The research was carried out in order to assess, at an early stage, the potential barrier in mother-child relationship in the childs' development by using the screening tool. Consequently, after modifying the Neonatal Perception Inventory developed by Broussard and testing its reliability. It has been applied to 152 mothers at the hospital of three universities in Seoul from August 1 to September 30, 1986. The data was analyzed by S.P.S.S. pro-gram and the results were as follows: 1) Mothers perceived the newborn as an individual, and evaluated their baby higher than other babies. 2) Mothers indicated difficulties in their role performance in the fellowing order-bathing, safety, detecting pain or suffering, and feeding. 3) Mothers' perception of a newborn was positive for 75% of the mothers. 4) The factor analysis of the modified tool using principal components analysis and Varimax rotation resulted in the two factors: Factor 1, Baby as an individual: Factor Ⅱ, Performances are required to meet the new-born's needs. 5) The difference in mothers' individual characteristics such as the number of childbirth, the desire to get pregnant or not, the type of deli-very, and the sex of the newborn did not influence on a mothers' perception of her newborn. As seen above, most of the mothers perceived their newborns as able individuals and expressed difficulties involved in taking care of the newborn. Also most of the mothers perceived their babies positively. My point here is, we ought to observe those mothers who perceive their babies negatively, and then compare them with those mothers who perceive their babies extremely positive or extremely negative. In the future, for more comprehensive assessment tool for maternal perception of the newborn, a repetitive verification and modification of this tool is demanded. At the same time, father's perception of the newborn should be included.
Toxoplasma gondii is a zoonotic parasite resulting in human infections and one of the infectious pathogens leading to uveitis and retinochoroiditis. The present study was performed to assess T. gondii infection in 20 ocular patients with chronic irregular recurrent uveitis (20 aqueous humor and 20 peripheral blood samples) using PCR. All samples were analyzed by nested PCR targeting a specific B1 gene of T. gondii. The PCR-positive rate was 25% (5/20), including 5% (1) in blood samples, 25% (5) in aqueous humor samples, and 5% (1) in both sample types. A molecular screening test for T. gondii infection in ocular patients with common clinical findings of an unclear retinal margin and an inflammatory membrane over the retina, as seen by fundus examination, may be helpful for early diagnosis and treatment.

CDC48 is a member of the AAA ATPase superfamily. Yeast CDC48 and its mammalian homolog p97 are implicated in diverse cellular processes, including mitosis, membrane fusion, and ubiquitin-dependent protein degradation. However, the cellular functions of plant CDC48 proteins are largely unknown. In the present study, we performed virus-induced gene silencing (VIGS) screening and found that silencing of a gene encoding a tobacco CDC48 homolog, NgCDC48, resulted in severe abnormalities in leaf and shoot development in tobacco. Furthermore, transgenic tobacco plants (35S:anti-NgCDC48), in which the NgCDC48 gene was suppressed using the antisense RNA method, exhibited severely aberrant development of both vegetative and reproductive organs, resulting in arrested shoot and leaf growth and sterile flowers. Approximately 57-83% of 35S:anti-NgCDC48 plants failed to develop mature organs and died at early stage of development. Scanning electron microscopy showed that both adaxial and abaxial epidermal pavement cells in antisense transgenic leaves were significantly smaller and more numerous than those in wild type leaves. These results indicate that NgCDC48 is critically involved in cell growth and development of tobacco plants. An in vivo targeting experiment revealed that NgCDC48 resides in the endoplasmic reticulum (ER) in tobacco protoplasts. We consider the tantalizing possibility that CDC48-mediated degradation of an as-yet unidentified protein(s) in the ER might be a critical step for cell growth and expansion in tobacco leaves.

