter ScGAL2-N376F was overexpressed in Kluyveromyces marxianus for efficient utilization of xylose, which is one of the main components of cellulosic biomass. K. marxianus ScGal2_N376F, the ScGAL2-N376F-overexpressing strain, exhibited 47.04 g/l of xylose consumption and 26.55 g/l of xylitol production, as compared to the parental strain (24.68 g/l and 7.03 g/l, respectively) when xylose was used as the sole carbon source. When a mixture of glucose and xylose was used as the carbon source, xylose consumption and xylitol production rates were improved by 195% and 360%, respectively, by K. marxianus ScGal2_N376F. Moreover, the glucose consumption rate was improved by 27% as compared to that in the parental strain. Overexpression of both wild-type ScGAL2 and mutant ScGAL2-N376F showed 48% and 52% enhanced sugar consumption and ethanol production rates, respectively, when a mixture of glucose and galactose was used as the carbon source, which is the main component of marine biomass. As shown in this study, ScGAL2-N376F overexpression can be applied for the efficient production of biofuels or biochemicals from cellulosic or marine biomass.

Selective adsorption of Ba2+ using chemically modified alginate beads with enhanced Ba2+ affinity and its application to 131Cs production

  • The 131Cs radioisotope with a short half-life time and high average radiation energy can treat the cancer effectively in prostate brachytherapy. The typical 131Cs production processes have a separation step of the cesium from 131Ba to obtain a high specific radioactivity. Herein, we suggested a novel 131Cs separation method based on the Ba2+ adsorption of alginate beads. It is necessary to reduce the affinity of alginate beads to cesium ions for a high production yield. The carboxyl group of the alginate beads was replaced by a sulfonate group to reduce the cesium affinity while reinforcing their affinity to barium ions. The modified beads exhibited superior Ba2+ adsorption performances to native beads. In the fixed-bed column tests, the saturation time and adsorption capacity could be estimated with the Yoon-Nelson model in various injection flow rates and initial concentrations. In terms of the Cs elution, the modified alginate showed better performance (i.e., an elution over 88%) than the native alginate (i.e., an elution below 10%), indicating that the functional group modification was effective in reducing the affinity to cesium ions. Therefore, the separation of cesium from the barium using the modified alginate is expected to be an additional option to produce 131Cs.

Expatriate Staffing and Foreign Affiliate's Labor Productivity: Contingent on Foreign Production Intensity and Cultural Distance

  • Purpose - This study examines the effects of expatriate transfer on foreign affiliate's labor productivity. Design/methodology - Using Korean-owned foreign affiliate-level data, we estimate the effect of expatriate transfer on foreign affiliate's labor productivity using the system generalized method of moments model. We also consider foreign affiliate- and host country-specific contingencies and test how they are associated with expatriates in enhancing foreign affiliate's labor productivity. Findings - We consider foreign production intensity and cultural distance between the home (i.e., South Korea) and host countries as key contingencies that influence the effect of expatriates on foreign affiliate's labor productivity. We find that expatriates are effective in enhancing the labor productivity of less production-intensive foreign affiliates. This effect is strengthened as expatriates are deployed to countries that share cultural similarities with the home country. Originality/value - Considering that previous studies provide mixed results on the effect of expatriates, our findings suggest that foreign affiliate-specific operational orientation and cultural distance should be considered jointly to understand the true effect of expatriate staffing on foreign affiliate performance.

Effect of Spore-Forming Probiotics on the Poultry Production: A Review

  • Due to the bad aspects associated with the use of antibiotics, the pressure on poultry production prompted the efforts to find out suitable growth-promoting and disease-preventing alternatives. Although many cost-effective alternatives have been developed, currently, one of the most auspicious alternatives for poultry feed is spore-forming probiotics, which can exert more beneficial effects as compared to normal probiotics, because of their ability to withstand the harsh external and internal conditions which result in increased viability. Many studies have already used spore-forming probiotics to improve different parameters of poultry production. Our laboratory has recently isolated a spore-forming bacterial strain, which has the potential to be used as a probiotic. So, to provide a detailed understanding, the current review aimed to collect valuable references to describe the mechanism of action of spore-forming probiotics and their effect on all the key aspects of poultry production.
