Inhibitory Effects of Histamine Production in Mackerel Muscle by Medicinal Herbs and Seaweed Extracts (한약재 및 해조류 추출물에 의한 고등어 육중의 Histamine 생성 억제 효과)

  • This study was conducted in order to investigate the inhibitory effects of natural materials on histamine production in mackerel. Antimicrobial activities on Photobacterium phosphoreum of medicinal herbs and seaweeds were investigated using the paper disc assay and MIC (minimum inhibitory concentration) test. According to the results, Sargassum sagamianum and Ecklonia cava ethanol extracts exhibited antibacterial activity. In particular, Sargassum sagamianum ethanol extract showed excellent antibacterial activity at 0.015625 mg/mL by the MIC test. Anti-histamine release activities of natural materials were further investigated by examining their inhibitory effects on histidine decarboxylase (HDC) activity in the crude enzyme preparation from Photobacterium phosphoreum. The ethanol extracts of Ecklonia cava and Eisenia bicyclis exhibited the strongest HDC inhibitory activity, with 32% and 22%, at a concentration of 1 mg/mL, respectively. Therefore, natural materials may reduce histamine poisoning through decrease of histamine production in mackerel.

Evaluation of In-vitro Anticoagulation Activity of 35 Different Seaweed Extracts (35종 해조류 추출물의 in-vitro 항혈전 활성 평가)

  • Seaweeds have been recognized as a health food, having anti-obesity, anti-constipation and anticoagulation activities, and the use of seaweeds in the food, medicine, and cosmetic industries have recently significantly increased. In this study, methanol extracts were prepared from 35 different seaweeds (17 phaeophyta, 11 rhodophyta and 7 chlorophyta), and thrombin time (TT), prothrombin time (PT) and activated partial thromboplastin time (aPTT) were determined in order to develop safe and novel anticoagulation agents from natural products. In TT experiments, Ecklonia cava, Ecklonia stolonifera, Eisenia bicyclis (Kjellman) Setchell, Ishige foliacea, I. okamurai, Sargassum confusum and S. yamade showed strong thrombin inhibition activity among the 35 different seaweeds. In PT experiments, the inhibitions of prothrombin were identified in the selected seaweeds from TT experiment, with the exception of S. yamade. In aPTT experiments, the seaweeds with blood coagulation inhibition factors were E. cava, E. stolonifera, E. bicyclis (Kjellman) Setchell, I. foliacea, I. okamurai, S. confusum and Hixikia fusiforme Okamura. Further anticoagulation assay with the selected 8 seaweeds suggested that S. confusum is most effective in antithrombosis, and E. stolonifera, E. bicyclis (Kjellman) Setchell, and I. foliacea have high potential as antithrombosis agents. Based on components-activity correlation analysis, flavonoids are considered as active anticoagulation components of seaweeds These results suggest that edible seaweeds, especially S. confusum, have potential as safe and novel anticoagulants, and S. yamade and H. fusiforme Okamura could be used as a thrombin-specific and coagulation factor-specific inhibitors.

Antioxidant Activities and Acetylcholinesterase Inhibitory Activities from Seaweed Extracts (해조류의 항산화 활성 및 아세틸콜린에스테라제 저해 활성)

  • This study was conducted to investigate the antioxidant and acetylcholinesterase (AChE) inhibitory activities of extracts from various seaweed. The extracts of $Sargassum$ $thunbergii$ (91.3%), $Polysiphonia$ $morrowii$ (90.7%), $Ecklonia$ $cava$ (89.9%), and $Artemisia$ $fukudo$ (85.9%) showed over 80% high radical scavenging activities at the final concentration of 40 ${\mu}g$/mL. The $Artemisia$ $fukudo$ extract showed the highest inhibition activity of 30.2% on AChE at the final concentration of 10 ${\mu}g$/mL. The extract of $Porphyra$ $tenera$, $Costaria$ $costata$, $Monostroma$ $nitidum$, $Ecklonia$ $cava$, and $Agarum$ $clathratum$ against AChE at a concentration of 10 ${\mu}g$/mL exhibited inhibition of 26.6%, 25.3%, 23.4%, 21.7%, 20.4% and 19.9%, respectively. The bioautography results showed that the mixtures of structurally diverse compounds were thought to affect AChE inhibitory activity. These results suggest that extracts from seaweed with their high quality components may be effective in the prevention of Alzheimer's disease and may be used to develop various functional food products.

