Streptomyces 속균이 생산하는 Pretense에 관한 연구 (Studies on the Protease procuced by Streptomyces sp.)

  • 토양에서 분리한 Streptomyces 속 중에 금속을 함유하고 있는 protease를 강하게 분필하는 균일 주를 선별하여 그 효소학적 성질 멸가지를 알아본 결과는 다음과 같다. 1 본 효소의 작용 최적 pH는 중성부근이며 최적온도는 37$^{\circ}C$ 부근이다. 2. 본 효소는 중성부근에서 비교적 안정하며 열에는 비교적 불안정하며 37$^{\circ}C$에서 100분간 열처리했을때 약 50% 가량 실활하였다. 3. Hg$^{++}$, Cu$^{++}$, Cd$^{++}$, Ag$^{+}$ 등에 의하여 강하게 조해되며, Mn$^{++}$, $Mg^{++}$, $Ca^{++}$, Pb$^{++}$, $Ba^{++}$ 등에는 별 영향을 받지 않는다. 4. oxalate, citrate, 2-4-dinitrophenol, $\varepsilon$-amino caproic acid, thiourea, cysteine에 의해서는 강하게 조해되었다.

Cloning, Expression, and Characterization of a Family B-Type DNA Polymerase from the Hyperthermophilic Crenarchaeon Pyrobaculum arsenaticum and Its Application to PCR

  • The gene encoding Pyrobaculum arsenaticum DNA polymerase (Par DNA polymerase) was cloned and sequenced. The gene consists of 2,361 bp coding for a protein with 786 amino acid residues. The deduced amino acid sequence of Par DNA polymerase showed a high similarity to archaeal family B-type DNA polymerases (Group I), and contained all of the motifs conserved in the family B-type DNA polymerases for $3'{\rightarrow}5'$ exonuclease and polymerase activities. The Par DNA polymerase gene was expressed under the control of the T7lac promoter on the expression vector pET-22b(+) in Escherichia coli BL21-CodonPlus(DE3)-RP. The expressed enzyme was purified by heat treatment, and Cibacron blue 3GA and $Hirap^{TM}$ Heparin HP column chromatographies. The optimum pH of the purified enzyme was 7.5. The enzyme activity was activated by divalent cations, and was inhibited by EDTA and monovalent cations. The half-life of the enzyme at $95^{\circ}C$ was 6 h. Par DNA polymerase possessed associated $3'{\rightarrow}5'$ proofreading exonuclease activity, which is consistent with its deduced amino acid sequence. PCR experiment with Par DNA polymerase showed an amplified product, indicating that this enzyme might be useful in DNA amplification and PCR-based applications.

Use of FT-IR to Identify Enhanced Biomass Production and Biochemical Pool Shifts in the Marine Microalgae, Chlorella ovalis, Cultured in Media Composed of Different Ratios of Deep Seawater and Fermented Animal Wastewater

  • Growth rates, photosystem II photosynthesis, and the levels of chlorophyll $\alpha$ and secondary metabolites of Chlorella ovalis were estimated to determine if they were enhanced by the addition of swine urine (BM) or cow compost water (EP) that had been fermented by soil bacteria to deep seawater (DSW) in an attempt to develop media that enabled batch mass culture at lower costs. Growth of C. ovalis in f/2, f/2-EDTA+BM60%, DSW+BM30%, and DSW+EP60% was enhanced and maintained in the log phase of growth for 16 days. The cell densities of C. ovalis in DSW+EP60% ($4.1{\times}10^6$ Cells/ml) were higher than those of f/2 ($2.9{\times}10^6$ Cells/ml), f/2-E+BM60% ($3.7{\times}10^6$ Cells/ml), and DSW+BM30% ($2.7{\times}10^6$ Cells/ml). The growth rate was also more favorable for C. ovalis cultured in DSW+EP60% ($0.15\;day^{-1}$) than that of C. ovalis cultured in the control medium (f/2) ($0.12\;day^{-1}$). Furthermore, the chlorophyll a concentration of C. ovalis cultured in DSW+EP60% (4.56 mg/l) was more than 2-fold greater than that of C. ovalis cultured in f/2 (2.35 mg/l). Moreover, the maximal quantum yields of photo system II at 470 nm (Fv/Fm) were significantly higher in organisms cultured at f/2-E+BM60% (0.53) and DSW+EP60% (0.52) than in the other treatment groups. Finally, Fourier transformation infrared (FT-IR) spectroscopy revealed that C. ovalis grown in DSW+EP60% had more typical peaks and various biochemical pool shifts than those grown in other types of media. Taken together, the results of this study indicate that the use of DSW+EP60% to culture C. ovalis can reduce maintenance expenses and promote higher yields.

