Suppression of Transglutaminase-2 is Involved in Anti-Inflammatory Actions of Glucosamine in 12-O-Tetradecanoylphorbol-13-Acetate-Induced Skin Inflammation

  • Glucosamine (GS) is well known for the treatment of inflammation. However, the mechanism and efficacy of GS for skin inflammation are unclear. The aim of this study was to evaluate the effects and mechanism of GS in the mouse 12-O-tetradecanoyl 13-acetate (TPA)-induced ear edema model. TPA-induced ear edema was evoked in ICR or transglutaminase 2 (Tgase-2) (-/-) mice. GS was administered orally (10-100 mg/kg) or topically (0.5-2.0 w/v %) prior to TPA treatment. Orally administered GS at 10 mg/kg showed a 76 or 57% reduction in ear weight or myeloperoxidase, respectively, and a decreased expression of cyclooxygenase-2 (COX-2), NF-${\kappa}B$ and Tgase-2 in TPA-induced ear edema by western blot and immunohistochemistry. Role of Tgase-2 in TPA ear edema is examined using Tgase-2 (-/-) mice and TPA did not induce COX-2 expression in ear of Tgase-2 (-/-) mice. These observations suggested that Tgase-2 is involved in TPA-induced COX-2 expression in the inflamed ear of mice and antiinflammatory effects of glucosamine is mediated through suppression of Tgase-2 in TPA ear edema.

Anti-inflammatory Effects of Enzymatic Extract from Ecklonia cava on TPA-induced Ear Skin Edema

  • Anti-inflammatory potential of the enzymatic extract prepared by Kojizyme (ECK), a component of brown seaweeds Ecklonia cava (Alariaceae, Phaeophyta) in vivo was investigated. For the application of mouse ear edema model, 12-O-tetradecanoylphorbol acetate (TPA) was used, a topical inducer of a long-lasting inflammatory response. Our results demonstrated that ECK inhibited ear edema when topically applied to mouse ear skin. In histological evaluation, the inhibition activity of ECK on TPA-induced inflammation is similar to that of dexamethasone, although less strong. In addition, the mRNA expression levels of IL-$1{\beta}$, IFN-$\gamma$, TNF-$\alpha$, and cyclooxygenase-2 (COX2) and the immunoreactivity to inducible nitric oxide synthase (iNOS) and COX2 expressed mainly in inflammatory cells were down-regulated by ECK. These results indicate that ECK has anti-inflammatory effects through the inhibition of Th1 cytokines and 2 inducers of inflammation in TPA-induced ear skin edema.

Synthesis and Anti-inflammatory Activity of Fructigenine A Derivatives

  • Several derivatives were synthesized from fructigenine A, which was isolated from Penicillium fructigenum. The anti-inflammatory properties of fructigenine A was evaluated in vivo with a 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced mouse ear edema model and a carrageenan-induced rat paw edema model. Results showed that the anti-inflammatory activity was significantly higher with fructigenine derivatives than with indomethacin, which was used as a standard. We concluded that fructigenine derivatives could exert an anti-inflammatory effect.

Ginsenoside Rg3 attenuates skin disorders via down-regulation of MDM2/HIF1α signaling pathway

  • Background: Thymic stromal lymphopoietin (TSLP) acts as a master switch for inflammatory responses. Ginsenoside Rg3 (Rg3) which is an active ingredient of Panax ginseng Meyer (Araliaceae) is known to possess various therapeutic effects. However, a modulatory effect of Rg3 on TSLP expression in the inflammatory responses remains poorly understood. Methods: We investigated antiinflammatory effects of Rg3 on an in vitro model using HMC-1 cells stimulated by PMA plus calcium ionophore (PMACI), as well as an in vivo model using PMA-induced mouse ear edema. TSLP and vascular endothelial growth factor (VEGF) levels were detected using enzyme-linked immunosorbent assay or real-time PCR analysis. Murine double minute 2 (MDM2) and hypoxia-inducible factor 1α (HIF1α) expression levels were detected using Western blot analysis. Results: Rg3 treatment restrained the production and mRNA expression levels of TSLP and VEGF in activated HMC-1 cells. Rg3 down-regulated the MDM2 expression level increased by PMACI stimulation. The HIF1α expression level was also reduced by Rg3 in activated HMC-1 cells. In addition, Rg3-administered mice showed the decreased redness and ear thickness in PMA-irritated ear edema. Rg3 inhibited the TSLP and VEGF levels in the serum and ear tissue homogenate. Moreover, the MDM2 and HIF1α expression levels in the ear tissue homogenate were suppressed by Rg3. Conclusion: Taken together, the current study identifies new mechanistic evidence about MDM2/HIF1α pathway in the antiinflammatory effect of Rg3, providing a new effective therapeutic strategy for the treatment of skin inflammatory diseases.

