Anxiolytic-like activity of leaf extract of traditionally used Indian-Mustard (Brassica juncea) in diabetic rats

  • Brassica juncea is a polyphenols enriched edible plant, with diverse medicinal uses of different parts of which have been mentioned in the Ayurveda. The effects of 10 daily oral doses (100, 200, and 400 mg/kg/day) of a methanolic Brassica juncea leaf extract in rat models of anxiety using nondiabetic and alloxan-diabetic rats were quantified. In all the three behavioural tests used, i.e. elevated plus maze, open field, and social interaction tests, anxiolytic-like activity of the extract was observed in the diabetic animals only. Quantitatively, the efficacy of the highest tested dose of the extract in these tests was always less than those observed after its lower ones. These observations provide further experimental evidences for the conviction that Brassica vegetables could as well be useful for combating diabetes associated mental health problems.

식용 식물로부터 얻은 추출물의 두부, 어묵, 막걸리 변질균에 대한 항균성 검색 (Screening of Natural Antimicrobial Edible Plant Extract for Dooboo, Fish paste, Makkoli Spoilage Microorganism)

  • 저온유통이 불가피하면서도 저장 가능 기간이 짧은 두부, 어묵 및 막걸리의 보존성 향상을 위하여 이들의 부패에 관여하는 균을 순수분리, 이들 균주틀 대상으로 각종 식용 식물의 에탄올 및 물 추출물의 향균 효과를 비교하였다. 두부와 막걸리 변질 관여균 모두에 황백 에탄올 추출물이 확실한 향균 효과틀 보였고 어묵의 경우는 황백, 정향, 오배자, 리기다, 백급 및 적작약 에탄을 추출물이 대상 균주 모두에 뚜렷한 항균성을 보였으며 에탄올 추출물이 물 추출물보다 높은 향균성을 나라내었다. 어묵 변질 관여균 모두는 황백 에란올 추출물의 첨가 농도에 따라 중식 저해를 받았으며 2000 ppm에서는 확실한 중식 저지 현상을 보였고 백급 에탄올 추출물은 한 균주에 대해서만 효과가 뚜렸하였다. 막걸리 변질 관여균은 황백 에탄을 추출물 1000 ppm 이상을 첨가시 2균주에서 뚜렷한 중식 억제 현상을 보였다.

국내산 식용식물체의 멜라토닌 함량 분석 (Analysis of Melatonin Content from Domestic Edible Plants)

  • 뇌의 송과선에서 분비되는 호르몬인 melatonin은 생체의 산화적 손상을 방어하는 효능이 있다고 알려져 있는데, 26종의 식물체로부터 그 존재 및 함량을 분석하였다. Melatonin 함유 분획을 조제하기 위하여 동결건조된 식물체 분말을 50% ethanol로 추출한 다음 여과 후 TLC 상에서 acetone으로 전개시켜 melatonin을 분리하였다. 형광하에서 확인된 melatonin spot은 methanol로 추출한 다음 HPLC로 분석하였다. HPLC 분석시 ODS-A column을 사용하였으며 이동상은 70% ammonium acetate와 메탄올을 30 : 70(v/v)으로 섞어서 1.0 mL/min의 속도로 흘려주었다. Melatonin peak의 확인은 형광 detector를 이용하였으며 retention time은 17분이었다. 이같은 분석조건에서 셀러리, 부추, 브로콜리, 컬리플라워 등에서 높은 melatonin 함량이 확인되었다.

Ecklonia cava Extract Containing Dieckol Suppresses RANKL-Induced Osteoclastogenesis via MAP Kinase/NF-κB Pathway Inhibition and Heme Oxygenase-1 Induction

