경관지수를 활용한 채소 주산지 공간적 분포 특성 분석 (Spatial Characteristic Analysis for the Main Production Areas of Vegetables based on Landscape Indices)

  • The objective of this study is to accurately understand the spatial distribution characteristics of the main production area for the three vegetable crops such as Chinese cabbage, radish, and hot pepper. We applied the 8 landscape indices such as TA, NP, PD, LPI, LSI, PLADJ, COHESION, and CONNECT to 35 cities and counties using FRAGSTATS. In the case of main production area for Chinese cabbage, six cities and counties in Gangwon province were revealed as a relatively high degree of aggregation by cultivation parcels than other area. In addition, Gangneung city and Hongcheon county have been analyzed to be the most aggregated area in the case of radish and hot pepper, respectively. In the future, the spatial analysis method used in this study would be helpful to develop an effective regional plan of the main production area.

Optimal Culture Conditions for Production of Polygalacturonase from Bacillus sp. DFN-75 Isolated from Kimchi

  • A bacterium capable of producing polygalacturonase was isolated from kimchi, and identified as a strain of Bacillus. The effects of culture conditions and medium composition on enzyme production were investigated. Among the tested carbon sources, polygalacturonic acid or pectin was most effective for the production of the enzyme. Therefore, it seemed that the enzyme was induced when pectin or polygalacturonic acid was used as a sole source of carbon. The optimal concentration of polygalacturonic acid was 0.5%. For nitrogen sources, yeast extract was best for the production of the enzyme, at a level of 0.25%. The enzyme was maximally produced by cultivating the isolated Bacillus sp. at an initial pH of 88.0 and temperature of 45$^{\circ}C$ for 20 hours.

Erwinia sp.에 의한 Gallic Acid로부터 Pyrogallol의 생산 (Production of Pyrogallol from Gallic Acid by Erwinia sp.)

  • For the production of pyrogallot from gallic acid, about 100 strains of bacteria capable of assimilating gallic acid as a sole carbon and energy source were isolated from the soil. JH- 004 strain showing the highest activity of gallate decarboxy#lase was selected from them and identi- fied as Erwinia sp. The optimal conditions for the production of pyrogallol from gallic acid were examined. The resting cells of JH-004 cultured in a complex medium containing 0.2%(w/v) gallic acid were prepared after the treatment of the pellet with a freezing and thawing, and used as a enzyme source. The reaction mixtures for the maximal production of pyrogallol were shown to be 6 g/l of resting cells and 15 g/l of gallic acid in 25 mM potassium phosphate buffer. The optimal pH for the reaction was 5.0 and the optimal temperature was 35$\circ$C . Additionally, Triton X-100(0.01%, w/v) was found to be most effective for the production of pyrogallol. Under the above conditions, 10.27 g/l of pyrogallol was produced from 15 g/l of gallic acid after incubation of 35 hrs. This amount of pyrogallol corresponds to a 92.37% yields, based on gallic acid.

  • Sound nutrition is the basic foundation for the health of a population. Proper nutrition education through the school system can actualize the building of sound nutrition being provided at school lunch programs along with nutrition education can have far reaching impact to the health of the whole nation. In Hong Kong, the recent change over from a half day to full day school program signaled the need for a school lunch program. However, there has been no organized effort so far in Hong Kong to provide such service. School lunches are being provided by caterers who are more concerned about profits than food safetly. Alarming rates of food poisoning occurring recently in Taiwan school has further accentuated the importance of fool safety. With the advent of centralized food production technologies, Dietirians can apply their nutrition knowledge and integrate with food production processes such that safe, healthy and nutritious mcals can be produced in large quantities in a cost-effective manner serving he needs of the school children. This paper introduces the centralized food production concept, using some of the cool chill production centers for school lunch programs in the United States as examples to discuss how new technologies and management system can be integrated to yield the best outcome in food quality and safety.

An Observation Supporting System for Predicting Citrus Fruit Production

  • The purpose of this study is to develop a growth prediction model that can predict growth and development information influencing the production of citrus fruits: the growth model algorithm that can predict floral leaf ratio, number of fruit sets, fruit width, and overweight depending on the main period of growth and development with consideration of the applied weather factors. Every year, large scale of manpower was mobilized to investigate the production of outdoor-grown citrus fruits, but it was limited to recycling the data without an observation supporting system to systemize the database. This study intends to create a systematical database based on the basic data obtained through the observation supporting system in application of an algorithm according to the accumulated long term data and prepare a base for its continuous improvement and development. The importance of the observed data is increasingly recognized every year, and the citrus fruit observation supporting system is important for utilizing an effective policy and decision making according to various applications and analysis results through an interconnection and an integration of the investigated statistical data. The citrus fruit is a representative crop having a great ripple effect in Jeju agriculture. An early prediction of the growth and development information influencing the production of citrus fruits may be helpful for decision making in supply and demand control of agricultural products.