Purification and Characterization of a Novel Fibrinolytic Enzyme from Culture Supernatant of Pleurotus ostreatus

  • A fibrinolytic enzyme was produced by an edible mushroom of Pleurotus ostreatus using submerged culture fermentation. The enzyme was purified from the culture supernatant by applying a combination of freeze-thaw treatment, ammonium sulfate precipitation, hydrophobic interaction, and gel filtration chromatographies. The enzyme was purified by a 147-fold, with a yield of 7.54%. The molecular masses of the enzyme an determined by gel filtration and SDS-PAGE were 13.6 and 18.2 kDa, respectively. The isoelectric point of the enzyme was 8.52. It hydrolyzed fibrinogen by cleaving the ${\alpha}$ and ${\beta}$ chains of fibrinogen followed by the ${\gamma}$ chains, and also activated plasminogen into plasmin. The enzyme was optimally active at $45^{\circ}C$ and pH 7.4. The enzyme activity was completely inhibited by EDTA, whereas protease inhibitors of TPCK, SBTI, PMSF, aprotinin and pepstatin showed no inhibition on its activity. The partial amino acid sequences of the enzyme as determined by Q-TOF2 were ATFVGCSATR, GGTLIHESSHFTR, and YTTWFGTFVTSR. These sequences showed a high degree of homology with those of metallo-endopeptidases from P. ostreatus and Armillaria mellea. The purified enzyme can also be applied as a natural agent for oral fibrinolytic therapy or prevention of thrombosis.

  • DNA's were extracted from Saccharomyces cerevisiae and Mycobacterium phlei and the damaging cell walls of these microoragnisms were examined under an electron microscope in the extraction process in which a number of physico-chemical tratments of cells was involved. While the DNA was easily extracted from S. cerevisiae using conventional meylelded very little DNA, of M. phlei was extremely difficult to isolate and yielded very little DNA, applying various methods of isolation published earlier. When the cell walls of S. cerevisiae were examined with the electron microscope, they were not yet damaged even after the cells were treated with sodium lauryl sulfate(SLS) and ethylene diamine tetracetic acid(EDTA), but they were completely destroyed by the treatment of sodium perchlorate followed by the addition of chloroform and a vigorous agitation. Oozing cytoplasm through the broken cell walls was also observed. In the extraction of DNA from M.phlei, the pronase was not effective at the aerobic environment of the sample. When phenol was applied at the last step of DNA isolation, an extreme damage mass yielding little DNA into the solution. Unlike the cells of S.cerevisiae.M.phlei cells showed a tendency of aggregation, thus the destruction of cell walls by sodium hydroxide was seen only on the walls of peripheral cells in the aggregated mass, leaving the walls of the inner cells undamaged.

  • The biochemical properties of the fibrinolytic protease of 50,800 Da isolated from the venom of Kgdistrodon blomhoffi brevicaudus were characterized. The enzyme hydrolyzed the carboxyl side of arginine in the synthetic chromogenic peptides, N-Benzoyl-Phe-Val-Arg-pNA and N-p-Tosyl-Gly-Pro-Arg-pNA, and the enzyme activity was inhibited by phenylmethylsulfonylfluoride indicating that the enzyme belongs to the serine protease family. The pretense showed maximum activity at pH 7.5 and inhibited by ZnCl$_2$, CuSO$_4$, but not by soybean trypsin inhibitor, pepstatin A, 2-mercaptoethanol and EDTA. The fm value determined with N-p-Tosyl-Gly-Pro-Arg-pNA was 0.2 mM. The thrombolytic activity of the purified enzyme was evaluated by platelet aggregation test in rabbits. While the platelet count ratio in blood of the rabbits injected with thrombin alone declined from 1.0 to 0.6 within 7 min and maintained around 0.6 for 24 hours thereafter, the ratio rapidly recovered from around 0.6 to 0.8 in 1 hr, to 1.0 in 24 hrs when the rabbits were sequentially treated with thrombin and the purified enzyme. The result showed that the serine protease from A. blomhoffi brevicoudus of 50,800 Da had a thrombolytic activity in vivo and the enzyme might be developed as a therapuetic agent for the treatment of thrombic disease.

원자흡광법을 이용한 요중 연 배설량의 정량 (Determination of Lead in Urine by Atomic Absorption Spectrophotometry)

  • Determination of lead in urine is important in industrial hygiene and toxicology. Dithizone method has been principally used for the determination of lead in urine, which gives accurate results in skilful hands but is usually complex and time-consuming. Atomic absorption spectrophotometry is a new simple method and several procedures have been described. However, the influences of pH and the presence of chelating agents during treatment of lead poisoning are not clear. The purpose of this study was to find out the effect of pH and chelating agents on the determination of lead using Shimadzu atomic absorption/flame spectrophotometer, model AA-610. The results obtained were as follows: 1. The atomic absorption spectrophotometry(AAS) could be applied without prior acid digestion to specimens in the absence of chelating agents. The absorbance at $2,170\;{\AA}$, though more sensitive, was more noisy electronically. Therefore, we selected the wavelength of $2,833\;{\AA}$ plus scale expansion. 2. The optimal pH was in the range from 2 to 3. 3. The sensitivity was $0.075{\mu}g/ml/%$ and detection limit was about $0.2{\mu}g/ml$. 4. In the presence of EDTA, lead could not be completely determined without prior acid digestion. 5. On specimens from patients receiving penicillamine therapy, a comparison was made between the values obtained with dithizone method and AAS method with prior acid digestion. The results of comparison showed a very good agreement.

