The fpr gene, which encodes a ferredoxin-$NADP^+$ reductase, is known to participate in the reversible redox reactions between $NADP^+$/NADPH and electron carriers, such as ferredoxin or flavodoxin. The role of Fpr and its regulatory protein, FinR, in Pseudomonas putida KT2440 on the oxidative and osmotic stress responses has already been characterized [Lee at al. (2006). Biochem. Biophys. Res. Commun. 339, 1246-1254]. In the genome of P. putida KT2440, another Fpr homolog (FprB) has a 35.3% amino acid identity with Fpr. The fprB gene was cloned and expressed in Escherichia coli. The diaphorase activity assay was conducted using purified FprB to identify the function of FprB. In contrast to the fpr gene, the induction of fprB was not affected by oxidative stress agents, such as paraquat, menadione, $H_2O_2$, and t-butyl hydroperoxide. However, a higher level of fprB induction was observed under osmotic stress. Targeted disruption of fprB by homologous recombination resulted in a growth defect under high osmotic conditions. Recovery of oxidatively damaged aconitase activity was faster for the fprB mutant than for the fpr mutant, yet still slower than that for the wild type. Therefore, these data suggest that the catalytic function of FprB may have evolved to augment the function of Fpr in P. putida KT2440.
Human genome projects have enabled whole genome mapping and improved our understanding of the genes in humans. However, many unknown genes remain to be functionally characterized. In this study, we characterized human chromosome 4 open reading frame 34 gene (hC4orf34). hC4orf34 was highly conserved from invertebrate to mammalian cells and ubiquitously expressed in the organs of mice, including the heart and brain. Interestingly, hC4orf34 is a novel ER-resident, type I transmembrane protein. Mutant analysis showed that the transmembrane domain (TMD) of hC4orf34 was involved in ER retention. Overall, our results indicate that hC4orf34 is an ER-resident type I transmembrane protein, and might play a role in ER functions including $Ca^{2+}$ homeostasis and ER stress.
An endosulfan degrading bacterial strain, K-1321, was isolated by endosulfan-enrichment culture from a seaside sediment collected at Dadaepo Beach, Busan, Korea. The strain was identified as a Serratia sp. based on the results of morphological, biochemical and 16S rDNA homology analyses. Serratia sp. K-1321 was able to completely degrade 50 ppm endosulfan in culture media and soil within 6 weeks at $25^{\circ}C$. GC/MS analysis revealed that endosulfan diol was an intermediate of the bacterial endosulfan degradation. Considering the above results, we concluded that Serratia sp. K-1321 utilized endosulfan as a carbon source and metabolized endosulfan via a less toxic pathway, such as the formation of endosulfan diol as an intermediate.
The extent of biological diversity being revealed by molecular techniques accentuates the need to develop methods to isolate and culture the large numbers of microorganisms that remain to be studied. The discovery and characterization of novel microorganisms will provide information useful in understanding microbial ecosystems and have the potential to lead to new products for the biotechnology industry. In this review, the use of innovative techniques and exploration of unusual ecosystems, that have begun to address the challenge of isolating the "uncultured" members of the microbial population, are examined.
Activated carbons (ACs) have been used as EDLC (electric double-layer capacitor) electrode materials due to their high specific area, stability, and ecological advantages. In order to prepare ACs with high density and crystallinity, coal tar pitch (CTP) was activated by $K_2CO_3$ and the textural and electrochemical properties of the obtained ACs were investigated. Although the CTP ACs formed by $K_2CO_3$ activation had much smaller specific surface area and pore volume than did the CTP ACs formed by KOH activation, their volumetric specific capacitance (F/cc) levels as electrode materials for EDLC were comparable due to their higher density and micro-crystallinity. Structural characterization and EDLC-electrode performance were studied with different activation conditions of $CTP/K_2CO_3$ ratio, activation temperature, and activation period.
동식물의 서식지 제공 및 수문학적 조절 등 다양한 생태계 서비스를 제공하는 습지는 경관 내 습지간의 상호작용을 통해 생태계를 구성한다. 따라서 습지경관에서의 생태계의 리질리언스 등의 기능을 평가하기 위해서는 습지의 동역학적 특성과 더불어 습지간에 형성되는 생태적 연결성 분석이 필요하다. 본 연구에서는 습지를 생태학적 노드로 정의하여 습지 서식종의 분산 모형에 따라 발생하는 습지간 연결을 통해 생태 네트워크를 생성하고 네트워크 분석 방법을 통해 생태 네트워크의 특성을 비교, 분석하였다. 임계거리를 활용한 분산모델 (threshold distance)의 경우 확률적인 분산모델(exponential dispersal kernel, heavy-tailed dispersal model)과 비교하여 국지적인 군집화가 높으나 습지 간의 이동에는 비효율적인 것으로 나타났다. 반면 확률적 분산모델의 경우 국지적인 군집화는 낮게 나타나나 습지 경관 내에서의 이동은 효율적인 것으로 나타남에 따라 분산 모델에 따른 생태 네트워크 특성에 차이가 발생함을 확인하였다. 본 연구는 향후 습지경관 리질리언스 분석을 위한 생태 네트워크 구축의 기초자료로 활용될 수 있을 것으로 사료된다.