Antley-Bixler syndrome (ABS) is a rare form of syndromic craniosynostosis with additional systemic synostosis, including radiohumeral or radioulnar synostosis. Another characteristic feature of ABS is mid-facial hypoplasia that leads to airway narrowing after birth. ABS is associated with mutations in the FGFR2 and POR genes. Patients with POR mutations present with either skeletal manifestations or congenital adrenal hyperplasia with ambiguous genitalia. We report here two cases of ABS caused by mutations in FGFR2 and POR. Although the patients had craniosynostosis and radiohumeral synostosis in common and cranioplasty was performed in both cases, the male with POR mutations showed an elevated level of $17{\alpha}$-hydroxyprogesterone during newborn screening and was diagnosed with congenital adrenal hyperplasia by adrenocorticotropic hormone stimulation. This patient has been treated with hydrocortisone and fludrocortisone. He had no ambiguous genitalia but had bilateral cryptorchidism. On the other hand, the female with the FGFR2 mutation showed severe clinical manifestations: upper airway narrowing leading to tracheostomy, kyphosis of the cervical spine, and coccyx deformity. ABS shows locus heterogeneity, and mutations in two different genes can cause similar craniofacial and skeletal phenotypes. Because the long-term outcomes and inheritance patterns of the disease differ markedly, depending on the causative mutation, early molecular genetic testing is helpful.
Purpose: If early diagnosis is not made, patients with metabolic disorders as homocystinemia rapidly progress to physical defect or mental retardation resulted in storage of the toxic material into the brain. Therefore, it is necessary to develop an analytical method for a rapid screening and/or correct confirmation diagnosis. Methods: The standard solution of sulfur amino acids spiked plasma was subjected to protein precipitation with methanol, and then consecutively derivatized with trimethylsilyl (TMS) and trifluoroacyl (TFA) and determined by GC-MS. The formation of TMS derivative of the hydroxyl and TFA derivative of amino functional group was performed by BSTFA and MBTFA, respectively. Selective ion monitoring (SIM) mode was used for quantification with selected specific ions. Results: A calibration curve on standard spiked pooled plasma showed a linear relationship with correlation coefficient of 0.9936-0.9992 for all compounds over the range of 0.1-300 ng. The precision and accuracy were within S.D. of 1-15% and RSD of 1-15% for intra-day assay at 2 ng/mL, 15 ng/mL and 30 ng/mL. LOD and LOQ was 0.4 ng/mL and 4 ng/mL respectively. Conclusion: A rapid analytical method was developed to quantify sulfur amino acids and methyl malonic acid, after two-step derivatization procedure with good sensitivity and specificity on human plasma. Advantages of a new method are simplicity and rapidity. The method could be useful for routine analysis, diagnosis of homocysteinemia.
Background and Objectives : The purpose of this study was to investigate the criterion-related concurrent validity of two standardized auditory-perceptual assessments and the Acoustic Voice Quality Index (AVQI) for measuring dysphonia severity in patients with vocal cord paralysis (VCP). Materials and Methods : Total 210 patients with VCP and 236 normal voice subjects were asked to sustain the vowel [a:] and to read aloud the Korean text "Walk". A 2 second mid-vowel portion of the sustained vowel and two sentences (with 26 syllables) were recorded. And then voice samples were edited, concatenated, and analyzed according to Praat script. Two standardized auditory-perceptual assessment (GRBAS and CAPE-V) were performed by three raters. Results : The VCP group showed higher AVQI, Grade (G) and Overall Severity (OS) values than normal voice group. And the correlation among AVQI, G, and OS ranged from 0.904 to 0.926. In ROC curve analysis, cutoff values of AVQI, G, and OS were <3.79, <0.00, and <30.00, respectively, and the AUC of each analysis was over .89. Conclusion : AVQI and auditory evaluation can improve the early screening ability of VCP voice and help to establish effective diagnosis and treatment plan for VCP-related dysphonia.