다발구멍장이버섯으로부터 분리한 혈전용해 세린분해효소의 특성 연구 (Characterization of a Fibrinolytic Serine Protease from an Edible Mushroom, Albatrellus confluens)

  • 3단계를 거처 식용 가능한 다발구멍장이버섯으로부터 분리한 혈전용해효소의 비활성은 30.49 U/mg이었으며, MALDI-TOF/MS로부터 분자량이 30086.41 Da이었다. 일차 아미노산 구조는 Glu-Thr-Val-Thr-Glu-Thr-Thr-Ala-Pro-Trp-Gly-Leu-Ser-Arg-Ile으로 밝혀졌으며, pH 8.0에서 pH 10.0의 넓은 범위에서 큰 활성을 나타내는 alkaline protease로, 최적온도는 $50^{\circ}C$이며, $30^{\circ}C$까지는 대체로 열에 안정한 효소였다. 이 효소는 합성 기질 N-Suc-Ala-Ala-Pro-Phe pNA을 강하게 분해하였으며, $Hg^{2+}$ 금속이온의 첨가로 효소 활성이 완전히 사라졌다. 효소저해제인 PMSF의 첨가로 혈전용해활성이 사라지는 결과로부터 serine 분해효소임을 알 수 있었다.

Cloning, Expression, and Characterization of a Hyperalkaline Phosphatase from the Thermophilic Bacterium Thermus sp. T351

  • The gene encoding Thermus sp. T351 alkaline phosphatase (T351 APase) was cloned and sequenced. The gene consisted of 1,503 bp coding for a protein with 500 amino acid residues including a signal peptide. The deduced amino acid sequence of T351 APase showed relatively low similarity to other Thermus APases. The T351 APase gene was expressed under the control of the T7lac promoter on the expression vector pET-22b(+) in Escherichia coli BL21 (DE3). The expressed enzyme was purified by heat treatment, and $UNO^{TM}$ Q and $HiTrap^{TM}$ Heparin HP column chromatographies. The purified enzyme exhibited high activity at extremely alkaline pHs, reaching a maximum at pH 12.0. The optimum temperature of the enzyme was $80^{\circ}C$, and the half-life at $85^{\circ}C$ was approximately 103 min. The enzyme activity was found to be dependent on metal ions: the addition of $Mg^{2+}$ and $CO^{2+}$ increased the activity, whereas EDTA inhibited it. With p-nitrophenyl phosphate as the substrate, T351 APase had a Michaelis constant ($K_{m}$) of $3.9{\times}10^{-5}M$. The enzyme catalyzed the hydrolysis of a wide variety of phosphorylated compounds.

생쥐 체외수정난의 초급속동결 및 이식에 관한 연구 II. pH, 삼수압 및 정자 전배양처리가 생쥐 처리수정난의 발달에 미치는 영향 (Studies on Transfer of In Vitro Fertilized Mouse Embryos Following Ultrarapid Freezing II. Effect of Treatment of pH, Osmolality and Sperm Preincubation on Development of In Vitro Fertilization Mouse Embryos)

  • These studies were carried out to overcome 2-cell block and in vitro development to blastocysts in vitro fertilization of mouse embryos. The unfertilized ova were obtained by superovulation in ICR mice of 4 to 6 weeks old. Tyrode's 280 solution was used as basal media, and the pH range of media examined was designed from 6.5 to 7.5 with 0.2 interval and the range of osmolality from 250 to 370 mOsm with 20 interval, and the period of sperm preincubation examined was 30, 60, 120, and 180 minutes. The ova developed to 2-cell embryos after 26hrs of incubation with preincubated sperm were evaluatated as in vitro fertilized ones. The results obtained were summarized as follows: 1. The optimal ranges of pH and osmolality of culture media and of sperm preincubation time for in vitro development of in vitro fertilized ova to blastocyst were pH 7.1 to 7.3, 250 to 350 mosmol and 60 to 180 min, respectively. 2. With the media of pH 7.1, 310 mOsm and sperm preincubation period of 120min in another experiment of large sample size, the in vitro fertilized ova was found 66.5% and the in vitro development of in vitro fertilized ova to blastocyst was found 35.8%. From the above results it was concluded that the optimal conditions of pH and osmolality of the media for mouse IVF and embryo culture, and the period of sperm preincubation might be 7.1, 310 mOam and 120min, respectively.