Macroalgal Community Structure on the Subtidal of Southern Six Islands, Korea (남해안 6개 도서의 조하대 해조류 군집구조)

  • The macroalgal community structure was examined at the subtidal zones of six study sites, on the Southern coast of Korea from between May and August 2015. A total of 132 seaweeds, comprising 10 green, 28 brown and 94 red Seaweed, were identified. The seaweed biomass was $80.32g\;dry\;wt.\;m^{-2}$ in average, and it was maximal at Geomundo ($166.94g\;dry\;wt.\;m^{-2}$) and minimal at Byeongpungdo ($14.52g\;dry\;wt.\;m^{-2}$). On the basis of the biomass, the Ecklonia cava was a representative species, distributed widely in the subtidal zone of the three islands (Yeoseodo, Geomundo, Baekdo). Also, the Sargassum sp. was dominant at Sejondo and Hongdo. The turbidity and light transmittance was divided into two groups. The seaweed community structure of group A (Byeongpungdo, Sejondo, Geomundo) was characterized by high turbidity, low light transmittance and a lower habitat depth than were observed in group B (Hongdo, Baekdo, Yeoseodo). As the water depth increased, the biomass decreased due to the lowered light transmittance. In Byeongpungdo and Sejondo, which showed high turbidity and low light transmittance, the degree of seaweed coverage was decreased with the depth of water. The ESGII ratio of the Ecological Status Group was higher than fourty percentage in Byeongpungdo and Baekdo. Community indices were as follows: dominance index (DI) 0.35-0.90, richness index (R) 7.03-17.93, evenness index (J′) 0.22-0.60, and diversity index (H′) 0.79-2.18. The Macroalgal zonation of the subtidal zone was calculated by the Ecklonia cava and Brown algal population on five islands(Byeongpungdo, Yeoseodo, Geomundo, Beakdo, Hongdo). On the other hand, Undaria pinnatifida and Sargassum sp. dominated at Sejondo. Additionally, the biomass ratio and the species richness of green algae was lower in group A. These differences in the seaweed community structure may have resulted from the effects of turbidity and light transmittance.

Characteristics of Summer Marine Algal Community and Barren Ground in the Southern Coast of Jeju, Korea (제주 남부해역 조하대 하계 해조군집 및 갯녹음 특성)

  • This study was conducted to investigate marine algal community characteristics and the status of barren ground in the summer at study sites on Jeju Island, Korea. Sampling was carried out from July to September 2017 using a qualitative and quantitative survey (including coverage of non-geniculated coralline algae and density of grazer) by scuba diving. A total of 121 species were identified, including 11 (9.1 %) green algae, 24 (19.8 %) brown algae, and 86 (71.1 %) red algae. Hyeongjeseom had the greatest diversity, with 60 species, and Harye the least, with 18 species. The mean biomass at the study sites was $1,503.0g{\cdot}m^{-2}$, while the mean for the neighboring islets ($3,268.7g{\cdot}m^{-2}$) was higher than that of the main island ($914.7g{\cdot}m^{-2}$). Also, dominant species was identified: Sargassum macrocarpum at the neighboring islets, and Ecklonia cava at the main island, with differences showing not only in biomass but also species composition. In conclusion, the marine algal community status in summer at the study sites was evaluated based on the algal community characteristics (species composition, biomass, biomass ratio of kelp species), coverage of non-geniculated coralline algae, and density of grazer. As a result, both Hyeongjeseom and Marado require preservation and management to maintain their excellent marine algal communities, and other sites on the main island require the creation and/or restoration of marine algal communities. In addition, as the generation of barren ground accelerates, it is urgent not only to grasp existing monitoring research but also to identify the status of the marine algal community where it is not known at present.

Evaluation of Effective MMP Inhibitors from Eight Different Brown Algae in Human Fibrosarcoma HT1080 Cells

  • Matrix metalloproteinases (MMPs) are crucial extracellular matrices degrading enzymes that have important roles in metastasis of cancer progression as well as other significant conditions such as oxidative stress and hepatic fibrosis. Marine plants are on the rise for their potential to provide natural products that exhibit remarkable health benefits. In this context, brown algae species have been of much interest in the pharmaceutical field with reported instances of isolation of bioactive compounds against tumor growth and MMP activity. In this study, eight different brown algae species were harvested, and their extracts were compared in regard to their anti-MMP effects. According to gelatin zymography results, Ecklonia cava, Ecklonia bicyclis, and Ishige okamurae showed higher inhibitory effects than the other samples on MMP-2 and -9 activity at the concentrations of 10, 50, and $100{\mu}g/mL$. However, only I. okamurae was able to regulate the MMP activity through the expression of MMP and tissue inhibitor of MMP observed by mRNA levels. Overall, brown algae species showed to be good sources for anti-MMP agents, while I. okamurae needs to be further studied for its potential to yield pharmaceutical molecules that can regulate MMP-activity through cellular pathways as well as enzymatic inhibition.

The Study on Antimicrobial and Antifungal Activity of the Wild Seaweeds of Jeju Island (제주 자생 해양식물들의 항균 및 항산화 활성 연구)

  • In this study, we investigated the antimicrobial and antifungal activity from the wild seaweeds of Jeju island. The active ingredients of the seaweeds were prepared by 80% methanol extraction. Antimicrobial and antifungal activity of seaweed extracts was examined. We found that 6 plant extracts among 45 plants, namely, Codium contractum, Undaria pinnatifida, Ishige sinicola, Ishige okamurai, Ishige okamuriai, Ecklonia cava, Hizikia fusiformis, Ulva fasciata, Ulva pertusa, Sargassum siliquastrum, Ecklonia kurome, Gracilaria textorii, significantly inhibited growth of harmful microorganisms. Additionally, according to DPPH assay. 2 plant extracts were found to have antioxidant activities. Taken together, these results suggest the possibility that 11 plant extracts can be utilized as an antimicrobial agent.