Balb/c 생쥐에 대한 어싱 매트리스에 의한 항염 효과 (Anti-inflammatory Effects of Earthing Mattress in Mouse)

  • Earthing, caused by direct skin contact with the Earth's surface, is used to reduce the symptoms of inflammation (fever, fever, swelling and pain). However, there is little evidence to support the anti-inflammatory effects of earthing mattresses. Therefore, this study was conducted to investigate whether anti-inflammatory effect of earthing mattress using an in vivo animal model. The anti - inflammatory effect was evaluated by measuring ear thickness and foot volume in 12-O-tetradecanoylphorbol-13 acetate (TPA) - induced ear edema and carrageenan - induced paw edema model, respectively. Balb/c mouse in carrageenan paw edema model showed significant anti - inflammatory effect in the group treated with earthing mattress for 4 hours or 24 hours for 3 days. For females, the anti-inflammatory effect was greater when the earthing mattress was added to the mattress than the mattress alone treatment. From the above results, it was found that the female responds more to the effect of the earthing as well as the mattress effect. In addition, when the male and female Balb/c mice were exposed to mattresses and earthing mattresses for 24 h for 3 days, respectively, the mattress and earthing mattresses showed significant inhibition of IL (Interleukin)-1β levels compared to the control. In the TPA ear edema model, Balb/c mouse showed significant anti - inflammatory effect in the group treated with the earthing mattress for 4 hours or 24 hours for 3 days. Both males and females showed more anti-inflammatory effects when they were exposed to earthing mattresses with mattresses added to the mattresses. From the above results, it was found that both male and female respond to the effect of earthing as well as the mattress effect in the TPA ear edema model. In conclusion, in this study, we have verified that earthing mattress shows inhibitory effects on TPA and carrageenan-induced inflammation. From these results, it is suggested that the anti-inflammatory effect can be expected by applying the earthing mattress to patients suffering from inflammatory diseases. However, there is a need to pinpoint exactly how the earthing mattress relieves inflammation, and further research is needed to investigate the mechanism.

Anti-Inflammatory Activity of Elsholtzia splendens

  • Elsholtzia splendens Nakai has been used in North-East Asia as an ingredient of folk medicines for treating cough, headache and inflammation. The present investigation was carried out to establish its in vivo anti-inflammatory activity using several animal models of inflammation and pain. The 75% ethanol extract of the aerial part of E. splendens significantly inhibited mouse croton oil-induced, as well as arachidonic acid-induced, ear edema by oral administration (44.6% inhibition of croton oil-induced edema at 400 mg/kg). This plant material also showed significant inhibitory activity against the mouse ear edema induced by multiple treatment of phorbol ester for 3 days, which is an animal model of subchronic inflammation. In addition, E. splendens exhibited significant analgesic activity against mouse acetic acid-induced writhing (50% inhibition at 400 mg/kg), while indomethacin (5 mg/kg) demonstrated 95% inhibition. E. splendens ($5-100{\;}{\mu}g/mL$) significantly inhibited $PGE_2$ production by pre-induced cyclooxygenase-2 of lipopolysaccharide-treated RAW 264.7 cells, suggesting that cyclooxygenase-2 inhibition might be one of the cellular mechanisms of anti-inflammation.

In vivo 및 in vitro 시험을 통한 페그마타이트의 항염증 효과 (The Anti-Inflammatory Effect of Pegmatite by in Vivo and in Vitro Study)

  • Purpose: This work aimed to elucidate the anti-inflammatory effect of pegmatite in vitro and in vivo. Methods: Author evaluated the suppressive effects of pegmatite on lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production, TNF-${\alpha}$ and IL-6 release in the RAW 264.7 murinemacrophages. Results: Treatment of RAW 264.7 cells with pegmatite significantly reduced LPS-stimulated NO production and inflammatory cytokine such as TNF-${\alpha}$ and IL-6 secretion in a concentration-dependent manner. Also pegmatite showed topical anti-inflammatory activity in the arachidonic acid (AA)-induced ear edema and acetic acid-induced increase in capillary permeability assessment in mice. It was also found that pegmatite (10 mg per ear in DW) inhibited arachidonic acid induced edema at 24 h more profoundly than 1 h by topical application. Furthermore, the vascular permeability increase induced by acetic acid was significantly reduced in mice that received pegmatite in 50 mg per mouse. Conclusion: Therefore the results of the present study suggest that pegmatite is a potent inhibitor of the LPS-induced NO and inflammatory cytokine in RAW 264.7 macrophages and showed anti-inflammatory activities in vivo animal model.

