Characterization of ovarian culture in vitro and sex steroids in vivo by recombinant eel gonadotropin treatments in the eel Anguilla japonica

  • In the present study, we investigated the effects of recombinant eel gonadotropins (rec-GTHs) on maturation induction in immature ovarian culture in vitro and sex steroid hormones in vivo in the Japanese eel Anguilla japonica. To study the in vitro effects of rec-GTHs on estradiol-17β (E2) production in immature ovarian tissues, ovarian tissues were incubated with different doses of rec-follicle-stimulating hormone (rec-FSH) or rec-luteinizing hormone (rec-LH). The results revealed that the E2 levels in the rec-FSH (0.1, 0.5, or 1 ㎍/mL)- and rec-LH (0.1 or 0.5 ㎍/mL)-treated groups were significantly higher than those in the female eels from the control group. Furthermore, to investigate the in vivo effects of rec-GTHs on the gonadosomatic index (GSI) and plasma sex steroid hormone levels, the eels were injected intraperitoneally with eel's ringer (control), salmon pituitary extract (SPE; for female eels), human chorionic gonadotropin (hCG; for male eels), rec-FSH, rec-LH, and rec-FSH + rec-LH once a week. The results revealed that except for the SPE and the hCG groups, none of the groups exhibited a significant difference in GSI values. However, in vivo plasma E2 levels increased at the end of 4 weeks after rec-FSH treatment in female eels. Based on these results, it is suggested that rec-GTHs may have a positive effect on sexual maturation in female eels; however, further studies on complementary rec-protein production systems and additional glycosylation of rec-hormones are needed to elucidate hormone bioactivity in vivo and in vitro.

Effect of Environmetally-friendly Red Clay-Processed Materials on Quality Characteristics of Eel (친환경 황토 가공 신소재가 장어의 품질 특성에 미치는 영향)

  • The objectives of this study were to examine the effects of red clay-processed nano-materials (RCPM) on the quality characteristics of eel (Anguilla japonica). Both eels treated with RCPM and control were farm-raised under identical environments and were commercially processed. General components, texture, nutrients, antioxidant activities, and sensory analysis were performed. RCPM-treated eels showed significantly higher DPPH radical scavenging activities and FRAP values than the of control, indicating higher antioxidant activities of eels raised by RCPM. The amounts of niacin and tocopherol in RCPM-treated eels were higher than those of the control. For analysis of freeze-thaw stability, RCPM-treated eels showed more stable texture over freeze-thaw three treatment cycles. Descriptive panelists perceived eels raised by RCPM to be less oily than the control. Overall, RCPM exhibited positive effects on the quality of farm-raised eels. Therefore, RCPM would be of benefit to produce high value-added eels of premium quality.

  • The behavior of conger eel, Astroconger myriaste(Brevoort), to the net pots with baits was investigated alternately in two experimental water tanks. One of the pots being dropped on the tank bottom, the eels increased rapidly in number of touching the pot to show a maximum whithin 30 minutes and then decreased gradually. The enterings of eels into the pots were also made mainly within 30 minutes and easier in pots with lower entrances. But the complete enterings were decided by the passage of eel tails through the pot entrances and so the eels retreated frequently after attemping to enter the pots, especially in pots small in the interval between the entrances and the baits or in pots with flappers. However, most of enterings encouraged the touches and earlier enterings ascribed earlier production of maximum touch. The eels, if entered the pots without flappers, got out of the pots relatively easily and the getting-out was easier in pots with lower entrances. On the other hand, the eels entered the pot with flappers could not get out of the pots. A comparison between the circular and square pots gave that the guidance of eels to the entrances was easier in the former and the passing of eels through the entrances in the latter, not only at the inside of pots but also at the outside. The general behavior of eels in touching or entering the net pots in the above results was almost equal to that on the bamboo and plastic pots in the previous paper.