Residue of the Organophosphorus Pesticides in the Coastal Environment on the Cheju island (제주도 연안 해양환경 중에서 유기인계 농약의 잔류)

  • To investigate the residue of organophosphorus pesticides such as Monocrotophos[Dimethyl-1-methyl-2-methyl carbamoylvinylphosphate]and EPN[O-ethyl-O-4-nitrophenyl phenylphosphonothioate]in the coastal environment of Cheju island, samples of sea organism, water and sediment were collected at the Sackdaldong and Daepodong near the Jungmoon golf course in July and Aug. 1997. The qualified detection limit of mono-crotophos and EPN by GC -FPD were 0.024 ng/g and 0.020 ng/g in the sea organism, respectively Neither Monocrotophos nor EPN was detected in seaweed cava (Ecklonia cava), Agar (Gelidium amansii), turban sell (Batillus cornutus) and sea urchin (Anthocidaris Crassispina). They were not detected in seawater and sediment, either. These results showed that the coastal area near the Jungmoon golf course in the Cheju island was not polluted by the organophosphorus pesticides such as Monocrotophos and EPN.

SEASONAL VARIATION IN THE CHEMICAL COMPOSITION OF BROWN ALGAE WITH SPECIAL REFERENCE TO ALGINIC ACID (갈조류의 알긴산 함량의 계절적인 변화에 관하여)

  • The present investigations were carried out with the purpose of making clear the fundamental features of the seasonal variations in chemical composition of the brown algae. Three species of samples, Ecklonia cava, Sargassum sagamianum and Hizikia fusiforme have been analyzed monthly for their contents of total ash, crude protein, alginic acid, mannitol, and laminarin over a year period. Three kinds of samples were collected from the same locality, situated on the southern coast of the Che-ju Island, from September 1966 to August 1967. In addition, the comparative analysis was made on fronds and stipes of the plant for their chemical composition. The results obtained are summarized as follows: 1, In general, the three species examined underwent a similar mode of seasonal variation, and no essential difference was detected among them. 2. The chemical composition of the plant exhibited a considerable difference between the species. The content of total ash in H. fusiforme was remarkably higher than those in the two other species respectively. The alginic acid content was relatively high in S. sagamianum and low in H. fusiforme. The contents of crude protein, mannitol and laminarin were appreciably high in E. cava and low in H. fusiforme in general. 3. The most outstanding feature in the seasonal variation was that, in general, the total ash, crude protein, and alginic acid contents were at a maximum in the winter months while laminarin and mannitol contents were at a minimum. The converse was true in summer. Total ash-Maximum values were observed from December to February and minimum from August till October. Crude Protein-All species exhibited maxima in January and February, and minima from August to October. Alginic acid-Maximum contents occured from January to March and minimum from September to November. Mannitol-The maximum content of mannitol was In May and lune and minimum in January, February and March. Leminerin-Maximum content was in September and October, and minimum in January and February. 4. The wide seasonal variation in chemical constitution occured in the fronds, but the stipes showed a slight seasonal variation. In the chemical composition, the stipe was high in alginic acid, low in mannitol and laminarin. The reverse was in the frond.

Monitoring of Macroalgal Flora and Community Structure in the Subtidal Zone around Jeju Coasts and Gapado Island, Korea (2013-2015) (한국 제주 연안 및 가파도 해역의 조하대 해조상 및 군집구조 모니터링 (2013-2015))

  • We seasonally examined marine macroalgal community structures and ocean environmental characteristics in subtidal zones at five sites on and around Jeju Island, Korea, from February 2013 to November 2015. A total of 186 macroalgal species were identified, including 18 green, 33 brown, and 135 red algae. During the study period, the number of species was highest at Gapado Island (125 species) followed by Bukchon (123 species), Sagye (122 species), Sinheung (122 species) and Sinchang (97 species). Ecklonia cava, Peyssonnelia caulifera, Synarthrophyton chejuense, Corallina aberrans and Corallina crassisima occurred at all study sites and in all seasons. The average annual biomass of seaweed was $1,125.10g\;wet\;wt./m^2$ and ranged from $899.77g/m^2$ at Sinheung to $1,452.00g/m^2$ at Gapado. A brown alga E. cava was the most dominant species, accounting for 49.84% ($560.78g/m^2$) of the total seaweed biomass. Subdominant species were C. aberrans and C. crassissima, comprising 6.83% ($76.79g/m^2$) and 5.98% ($67.28g/m^2$) of total biomass, respectively. Cluster analysis revealed three distinct groups: the Sagye/Sinheung group (group A), the Bukchon/Sinchang group (group B), and the Gapado group (group C), indicating significantdifferences in macroalgal communities between sites.