고온성 방선균에 의한 효소생산에 관한 연구 제 1보 $\alpha$-Amylase생산균의 분이 및 효소생산 조건 (Studies on the Production of Enzymes by Thermophilic Actinomycetes Part I. Isolation and Culture Conditions of Thermostable $\alpha$-Amylase Producing Actinomycete)

  • In the course of studies on the production of thermostable amylases by thermophilic actinomycetes isolated from soils the investigation was carried out on the production of $\alpha$-amylase by G-1011 strain which had presented the most remarkable $\alpha$-amylase formation ability among 128 amylolytic isolates. The results were as follows : 1. Characteristics of G-1011 strain were compared with those descriptions of thermophilic actinomycetes given in Bergey's Manual. The strain was identical to these species of actinomycetes. The details of physiological properties of the strain luould he published in near future. 2. The optimum temperature for incubation of the cell growth of G-1011 strain and $\alpha$-amylase production by the strain was revealed to 5$0^{\circ}C$. 3. The effective medium for $\alpha$-amylase formation by the strain was consisted of 3.0%, soluble starch, 1.0%, peptone, 0.5%, yeast extract, 0.5%, NaCl, 0.1%, MgSO$_4$ㆍ7$H_2O$ 0.02%, $K_2$MPO$_4$and 0.002% FeSO$_4$ㆍ7$H_2O$. The pH of the medium was ajusted to 7.0 with phosphate buffer solution. 4. The maximum production of $\alpha$-amylase (3420 D. U/ml) by G-1011 strain resulted when it was grown for 16 hours with the culture of reciprocal shaking.

Candida parapsilosis 돌연변이주에 의한 Xylitol 생산의 배지조건 최적화

  • Medium optimization for xylitol production from xylose by Candida parapsilosis ATCC 22019 mutant was performed. Effect of various nitrogen sources on xylitol production was investigatied. Of inorganic nitrogenous compounds, ammonium sulfate was effective for xylitol production and yeast extract was the most suitable orangic nitrogen nutrient for enhancement of xylitol production. Effect of inorganic salts such as KH$_{2}$PO$_{4}$ and MgSO$_{4}$-7H$_{2}$0 on xylitol production was also studied. Optimal medium was selected as xylose of 50 g/l, yeast extract of 5 g/l, (NH4$_{4}$)$_{2}$SO$_{4}$ of 5 g/l, KH$_{2}$PO$_{4}$ of 5 g/l, MgSO$_{4}$-7H$_{2}$O of 0.2 g/l. In a fermentor by using the optimal medium, a final xylitol concentration of 37 g/l could be obtained from 50 g/l of xylose with a xylitol yield of 74% and a xylitol productivity of 0.58 g/1-hr. At 300 g/l xylose, fermentation was also carried out and then a final xylitol concentration of 242 g/l was obtained at 272 hours. It was corresponding to xylitol yield of 80.7% and xylitol productivity of 0.58 g/1-hr.

3D 매트릭스 개질기를 활용한 모사 바이오가스 부분산화 및 수증기 영향 연구 (The Study of Effect of Steam on Partial Oxidation for Model Biogas using 3D Matrix Reformer)

  • New type of syngas generator based on the partial oxidation of biogas in volumetric permeable matrix reformers was suggested as an effective, adaptable and relatively simple way of syngas and hydrogen production for various low-scale applications. The use of biogas as an energy source reduces the chance of possible emission of two greenhouse gases, $CH_4$ and $CO_2$, into the atmosphere at the same time. Its nature of being a reproducible energy source makes its use even more attractive. Parametric screening studies were achieved as air ratio, biogas component ratio, input gas temperature, Steam/Carbon ratio. As the air ratio was low, the production of the hydrogen and carbon monoxide increased in the condition that 3D matrix reformer maintains the stable driving. As it was the simulation biogas in which the carbon dioxide content is high, the flammable range became narrow. And the flammable range was extended if the injected gas was preheated. The stable driving was possible in the low air ratio. The amount of hydrogen production was increased as S/C ratio increased.

Camptotheca acuminata 현탁배양에서 세포응집이 Camptothecin 생산에 미치는 영향 (Effect of Cell Aggregation on Camptothecin Production in Suspension Cultures of Camptotheca acuminata)

  • 최근 항암제로서 관심을 끌고있는 camptothecin올 생산하기 위한 희수(Camptotheca acuminata) 현탁세포 배양과 cam­p ptothecin의 생산을 촉진하는 연구를 수행하였다. 장기간 배양 으로 발생되는 이차대사산물 생산능력의 변이가 관찰되어 이 를 극복하고자 세포의 응집을 통한 camptothecin의 생산을 촉진하고자 하였다. 세포의 응집은 hybrid 배지와 sucrose 4 %일 때에 가장 많이 유도되었고, 이때 camptothecin의 생산량도 $18.04{\times}10^{-4} mg/L$로 가장 높은 생산성을 가졌다. 또한, 세포의 응집을 유도하기 위한 일환으로 교반속도를 변화시켜 이에 대한 효과를 알아보았는데, 100rpm에서 가장 많은 세포응집이 유도되었으며, camptothecin의 생산 역시 $19.4{\times}10^{-4} mg/L$로 가장 높은 결과를 나타내었다.

Inhibitory Effect of Korean Fermented Vegetable (Kimchi) on the Growth and Aflatoxin Production of Aspergillus parasicitus-Part 1.

  • Aflatoxin $B_1$ is a mycotoxin produced by Aspergillus flavus and A. parasiticus and is a human carcinogen. This study was performed to investigate reduction of growth and aflatoxin production of A. parasiticus by kimchi. A. parasiticus was grown in a modified APT broth with the juice of kimchi (at a concentration of 7%) at $28^{\circ}C$ for 9 days. Aflatoxin $B_1$ was determined by use of high performance liquid chromatography (HPLC). The addition of the juice of kimchi significantly reduced mycelial growth and aflatoxin production during the incubation period (p<0.05). Reduction of mycelial growth of A. parasiticus as the result of addition of the juice of kimchi was observed to range between 64.8 to 83.4% while reduction of aflatoxin production ranged from 62.2 to 73.0%. This study indicates that kimchi could be an effective inhibitor of aflatoxin production although mycelial growth may be permitted. More research is needed to study the inhibitory effects of the metabolites of kimchi.