Effects of Estradiol-17β on the Feminization of Japanase Eel, Anguilla japonica (극동산 뱀장어, Anguilla japonica 자성화에 미치는 estradiol-17β 유도 효과)

  • The effects of oral administration of estradiol-$17{\beta}$ (E2) on glass eels (Body weight: $0.16{\pm}0.05g$, Total length: $6.2{\pm}0.9cm$) and young eels (Body weight: $2.6{\pm}0.6g$, Total length: $13.2{\pm}0.6cm$) on gonadal sex and growth were examined, respectively. Glass eels were fed a diet containing E2 at a dose of 10 mg/kg or 25 mg/kg, respectively, for five months. The female ratio significantly increased in all E2-treated groups (10 mg/kg diet group: 70%; 25 mg/kg diet group: 90%) when compared to the control group (10%). Young eels were fed a diet containing E2 at a dose of 25 mg/kg for four months. The female ratio also significantly increased in the E2-treated groups (60%) compared to the control group (20%). The highest female ratio was observed in the stage of glass eels rather than young eels. In all experiments, however, the growth of eels treated with E2 was similar to that of controls to the end of the experiment. Thus, oral administration of E2 could be a good approach to controlling sex differentiation.

  • The chemical attractants for sea eels, Astroconger myriaster, were identified by using fish traps in their natural habitat around namhae island, southern part of Korea. The ethyl alcohol extracts of anchovy, Engraulis japonica, which was a very good baits for sea eels showed higher attraction activities than residuals of it. Because sea eels showed high attraction activities to the free amino acid or nucleotides of synthetic anchovy extract, the succeeding experiment was conducted to identify the major chemical attractants among 9 free amino acid and 2 nucleotides by omission test. L-Glutamic acid, L-glycine, taurine, L-methionine or IMP showed highest attraction activities among 11 chemical compounds. But sea eels showed significantly weaker attraction activities to the synthethic extracts and single component than to the natural extract.

Effects of Aspirin on Nitrite Toxicity in Cultured Eel (Anguilla japonica) (양식 뱀장어 (Anguilla japonica)의 아질산 독성에 미치는 aspirin의 투여 효과)

  • The effect of aspirin on the recovery of nitrite-induced methemoglobinemia in cultured eels (Anguilla japonica) was studied. Methemoglobinemia was induced by exposing eels to nitrite (120 ㎎ NO2-N/ℓ) for 24 hr. The nitrite exposed eels were bathed in 20 ppm aspirin solution (Aspirin), 0.8 % NaCl solution (NaCl), 20 ppm aspirin plus 0.8% NaCl solution (NaCl + Aspirin) and 50% nitrite free water(control) for 24 hr to recover from nitrite toxicity. Peripheral blood was taken from the arterious bulb from all groups to analyse hematocrit value, hemoglobin concentration, and nitrite concentration of the blood. Histopathological features of gill were also observed. Aspirin and control groups were more effective than NaCl and NaCl + Aspirin groups in recovery of hematocrit value and hemoglobin concentration, methemoglobin rate and nitrite concentration. The histopathological features on the gill of aspirin group were similar to those of normal eels, but other groups showed focal hyperemia in the lamellar carpillaries, epithelial hyperplasia. These results suggested that aspirin was very effective to recover from methemoglobinemia in nitrite-induced cultured eels.

  • Specimens of eels, collected in the lagoons of Arcachon Bay from April 1977 to October 1978, were analysed for age and growty. The structure of the otolith was described based on examination of the decalccified surface using an scanning electron microscope, and was compared with obcervations made under an optical microscope. The otolith of Arcachon eels show a narrow summer growth stop in addition to a growth stop formed in winter. The narrow summer growtu in length of sedentsry eels was well represented removal of the large memgers by the migration to the sea decreases the growth ratio of sedentary eels as the fish grows older. This size selective migrationin part contributes to satisfy Von Bertalanffy's assumption. Length distribution of the exceptionally large eels in relation to age was analysed ty comparing them with "normal" sedentary eesl and with silver migsating, and it is suggested that thew exceptionally large eesl consist of the anmals which probably the [apacity for growth wen alredy young.