Volatile compounds (VOCs) are an important factor affecting meat quality. However, the characteristic VOCs in different parts of donkey meat remain unknown. Accordingly, this study represents a preliminary investigation of VOCs to differentiate between different cuts of donkey meat by using headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS) combined with chemometrics analysis. The results showed that the 31 VOCs identified in donkey meat, ketones, alcohols, aldehydes, and esters were the predominant categories. A total of 10 VOCs with relative odor activity values ≥1 were found to be characteristic of donkey meat, including pentanone, hexanal, nonanal, octanal, and 3-methylbutanal. The VOC profiles in different parts of donkey meat were well differentiated using three- and two-dimensional fingerprint maps. Nine differential VOCs that represent potential markers to discriminate different parts of donkey meat were identified by chemometrics analysis. These include 2-butanone, 2-pentanone, and 2-heptanone. Thus, the VOC profiles in donkey meat and specific VOCs in different parts of donkey meat were revealed by HS-GC-IMS combined with chemometrics, whcih provided a basis and method of investigating the characteristic VOCs and quality control of donkey meat.

This study conducted a comparative analysis of benthic macroinvertebrate communities in the Fosu and Benya lagoons in Ghana, based on the anthropogenic effect on the two lagoons. Salinity, oxygen, temperature, conductivity, turbidity and pH were measured, invertebrate richness and species densities were determined. The AZTI Marine Biotic Index (AMBI) and multivariate statistics were used to determine the different responses of fauna to pollution. The fauna were categorized into five ecological groups based on the degree of tolerance of the different species to pollution: disturbance-sensitive species; disturbance-indifferent species, disturbance-tolerant species, second-order opportunistic species; and first-order opportunistic species. The Fosu Lagoon supported more pollution tolerant species, whereas the Benya Lagoon had more species that were sensitive to organic enrichment under relatively unpolluted conditions. Chironomus sp., which is adapted to virtually anoxic conditions, was the most abundant in the Fosu Lagoon whereas Nemertea sp. was the most abundant in the Benya Lagoon. The numerical and relative abundance (%) of all 7 taxa in the Fosu Lagoon was 1,359 and 92.35%, respectively. The numerical and relative abundance (%) of all 34 taxa in the Benya Lagoon was 2,459 and 87.52%, respectively. Expectedly, the level of dissolved oxygen in the less saline Fosu Lagoon was higher than that in the more saline Benya Lagoon. The reduced photoperiod and photosynthetic activities of aquatic plants might account for this trend. There is a need to implement comprehensive monitoring and management initiatives for sustaining the ecological health of coastal lagoons in Ghana in order to support the many people that depend upon these ecosystems for their livelihood.

Because an ecological community consists of diverse species that vary nonlinearly with environmental variability, its dynamics are complex and difficult to analyze. To investigate temporal variations of benthic macroinvertebrate community, we used the community data that were collected at the sampling site in Baenae Stream near Busan, Korea, which is a clean stream with minimum pollution, from July 2006 to July 2013. First, we used a self-organizing map (SOM) to heuristically derive the states that characterizes the biotic condition of the benthic macroinvertebrate communities in forms of time series data. Next, we applied the hidden Markov model (HMM) to fine-tune the states objectively and to obtain the transition probabilities between the states and the emission probabilities that show the connection of the states with observable events such as the number of species, the diversity measured by Shannon entropy, and the biological water quality index (BMWP). While the number of species apparently addressed the state of the community, the diversity reflected the state changes after the HMM training along with seasonal variations in cyclic manners. The BMWP showed clear characterization of events that correspond to the different states based on the emission probabilities. The environmental factors such as temperature and precipitation also indicated the seasonal and cyclic changes according to the HMM. Though the usage of the HMM alone can guarantee the convergence of the training or the precision of the derived states based on field data in this study, the derivation of the states by the SOM that followed the fine-tuning by the HMM well elucidated the states of the community and could serve as an alternative reference system to reveal the ecological structures in stream communities.