This research was designed to determine what components of Gardenia jasminoides play a major role in inhibiting the enzymes related antidepressant activity of this plant. In our previous research, the ethyl acetate fraction of G. jasminosides fruits inhibited the activities of both monoamine oxidase-A (MAO-A) and monoamine oxidase-B (MAO-B), and oral administration of the ethanolic extract slightly increased serotonin concentrations in the brain tissues of rats and decreased MAO-B activity. In addition, we found through in vitro screening test that the ethyl acetate fraction showed modest inhibitory activity on dopamine-${\beta}$ hydroxylase (DBH). The bioassay-guided fractionation led to the isolation of five bio-active compounds, protocatechuic acid (1), geniposide (2), 6'-O-trans-p-coumaroylgeniposide (3), 3,5-dihydroxy-1,7-bis(4-hydroxyphenyl) heptanes (4), and ursolic acid (5), from the ethyl acetate fraction of G. jasminoides fruits. The isolated compounds showed different inhibitory potentials against MAO-A, -B, and DBH. Protocatechuic acid showed potent inhibition against MAO-B ($IC_{50}$$300{\mu}mol/L$) and DBH ($334{\mu}mol/L$), exhibiting weak MAO-A inhibition (2.41 mmol/L). Two iridoid glycosides, geniposide ($223{\mu}mol/L$) and 6'-O-trans-p-coumaroylgeniposide ($127{\mu}mol/L$), were selective MAO-B inhibitor. Especially, 6'-O-trans-p-coumaroylgeniposide exhibited more selective MAO-B inhibition than deprenyl, well-known MAO-B inhibitor for the treatment of early-stage Parkinson's disease. The inhibitory activity of 3,5-dihydroxy-1,7-bis (4-hydroxyphenyl) heptane was strong for MAO-B ($196{\mu}mol/L$), modest for MAO-A ($400{\mu}mol/L$), and weak for DBH ($941{\mu}mol/L$). Ursolic acid exhibited significant inhibition of DBH ($214{\mu}mol/L$), weak inhibition of MAO-B ($780{\mu}mol/L$), and no inhibition against MAO-A. Consequently, G. jasminoides fruits are considerable for development of biofunctional food materials for the combination treatment of depression and neurodegenerative disorders.
Objective : The purpose of our study was to investigate the relationship between job stress and depressive disorder among emotional laborers in a city. Methods : A total of 677 emotional laborers living in Jeonju-city participated in this study. The participants completed survey questions regarding demographic characteristics, job stress (using the Korean Occupational Stress Scale-24, KOSS-24), and depression (the Patient Health Questionnaire-9, PHQ-9). Logistic regression was performed to estimate the association between job stress and depression. Results : Our results indicated that 34.9% of the participants had a high risk of total job stress and 21.7% of participants had depression. In the multivariate logistic regression analysis, female sex (OR=1.66, 95% CI=1.01-2.72) and total job stress (OR=3.84, 95% CI=2.57-5.75) were significantly associated with depression after adjusting for confounding factors. Among the subscales of job stress, job demand (OR=3.06, 95% CI=1.99-4.72) and occupational climate (OR=1.88, 95% CI=1.19-2.97) were significantly associated with depression. But insufficient job control, interpersonal conflict, job insecurity, the organizational system and lack of rewards were not. Conclusion : Among the emotional laborers, job stress, particularly job demand and occupational climate, were major contributing factors associated with depression. Early screening and preventive strategies focusing on job stress could reduce the impact of depression in emotional laborers.
This research was performed to screen eye health status for pre-school children (4~5 age) and establish as eye health checking system m community health center. This data was collected from 1st February, 1995 to 31st October, 1995. To screen the state of their vision, the Developed Visuual Test (D.V.T) was used for pre-school children. This is used at home by their parents and then it is also used among the kindergarten health team at Jung-gu health center in Seoul. This was done with the and of Randot and Han Choun Souk tests. The total number of children who were tested was 1441. Among the children, children aged 5 were 707, and those aged 4 were 734. The results of this study were as follows; * Description of the health status for the pre-school children. 1. The rate of children average vision in both eyes, whose was below 05 by Han Choun Souk are 7.6% (age 4) and 4.8% (age 5). 2. Only 20 4% (age 5) and 30 8% (age 4) of the pre-school children had been tested through the ophthalmic department. 3. The children with a level below 05 (Han Choun Souk test) had been tested only 43% (age 4), and 12% (age 5) 4. There was no particular difficulty in understanding the D.Y.T: 13.9% (age 4), 11.6% (age 5) of the tested children had eye problems. 5. 231 cases were trichiasis, entropin, strabismus, and amblyopia. * The different visual tests.