  • Ecklonia cava, an edible marine brown alga (Laminariaceae), is a rich source of bioactive compounds such as fucoidan and phlorotannins. Ecklonia cava extract (ECE) was prepared using 70% ethanol extraction and ECE contained 67% and 10.6% of total phlorotannins and dieckol, respectively. ECE treatment significantly inhibited receptor activator of nuclear $factor-{\kappa}B$ ligand (RANKL)-induced osteoclast differentiation of RAW 264.7 cells and pit formation in bone resorption assay (p <0.05). Moreover, it suppressed RANKL-induced $NF-{\kappa}B$ and mitogen-activated protein kinase signaling in a dose dependent manner. Downregulated osteoclast-specific gene (tartrate-resistant acid phosphatase, cathepsin K, and matrix metalloproteinase-9) expression and osteoclast proliferative transcriptional factors (nuclear factor of activated T cells-1 and c-fos) confirmed ECE-mediated suppression of osteoclastogenesis. ECE treatment ($100{\mu}g/ml$) increased heme oxygenase-1 expression by 2.5-fold and decreased intercellular reactive oxygen species production during osteoclastogenesis. The effective inhibition of RANKL-stimulated osteoclast differentiation and oxidative stress by ECE suggest that ECE has therapeutic potential in alleviating osteoclast-associated disorders.

The protective effect of Halal food extract in pancreatic beta cell lines.

  • In Islamic dietary guidelines, Halal foods are allowed as edible blessed food. Most foods were categorized within halal for Muslims. The main point of Halal food is that foods are clean in every process and based on Halal standard which might be different in each country. Most pancreatic ${\beta}$ cells synthetize, store, and release insulin. Specific molecular, functional as well as ultrastructural traits of pancreatic ${\beta}$ cells could control their insulin secretion properties and survival phentoype. Insulin-secreting pancreatic ${\beta}$-cells are essential regulators of mammalian metabolism. In addition, the pancreatic ${\beta}$ cell plays an important role in the pathogenesis of type 1 and type 2 diabetes as improving glucose homeostasis by preserving, expanding and improving the function of this key cell type. However, the pharmacological effect of halal food has not been unclear yet, especially food habit-dependent diabetes. The aim of the this study was to determine the preventive effect of Iran plants extract (Almond, Garlic, Cumin, Ginkgo biloba, Holy basil, Psyllium, Satureja khuzistanica, Fenugreek, Green tea, Ipomoea betatas, Blueberry) on RINm5F cells and MIN6 cells as pancreatic ${\beta}$ cell line. The cytotoxicity of the extracts of Iran plants on RINm5F cells and MIN6 cells were measured by using MTT assays. The preventive effects of Iran plant extracts were measured by WST-8 cell proliferation assay on streptozotocin (STZ)-induced cell death in MIN6 cells. In presented result showed that all extract of Iran plants (0.01-10mg/ml) did not show cytotoxicity in RINm5F cells and MIN6 cells. Among non-cytotoxic extract, the protective effects could be detect in high dose concentration. These results suggest that the extract of Iran plants may serve as a potential therapy for diabetes.

국내산 식물의 항균활성 검색 (Screening for Antimicrobial Activity from Korean Plants)

  • 한국에서 자생하는 125종의 식물에서 추출한 141가지 메탄을 추출물들에서 식품부패미생물인 Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Enterobacter aerogenes 그리고 Pseudomonas aeruginosa에 대한 항균 활성도를 조사하였다. Bacillus subtilis에 대해 가장 강한 항균력을 보인 것은 오미자였으며 Staphylococcus aureus에 대해서 대황이, Pseudomonas aeruginosa에 대해서는 산사가 가장 강한 항균력을 나타냈다. 금은화는 Bacillus subtilis를, 산사는 Staphylococcus aureus를, 모과와 황금은 Pseudomonas aeruginosa를, 모감주나무꽃, 밤나무꽃, 장미꽃과 파꽃은 Enterobacter aerogenes를, 오미자는 Escherichia coli를 제외한 나머지 4가지 균주에 모두 항균력이 있었다.

