Algal Flora and Ecklonia stolonifera Okamura (Laminariaceae) Population of Youngdo in Busan, Korea (부산 영도의 해조상과 곰피(Ecklonia stolonifera Okamura) 군락)

  • Studies have been made of the species composition and community structure of benthic marine algae at intertidal zone and subtidal zone of Youngdo at Busan. In this area, a total of 69 marine algal species including 10 Chlorophyta, 17 Phaeophyta and 42 Rhodophyta was identified. Dominant species in coverage were Enteromorpha linza, Ulva pertusa, Gelidium divaricatum, Hildenbrandtia rubra and Chondrus ocellatus throughout the year. Enteromorpha linza, Ulva pertusa, Porphyra yezoensis, Gelidium divaricatum, Corallina pilulifera, Grateloupia elliptica and Chondrus ocellatus vertically distributed in intertidal zone, while Ecklonia stolonifera, Gelidium amansii, Prionitis cornea, Gracilaria textorii, Acrosorium polyneurum and Phycodrys fimbriata in subtidal zone. The vertical distribution of subtidal zone were characterized by Ecklonia stolonifera at 1 to 5 m depths. During a year, biomass and length of Ecklonia stolonifera gradually decreased from March to January, but increased from February. Zoosporangial sori were observed from September to December. In conclusion, number of species in this area was remarkably reduced as compared to the previous data at Busan and it’s vicinity.

Inducing the Regeneration and Maturation of Free-living Gametophytes of Ecklonia stolonifera Okamura (Laminariales, Phaeophyta) (대형갈조류 곰피 (Ecklonia stolonifera Okamura) 유리배우체의 재생 및 성숙 유도)

  • The Induction of regeneration and maturation in the free-living gametophytes of Ecklonia stolonifera Okamura was studied at four temperatures (5, 10, 15, and $20^{\circ}C$), four levels of irradiance (5, 10, 20, and 40 ${\mu}mol\;m^{-2}s^{-1}$) and three photoperiods (14:10, 12:12, and 10:14 h L:D). Female gametophyte fragments were maintained in active regeneration without reaching sexual maturity under $5{\sim}10^{\circ}C$, 10 ${\mu}mol\;m^{-2}s^{-1}$, 10:14 h (L:D), whereas the conditions for male gametophytes were slightly different at $20^{\circ}C$, 40 ${\mu}mol\;m^{-2}s^{-1}$, 10:14 h (L:D). The sexual maturation of female and male gametophytes was facilitated at $15^{\circ}C$, 20 ${\mu}mol\;m^{-2}s^{-1}$, 14:10 h (L:D). These results provide basic information for controlling the regeneration and maturation of the free-living gametophytes for artificial seed production of E. stolonifera.

In vitro Effect of Ecklonia stolonifera Okamura Extract on the Cell Growth in CCD-986sk Human Fibroblast and Melanin Formation Inhibition in Clone M-3 Mouse Melanocyte Cell Line (청정해역 곰피추출물의 세포생리활성 연구)

  • In order to investigate whether or not CCD-986sk cell line can be affected by Korean Ecklonia stolonifera Okamura, we examined the MTT assay when we treated Korean Ecklonia stolonifera Okamura extract in CCD-986sk human fibroblast cell line. The sample were tested for cell proliferation activity by means of a modification of the MTT assay. Ecklonia stolonifera Okamura extract showed significantly strong cell proliferation activity at the range of from 6.25 mg $mL^{-1}$ to 1.56 mg $mL^{-1}$ compared with control group. And in order to search for inhibition agents of skin melanin formation, we tested for inhibition effect of melanin pigmentation of Korean Ecklonia stolonifera Okamura using Clone M-3 mouse melanocyte cell lines. when we treated the extracts of Ecklonia stolonifera Okamura to the mouse melanocyte cell lines, the sample showed a significantly little formation of melanin pigments compared with control group at the only range of 200 mg $mL^{-1}$. These results suggest that extract of Korean Ecklonia stolonifera Okamura may represents an excellent candidate for inhibition of melanin pigmentation and for protection of human skin aging at in vitro level.