  • The Ministry of Oceans and Fisheries promoted the installation of eel-ladder for the purpose of creating inland water resources. Currently, eel-ladder have been installed and operated at the Geumgang Estuary Bank (2018), Yeongam Embankment (2019), and Asanman Embankment (2020). In this study, the number of glass eels in eel-ladder in 2021 was monitored and factors affecting the rise that from ocean to river of eels were investigated. Glass eels in eel-ladder were found when the salinity was relatively low, and they started when the freshwater and seawater temperatures were above 20℃. Comparing the number of occurrences by year, the largest number of glass eels was observed in 2021, but it is judged that this is not according to the distribution of glass eels in sea, but rather as a result of the investigator's eel-ladder repair and guidance on illegal fishing.

Feeding Habits of the Glass Eel Anguilla japonica Determined by C and N Stable Isotopes in the Nakdong River Estuary of the Korean Peninsula (안정동위원소를 이용한 낙동강 하구 실뱀장어의 먹이 습성 분석)

  • Wild glass eels found in the Korean peninsula are thought to migrate along the North Equatorial Current and undergo the processes of spawning and incubation in the Mariana Trench. Juveniles of the wild glass eels are collected from the southern and western coasts of the Korean peninsula and used as seeds for aquaculture. To investigate the feeding behavior of wild glass eels, we collected glass eels from the Nakdong River estuary during March and April 2014 and analyzed the total length, wet weight, dry weight, carbon and nitrogen contents, and stable isotope ratio of eels as well as water temperature and salinity. Water temperature in the Nakdong River estuary was $13.2{\pm}1.0$ (mean ${\pm}$ 1SE; range, $10.1{\sim}15.7)^{\circ}C$ and salinity was $24.8{\pm}2.4$ (13.2~34.0), showed a lower range from 13.2 to 30.0 (a mean of 21.2) when the floodgates were opened. The total length of glass eels was $56.5{\pm}0.2$ (51.0~63.6) mm, wet weight was $70.9{\pm}1.4$ (33.6~133.2) mg, and dry weight was $16.5{\pm}0.3$ (10.1~29.1) mg. Carbon and nitrogen contents of glass eels were $51.0{\pm}0.8%$ and $13.9{\pm}0.1%$, respectively. Mean ${\delta}^{13}C$ and ${\delta}^{15}N$ values of glass eels collected from the Nakdong River estuary were -20.9±0.2‰ and 6.1±0.1‰, displaying similar values to those of leptocephalus, glass eel larvae collected from the North Equatorial Current. Therefore, this result suggest that the glass eels collected from the Nakdong River estuary do not feed on prey after metamorphosis from eel larvae to glass eels their migration.

An investigation of the congestion on the gills of eel (뱀장어 아기미 울혈증에 관한 연구)

  • In order to elucidate the outbreak mechanisms of a new disease which is characterized by an intense congestion in central venus sinuses(CVS) of gill filaments in cultured eel. these experiments were carried out; epidemically surveyed on the cultured eel farms in the vicinity of Kunsan city and experimentaliy outbreaked the disease in the stressful condition such as thermal and handling shock and innoculated the supernatant from the homogenate of naturally severe congested gill into eels and onto the monolayer of the CHSE-214. Although the frequency of congestion in eels of B, C, D and E farms were higher than in eels of A farms, the water qualities(stocked and cultured water) among farms were not a great difference. In eels of B, C, D and E farms, the values of haematocrit(Ht), haemoglobin(Hb), total protein(Tp), albumin(Alb), glucose(Glu), magnesium(Mg) were lower and the values of calcium(Ca), methemoglobin(Met-Hb), glutamic pyruvic transminase(GPT), glutamic oxalacetic transminase(GOT) higher than in eels of A farms. These valules have not related to the frequency of congestion. An intensive congestion and dilataton in CVS of gill filaments in experimentally handling-stressed eels produced similar histopathological changes to those observed in the spontaneously diseased eel, but not in eels experimentally injected with filtering contents. The cytopathic effect on the CHSE-214 was not observed. In stressed eels the congestion of gill was increased in relation to either the decrease ranges of water temperature or the incerase in accllimated times. And increase in Ht, Met-Hb, Alb, Glu, GOT and GPT and decrease in Mg, Hb and Tp were found, which had a close relationship to congestion of gill. Cortisol were increased according to the decrease ranges in acclimated water temperature. From these results, decrease in water temperature during selection placed eels upon the stressed condition, made increase in ionic strength in blood stream, and CVS was dilatated owing to the increased blood inflow.