Synergistic Antifungal Activity of Magnoliae Cortex and Syzyii Flos against Candida albicans

  • Candida albicans is a dermal fungus of the human body that is known to cause oral candidiasis, vaginal candidiasis, and bloodstream infections in immunocompromised people or in certain environmental conditions. As cases of strains resistant to antifungal agents in C. albicans have been reported, studies using plant materials as safe antifungal agents are being actively conducted. In this study, a total of 17 edible plant extracts showed antifungal activity against C. albicans as a result of evaluating a 280-plant extract library using paper disk diffusion method. Among them, the four extracts with the strongest antifungal activity (Cinnamomi Cortex, Cinnamomi Ramulus, Magnoliae Cortex, and Syzygii Flos) were selected and evaluated for synergistic antifungal activity against C. albicans. The combination of Magnoliae Cortex and Syzygii Flos showed a synergistic activity. The antifungal activity was evaluated based on the concentrations of magnolol and eugenol, the respective components of Magnoliae Cortex and Syzygii Flos. Magnolol and eugenol showed synergistic antifungal activities at the concentration ratio of 1:25 - 1:61. The antifungal activity of these two compounds contributes 28 to 48% to the synergistic antifungal activity of the combination of Magnoliae Cortex and Syzygii Flos extract. In this study, we propose that a combination of Magnoliae Cortex and Syzygii Flos can effectively inhibit the growth of C. albicans and that magnolol and eugenol are the responsible inhibitory compounds.

  • The estrogenic activity of 47 plant extracts was assessed by reporter gene assay using MCF-7 breast cancer cell lines stably transfected with luciferase reporter gene. The estrogenic activity of food extracts was expressed as 17${\beta}$-estradiol(E2) equivalent concentration(EEQ), the concentration of E2 that resulted in the same relative luciferase unit(RLU) of the food extract of 0.2mg/$m\ell$.(omitted)

쐐기풀(Urtica Dioica L.추출물의 염색성 연구) (A Study on the Dyeability of Urtica Dioica L. Extract)

  • In this study, dyeability of Urtica Dioica L. extract, which is relatively less studied, was measured. The extract of this plant was used to dye cellulose and protein fiber to check its usage as a natural green dye. Three different methods were used to produce extract. Dried Urtica Dioica L. was extracted with 100% ethanol, 50% ethanol with 50% distilled water and 100% distilled water. Then dyeing solution was obtained by blending with distilled water at 1-to-1 ratio. The maximum dyeability was obtained when 100% ethanol extract of dried Urtica Dioica L. used to dye fabrics at 60 degrees celsius for 60 minutes without mordant treatment. Cotton, rayon, wool and silk were dyed and dyeability for each fiber was measured for color difference value then compared to its control. The results show that dyeability of rayon and dyeability of wool are stronger, and that when color position for each mordant is measured, color difference is most diverse on cotton with pre-mordant treatment. Color fastness to wash, perspiration and rubbing crockmeter were superb, but color fastness to light was low, therefore, additional study on this is needed to improve. Urtica Dioica L. is now expected to be used practically as green color dye as well as medicinally and edible.

High-Performance Liquid Chromatographic Quantification and Validation of Luteolin Glycosides from Sonchus brachyotus and Their Peroxynitrite-Scavenging Activity

  • In Korea, the leaves of Sonchus brachyotus (Compositae), an edible mountainous vegetable, are traditionally used to treat hepatitis and hemorrhage and are known to have diuretic action. The aqueous ethanolic extract of this plant was selected in our screening experiment using the peroxynitrite ($ONO_2^-$)-scavenging assay, and the present study was performed to qualitatively and quantitatively identify the active compounds from S. brachyotus and validate the present high-permormance liquid chromatography (HPLC) coupled with ultraviolet absorption detection method based on accuracy, precision and repeatability. Five phenolic substances including the main compound, luteolin $7-O-{\beta}-D$-glucuronopyranoside, as well as chlorogenic acid, luteolin 7-O-rutinoside, luteolin $7-O-{\beta}-D$-glucopyranoside, and luteolin, were found in the aqueous ethanolic extract of S. brachyotus. In the HPLC validation experiment, the linearity of the four compounds was established by $R^2$ values of more than 0.999 within the test ranges, and the recovery rate ranged from 98.2 - 105.3%. Luteolin 7-O-glucuronide was a predominant compound (143 mg/g of extract and 18.3 mg/g of the dry weight of plant material) with a potent peroxynitrite-scavenging effect ($IC_{50}$, $1.02{\pm}0.08{\mu}M$). Luteolin and its three glycosides together with chlorogenic acid were qualitatively and quantitatively determined using an HPLC method validated in the present study.