Seed Production and Cultivation of Ecklonia stolonifera Okamura, Phaeophyta (갈조 곰피(Ecklonia stolonifera Okamura)의 종묘생산과 양성)

  • The seed production of Ecklonia stolonifera Okamura was studied under laboratory conditions through the embryonic sporophyte stage and the field cultivation was conducted in eastern coast of Korea. The germination of zoospores occurred within 3 days and the growth of gametophytes was most rapid at $25^{\circ}C$ and $20\;{\mu}mol{\cdot}m^{-2}s^{-1}$. Sporophyte growth was highest at $20^{\circ}C$ and $20\;{\mu}mol{\cdot}m^{-2}s^{-1}$ and lowest at $25^{\circ}C$ and $80\;{\mu}mol{\cdot}m^{-2}s^{-1}$. In the nursery culture of E. stolonifera lasting for 2 weeks in January, the initial blade length of E. stolonifera (about $500\;{\mu}m$) grew to $526.3{\pm}176.0\;{\mu}m$ at water temperature of $12.05^{\circ}C$. The blade length and width reached their maxima in July, after which the ends of blade and stem began to degrade with the increase in water temperature. The degraded end of the blade started to regenerate in October, when water temperature began to decline. This species can be considered a potential candidate for aquaculture, increasing in the availability of raw material and aiding in recovery of seaweed bed.

Inhibitory Phlorotannins from the Edible Brown Alga Ecklonia stolonifera on Total Reactive Oxygen Species (ROS) Generation

  • Reactive oxygen species (ROS) play an important role in the pathogenesis of many human degenerative diseases such as cancer, aging, arteriosclerosis, and rheumatism. Much attention has been focused on the development of safe and effective antioxidants. To discover sources of antioxidative activity in marine algae, extracts from 17 kinds of seaweed were screened for their inhibitory effect on total ROS generation in kidney homogenate using 2',7'-dichlorofluorescein diacetate (DCFH-DA). ROS inhibition was seen in three species: UIva pertusa, Symphyocladia latiuscula, and Ecklonia stolonifera. At a final concentration of 25 $\mu\textrm{g}$/mL, U. pertusa inhibited 85.65$\pm$20.28% of total ROS generation, S. latiscula caused 50.63$\pm$0.09% inhibitory, and the Ecklonia species was 44.30$\pm$7.33% inhibition. E. stolonifera OKAMURA (Lam-inariaceae), which belongs to the brown algae, has been further investigated because it is commonly used as a foodstuff in Korea. Five compounds, phloroglucinol (1), eckstolonol (2), eckol (3), phlorofucofuroeckol A (4), and dieckol (5), isolated from the ethyl acetate soluble fraction of the methanolic extrclct of E. stolonifera inhibited total ROS generation.

Effect of Transplantation of Ecklonia stolonifera Okamura with Adhesive Glue (순간접착제를 이용한 곰피 (Ecklonia stolonifera Okamura)의 이식효과)

  • Experimental transplantation of Eklonia stoionifera to natural rock was carried out using adhesive Blue. A plant was attached to an architecture tile ($10\times$10 cm) with the glue, and then the tile was attached to natural rock with underwater glue. Within one month, the tile were fixed to natural rock with an attachment rate of $75\%$, and 5. stoionifera grew up with an insertion rate of $66.7\%$ on the tile. After two months, number of E. stolonifera on the tile decreased. However, the attached plants were regenerated by new stolons, maintaining good growing condition. After seven months,5. stoloaifera increased 6 to 71 individuals. These results indicate that regeneration of plants was mainly affected by the recruitment of stolen around the plant.

Hepatoprotective Constituents of the Edible Brown Alga Ecklonia stolonifera on Tacrine-induced Cytotoxicity in Hep G2 Cells

  • In this study, ethanolic extracts from 18 seaweed variants were assessed for hepatoprotective activity against tacrine-induced cytotoxicity in Hep G2 cells. Only one of these, Ecklonia stolonifera Okamura (Laminariaceae), a member of the brown algae, exhibited promising hepatoprotective activity. Bioassay-guided fractionation of the active ethyl acetate (EtOAc) soluble fraction obtained from the ethanolic extract of E. stolonifera, resulted in the isolation of several phlorotannins [phloroglucinol (1), eckstolonol (2), eckol (3), phlorofucofuroeckol A (4), and dieckol (5)]. Compounds 2 and 4 were determined to protect Hep G2 cells against the cytotoxic effects of tacrine, with $EC_{50}$ values of 62.0 and 79.2 $\mu$g/mL, respectively. Silybin, a well characterized hepatoprotective agent, was used as a positive control, and exhibited an $EC_{50}$ value of 50.0 $\mu$g/mL. It has been suggested that the phlorotannins derived from marine brown algae might prove useful sources in the development of novel hepatoprotective agents.

  • Ecklonia stolonifera Okamura is a member of the family Laminariaceae, belonging to the order Laminariales. Previously we reported that the methanolic extract of the brown alga E stolonifera exerts antioxidative activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical.