만성 및 급성 염증 모델에서 강황추출물의 효과 (Anti-inflammatory Effects of Turmeric (Curcuma longa L.) Extract on Acute and Chronic Inflammation Models)

  • 본 연구에서는 강황 유래의 curcuminoids가 없는 다당체를 이용하여 급성 및 만성 염증 모델에서의 항염증 효과를 검증하고자 하였다. 급성염증에 대한 효과를 알아보기 위하여 carrageenan에 의한 족부종 유발과 xylene에 의한 귀부종 유발을 일으킨 모델을 이용하여 수행하였다. Carrageenan에 의한 족부종 유발 실험에서 강황추출물과 관절염 치료제로 알려진 diclofenac sodium을 처리하였을 때는 부종의 증가도가 낮은 것으로 나타났다($P{\leq}0.05$). 또한 다른 급성염증 모델인 xylene application 마우스 모델에서도 대조군에 비하여 diclofenac sodium과 강황추출물(31.5, 63, 126 mg/kg)을 농도별로 경구투여 하였을 때 염증 억제율이 각각 78.13%, 68.75%, 62.50% 및 59.38%로 나타나 염증 억제율이 높은 것으로 나타났다($P{\leq}0.05$). 만성염증에 대한 효과를 알아보기 위한 cotton pellet 육아종 실험에서는 대조군과 비교하여 삼출물과 염증 육아종이 적게 발생된 것으로 나타났다($P{\leq}0.05$). 따라서 강황 유래의 다당체를 경구투여 하였을 때 만성 및 급성 염증에 대한 저해활성이 있는 것으로 나타나 항염증 소재로서 개발 가능성이 있음을 제시하였다.

LPS로 유도된 RAW264.7 Cell과 마우스모델에 대한 잘피 에탄올 추출물의 항염증 효과 (Anti-inflammatory Effect of Zostera marina Ethanolic Extract on LPS-induced RAW264.7 Cells and Mouse Model)

  • The Zostera marina ethanolic extract (ZMEE) was tested in this study to investigate the anti-inflammatory activity in LPS-induced RAW 264.7 cells and mouse model. Nitric oxide production and inducible nitiric oxide synthase expression in cells treated with ZMEE was reduced significantly in a dose-dependent manner. Similarly, the secretion of pro-inflammatory cytokines such as interleukin (IL)-6, IL-$1{\beta}$, and TNF-${\alpha}$ was inhibited markedly. In addition, the expression of nuclear factor kappa B (NF-${\kappa}B$) and the phosphorylation of JNK, ERK, and p38 MAPKs was suppressed by ZMEE as well. In vivo test, ZMEE attenuated the croton oil-induced mouse ear edema and there were no mortalities in mice administered 5,000 mg/kg body weight of ZMEE during the observation periods. The results in photomicrograph of mice ear tissue showed the reduction of dermal thickness and the number of infiltrated mast cells. These results indicate that ZMEE inhibits the production of LPS-induced pro-inflammatory mediators, suggesting that ZMEE may be a potential material for anti-inflammatory therapies.

정천화담강기탕가녹용(定喘化痰降氣湯加鹿茸)이 Xylene으로 유발된 마우스의 급성 염증에 미치는 영향 (Anti-Inflammatory Effect of Jungcheonhwadamgangki-tang ga Antler in Xylene-Application Mouse Ear Acute Inflammation Model)

  • In the present study, the anti-inflammatory effect of "Jungcheonhwadamgangki-tang ga Antler" water extract was tested in Xylene-Application mouse ear acute inflammation model. The test articles were once dosed before Xylene-Application, and the changes on body weight and ear weights and histopathological observation of induced ear were conducted with ear histomorphometry. The obtained results were as follows. The increases of absolute and relative ear weight detected in vehicle control compared to that of sham, were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang ga Antler in the present study. A classic acute inflammatory histological changes such as subcutaneous edema, thickness and infiltration of inflammatory cells, was detected in vehicle control. However, these histological changes were significantly and dose-dependently inhibited by Jungcheonhwadamgangki-tang ga Antler. In addition, the increases of ear thickness half and thickness full detected in the vehicle control were also dose-dependently decreased in the all Jungcheonhwadamgangki-tang ga Antler-dosing groups. Base on these results, it is concluded that Jungcheonhwadamgangki-tang ga Antler extracts has clear anti-inflammatory effect on the acute inflammation such as bronchial asthma.
